멸치 젓갈로부터 분리된 젖산세균의 프로바이오틱 특성 및 안전성 평가 (Probiotic properties and safety assessment of lactic acid bacteria isolated from salt-fermented anchovy)

  • 멸치 젓갈로부터 분리한 젖산세균을 대상으로 프로바이오틱 균주로서의 기능적 특성과 안전성을 평가하였다. 분리된 균주는 생화학적 특성과 당 발효능 및 염기순서 분석을 통해 Enterococcus faecium AJ06, Leuconostoc mesenteroides AJ13, Pediococcus halophilus AJ22, Lactobacillus sakei AJ29 및 Pediococcus pentosaceus AJ35로 동정되었다. AJ06, AJ22 및 AJ29 균주들은 펩신이 첨가된 인공 위액 및 쓸개즙액에서 강한 저항성을 나타내었고, taurocholic acid 혹은 taurodeoxycholic acid가 첨가된 MRS 한천 평판 배지 상에서 쓸개즙분해효소를 생산하였다. 특히, Caco-2 세포에 대한 높은 부착능과 다양한 항생제에 대한 저항성을 나타낸 AJ22와 AJ29 균주는 항균물질 생산으로 인해 식중독균의 증식을 효과적으로 저해하였다. 이들 균주는 혈액 한천 평판배지 상에서 알파 및 베타용혈독을 나타내지 않았고, 아미노산 전구체가 함유된 MRS 액상배지 내에서 생체 아민을 생산하지 않았으며, Salmonella Typhimurium TA98 and TA100 균주에 대한 돌연변이도 유발하지 않았다.

Physicochemical Characteristics of Beef Jerky Cured with Salted-fermented Anchovy and Shrimp

  • The aim of this study is to evaluate the availability of salted and fermented fish (SFF) including salted and fermented anchovy (SFA) and shrimp (SFS) as a marinade of beef jerky. In curing solutions, half (SFA 1 and SFS 1) or whole (SFA 2 and SFS 2) salt-water was replaced with SFF juices. Higher water activity ($a_w$) was found in the beef jerky cured with SFFs than the control (C) (p<0.05). The SFFs had the effect of causing a decrease in hardness and an increase in cohesiveness (p<0.05). Among the treatment samples, springiness was the highest in SFA2 and SFS2 (p<0.05) and the lowest values of Warner-Bratzler shear force were found in SFA1 and SFA2 (p<0.05). The SFFs also had the effect of increasing the flavor of the sensory properties; however, color measurements from both the instrumental surface color ($L^*$, $a^*$, $b^*$, chroma, and hue angle) and color of sensory evaluation were decreased by addition of SFFs (p<0.05). Therefore, we conclude the SFFs can improve the texture and sensory properties of the beef jerky. In particular, the SFS is a good ingredient for the curing solution. However, studies are still needed on improving the $a_w$, pH, and surface color of the beef jerky to apply the SFFs for making beef jerky.

깍두기 숙성 중 N-Nitrosamine의 생성 (The Formation of N-Nitrosamine during Fermentation of Kakdugi)

  • Traditional kakdugi prepared from salted radish by the addition of fermented shrimp or anchovy sauce along with seasonings and spices, e.g., red pepper powder, garlic or ginger, which has been examined for the formation of volatile nitrosamine(NA) during its fermentation at 5oC and 17oC. Changes of acidity were increased; while, those of ascorbic acid and pH were continually de creased and besides concentrations of salt were scarcely observed during the fermentation of kakdugi at 5oC and 17oC. High initial levels of nitrate in radish followed by a gradual decreased in nitrate concentration, along with increasing levels of nitrite, during its fermentation. During the fermentation of kakdugi, NA such as N nitrosodimethylamine(NDMA) and N nitrosodiethylamine(NDEA) was identified; these NA in kakdugi that had been prepared with shrimp sauce contained in the higher levels than kimchi that had been prepared with anchovy sauce, and both NDMA and NDEA were detected higher levels in the fermented sample at 17oC than in the fermented sample at 5oC. In kakdugi, NDMA and NDEA levels were continually increased during its fermentation; its levels after 35 days fermentation at 17oC were 0.5~16.7 and 0~5.2 g/kg in added shrimp sauce sample, 0. 5~8.7 and 0~4.7 g/kg in added anchovy sauce sample and 0.4~4.8 and 0~2.4 g/kg in control, respectively. The occurrence of NA such as NDMA and NDEA appeared to arise from the fermented sauce and radish used in preparation of the kimchi.

