ELECTRICAL BREAKDOWN INITIATION OF ANODIC FILMS DURING ANODIZING IN MOLTEN BISULPHATE MELT

  • The morphology and composition of anodic films, formed on aluminium at various current densities, in the range $1-100{\;}Am^{-2}$, in the molten bisulphate melt at different temperatures (418-498K), have been studied using transmission electron microscopy of ultramicrotomed film sections, and ion beam thinned films. The first sign of incipient breakdown revealed by transmission electron microscopy of stripped films, is always the appearance of dark regions about 1,000 nm in diameter, representing local overgrowth of the film. The breakdown mechanism is closely related to thermal effects, because temperature rises at regions representing local overgrowth in the stripped films were observed at voltages close to the breakdown voltage, likely arising through impact ionization.

Development of a Novel Yeast Strain Which Ferments Soy Sauce by Protoplast Fusion

  • In order to develop a novel yeast which produces the charateristic aroma of soy sauce, a protoplast fusion between Zygosaccharomyces rouxii WFS4 and Torulopsis versatilis IAM 4993 was carried out. Auxotrophic mutants as selective markers were obtained from Zygosaccharomyces rouxii and Torulopsis versatilis by treatment of N-methyl-N -nitro-N-nitrosoguanidine. The conditions of the protoplast formation and the regeneration for fusion were examined. The protoplast fusion using polyethylene glycol 4000 led to the fusion frequency of $4~5{\times}10^{-7}\;cells/ml$. Among fusants, a fusant ST723-F31 presented the best results in terms of the aromaticity of fragrance, the growth pattern, the resistance against salt and the degree of growth according to pH. It makes easy to control the production and the balance of aroma components so that it gives a good flavor, shortens the fermentation period and, simplifies the preparation process when using a bioreactor into which fusant is immobilized.

  • In response to the global interest and efforts towards reducing plastic use and promoting resource recycling, there is a growing need to establish methods for recycling discarded fishing gear. In Korea, various technologies are being developed to recycle discarded fishing gear, but significant technical and policy challenges still remain. In particular, biodegradable gill nets require a pre-treatment process to separate biodegradable materials from other substances and to remove salt before recycling. Therefore, this study aims to develop a pre-treatment device for recycling biodegradable gill nets and to evaluate the feasibility of recycling them.

Opportunities for Agricultural Water Management Interventions in the Krishna Western Delta - A case from Andhra Pradesh, India

  • Agricultural water management has gained enormous attention in the developing world to alleviate poverty, reduce hunger and conserve ecosystems in small-scale production systems of resource-poor farmers. The story of food security in the $21^{st}$ century in India is likely t o be closely linked to the story of water security. Today, the water resource is under severe threat. The past experiences in India in general and in Andhra Pradesh in particular, indicated inappropriate management of irrigation has led to severe problems like excessive water depletion, reduction in water quality, water logging, salinization, marked reduction in the annual discharge of some of the rivers, lowering of ground water tables due to pumping at unsustainable rates, intrusion of salt water in some coastal areas etc. Considering the importance of irrigation water resource efficiency, Krishna Western Delta (KWD) of Andhra Pradesh was purposively selected for this in depth study, as the farming community in this area are severely affected due to severe soil salinity and water logging problems and hence, adoption of different water saving crop production technologies deserve special mention. It is quite disappointing that, canals, tube wells and filter points and other wells could not contribute much to the irrigated area in KWD. Due to less contribution from these sources, the net area irrigated also showed declining growth at a rate of -6.15 per cent. Regarding paddy production, both SRI and semi-dry cultivation technologies involves less irrigation cost (Rs. 2475.21/ha and Rs. 3248.15/ha respectively) when compared to transplanted technology (Rs. 4321.58/ha). The share of irrigation cost in Total Operational Cost (TOC) was highest for transplanted technology of paddy (11.06%) followed by semi-dry technology (10.85%) and SRI technology (6.21%). The increased yield and declined cost of cultivation of paddy in SRI and semi-dry production technologies respectively were mainly responsible for the low cost of production of paddy in SRI (Rs. 495.22/qtl) and semi-dry (Rs. 532.81/qtl) technologies over transplanted technology (Rs. 574.93/qtl). This clearly indicates that, by less water usage, paddy returns can be boosted by adopting SRI and semi-dry production technologies. Both the system-level and field-level interventions should be addressed to solve the issues/problems of water management. The enabling environment, institutional roles and functions and management instruments are posing favourable picture for executing the water management interventions in the State of Andhra Pradesh in general and in KWD in particular. This facilitates the farming community to harvest good crop per unit of water resource used in the production programme. To achieve better results, the Farmers' Organizations, Water Users Associations, Department of Irrigation etc., will have to aim at improving productivity per unit of water drop used and this must be supported through system-wide enhancement of water delivery systems and decision support tools to assist farmers in optimizing the allocation of limited water among crops, selection of crops based on farming situations, and adoption of appropriate alternative crops in drought years.

