Nutritional Characterization of the Uncooked Skipjack Tuna Katsuwonus pelamis Frame as a Basic Resource for Flavor-enriching Concentrates (향미강화농축물 소재로서 비가열 가다랑어(Katsuwonus pelamis) 프레임의 영양학적 품질 특성)

  • We investigated the nutritional characteristics of the uncooked Skipjack tuna Katsuwonus pelamis frame (U-STF) as a basic resource for preparing flavor-enriching concentrates. The bone rate based on the fish frame (FF) was 53.2% in U-STF, thus lower than those of cooked skipjack tuna frame (C-STF), the uncooked salmon frame (U-SF), and beef leg bone (BLB). The composition of FF had 48.6-58.0% moisture, 18.4-21.3% crude protein, 7.0-15.0% crude lipid, and 12.8-22.1% ash. Compared to the proximate composition of BLB, that of FF was higher moisture and crude protein but lower solids, crude lipids, and ash. The total amino acid contents of FF was 17.6-20.8 g/100 g, which was higher than that of BLB. The levels of calcium and phosphorus of FF were 4.7-8.5 g/100 g and 2.4-4.2 g/100 g, respectively, thus lower than those of BLB. The brix concentration of FF extract was $5.0-8.2^{\circ}$, being highest in U-STF extracts, followed by extracts of U-SF and then extracts of C-STF. Our results indicate that U-STF is an optimal resource for preparing flavor-enriching concentrates.

Identification of an Embryonic Growth Factor IGF-II from the Central Nervous System of the Teleost, Flounder, and Its Expressions in Adult Tissues

  • The insulin-like growth factor (IGF) is found in all vertebrates and its type-II molecule is regarded as a fundamental embryonic growth factor during development. We have firstly identified, in this study, a cDNA clone corresponding to IGF-II (flIGF-II) from the adult brain of the teleost, Paralichthys olivaceus. We also examined the tissue expression of flIGF-II in several adult tissues by RT-PCR. The flIGF-II cDNA contained a complete ORF consisting of 215 amino acids and one stop codon. Its molecular characteristics appear to be similar to the previously identified IGF-II molecules, in which a common primary structure exhibiting B, C, A, D, and E domains is evidently observed. This cDNA clone seems to be cleaved at $Ala_{52}$ for the $NH_2$-end signal peptide and appears to produce a 98 amino acid-long E-peptide from the $Arg^{118}$. The functional B-D domain regions, therefore, include 65 amino acids and is able to encode a 7.4-kDa protein. The most prominent structural difference between IGF-I and IGF-II was that the D domain of IGF-II exhibits a two-codon-deleted pattern compared to the 8 amino acid-containing IGF-I. The insulin family signature in the A domain and six cysteins forming three disulfide bridges between the B and A domains were evolutionary-conserved from teleosts to mammalian IGF-II. Interestingly, the E-peptide region appears to provide a distinct hallmark between teleosts in amino acid composition. The flIGF-II shows 85.1% of sequence identity to salmon and trout, 90.6% to tilapia, and 98.4% to perch in amino acid level. In tissue expressions of IGF-II, it is very likely that flIGF-II has a significant expression in the adult brain. However, liver seems to be the main source for IGF-II production, and relatively low signals were observed in the adult muscle and kidney. Taken together, it would be concluded that the functional region for IGF-II mRNA is highly similar in phylogeny and is evolutionary, conserved as a mediator for the growth of vertebrates.