배추김치 부재료 혼합의 모델화와 품질 (Studies on Modelization of Subingredients of Chinese-Cabbage Kimchi)

  • This study was conducted to ivestigate the quality of modeled chinese-cabbage Kimchi. The mutual action of sub-ingredients and optimum mixing conditions were examined by analyses of pH, acidity, content of vitamins, carotenes, number of total microorganism, and Lactobacilli, the activities of softening related to enzymes (polygalacturonase and ${\beta}$-galactosidase), and sensory quality during fermentation. The result about the mutual action of red pepler powder and garlic showed that the pH was decresed as concentration of garlic was higher and that of red peppler powder became lower. Kimchi had unpleasant taste and undesirable odor when the adding ratio of the peper powder and garlic was unbalanced. As the concentration of garlic became higher, the content of vitamin C in Kimchi tissue decreased while that of vitamin C in Kimchi juice increased. The contents of vitamin B1, B2 and carotene increased as the concentration of garlic became lower. The optimum mixing ratio of red pepper powder and garlic evaluated by overall eating quality was 4.66% and 3.45%, respectively. The result about the mutual action of ginger and green onion was that pH became higher as the concentration of ginger was 1.84% and that of green onion was 5.96%. The intensity of fresh taste and odor became stronger as the concentration of ginger increaed to 2.76% and that of green onion decreased to 2.98%. The balance of sour, salty and hot taste became better as the concentration of ginger increased and that of green onion decreased. The optimum mixing condition evaluated by overall eating quality was 2.94% in ginger + 2.78% in green onion. The contents of vitamin B1, B2 and C were high in this condition. The result about the mutual action of leek and salted anchovy juice was that pH became lower as the concentration of leek increased and that of salted anchovy juice decreased. Sour, hot and sweet tastes were good in the condition of leek 12%, salted anchovy juice 4.69%. Palatable, fresh taste and odor became weaker when the one of both sub-ingredients was too much little. The optimum mixing condition evaluated by overall easting quality was 9.76% in leek + 7.32% in salted anchovy juice. The contents of vitamin B1, B2, C and carotene were high in this condition and other sensory qualities were good. The result about the mutual action of salt and sugar was that pH became lower as the concentration of sugar became higher and that of salt became lower. The sweet taste was reduced of hot taste than salty taste. Palatable and fresh tastes were desirable when Kimchi fermented with less salt and more sugar. The optimum mixing condition evaluated by overall eating quality was 3.29% in sugar + 4.80% in salt. The contents of vitamin B1, B1, C and carotene were high in this condition. The result of model Kimchi fermented at 20$^{\circ}C$ was that the number of Lactobacilli in model Kimchi was higher than that of in control Kimchi. The fermentation period was extended under pH 4.0 and the contents of vitamin B1, B1,C and carotene were high through the whole edible period. The activity of polygalacturonase was low, but that of ${\beta}$-galactosidase was high during fermentation.

