MORPHOLOGICAL STUDY BY SCANNING ELECTRON MICROSCOPY OF RUMEN DEGRADATION OF WHEAT STRAW TREATED WITH AMMONIA AND SULPHUR DIOXIDE

  • Ammonia and/or sulphur dioxide treated and untreated wheat leaf sheaths were compared for cell wall digestion by incubation with rumen liquor for 24 and 48 hours. Scanning electron microscope (SEM) was used to study the relative rate and extent of cell wall digestion. Treated wheat straw leaf sheaths were distorted, with more distortion observed in ammonia and sulphur dioxide combined treatment than any other treatment. Rumen liquor digestion for 24 hours of untreated leaf sheath showed disrupted phloem, partially ruptured parenchyma and vascular tissues and further partially distorted inner bundle sheaths and vascular bundle after 48 hours incubation. Sulphurated leaf sheaths showed extensive degraded parenchyma and sclerenchyma material in 24 hours incubation, however, all tissues were irregulary shaped in 48 hours incubation. In ammoniation, epidermal cell walls and small vascular bundles began to disintegrate by 24 hours incubation, extensively changed structure and degraded epidermal tissue by 48 hours incubation. Combination treatment of leaf sheaths degraded all cell walls of parenchyma, phloem and vascular bundle by 24 hours incubation, however, structures only of inner bundles sheath with extended land, sclerenchyma and cutinized epidermal cell walls remained.

Scanning electron microscopical study on the uterine development of fetuses and neonates in Korean native goats (한국재래산양의 태아 및 신생아의 자궁발달에 관한 주사전자현미경적 연구)

  • The development of uterus in fetuses on 60, 90 and 120 days of gastation and neonates of Korean native goats was investigated by scanning electron microscopy. The results were summarized as follows ; 1. In the 60-day-old fetuses, the short microvilli were sporadically observed on the luminal surface of the endometrium. 2. In the 90-day-old fetuses, the mucosal folds, polygonal microridges, numerous microvilli, flower-like-buds, and domeshaped or crateriform area were also observed on the luminal surface of endometrium. 3. In the 120-day-old fetuses, the primordial caruncles(nodules) of the endometrium were developed conspicuously and long microvilli were developed densely. 4. In the neonates, the caruncles and microvilli of the endometrium were more developed than those of 120-day-old fetuses.

Investigation of Domain Structure in (001) PMN-x%PT Crystals by Scanning Force Microscope (Scanning Force Microscope에 의한 (001) PMN-x%PT 단결정의 도메인 구조에 대한 연구)

  • The domain structures of annealed (001)-oriented $Pb(Mg_{1/3}Nb_{2/3})O_3-x%PbTiO_3$ (PMN-x%PT) crystals for x = 10, 20, 30, 35, and 40 at% were investigated by Polarized Optical Microscopy (POM) and Scanning Force Microscopy (SFM) in the piezoresponse mode. Both Polar Nano-Domains (PND) and long strip-like domains were clearly observed. The results also showed how the domain structure changed between phases with an increasing x in the PMN-x%PT crystals and the domain hierarchy on various length scales ranging from 40 nm to 0.1 mm. Distorted pseudo-cubic phase (x < 20%) consisted of PNDs that did not self-assemble into macro-domain plates. The rhombohedral phase (x = 30%) consisted of PNDs that began to self-assemble into colonies along preferred {110} planes. The monoclinic phase (x = 35%) consisted of miniature polar domains on the nm scale, whereas, the tetragonal phase (x = 40%) consisted of {001} oriented lamella domains on the mm scale that had internal nano-scale heterogeneities, which self-assembled into macro-domain plates oriented along {001} the mm scale.
