The thymus weight of the mouse was 54.1% in protein malnutritive diet group and 39.2% in group treated with saponin fraction of Panax ginseng in comparison to normal diet group. This decreasing effects of protein malnutritive diet and saponin fraction on the thymus weight practically disappeared after four weeks. The saponin fraction showed no effect on the spleen weight of the mouse. The supplement of the saponin fraction enhanced total peritoneal exudate cells, content of total serum protein and albumin content of the mouse, each 45, 8 and 10% respectively in comparision to that of normal diet group. And these values in protein malnutritive diet group were 61.2, 83.6 and 87.0% respectively in comparision to that of normal diet group, and recovered to the level of normal diet group by the supplement of the saponin fraction. The electrophoregram of the serum protein of the mouse fed with protein malnutritive diet was different from that of the mouse fed with normal diet, but this difference practically disappeared by the supplement of the saponin fraction.
Crude saponin and ginsenosides in commercial white ginsengs such as six kinds of whole ginsengs, five kinds of tail roots and one undergrade raw ginseng were analyzed. The contents of crude saponin and ginsenosides in whole ginsengs were found to be 2.7 to 4.6% and 1.0 to 1.7%, respectively. On the other hand, those of tail roots were 3.3 to 7.2% and 1.3 to 4.4%, respectively. The content ratio of PD to PT saponin in whole ginsengs showed a little variation as 0.73 to 0.92, while those of tail roots showed a greater variation in the range of 0.72 to 1.75, indicating that tail roots contain higher content of PD saponin than whole ginsengs did.

This study was carried out to isolate saponin compounds from red-ginseng efflux, which was produced during the industrial processing of red-ginseng from fresh ginseng. We isolated crude saponin from the efflux extract (moisture content 35.0%) by using Diaion HP-20 adsorption method. Non-saponin fraction, which was adsorbed on Diaion HP-20 resin, was removed by eluating with $H_{2}O$ and 25% spirit. Then crude saponin was eluated with 95% spirit, continuously. Saponin in the eluated fractions was confirmed by TLC analysis. Crude saponin isolated from red ginseng efflux extract contained 12.10% of saponin. whereas those of white ginseng and red-ginseng were 3.30 and 3.39%, respectively. Ginsenoside contents showed the highest contents kin crude saponin from red ginseng efflux extract. Expacilly, the ginsenoside-$Rb_{1}$ and Re showed the highest contents in red-ginseng efflux extract when compared with those of white ginseng and red ginseng crude saponins. And the other ginsenosides except ginsenoside-$Rb_{1}$ and -Re also showed the highest contents in red ginseng efflux extract. However, the ratio of PD saponin (Panaxadiol saponin: $Rb_{1}+Rb_{2}$+Rc+Rd) to PT saponin (panaxatriol: $Re+Rg_{1}$) showed almost the same level when compared with those of ginseng saponin fractions. Ratio of PD/PT from red ginseng efflux extract was 1.99. Ratios of PD/PT from white ginseng and red ginseng were 1.85 and 1.84, respectively. Saponin purity, which was calculated by ratio percent of total ginsenoside to curde saponin content, was 45.90%. In case of white ginseng and red ginseng, the purities were 35.50 and 36.00%, respectively. However, by PHLC analysis, we confirmed that crude saponin isolated from red ginsengs. It suggested that crude saponin isolated from red ginseng ellux also would be useful component as ginseng saponins.
On the cAMP content in isolated gastric glands from rabbit stomach, the effect of ginseng components (total saponin, diol saponin, triol saponin) with pepsinogen secretion regulatory agents (cholecystokinin, isoproterenol, carbachol, propranolol, atropine, DECAMP, DBcGMP) were studied in vitro. According to the results, ginseng components may have the effect of stimulation or inhibition on cAMP production, and both dial saponin and triol saponin may be reciprocal effect to pepsinogen secretion regulatory agents. It seemed that the ginseng components may have the normalization action to pepsinogen regulatory agents on cAMP content in isolated rabbit gastric glands.
It has generally been accepted that quality of ginseng should be determined not by the content of a single component but by composition and balance of total active principles. However, there still can be an exception with a product in which a given ginsenoside is used for the treatment of a specific disease. Although ginsenosides have been regarded to be major active components of ginseng and employed as index components for the quality control, it does not consistent with the traditional concept on ginseng quality creterion; main root has been more highly appreciated than the lateral or fine root. Content of ginsenosides in the lateral or fine root is much higher than that in main root. However, the ratio of protopanaxadiol (PD) and protopanaxatriol (PT) saponins existing in various part of ginseng root is greatly different. The ratio of PD/PT saponins in main root is well balanced but the thinner the root is the higher the ratio. Thus far, a total of 34 different kinds of ginsenosides have been isolated from Korean (red) ginseng, and their pharmacological activities were elucidated partly. Interestingly, different ginsenoside shows similar or contrary effects to each other in biological systems, thus indicating the significance of absolute content of single ginsenoside as well as compositional patterns of each ginsenoside. Therefore, pharmacological activities of ginseng should be determined as a wholly concept. In these regards, standardization of ginseng material (fresh ginseng root) should be preceded to the standardization of ginseng products because ginsenoside content and non-saponin active principles such as polysaccharides and nitrogen (N)-containing compound including proteins are significantly different from part to part of the root. In other words, the main root contains less ginsenosides than other lateral or fine roots. Contents of polysaccharides and N-containing compound in main root is higher. However, the quality control of ginseng products focused on non-saponin compounds has limitation in applying to the analytical method, because of the difficult chemical analysis of these compounds. Content of ginsenosides, and ratios of PD/PT and ginsenoside Rb,/Rg, are inversely proportional to the diameter of ginseng root. Therefore, these can be served as the chemical parameters for the indirect method of evaluating from what part of the root does the material originate. Furthermore, contents of polysaccharides and N-containing compounds show inverse relationship to saponin content. Therefore, it seems that index for analytical chemistry of saponin can be applied to the indirect method of evaluating not only saponin but also non-saponin compounds of ginseng. From these viewpoints, it is strongly recommended that quality of ginseng or ginseng products be judged not only by the absolute content of given ginsenoside but also by varieties and compositional balance of ginsenosides, including contents of non-saponin active principles.
