The present study undertaken to investigate the interrelationship among their regular dietary fatty acid intake, body fat content(BFC), and plasma lipid fractions(PLF) of middle-aged Korean women. Forty-nine women (age 30 to 49) participated in this study in which the aim was to determine their total nutrient intake per day, BFC and PLF, and to analyse the intercorrelations of these. Strong correlation was seen among their body mass index, mid-upper-arm-circumference, and BFC which were positively correlated with plasma total lipids(TL) and triglycerides(TG). Dietary myristic acid and lauric acid were positively correlated with both total cholesterol(TC) and LDL-cholesterol (LDL-C) in plasma. Palmitic acid, stearic acid, oleic acid, and total monounsaturated fatty acid(MUFA) were negatively correlated with HDL-cholesterol(HDL-C). Although dietary total saturated fatty acid (SFA) was not correlated with TC and LDL-C, it was positively correlated with TL and TG in plasma. However, dieary n-3 polyunsaturated fatty acid(PUFA) in regular Korean diet was not correlated with any of lipid fractions in plasma which might be due to the high ratio of n-6/n-3 in their diet. From the above results, it was concluded that plasma lipid fractions were modulated by their dietary nutrients, especially the types of the fatty acids. Dietary SFA and MUFA intake not PUFA are strongly correlated with LDL-C and HDL-C in the Korean diet This may relate to the frequency of coronary heart disease in the age group.
Skeletal myogenesis is a complex process that is finely regulated by myogenic transcription factors. Recent studies have shown that saturated fatty acids (SFA) can suppress the activation of myogenic transcription factors and impair the myogenic differentiation of progenitor cells. Despite the increasing evidence of the roles of miRNAs in myogenesis, the targets and myogenic regulatory mechanisms of miRNAs are largely unknown, particularly when myogenesis is dysregulated by SFA deposition. This study examined the implications of SFA-induced miR-183-5p on the myogenic differentiation in C2C12 myoblasts. Long-chain SFA palmitic acid (PA) drastically reduced myogenic transcription factors, such as myoblast determination protein (MyoD), myogenin (MyoG), and myocyte enhancer factor 2C (MEF2C), and inhibited FHL1 expression and myogenic differentiation of C2C12 myoblasts, accompanied by the induction of miR-183-5p. The knockdown of FHL1 by siRNA inhibited myogenic differentiation of myoblasts. Interestingly, miR-183-5p inversely regulated the expression of FHL1, a crucial regulator of skeletal myogenesis, by targeting the 3'UTR of FHL1 mRNA. Furthermore, the transfection of miR-183-5p mimic suppressed the expression of MyoD, MyoG, MEF2C, and MyHC, and impaired the differentiation and myotube formation of myoblasts. Overall, this study highlights the role of miR-183-5p in myogenic differentiation through FHL1 repression and suggests a novel miRNA-mediated mechanism for myogenesis in a background of obesity.

Buffaloes usually maintain higher body condition and do not produce milk at the cost of their own body reserves under tropical conditions. The mobilization of body reserves for fulfilling the demands of lactation has been extensively studied in dairy cows while limited work is available on this aspect in dairy buffaloes. Therefore, the present study was conducted to examine variations in milk fatty acid profiles with body condition in Nili-Ravi buffaloes. A total of 24 Nili-Ravi buffaloes within 60 days after parturition, were selected from a private dairy farm in the district of Peshawar. All animals consumed the same diet during the experimental period. A total of 576 raw milk samples were collected for laboratory analysis. The study continued up to 6 months during 2008. Body condition score (BCS), milk yield and composition were recorded once a week. Means for milk fatty acid profile were compared for various levels of BCS. The mean milk yield and fat content were 9.28 kg/d and 5.36%, respectively. The total saturated fatty acids (SFA) were 64.22 g/100 g and the unsaturated fatty acids (UFA) were 35.79 g/100 g. Of the SFA the highest amount was recorded for $C_{16:0}$, followed by $C_{18:0}$, and $C_{14:0}$. The total sum of hypercholesterolemic fatty acids (HCFA, $C_{12:0}$, $C_{14:0}$ and $C_{16:0}$) was 43.33 g/100 g. The concentrations of UFA were greater for moderate BCS followed by poor and highest BCS while SFA showed the opposite trend. The correlation analysis showed that milk yield was negatively affected by BCS and milk fat positively affected, though non-significantly. The present study suggests that Nili-Ravi dairy buffaloes produce similar milk to dairy cows regarding availability of cardioprotective fatty acids, with the highest concentration of $C_{18:1\;cis-9}$. Two HCFA ($C_{12:0}$ and $C_{14:0}$) were associated with higher body condition. Buffaloes with moderate body condition yielded milk containing healthier fatty acids.