멸치액젓의 품질특성에 미치는 여과조제 처리의 효과 (Effect of Filter Aid Treatment on the Quality Characteristics of Salt-Fermented Anchovy Sauce)

  • 멸치액젓제품의 풍미, 색택개선 및 유통중 불용성 침전생성 억제를 위한 기초자료를 얻기 위하여 멸치액젓을 활성탄, 활성백토. 알루미늄옥사이드, 규조토 및 celite의 농도를 달리하여 첨가하고 상온에서 60분간 교반한 후 0.45$\mu$m membrane filter로 감압여과 한 다음 탈색률, 총질소, 아미노태질소, 불용성질소, TMA 및 침전생성 억제율을 측정하였다. 탈색율은 활성탄과 활성백토를 첨가하였 때 높았으며 $5\%$ 첨가농도에서 $32\~33\%$의 탈색효과를 나타내었으나 TMA 함량은 각 처리구에서 $7\%$ 이하의 감소를 나타내어 탈취효과는 기대할 수 없었다. 멸치액젓 저장중 침전물의 생성억제 효과는 $5\%$ 처리농도에서 활성백토 $82\%$, 규조토 $73\%$$Al_2O_3$ $70\%$로 나타났다.

소금 및 멸치적갈이 Drosophila 돌연변이유발에 미치는 영향 (Effects of Salt and Fermented Anchovy Extract on the Somatic Mutagenicity in Drosophila Wing Spot Test System)

  • In vivo Drosophilla 돌연변이 검출계에서 발생 단계상의 독성효과가 없는 범위에서 소금 및 젓갈을 단독 처리했을 때 모든 시료가 자연발생 빈도가 유사하게 나타났다. MNNG의 체세포 돌연변이 유발에 미치는 영향을 살펴본 결과 생멸치, 6개월간 속성된 멸치 12개월된 멸치액젓은 항돌연변이효과를 보였고, 발효하지 않은 소금에 절인 멸치는 small muh spots 와 large mwh spots 의 출현빈도에 대해 모두 보돌연변이 효과를 보였다. 따라서 멸치 젓갈 제조시 멸치에 소금이 첨가되어 (생젓갈) 돌연변이 유발이 일어날 가능성이 있지만 숙성기간이 길어지면 (12개월, 익은 젓갈) 오히려 항돌연변이효과가 있는 것으로 나타났다.

멸치젓 첨가량이 부추김치의 발효숙성에 미치는 영향 (Effect of Various Level of Salted Anchovy on Puchukimchi Fermentation)

  • This study was undertaden to investigate the effect of salted anchovy during Puchukimchi fermentation, Puchukimchi is a kind of Korean kimchi made with Puchu(Allium odorum L.). For this purpose, the Puchukimchi with 10%, 15% and 30% salted anchovy was investigated by measuring physicochemical, microbiological and sensory characteristics during fermentation at 5$\pm$2$^{\circ}C$ up to 60 days, In the palatable period, pH was remarkably decreased to the range of 4.8-5.16 and total acidity was increased 2-3 times more than that of initial period. Reducing sugar contents in Puchukimchi increased up to 22days and remarkably decreased. The changing pattern of vitamin C drawed sigmoidal curve, invreasing significantly in the palatable period. The content of lactic acid was a little in the initial fermentation period but increased 3-13 times more than that of the initial fermentation period in the palatable period. The content of malic acid and citric acid were decreased as the fermentation progressed. The contents of Ca, Mg and K was gradually decreased and Na was increased as the fermentation progressed in the same period. There were similiar changes in the number of lactic acid bacteria and total cell number. The score of overall taste, overall acceptability in evaluation during fermentation was high in order of 15%, 10%, 30% level of salt fermented anchovy.

숙성 및 저장 중 액젓의 색도 변화 (Changes in color value of salt-fermented fish sauces during fermentation and storage)