The Norwegian Model of Fisheries Bio-Resources Management (노르웨이 해역 수산생명자원 관리모델)

  • The Norwegian coastal area is the most efficient region for fishery production in the world's oceans, the Norway is the world's top 10 fisheries countries through efficient fishing and fishing aquaculture technology and its scientific management of fisheries bio-resources, with Norwegian salmon having attained the world's highest level. In the late 1980s, fisheries resources were depleted due to overfishing and fish diseases, resulting in a crisis in the fishing industry that lasted until the early 1990s. Since the national fishery emergency, people involved in the fishing industry, including fishermen, research scientists, and government officers, have tried to overcome the challenges facing the industry and identify an appropriate management model for fisheries bio-resources in the Norwegian coastal area. First, research vessels were used to monitor water and sediment conditions and fishery species, with the long-term aim of predicting fishery resources in real time and collecting information on species diversity, abundance, and distribution. Second, a "Healthy Fish Project" was promoted to counter natural disasters and fish disease problems with the development of vaccines against viruses and bacteria, eventually allowing for a decrease in the use of antibiotics and the production of notably healthier fish in the 2000s. Third, a systematic management model was developed to help with preparations for decreases in the total number of fishermen and increases in the proportion of elderly fishermen in the fishery industry using the development of automatic fishing aquaculture systems and short-chain systems. We could learn from the Norwegian model of fisheries bio-resources, management and could adopt it for the preparation of fishery bio-resources management policy for South Korean coastal areas in the near future.

Cultural Characteristics of Chromogenic and Teleomorphic Strains of Collectotricum gloeosporioides Isolated from Apple and Red pepper (사과와 고추에서 분리한 Colletotrichum gloeosporioides의 색소형성형 및 유성세대형 계통의 배양적 특징)

  • Conidia and cultural characteristics of isolates of chromogenic and teleomorphic strains of Colletotrichum gloeosporioides from apple and red pepper were compared. The mycelial growth of teleomorphic strains was faster than that of chromogenic strains in potato dextrose agar and V-8 agar. The chromogenic isolates from apple and red pepper developed. white gray to gray green mycelial rings interspersed with salmon to apricot colored conidial masses in colonies on potato dextrose agar and V-8 agar and none formed on ascigerous stage in cultures. The chromogenic isolates from red pepper produced conidia, most with one apex attenuated on apple and potato dextrose agar whereas fusiform and smaller conidia were produced in V-8 agar and water agar leaf medium. The chromogenic isolates from apple produced fusiform conidia in the media tested. The teleomorphic isolates from apple and red pepper produced cylindrical conidia, most with both apices rounded, developed white gray to dark olive green in a zonate pattern with small dark spots throughout colonies and formed the ascigerous stage in cultures.

Changes in Sex Steroid Hormones and Ovarian Development during Artificial Maturation of Female Eel, Anguilla japonica

  • The present study demonstrates the changes in body weight (BW) and plasma sex steroid hormone profiles during artificial maturation induced by human chorionic gonadotropin (HCG) or salmon pituitary extract (SPE) injections in cultured eel, Anguilla japonica, kept in seawater for 3 months. In the weekly SPE-injected female group, BW was relatively stable during vitellogenesis. Following induction of vitellogenesis, females exhibited a rapid increase of BW, and the oocytes were observed to be in the migratory nucleus stage at the end of the experiment. Plasma testosterone (T) and $estradiol-17{\beta}$ ($E_2$) levels increased slightly during vitellogenesis and peaked at an average of 5.82 ng/mL and 4.76 ng/mL, respectively, at the end of the experiment. In the weekly control and HCG-injected female groups, BW slowly decreased during the experimental period, and the oocytes of the two groups were observed to be at the primary yolk globule stage. In the weekly HCG-injected female group, plasma T and $E_2$ levels increased slightly during vitellogenesis and decreased afterward. In the control female group, however, plasma T and $E_2$ levels were not altered during the experimental period. Furthermore, plasma $17{\alpha},20{\beta}-dihydroxy-4-pregnen-3-one$ (DHP) was not detected in all experimental groups. Fertility and hatching rates of SPE-injected females were significantly higher in those that ovulated 15 h after DHP injection than 18 h. These results indicate that long rearing in seawater increases responsiveness to SPE in ovarian maturation of the Japanese eel, resulting in shortened period from completion of vitellogenesis by sex steroid hormone production.

