Objectives: The objective of this study was to evaluate antidiabetic activity of Chaturmukha rasa based on streptozotocin induced diabetes model, alpha amylase inhibitory activity, alpha Glucosidase inhibitory activity and inhibition of sucrase. Methods: Chaturmukha rasa was prepared as per Ayurvedic formulary. Antidiabetic activity was measured in experimentally streptozotocin induced rats. The dose was taken as 45 mg/kg, i.p. The antidiabetic activity of Chaturmukha rasa was compared Triphala Kwatha, a marketed formulation. Further In vitro $\acute{\alpha}$- Amylase Inhibitory Assay, In vitro salivary amylase Inhibitory Assay, In vitro ${\alpha}-Glucosidase$ Inhibitory Assay and In vitro Sucrase Inhibitory Assay was performed with respect to Chaturmukha rasa. The IC50 value was calculated for all the above activity. Results: Streptozotocin with Acarbose showed significant decrease in blood glucose level whereas streptozotocin with Triphala kwatha showed more decrease in blood glucose level than Streptozotocin with Acarbose. The combination of Streptozotocin + Triphala kwatha + Chaturmukha rasa showed a significant decrease in blood glucose level on 21st day. In vitro $\acute{\alpha}$- Amylase Inhibitory Assay the Chaturmukha rasa showed IC50 value $495.94{\mu}l$ when compared with Acarbose $427.33{\mu}l$, respectively. In the ${\alpha}-Glucosidase$ Inhibitory Assay Chaturmukha rasa showed IC50 value $70.93{\mu}l$ when compared with Acarbose $102.28{\mu}l$, respectively. In vitro Sucrase Inhibitory Assay Chaturmukha rasa showed IC50 value $415.4{\mu}l$ when compared with Acarbose $371.43{\mu}l$, respectively. Conclusion: This study supports that Chaturmukha rasa may inhibit diabetes by inhibition of salivary amylase or alpha Glucosidase or sucrase. This may be the mechanism by which Chaturmukha rasa inhibits diabetes. Further this study supports the usage of Chaturmukha rasa for the management of diabetes.
In general, the major causative factor of halitosis is thought to be a sulfated compounds. Clusterin, a sulfated glycoprotein-2(SGP-2), is frequently found in diabetic conditions and cold stress conditions. The same result is werum glucose level to diabeteic and cold stress conditions that founded Clusterin. Therefore, this study was performed to examine Clusterin in the slivary glands under stress conditions before insulin injection I.M. Fourty rats were diveded into 3 groups ; 1) 10 rats of gorup I were selected as a control 2) 15 rats beloning to group II were bathed in cold water for 30 seconds twice a day 3) 15 rats in group III received cold stress and injected I.M. with insulin. The rats were sacrifeced at day 0, 3, 5, 7 and 10 of the experiment and the submandibular glands and parotid galnds were removed. RNAs were purified from the salivary of the salivary glands were subjected to Hematoxillin-Eosin stainings and examined under the light microscope. The obtained results were as follows : 1. With immunohistochemistric method, in normal control goup, Clusterin was moderately stained in the intercalated ductal cell of the submandibualr glands, mild stained in the striated ductal cell of the submandibular glands, heavily stained on the cytoplasm of the intercalated ductal cell in the mucous submandibular glands nad slightrly stained in the intercalated ductal cell of the paroted gland, expressed negativity in the acina cell. 2. With immunohistochemistric method, Clusterin slightly increased in the acina cell of the submandibular glands under stress condition at 3 days after experement, moderately stained at 5 days after experiment so revealed positive response. And hearily in the intercalated ductal cell and mildly lin the acina celluar eytoplasm of the parotid glands under stress condition at 3 days experiment. 3. With immunohistochemistric method, no remarkable differences are found between the normal control group and stress conditioned group that insulin administration was performed before. 4. In the stressor-giving group, Clusterin mRNA was porminently expressed in submandibular gland after 5 days after experiment, in parotid gland after 3 days after experiment, performed in immunoelectrophoresis method. 5. In the insulin-injected nad stressor-giving group, Clusterin mRNA was not observed in all experimental submandibular and parotid gland, performed in immunoelectrophoresis method.
