This study was conducted to define the effects of color of polyethylene (P.E.) net shading on the meteorological conditions under the shading growth status, photosynthesis and saponin contents of ginseng plant. Red and blue polyethylene net shading (4 fold) showed a good light intensity of ginseng growth, but red one not only increased air temperature but also resulted in early defoliation. Photosynthetic rate of ginseng leaves was increased in order of red, blue and black shade, Root yield and saponin contents were significantly increased red and blue net as compared with those of common straw shading. Blue color appeared to be most recommendable.
The effects of the crude saponin extracted from ginseng leaves on carbon dioxide evolution, alcohol fermentation, and cell production by Saccharomyces cerevisiae were investigated. The results were summarized as follows : 1) In the process of fermentation, $CO_2$ evolution by yeast was faster in the order of 3%, 1.5%, 0.7%, 5%, 7%, 0.3% than in control in concentration of the crude saponin extracts. 2) In the course of fermentation, the content of alcohol increased in the order of 0%, 0.3%, 7%, 1.5%, 3%, 5%. Among all these concentration, the production of alcohol was remarkably much in 5% and 3%. 3) In the process of fermentation, pH in the low content (0-0.7%) of the crude saponin extracts was gradually decreased as tine goes by and in the high content (1.5-7%) dropped suddenly between 24 hrs. and 48 hrs., and after 48 hrs., increased. 4) Dried yeast cell weight increased more in all the above concentration than control (0%) and among these it increased visibly in 3%.
As a part of studies on the quality control of crude drug preparation drinks, ginseng saponins were identified by HPLC. Ginsenoside-Rb1 was determined quantitatively by HPLC. Ginsenoside MeOH/H2O(65:35:10, v/v) on Si-gel plate. Ginsenoside-Rb1 content determined by HPLC on Lichrosorbtract drinks was 57.5-70.4% compared to the content in the red ginseng extract.
Optimum condition for cultivation of Anemarrhena asphodeloides B$_{UNGE}$ and seasonal variation of saponin and sapogenin contents in the rhizome were investigated. It is confirmed that the best harvest time is 3rd year. Average contents of saponin in rhizome was 8.5% and 5.0%, in fibrous roots, being highest in winter. The facotrs of saponin content variation were seemed to be temperature and humidity. It was also proved that asphonin was the mixture of timosaponin A-III, timosaponin A-I and mangiferin. Mangiferin and isomangiferin were isolated from the flower.
This studies were conducted to investigate the changes of saponin pattern, pH and organic acid contents of malt wort added ginseng components during alcoholic fermentation by Sacch. uvarum. The results are as follows. Saponin patterns of fermented wort were same as that of the non- fermented wort, but the weight of former was decreased comparing to that of the latter. pH value of fermented wort contained 0.1∼0.5% of ginseng extract were almost same as that of control(PH 4.23). Lactate, pyruvate, succinate and fumarate, pyroglutarate and citrate contents of the fermented wort were increased by the addition of ginseng extract and pyruvate content, particularly, was increased from 28.4 to 214mg/100 ml while that of control was 33.2mg/100m1. Citrate content of fermented wort contained ginseng saponin was almost same as control (37. 5mg/100m1) . But pyruvate content was tower 4-8.6mg/100m1 than that of control(33.2mg/100m1) .
To find the monthly variation of kalopanaxsaponin contents in the leaves of Kalopanax pictus with thorns (KPT) and with no thorns (KPN), the leaves monthly collected from May to September were extracted with MeOH and then the kalopanaksaponin fractions were prepared. KPT collected on May showed the lowest saponin content of the KPTs whereas KPT on August exhibited the highest saponin content. From September, the saponin content in the loaves decreased. The highest saponin content was shown to be 7.3% in KPN collected on August. Evaluation of six kalopanaxsaponins A, I, J, B, H and K (KPA, KPI, KPJ, KPB, KPH, and KPK) were performed using TLC densitometer. In this measurement, considerably higher KPB and KPH, both hederagenin bisdesmosides, were found whereas very low contents In monodesrnosides KPI and KPJ were observed. In conclusion, it was shown that the leaves of KPN of August could be a biomaterial source for the kalopanaxsaponin fraction. It was also suggested that measurement of the weight of kalopanaxsaponin fraction and the content of KPB as the representative compound for kalopanaxsaponins will be used for the quantitative evaluation of the kalopanaxsaponins of K. pictus.