  • 안양천 공단 주변 슬러지를 미생물 접종원으로 무기염배지에 10 g/L의 자당을 첨가하여 수소 생산 균주 MeL 6-2을 분리하였다. 분리 균주 MeL 6-2은 호기성조건과 혐기성 조건에서 모두 생장하는 통성 혐기성 균주 Rhodopseudomonas sp.였다. 유기성 폐기물 내에 다량 함유되어있는 포도당과 자당의 농도변화가 수소 생산 속도 및 수소 생성효율에 미치는 영향에 대하여 알아보았다. 포도당을 1~12 g/L의 범위로 첨가할 경우 lag phase 없이 생장하였으며, 첨가량이 증가할수록 단위시간 및 단위부피당 수소 생산성 이 증가하여, 10 g/L에서 최대값인 $4.2\;mmol-H_2{\cdot}L^{-1}{\cdot}h^{-1}$을 보이고 그 이후 다소 감소하는 경향을 보였다. 균체량에 대한 수소생산수율은 $0.76{\sim}2.46\;L-H_2{\cdot}g-DCW^{-1}$의 값을 보였으며, 첨가된 기질인 포도당에 의한 수소생산수율은 $2.6{\sim}3.1\;mol-H_2{\cdot}mol-glucose^{-1}$의 범위였다. 자당을 1~12 g/L의 범위에서 첨가할 경우 약 10시간의 지체기 후 원할한 생장을 보였다. 단위시간 및 단위 세포무게 당 비수소 생산속도는 및 수소 생산수율은 자당의 첨가량이 증가할수록 증가하여 각각 $163\;mmol-H_2{\cdot}mg-DCW^{-1}{\cdot}h^{-1}$$4.5\;mol-H_2{\cdot}mol-glucose^{-1}$의 최대값을 보였다.

Pichia pastoris에서 메탄올 유도시 첨가물이 재조합 HBsAg 생산에 미치는 영향 (Effect of Various Additives on the Production of Recombinant HBsAg during Methanol Induction in Pichia pastoris)

  • 본 연구에서는 P. pastoris를 이용한 유전자 재조합 HBsAg 생산에서 메탄올 유도시 여러 가지 첨가물들의 영향에 대해서 알아보았다. 회분식 배양에서 탄소원으로 글리세롤을 사용하다가 유가식 배양의 공급 탄소원으로 글리세롤이 아닌 당알콜인 sorbitol로 대체하였을 때 단백질 발현이 향상된 결과를 보였다. 또한 메탄올 유도시에 적당량의 아미노산 혼합물 첨가는 세포증식에는 영향이 없었지만 단백질 발현율은 크게 증가시켰다. 계면활성제인 Trition X-100의 첨가는 세포증식과 단백질 발현을 현저히 감소시켰지만, Pluronic F-68를 첨가했을 경우 세포증식의 저해영향 없이 단백질 발현율을 향상시켰다. 배지 부피의 0.01%(v/v)으로 oleic acid를 첨가하면 플라스크 배양에서는 단백질 발현에 긍정적인 효과를 보였으나, 5 L 발효조 배양에서는 메탄올 유도 시간이 지속되면서 첨가하지 않은 경우에 비해 발현율이 낮아지는 결과를 보였다. 마지막으로 trace salts는 첨가량에 따라 세포증식에는 영향이 없으며 단백질 발현에는 소량 trace salts 첨가로 단백질 발현에 긍정적인 효과를 보였다. 하지만 첨가량이 많아질수록 단백질 발현에는 부정적인 영향을 보임을 확인할 수 있었다.

Candida tropicalis의 2단계 유가식 배양에 의한 Xylitol 생산의 최적화 (Optimization of Xylitol Production by Candida tropicalis in Two-stage Fed-batch Culture)

  • C. tropicalis ATCC 20336을 이용하여 높은 수율과 생산성으로 xylitol을 생산하기 위하여 glucose 배지에서 짧은 시간에 고농도로 세포를 배양한 후에 xylose를 첨가하여 xylitol로 전환하는 2단계 유가식 발효공정에 관한 연구를 수행하였다. Cell growth-stage에서의 배지공급 방법으로는 glucose가 모두 고갈된 후에 pH가 5.7 이상으로 올라가는 것을 신호로 glucose를 첨가하는 pH-stat 방법으로 18.5시간 배양에 의하여 67.9 g/L의 세포 농도를 얻었으며, ethanol 생성은 1.0 g/L로 매우 낮았다. Production-stage에서 최적의 초기 xylose 농도는 150 g/L이었으며, 이때에 2 g/L ($NH_4)_2SO_4$, 1 g/L $K_2HPO_4$, 0.2 g/L $MgSO_47H_2$O로 구성된 mineral salt를 2회 첨가에 의하여 xylitol의 생성이 향상되었다 그러나 0.2 vvm에서 1.0 vvm 사이의 통기조건에서는 xylitol의 생산에 큰 영향을 미치지 못하였다. 반면에 production-stage에서 yeast extract를 10 g/L가 되도록 첨가한 경우에 xylitol 수율과 생산성이 큰 폭으로 증가하였다. 즉 xylose 농도가 약 150 g/L이 되도록 232.5 g의 xylose를 2회 첨가한 경우에 배양액의 최종 부피는 1.67 L이 되었고, 이때의 xylitol 농도는 193 g/L이었으며, 수율은 70%이고 생산성은 3.55 g/L.h이었다.