Ichthyofauna and Fish Community Structure in Namdae Stream, Yangyang, Korea (양양 남대천의 어류상과 어류군집)

  • The ichthyofauna and fish community structure were investigated at eleven stations in Namdae stream, Yangyang during April, 2004 and March 2005 monthly. During the surveyed period thirty five species belonging to fifteen families were collected. Dominant species in number was Triboblodon hakonensis (relative abundance : 27.9%) and subdominant was Zacco koreanus (15.0%). There were five Korean endemic species including Rhynchocypris kumgangensis. Eleven species including Pungtungia herzi are first recorded in this study. There are stabilized fish community structure with dominant species R. kumgangensis in upstream, Z. koreanus and Z. platypus in midstream and T. hakonensis in downstream respectively. But, it is very worried the change of ichthyofauna in foreseeable future because of pumped storage power station construction in midstream.

  • This study is to analyze site layout traditional government office building focused on Chonui-hyon(全義縣) in Chungchong province with Chungchong-do regional maps(忠淸道地方地圖) and Eupjis(邑誌, topography) being compiled in the late Chosun dynasty. The conclusion of analysis can be summarized as follows; 1. Chonui-hyon(全義縣) in Chosun dynasty is gone with a planning principle of the capital city(Han-Seong, 漢城) with disposing Sa-Ji-Dan(社稷壇) and Gaek-Sa(客舍), which are placed to the left and right of Eupchi(邑治), and it is followed in the wake of the spatial structure of Chosun dynasty palace with disposing Dong-Heon(東軒) in front, Nae-A(內衙) at the back of it, or from side to side, also by the Dong-Heon of the center, with organizing each facility around it. 2. Dong-Heon and Gaek-Sa of Chonui-hyon is passed through three step gate, Mun-Ru(門樓), Woi-Sammun(外三門), Nae-Sammun(內三門), from Hong-Salmon(紅門), and are shown hierarchy by haying been placed at the end of approach axis. 3. Dong-Heon can be disposed by Feng-Shui(風水) which have influence on the southeast direction with Jin-San(鎭山) and An-Dae(案對), on the other hand, Gaek-Sa can be disposed by symbolic of the authority of a king. 4. Site layout by function of provincial government office is provided into four ferrite, Dong-Heon, Gaek-Sa, Nae-A, practical business territory, and eve territory is organized by official institution on a social position to radiation, by the compositive axis to be made Woi-Sammun Nae-Sammun Dong-Heon Nae-A 5. Approach process of Chnui-hyon government office is three door system similar to Kam-Young(監營) in a different way two door system in most provincial government office, also approach axis is formed by bending.

Comparison of chrysanthemum cultivars based on direct shoot regeneration rates in tissue culture

  • Direct shoot regeneration from leaf or internode or petiole segments was conducted in 33 cultivars of chrysanthemum. Shoot regeneration rates varied according to cultivars, culture media, and explant types. The high shoot regeneration rate of more than 70% in 15 cultivars (‘Pink Pangpang’, ‘Orange Memory’, ‘Relance’, ‘Zinba’, ‘Beakma’, ‘Innocence’, ‘Sunny Pangpang’, ‘Euro Yellow’, ‘Dublin’, ‘Boramae’, ‘Peak’, ‘Euro White’, ‘Vesuvio White’, ‘Linneker Salmon’ and ‘Pink Pride’) and 2 ones (‘Forward’ and ‘Agason’) was obtained from the segments of leaves and internodes, respectively, cultured on MS medium containing 1.0 mg-$L^{-1}$ BAP, 0.5 mg-$L^{-1}$ IAA and 30 g-$L^{-1}$ sucrose. That in 6 cultivars (‘Shuhonochikara’, ‘Hakunosen’, ‘Whitney Pangpang’, ‘Plaisir D’Amour’, ‘Grace’ and ‘Kumsu’) was observed from the segments of leaves or internodes cultured on 1/2 MS medium 1.0 mg-$L^{-1}$ BAP, 0.5 mg-$L^{-1}$ IAA and 15 g-$L^{-1}$ sucrose In case of 3 cultivars (‘Ilweol’, ‘Puma White’ and ‘Sharon’), when internode explants excised from mother plants, which were pre-cultured on MS medium containing 2 g-$L^{-1}$ activated charcoal and 30 g-$L^{-1}$ sucrose for two months in the dark, and cultured on MS medium containing 1.0 mg-$L^{-1}$ BAP, 0.5 mg-$L^{-1}$ IAA and 30 g-$L^{-1}$ sucrose, that was shown. Seven cultivars including ‘Puma Yellow’, ‘Argus’, ‘Yes Morning’, ‘Whiparam’, ‘Hakunohikari’, ‘Charming Eye’ and ‘Moon light’ requires more improved culture conditions. Tissues with the highest shoot regeneration rate were in descending order, leaf, petiole, and internode segments.