  • 액젓을 분광광도계로 최대 흡수파장을 scanning 하고, 12개월 숙성된 멸치액젓에 glucose, MSG, IMP, 카라멜색소 및 타르색소를 첨가하여 저장기간에 따라 색도의 변화를 측정하여 액젓의 품질 판정을 위한 지표로서의 사용가능성을 조사한 결과는 다음과 같다. 액젓의 종류별, 희석비별, 첨가색소별, 첨가물별에 따라서 scanning한 결과, 453nm에서 최대 흡광도를 나타내었고, 분광광도계로 453 nm에서 측정한 시판 멸치액젓의 색도는 직시색차계로 측정한 ${\delta}E$값 (색차)보다 감도가 더 좋았으므로, 액젓의 색도 측정시 직시색차계보다는 분광광도계로 453 nm에서 측정하는 것이 더 적합할 것으로 판단되었다. 숙성기간 중에는 색도값이 일정하게 증가하였으며, 까나리액젓이 멸치액젓보다 그 값이 높았다. 첨가물의 영향은, 당 (glucose)과 질소화합물 (MSG, IMP)을 첨가한 시료는 저장기간 동안 색도값이 일정하게 증가 하였으며, 농도별에 따른 큰 차이는 없었다. 색소의 영향은, 카라멜색소를 첨가한 액젓의 색도값은 첨가농도가 높을수록 높았으며, 저장기간중에 일정하게 증가하였다. 한편, 타르색소를 첨가한 액젓은 첨가농도 $0.003{\%}$$0.005{\%}$에서 저장 15일 후에 색도값이 급격히 증가하였으며, 저장기간 중에도 일정하게 증가하였다.

새우가공부산물을 이용한 속성 멸치액젓의 품질특성 (Quality Characteristics of Accelerated Salt-fermented Anchovy Sauce Added with Shrimp Pandalus borealis, Byproducts)

  • 새우가공부산물의 효율적 이용과 멸치 액젓의 영양성 및 기호성의 강화 목적으로 새우가공부산물 첨가 멸치액젓 (새우가공부산물 무첨가 및 $10\%$ 첨가 멸치액젓의 경우 270일 숙성, $20\%$$30\%$ 첨가 멸치액젓의 경우 180일 숙성)을 제조하여 엑스분 질소, 맛성분, 영양성분 및 관능검사에 대하여 분석 검토하였다. 멸치액젓의 엑스분 질소함량은 새우가 공부산물의 첨가량이 증가할수록 증가하는 경향을 나타내었으며, 맛의 주요성분을 살펴보기 위하여 omission test를 실시한 결과, 새우가공부산물의 첨가 유무에 관계없이 맛의 주성분은 유리아미노산이었고, ATP 관련물질 및 유기산 등의 성분은 맛에 보조적으로 관여하리라 판단되었다. 새우가 공부산물 첨가 멸치액젓의 유리아미노산 및 taste value의 결과, 멸치 액젓의 맛은 모두 glutamic acid와 aspartic acid에 의하여 좌우됨을 알 수 있었고, 첨가비율이 증가할수록 proline, valine 및 histidine의 함량과 조성비가 감소하는 경향인 반면에, methionine, isoleucine, leucine, phenylalanine 및 lysine은 증가하는 경향이었다. 총 아미노산은 새우가공부산물의 첨가량에 관계없이 $9,840\~10,325$ mg/100 g 범위였으며, 무기질 함량은 새우가공부산물의 첨가량이 증가할수록 칼슘, 인, 철 및 칼륨 모두가 증가하는 경향을 나타내어 새우가공부산물의 첨가에 기인함이 확인되었다. 또한 새우가공부산물 첨가 멸치액젓의 향, 맛 및 냄새에 대한 관능검사 결과, 쓴맛이 느껴지지 않으면서 새우 특유의 향을 느낄 수 있는 $20\%$ 첨가 제품이 가장 적절하였다. 멸치액젓의 제조시 새우가공부산물을 마쇄한 다음 첨가하여 숙성시키는 경우 맛, 향 및 영양 개선은 물론이고, 수산가공자원의 효율적인 이용 측면에서 의미 있는 결과를 얻었다.

  • A bacteial strain which can produce the extracellular fibrinolytic enzyme was isolated from Jeot-Gal (anchovy) that was Korean traditional salt-fermented fish. The isolated bacterium was identified to be a strain of Bacillus sp. The optimal medium for fibrinolytic enzyme production was determined to consist of 5 g maltose, 10 g defatted soybean, 20 g sodium chloride, 1.75 g K2HPO4 per liter (pH 7.0)