Two SNPs, i.e. L127V and T172M, of bovine growth hormone (GH) causing the presence of GH gene haplotypes A, B, and C was previously shown to alter intramuscular fatty acid (FA) composition in Japanese Black (JB) heifers. To determine the SNP effect on somatotropic hormone concentration and lipogenesis, we measured plasma GH, insulin, and insulin-like growth factor-1 (IGF-1) concentrations. We also measured mRNA levels of fatty acid synthase (FASN), stearoyl-coA desaturase (SCD), and sterol regulatory element binding proteins-1 (SREBP-1) and FA composition in diaphragm tissues. Heifers with genotype CC had the lowest plasma insulin concentration and FASN and SCD mRNA levels among genotypes. FASN mRNA levels in haplotype A tended to positively correlate with saturated FA (SFA) content and negatively correlated with C18:2 and unsaturated FA (USFA) contents. SCD mRNA levels in haplotype A positively correlated with monounsaturated FA (MUFA) contents and negatively correlated with C18:0 content. They also tended to positively correlate with C16:1, C18:1, and USFA contents and USFA/SFA ratio and negatively correlate with SFA content. Taken together, GH gene polymorphism affects the lipogenic genes expression levels and their relationships with fatty acid compositions in diaphragm tissues of JB heifers at 31 months of age.
Fatty acid compositions of seventy-two species of Korean fish muscle, 59 species of sea water fish and 13 species of fresh water fish, were studied. Polyunsaturated fatty acid (PUFA) was the richest fatty acid group in all fish samples, accounting for $38.0\pm10.3\%$ of total fatty acids. Monounsaturated fatty acids $(MUFA,\;31.4\pm9.67\%)$ and saturated fatty acids $(SFA, 30.5\pm3.81\%)$ showed a similar level. There was a positive correlation between the total lipid content and MUFA (r=0.7788, p<0.001) and a negative correlation between the total lipid content and PUFA (r= -0.7786, p<0.001) while there was no correlation between the total lipid content and SFA. The proportion of n-3 PUFA and n-6 PUFA was $29.7\pm8.73\%$ and $6,48\pm3.70\%$, respectively, in all fish samples. The n-3 PUFA was rich in sea water fish while n-6 PUFA was rich in fresh water fish. The migratory fish contained the highest level of the n-3 PUFA $(1.82\pm1.01g/100g\;muscle)$, followed by the fresh water fish $(1.09\pm1.04g/100g\;muscle)$, the reef fish $(0.90\pm0.60g/100g\;muscle)$ and the demersal fish $(0.77\pm0.38g/100g\;muscle)$. There was a positive correlation between the total lipid and n-3 PUFA content, $y=0.2083\times+0.05 (r=0.9352,\;p<0.0010)$.
This work was undertaken to study the effect of dietary fat source on the fatty acid profile of pork, and to evaluate the effect of inclusion of vitamin E in pig diets on lipid oxidation of pork tissue and processed pork products. Fifty-six pigs were allocated to four treatments, that included two dietary fat sources and two levels of vitamin E inclusion. Dietary fat was derived from either tallow, a source of saturated fatty acids (SFA), or from a mixture of soybean and linseed oils, which contain polyunsaturated fatty acids (PUFA). Vitamin E was included at either 0% or 0.011% of the diet. Growth and carcass characteristics were not affected by the dietary treatments. Dietary fat source affected the fatty acid profile of the longissimus muscle and subcutaneous fat tissue, with the PUFA diet resulting in significantly more polyunsaturated fatty acids in the tissues, and more favourable ratios of SFA to PUFA and C18:2 to C18:3 in terms of human health considerations. Lipid oxidation was significantly greater in tissues and processed products from PUFA-fed pigs. Inclusion of vitamin E in the diets, however, reduced the extent of lipid oxidation in the meat and meat products. Dietary manipulation of the fatty acid profile of pigs is an effective means of altering the fat composition of pork in order to provide human consumers with a healthy product. Vitamin E is effective as an antioxidant agent, particularly where processed products are concerned.

Pastirma is a dry-cured meat product, produced from whole beef or water buffalo muscles. This study was carried out to investigate the effect of production stages (raw meat, after curing, after $2^{nd}$ drying and pastirma) on the total lipid, neutral lipid, phospholipid and fatty acid composition of phospholipid fraction of pastirma produced from beef M. Longissimus dorsi muscles. The pH and colour ($L^*$, $a^*$ and $b^*$) analyses were also performed in raw meat and pastirma. It was found that pastirma production stages had significant effects (p<0.01) on the total amounts of lipid, neutral lipid and phospholipid, and the highest amounts of lipid, neutral lipid and phospholipid were detected in pastirma. In pastirma, neutral lipid ratio was determined as $79.33{\pm}2.06%$ and phospholipid ratio as $20.67{\pm}2.06%$. Phospholipids was proportionately lower in pastirma than raw meat. Pastirma production stages affected pentadecanoic acid (15:1) (p<0.01), linoleic acid (18:2n-6) (p<0.05), ${\gamma}-linoleic$ acid (18:3n-6) (p<0.05), erucic acid (22:1n-9) (p<0.05), docosapentaenoic acid (22:5n-6) (p<0.05), total unsaturated fatty acid (${\Sigma}USFA$) (p<0.05) and total saturated fatty acid (${\Sigma}SFA$) (p<0.05) ratios of phospholipid fraction and also the moisture content (p<0.01). Pastirma process also affected pH and colour ($L^*$, $a^*$ and $b^*$) values (p<0.01), and these values were higher in pastirma than raw meat.