A Case of Systemic Lupus Erythematosus Misdiagnosed as Adult-onset Still's Disease (성인 발병성 스틸병(Adult-onset Still's disease)으로 오인된 전신홍반루푸스(Systemic Lupus Erythematosus) 1예)

  • Adult-onset Still's disease (AOSD) is an inflammatory disorder that's characterized by daily, spiking high fever, arthritis and an evanescent, salmon-pink rash. AOSD is diagnosed purely on the basis of the typical clinical features of the illness. The symptoms commonly include swelling of the lymph nodes, enlargement of the spleen and liver, and a sore throat. AOSD is difficult to differentiate from systemic lupus erythematosus (SLE) due to the similar clinical manifestations. We report here on a case of a 16-year-old female patient with autism and epilepsy and who complained of daily spiking fever for 20 days. The patient had maculopapular skin rashes on the face and whole body and lymphadenopathy. The liver function tests were elevated mildly. The initial rheumatoid factor (RF) and antinuclear antibody (ANA) tests were negative. We diagnosed her as having adult-onset Still's disease according to the criteria of Yamaguchi. We successfully treated her with oral prednisolone. But her antinuclear antibody test was changed to positive after discharge. So we finally diagnosed her as having SLE.

The Control Mechanism of Gonadotropin-Releasing Hormone and Dopamine on Gonadotropin Release from Cultured Pituitary Cells of Rainbow Trout Oncorhynchus mykiss at Different Reproductive Stages

  • The mechanism by which gonadotropin-releasing hormone (GnRH) and dopamine (DA) control gonadotropin (GTH) release was studied in male and female rainbow trout using cultured pituitary cells obtained at different reproductive stages. The mechanisms of follicle-stimulating hormone (FSH) release by GnRH and DA could not be determined yet. However, basal and salmon-type GnRH (sGnRH)- or chicken-II-type GnRH (cGnRH-II)- induced luteinizing hormone (LH) release increased with gonadal maturation in both sexes. LH release activity was higher after sGnRH stimulation than cGnRH-II stimulation at maturing stages in both sexes. The GnRH antagonist ([Ac-3, 4-dehydro-$Pro^1$, D-p-F-$Phe^2$, D-$Trp^{3,6}$] GnRH) suppressed LH release by sGnRH stimulation in a dose-dependent manner, although the effect was weak in maturing fish. The role of DA as a GTH-release inhibitory factor differs during the reproductive cycle: the inhibition of sGnRH-stimulated LH release by DA was stronger in immature fish than in maturing, ovulating, or spermiated fish. DA did not completely inhibit sGnRH-stimulated LH release, and DA alone did not alter basal LH release. Relatively high doses ($10^{-6}$ or $10^{-5}M$) of domperidone (DOM, a DA D2 antagonist) increased LH release, which did not change with reproductive stage in either sex. The potency of DOM to enhance sGnRH-stimulated LH release was higher in maturing and ovulated fish than in immature fish. These data suggest that LH release from the pituitary gland is controlled by dual neuroendocrine mechanisms by GnRH and DA in rainbow trout, as has been reported in other teleosts. The mechanism of control of FSH release, however, remains unknown.