the purpose of this paper is to study on the proximate composition, mineral and saponin content in white sesame, black sesame, and perilla seed. For this purpose, we have compared six different materials: white-raw-sesame, white-roast-sesame, black-raw-sesame, black-roast-sesame, raw-perilla seed and roast perilla seed, and have come to the following results. The crude fat content was the highest in white-raw-sesame(55.3%). In all the six samples, the crude fat content in raw seeds was all higher than that in roast seeds. The crude protein content was the highest in the roast perilla seed(24.6%), and in the six samples, the crude protein content in roast seeds all higher than that in roast seeds. The total sugar content was found to be the highest in the roast perilla seed(8.29%). The reducing sugar content was higher in raw perilla seed(1.57%) than in other sample materials. The ash content was the highest in black raw-sesame(5.93%), and that percentage rates was the same as that of FAO and of Japan. Minerals like Cd. Mn. Cu. Na. Mg. Pb. and Ca. were found to be contained more in black sesame than in other sample materials. The minerals contained most in white sesame were Zn.(61.6ppm) and Fe(49.4ppm), and K was contained a little more in perilla seed than in the others. The sample materials which contain saponin most were white-roast-sesame(0.34%) and black-roast-sesame(029%).
The effects of Ginseng saponin on respiration and $Na^+$, $K^+$ content of rat cerebral cortex slices were investigated to determine the action of Ginseng saponin on brain cortex at cellular level. There are many reports for the study of Ginseng on central stimulatory action in experimental animals. The electrical stimulation of slices of cortex causes a loss of potassium. And the respiration is needed to maintain a supply of energy for active cation transport. The reduction in $Qo_2$ is a consequence of primary cessation of active cation transport. Ginseng saponin stimulated respiration which was depressed by Morphine. But there was no significant change of electrolyte. It is suggested that the Ginseng saponin act rather on metabolic process than neural excitatory mechanism in vitro.
The increased level of glucose, ketone bodies, non-esterified fatty acids and lactate in blood, decrease of glycogen content, phosphofructokinase activities and glucokinase activity and the increased level of glucose-6-phosphatase activity in the liver of streptozotocin injected rats were significantly moderated by ginseng saponin administration. It is not likely, however, that the hypoglycomic action of ginseng saponin might be due to their direct action on enzyme activities, since the saponin effect of servers enzymes in vitro was not enough to explain such an appreciable hypoglycemic activity of the saponin in streptozotocin induced diabetic rats, for which much work have to be done.
This stuffy was carried to establish a new efficient method for the preparation of edible crude ginseng saponin. The conventional butanol extraction and resin adsorption methods were compared for the contents of total crude ginseng saponin and major ginsenosides. Seventy- percent methanol extract was applied to Diaion HP-20 column and the resin was washed with Hn and eluted with absolute methanol. The methanol elute was dried in vivo and analyzed for its ginsenosides. Use of ethanol instead of methanol to make edible crude ginseng saponin gave a similar result. Butanol extraction was performed by the conventional method. The final aqueous layer from butanol extraction was passed through Diaion HP-20 column followed by elution with methanol and Diaion HP-20 passed fraction was extracted with butanol to recover remaining components, respectively, in order to determine saponin loss. TLC and HPLC qualitatively and quantitatively monitored Ginsenosides, respectively. Loss of ginsenosides was higher in butanol extraction method than in Diction HP-20 adsorption method. In addition, saponin fractions prepared by Diction HP-20 adsorption method showed higher content of each ginsenoside, showing 8.2% higher purity than that of butanol extracted fraction. From these results, we propose the resin adsorption method as a new efficient measure for the preparation of crude ginseng saponin, which is edible by using spirit instead of methanol.
Hairy root clones of Panax ginseng were established by selection of some hairy roots formed on the leaf, stem and root segments transformed with Agrobacterium rhizogenes strain $A_4$. The transformed roots grew well in MS medium under the dark condition. To confirm the transformation with Ri-T-DNA, dot blot hybridization and opine analysis were Performed. Among four hairy roots induced from different part of ginseng, the HB3 hairy roots were examined for selection of high-saponin-producing clones. Four clones isolated from HB3 hairy root cultures displayed various phenotypes characterized by growth and total saponin content. Maximum growth was obtained for cultures of HB3-10 clone and the content of total saponin was 0.55 wt%. However, higher amount of total saponin was obtained with HB3-2 clone cultures(0.74 wt%) in spite of lower growth. Dot blot hybridization confirmed the introduction of Ri-T-DNA in the plant genome. In the opine test, agropine and mannopine were detected from all hairy root clones.