Purpose: The purpose of this study was to examine the rate of salivation before and after olfactory stimulation using BPO (black pepper oil) and then, to collect basic data for screening the possibility of introducing olfactory stimulus to prevent and improve dry mouth. Methods: The subjects in this study were 60 college students in Jeon-Nam province. The subjects were randomly divided into 3 groups; the BPO group (n=20), the lavender oil (LVO) group (n=20), and the distilled water (DW) group (n=20). The amount of whole saliva before and after olfactory stimulation was measured 14 times for a 4-week period. Results: In the BPO and LVO groups, the amount of whole saliva increased after the olfactory stimulation and it was most prominent in the BPO group (p<0.05). The difference of whole salivation was higher in the BPO group (p<0.05) and there was no significant difference in the salivary flow rate in the DW group. After 14 times of measurement for 4 weeks, the BPO and LVO groups showed an increase in salivary rate at an early stage compared without stimulation (p<0.05), but there was no significant difference between the 2 groups in the middle. Since the twelfth measurement, whole saliva secretion increased and maintained the elevated level (p<0.05). Conclusions: The olfactory stimulation using BPO may contribute to stimulating salivation. Further studies are needed for subjects with dry mouth symptoms. In the future, qualitative and quantitative studies should be conducted for people who complains about oral dryness.
Background: The monocyte chemoattractant protein-1 (MCP-1/CCL2) is a potent chemoattractant for natural killer cells, monocytes, and memory T lymphocytes. However, any role in the genesis of salivary gland tumors (SGT) is unknown. To assess the diagnostic relevance of chemokines in SGT, MCP-1 levels in the serum of patients were investigated in association with tumor progression and clinical aggressiveness. Materials and Methods: Using an ELISA kit, we assessed and compared the circulating levels of MCP-1 in blood serum of 70 SGT patients with 44 healthy control samples. Results: The results of this study showed that the concentration of MCP-1 was significantly lower in patients with benign ($463.8{\pm}158.5pg/ml$, P=0.033) and malignant ($454.8{\pm}190.4pg/ml$, P=0.007) SGT than in healthy subjects ($645.7{\pm}338.9$). No significant difference in mean serum levels of MCP-1 was observed between the benign and malignant group (p=0.9). While MCP-1 levels were lower in patients with an advanced clinical stage, advanced tumor size, higher tumor grade, or lymph node involvement, but the mean MCP-1 level between groups showed no statistically significant difference (p>0.05). Conclusions: MCP-1 levels in the serum of patients with SGT were decreased, indicating that this might a good marker for discriminating patients with SGT from healthy people. However, no clear-cut relationship was detected between MCP-1 levels and clinicopathologic factors, and MCP-1 is not a good marker for evaluating tumor dissemination.

The purpose of this study was to investigate the effects of forest activities on the cortisol concentrations in the body and emotional vocabulary of kindergarteners. The participants were 36 kindergarten children aged 4-5, 17 in the control group and 19 in the experimental group. Forest activities consisted of six steps according to the level of kindergarteners, and the program was carried out at G City Environment Training Institute. Before and after the experiment, the children's emotional vocabulary was self-reported and salivary cortisol was analyzed to evaluate changes in the body's stress hormones. As a result, in the control group, positive words increased significantly from 0.14 to 1.53 (p=.003), but the decrease in negative words was not significant (p=.059). In the experimental group, positive words increased significantly from 0.47 to 2.84 after forest activities (p=.000). Negative words decreased significantly from 1.94 to 0.00 after forest activities (p=.000). In the analysis of salivary cortisol, the change in salivary cortisol concentrations was not significant (p=.667) in the control group, merely decreasing from 0.179 ㎍/dl to 0.161 ㎍/dl. However, the experimental group showed a significant decrease from 0.202 ㎍/dl to 0.161 ㎍/dl after forest activities (p=.049). These results suggest that forest play activities may be helpful in encouraging young children's emotional development by increasing positive vocabulary and decreasing negative vocabulary and reducing stress in kindergarteners.
The acinar cells and secretory granules of the parotid salivary gland were examined in the lesser white toothed shrew, Crocidura suaveolens and the big white-toothed shrew, C. lasiura. The parotid gland of both species were a serous gland having only one kind of serous acinar cells, and had conventional arrangement of acini and intercalated, granular and striated ducts. In case of C. suaveolens, serous acinar cells had well developed rER, prominent Golgi complex, several large mitochondria and abundant moderate dense secretory granules with various stages of the maturing or fusing process. Immature acinar secretory granules were only or mainly filled with fine strong dense specks and had an indistinct limiting membrane, and mature granules were filled with homogeneous pale large round center and had fine strong dense specks at the periphery of the homogeneous pale center and a distinct limiting membrane. In case of C. lasiula, serous acinar cells had well developed rER, prominent Golgi complex, several large mitochondria and abundant dense secretory granules with maturing or fusing process. Immature acinar secretory granules were only filled with pale rough specks and had an indistinct limiting membrane, and mature granules were only filled with rough dense specks and had a distinct limiting membrane. Eventually The acinar secretory granules of C. suaveolens were seen moderate at the light and ultrastuctural level, those of C. lasiura were strong dense at the light microscopic level and dense at the ultrastructural level.