Beef contains functional fatty acids such as conjugated linoleic acid and longchain fatty acids. This review summarizes results from studies comparing the fatty acid composition of beef from cattle fed either grass or grain-based feed. Since functional lipid components are contributed through dietary consumption of beef, the fatty acid composition is reported on mg/100 g of meat basis rather than on a percentage of total fat basis. Beef from grass-fed contains lesser total fat than that from grain-fed in all breeds of cattle. Reduced total fat content also influences the fatty acid composition of beef. A 100 g beef meat from grass-fed cattle contained 2,773 mg less total saturated fatty acids (SFA) than that from the same amount of grain-fed. Grass-fed also showed a more favorable SFA lipid profile containing less cholesterol-raising fatty acids (C12:0 to C16:0) but contained a lesser amount of cholesterol-lowering C18:0 than grain-fed beef. In terms of essential fatty acids, grass-fed beef showed greater levels of trans-vaccenic acid and long-chain n-3 polyunsaturated fatty acids (PUFA; EPA, DPA, DHA) than grain-fed beef. Grass-fed beef also contains an increased level of total n-3 PUFA which reduced the n-6 to n-3 ratio thus can offer more health benefits than grain-fed. The findings signify that grass-fed beef could exert protective effects against a number of diseases ranging from cancer to cardiovascular disease (CVD) as evidenced by the increased functional omega-3 PUFA and decreased undesirable SFA. Although grain-fed beef showed lesser EPA, DPA, and DHA, consumers should be aware that greater portions of grain-fed beef could also achieve a similar dietary intake of long-chain omega-3 fatty acids. Noteworthy, grain-fed beef contained higher total monounsaturated fatty acid that have beneficial roles in the amelioration of CVD risks than grass-fed beef. In Hanwoo beef, grain-fed showed higher EPA and DHA than grass-fed beef.

Fat composition of beef, taken here to mean marbling, can be manipulated by time on feed, finishing diet, and breed type. These three factors also strongly influence the fatty acid composition of beef. Both the amount of marbling and the concentration of monounsaturated fatty acids (MUFA) increase with time on feed in grain-fed and pasture-fed cattle, but much more dramatically in grain-fed cattle. High-concentrate diets stimulate the activity of adipose tissue stearoyl-CoA desaturase (SCD), which is responsible for the conversion of saturated fatty acids (SFA) to their $\Delta{9}$ desaturated counterparts. Also, grain feeding causes a depression in ruminal pH, which decreases those populations of ruminal microorganisms responsible for the isomerization and hydrogenation of polyunsaturated fatty acids (PUFA). The net result of elevated SCD activity in marbling adipose tissue and depressed ruminal isomerization/hydrogenation of dietary PUFA is a large increase in MUFA in beef over time. Conversely, pasture depresses both the accumulation of marbling and SCD activity, so that even though pasture feeding increases the relative concentration of PUFA in beef, it also increases SFA at the expense of MUFA. Wagyu and Hanwoo cattle accumulate large amounts of marbling and MUFA, and Wagyu cattle appear to be less sensitive to the effects of pastures in depressing overall rates of adipogenesis and the synthesis of MUFA in adipose tissues. There are small differences in fatty acid composition of beef from Bos indicus and Bos taurus cattle, but diet and time on feed are much more important determinants of beef fat content and fatty acid composition than breed type.
In this study, a method of heat adaptation was implemented in an attempt to increase the upper thermal threshold of two Streptococcus thermophilus found in South Korea and identified the alterations in membrane fatty acid composition to adaptive response to heat. In order to develop heat tolerant lactic acid bacteria, heat treatment was continuously applied to bacteria by increasing temperature from 60℃ until the point that no surviving cell was detected. Our results indicated significant increase in heat tolerance of heat-adapted strains compared to the wild type (WT) strains. In particular, the survival ratio of basically low heat-tolerant strain increased even more. In addition, the strains with improved heat tolerance acquired cross protection, which improved their survival ratio in acid, bile salts and osmotic conditions. A relation between heat tolerance and membrane fatty acid composition was identified. As a result of heat adaptation, the ratio of unsaturated to saturated fatty acids (UFA/SFA) and C18:1 relative concentration were decreased. C6:0 in only heat-adapted strains and C22:0 in only the naturally high heat tolerant strain were detected. These results support the hypothesis, that the consequent increase of SFA ratio is a cellular response to environmental stresses such as high temperatures, and it is able to protect the cells from acid, bile salts and osmotic conditions via cross protection. This study demonstrated that the increase in heat tolerance can be utilized as a mean to improve bacterial tolerance against various environmental stresses.