국산과 수입산 포장 배추 김치의 이화학적 특성 비교 (Evaluation of Physieochemical Characteristics of Domestic and Imported Kimchi)

  • This study evaluated the physicochemical characteristics of domestic and imported packaged Kimchi in Korea. Six types of domestic and imported Kimchi were collected from a supermarket. According to the results, the salt concentrations of the domestic and imported Kimchi were 3.83% and 3.06%, respectively. The total lactic acid content was 0.33% for domestic Kimchi, and 0.29% for imported Kimchi. The total vitamin C content was found to be higher in the domestic Kimchi (30.9mg%) than in the imported Kimchi(25.9mg%). These results suggest that both are within the standards of the Korea Food and Drug Administration in terms of food containment: 1% or lower for acid, and $1{\sim}4%$ for salt. For the number of aerobic bacteria present in each of them, both contain approximately $10^4{\sim}10^5\;CFU/g$g, which is safely under the FDA standard of $10^7{\sim}10^8 \;CFU/g$. Therefore, neither is harmful at all in terms of the number of bacteria but continuous monitoring will be necessary. from texture analysis, the hardness and chewiness of the domestic Kimchi was better than the imported Kimchi. The hardness and chewiness of the domestic Kimchi indicated a better production process than imported Kimchi. There may not be a particular difference in terms of the sensory evaluation apart from its crispy taste. However, this also needs to be continuously monitored while the Kimchi is undergoing the fermentation process.

The Role of Brain Somatostatin in the Central Regulation of Feed, Water and Salt Intake in Sheep

  • The physiological role of brain somatostatin in the central regulation of feed intake in sheep was investigated through a continuous intracerebroventricular (ICV) infusion of somatostastin 1-28 (SRIF) at a small dose of $5{\mu}g/0.2ml/hr$ for 98.5 hours from day 1 to day 5. Sheep (n=5) were fed for 2 hours once a day, and water and 0.5 M NaCI solution were given ad libitum. Feed, water and salt intake were measured during ICV infusion of artificial cerebrospinal fluid (CSF) and SRIF. The feed intake during SRIF infusion on days 2 to 5 increased significantly compared to that during CSF infusion. Water intake, when compared to that during CSF infusion, only increased significantly on day 4. NaCI intake during SRIF infusion was not different from that during CSF infusion. Mean arterial blood pressure (MAP) and heart rate during SRIF infusion were not different from those during CSF infusion. The plasma concentrations of Na, K, Cl, osmolality and total protein during SRIF infusion were also not different from those values during CSF infusion.There are two possible mechanisms, that is, the suppression of brain SRIF on feed suppressing hormones and the direct actions on brain mechanisms controlling feed intake, explaining how SRIF works in the brain to bring about increases in feed intake in sheep fed on hay. The results indicate that brain SRIF increases feed intake in sheep fed on hay.

Effect of the Addition of Antimicrobial Materials Before and After Aging on the Physicochemical Properties of Low-Salt Kochujang during Storage

  • To improve the shelf life of low-salt Kochujang, Korean hot pepper paste, antimicrobial materials were added at different times before and after aging. The kochujang was then packaged and stored at $30^{\circ}C$ for 15 weeks, and changes in microbiological and physiochemical properties were evaluated. Hunter a- and b-values decreased considerably during storage. The total color difference (${\delta}E$) was greater in the ethanol-chitosan (EC) treatment than in the control and after pasteurization (A-P) treatment. Gas was produced until the seventh week of storage. The control and the A-P treatments produced more gas than the other treatments, and these had the largest number of yeasts and aerobic bacteria. The pH of the EC treatment was higher than that of the other treatments, and the A-P treatment had the highest level of titratable acidity. During storage, the oxidation-reduction potential was lower in the EC and ethanol-mustard-chitosan (EMC) treatments. The reducing sugar content decreased remarkably in the control and A-P treatments, with high production of ethanol. There was a significant change in the content of amino-type nitrogen in the control and A-P treatments, and the content of ammonia-type nitrogen was lowest in the EMC treatment. In the sensory test of kochujang, the ethanol-mustard (EM) and ethanol (E) treatments were significantly higher than the EC, control, and A-P treatments (p <0.05). EM or E alone was effective in extending the shelf life of kochujang when added before aging.