A static, unilateral, and focal bone depression located lingually within the ascending ramus, identical to the Stafne's bone cavity of the angle of the mandible, is being reported. During development of the mandible, submandibular gland inclusion may lead to the formation of a lingual concavity, which could contain fatty tissue, blood vessels, or soft tissue. However, similar occurrences in the ascending ramus at the level of the parotid gland are extremely rare. Similar cases were previously reported in dry, excavated mandibles, and 3 cases were reported in living patients. A 52-year-old African American male patient was seen for pain in the mandibular teeth. Panoramic radiography showed an unusual concavity within the left ascending ramus. Cone-beam computed tomography confirmed this incidental finding. The patient was cleared for the extraction of non-restorable teeth and scheduled for annual follow-up.
The salivary gland undergoes complex process of growth and differentiation of the branching morphogenesis of ductal system during the prenatal and early postnatal periods which are regulated by various elements in the extracellular matrix. Extracellular matrix metalloproteinase inducer (EMMPRIN) is a cell adhesion molecule. In the present study, localization and expression of EMMPRIN in development and effects of chorda-lingual denervation and cyclosporine A (CsA) treatment on the EMMPRIN expression were investigated. Immunohistochemistry, RT-PCR and Western blot were used to determine expression level. Immunohistochemistry revealed that EMMPRIN was localized specifically in the cytoplasm of ductal cells, not acini of the submandibular gland all the postnatal periods. At prenatal day 18, when the formation of ducts was not definite, no immunoreactivity was observed. Both Western blot and RT-PCR analyses revealed that EMMPRIN expression was maintained up to postnatal day 7, decreased after postnatal day 10. The EMMPRIN expression was upregulated by the surgical denervation of the chorda-lingual nerve in the gland as well as by the CsA treatment. The present study suggests that EMMPRIN is a crucial molecule for maintaining physiological functions of the salivary gland.

Background: Oral squamous cell carcinoma (OSCC) is thought to develop from precancerous dysplastic lesions through multistep processes of carcinogenesis involving activation of oncogenes and loss of tumor suppressor genes. The human epidermal growth factor receptor 2 (Her-2/neu [erbB-2]), a cell membrane glycoprotein, is a growth factor receptor that has receptor tyrosine kinase activity. Her2/neu activation plays a central role in cell proliferation and survival. It has been shown that overexpression of Her2/neu increases the rate of cell division and growth, leading to precancerous changes. The aim of the present study was to compare the serum and salivary Her2/neu levels between cases with premalignant and malignant oral lesions. Materials and Methods: Fasting blood samples and unstimulated saliva by passive drooling were collected from three groups of healthy control (n=20), premalignant disorder (PMD) (n=20) and OSCC (n=25) subjects. The HER2 extracellular domain (HER2 ECD) levels were measured using ELISA. Results: The levels of serum Her2/neu showed no significant differences between any of the groups but on the other hand salivary Her2/neu levels were found to be significantly (p<0.05) higher when compared between control (median 68.7 pg/ml, range: 21.5 - 75.8) and OSCC (median 145.6 pg/ml, range: 45.1-191.1). A similar trend was observed when comparing between PMD (median 43.3, range: 22.1 -94.7) and OSCC with a statistical significance of p<0.05. Conclusions: Our study provided evidence of increased salivary Her2/neu in OSCC when compared to PMD and control which was not the case for serum levels. This suggests that probably Her2/neu is not highly amplified as in breast cancer so as to be reflected in serum. Since saliva is in local vicinity of the OSCC, even a mild increase might be mirrored. On the whole, this study proposes Her2/neu as marker for distinguishing premalignant and malignant conditions.
Purpose: This study investigated the effects of 1 year of training on imaging diagnosis, using static ultrasonography (US) salivary gland images of Sjögren syndrome patients. Materials and Methods: This study involved 3 inexperienced radiologists with different levels of experience, who received training 1 or 2 days a week under the supervision of experienced radiologists. The training program included collecting patient histories and performing physical and imaging examinations for various maxillofacial diseases. The 3 radiologists (observers A, B, and C) evaluated 400 static US images of salivary glands twice at a 1-year interval. To compare their performance, 2 experienced radiologists evaluated the same images. Diagnostic performance was compared between the 2 evaluations using the area under the receiver operating characteristic curve (AUC). Results: Observer A, who was participating in the training program for the second year, exhibited no significant difference in AUC between the first and second evaluations, with results consistently comparable to those of experienced radiologists. After 1 year of training, observer B showed significantly higher AUCs than before training. The diagnostic performance of observer B reached the level of experienced radiologists for parotid gland assessment, but differed for submandibular gland assessment. For observer C, who did not complete the training, there was no significant difference in the AUC between the first and second evaluations, both of which showed significant differences from those of the experienced radiologists. Conclusion: These preliminary results suggest that the training program effectively helped inexperienced radiologists reach the level of experienced radiologists for US examinations.