Background : Platycodon grandiflorum radix used as a therapeutic agent for lung and respiratory diseases in traditional Korean medicine. The saponin and sugar content in P. grandiflorum root is dependent on the drying temperature and method. In this study, we investigated saponins and antioxidant contents in dried P. grandiflorum roots either not cut or cut to different lengths. Methods and Results : P. grandiflorum roots were either cut to 0.5, 1.0 or 2.0 cm in length or left uncut. They were dried at $45^{\circ}C$ and the effect of antioxidant substances, antioxidant ability, and saponin content were investigated. P. grandiflorum roots rapidly dried out when cut to short lengths. The saponin contents (platycodin D, polygalacin D, and deapioplatycodin D) was highest in the uncut P. grandiflorum roots. When the cut lengths were long, the levels of polyphenolic compound increased. However, 2,2'-azinobis-3-ethylbenzo-thiazoline-6-sulfonic acid (ABTS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities were higher in the samples cut to shorter lengths. Conclusions : These results suggested that the saponin and antioxidant content of P. grandiflorum roots could be maximized by controlling the drying rate of the roots via adjustments to the root cutting length.
In this experiment, observations were made on the effects of ginseng saponin, one of the major components of Korean ginseng (Panax ginseng, C. A. Meyer) root, on the membranes of microorganism (E. coli K-12), the concentration of intracelluar and extracellular cycle AMP therein, and uptake of U-14C-glucose. When the E. coli were grown on media containing 0.1% ginseng saponin, the growth was faster than for that of the control by about 30 minutes. The lysis of E. coli grown on the ginseng saponin medium increased to about double that of the control in the stationary phase. And the amount of protein and lipopolysaccharides in the outer cell meberances increased 25% and 80% respectively in comparison with the control. By electron microscope observation, it was shown that the periplasmic region of the E. coli grown on the ginseng saponin medium was widened it was observed that the cellular cyclic AMP content of the E. coli increased significantly to the hightest levels between the late exponential phase and early stationary phase. The total cyclic AMP content of E. coli grown on the ginseng saponin medium decreased about 50% when compared to that of the control.
In the present study, We have tested the potential effects of ginseng saponin fractions on macrophage chemotaxis and intracellular calcium and F-actin mobilization. Peritoneal macrophages treated with various ginseng saponin fractions showed 28.4% to 71% of increasement of chemotaxis as compared with untreated cells. The activity of intracelluar calcium mobilization was increased up to 65% by treatment with saponins, and F-actin content also increased 10% in the cells loaded with NBD-phallacidin. When the cells were activated with calcium of PMA and treated with saponin fractions, the intracelluar F-actin content increased significantly and prolonged for 2 minutes. These results suggest that ginseng saponin fractions might be a chemoattractants.
For the elucidation of saponin synthesis during ontogeny changes of ginsenosides and free sugars in seeds during stratification and seedlings in early growth stage were investigated with high performance liquid chrom-atography. Embryo plus endosperm at 40-day stratification showed 80% decrease of total saponin, disappear-ance of Rc, Rb$_2$ and Rb$_1$ and appearance of Rg$_3$ (probable) and 20-Glc-Rf (probable). Leaf ginsenoside F$_3$ was found not in fruit plup but seed and decreased during stratification. Both decomposition and synthesis of saponin seemed to occure during stratification. Ginsenosides in endosperm and embryo might be originated from fruit pulp by penetration. In seedling saponin appeared first in shoot and in root about one month later. Ginsenoside Rc, Rb$_2$, Rb$_1$ appeared in root at the last investigation (June 30) indicating normal saponin synthetic capacity of root. Saponin synthetic rate was twice in leaf than in root. Leaf ginsenoside F$_3$ was found in seedling root. Root saponin Rg$_3$ and 20-Glc-Rf were found in leaf and stem in seedling and decreased with growth suggesting that rate saponin is not such in certain growth stage. Total saponin content was negatively correlated with PT/PD in seeds and arial parts of seedling due to greater change of PD. than PT. Seed at 70days stratification showed high sucrose content. In seedling glucose was main sugar in stem all the while and sucrose in root at early stage while glucose, fructose and sucrose were found in leaf.
