Water Quality and Heavy Metals in the Surface Seawaters of the Saemangeum Area during the Saemangeum-dike Construction (새만금 방조제 체절 과정 중 새만금 주변해역 표층수의 수질과 중금속 분포 특성)

  • In order to investigate spatial and temporal distributional characteristics of major water qualities in the Saemangeum area during the Saemangeum dike construction, salinity, COD, dissolved nutrients(DIN, Silicate) and heavy metals were analyzed from the surface water collected in April, May, August and November 2002. The overall value of Salinity, COD, DIN, and silicate in surface waters were in the range of $13.08{\sim}31.96\;psu$, $0.12{\sim}3.43\;mg/L$, $0.001{\sim}2.638\;mg/L$, and $0.010{\sim}3.181\;mg/L$, respectively. The COD and DIN in each survey showed the highest concentration at the mouth of Mangyeong river estuary(St. 1) where freshwater flow into the Saemangeum area. The concentrations of nutrients were high in the inner part of the Saemangeum dike with low-salinity, and low nutrients in the outer part of the dike with high-salinity, which strongly indicated that concentrations were adjusted by physical mixing. The ranges of dissolved metals and acid-soluble Hg in surface seawater were $0.006{\sim}0.115{\mu}g/L$ for Co, $0.26{\sim}0.114{\mu}g/L$ for Ni, $0.14{\sim}0.93{\mu}g/L$ for Cu, $0.04{\sim}0.53{\mu}g/L$ for Zn, $0.010{\sim}0.043{\mu}g/L$ for Cd, $0.010{\sim}0.795{\mu}g/L$ for Pb, and $0.25{\sim}4.16{\mu}g/L$ for Hg. The highest concentrations of some metals except for Cd were found at the estuary(Sts. 1 or 3). In most cases, a decreasing order of metal concentrations towards open sea(low-salinity$\rightarrow$high-salinity) was observed and showed positive relationship with DIN and silicate caused by land base pollutants input. On the other hand, due to Cd desorption from suspended solids in saline water, dissolved Cd concentrations were high in high-salinity area and low in low-salinity. In November, Co, Zn, Cu and Pb were relatively high in the northern area of the outer-side of Saemangeum, which was only influenced by the Geum river discharge. The concentrations of most dissolved metals of this study were lower than those of the past data in this area, but higher than those in Lena river estuary under the pristine environment.

  • The geothermal waters from the Busan area belong to Na-CI type and are characterized by much higher EC (921 ~6,520${\mu}$S/cm) and TDS (608-3,390 mg/L) than other geothermal waters in Korea. The concentration of majorions shows a weakly positive relationship with temperature except for Mg ion. The concentrations of the major cat ions have the order of Na>Ca>K>Mg. Ca ion is enriched and Mg ion is depleted compared with seawater. All Br concentrations of geothermal water are lower than those of seawater, showing a positive relationship with temperature. Generally geochemical characteristics of geothermal waters of the Busan area indicate that these waters have relatively increased Ca and Sr contents and depleted Mg, Na and K contents caused by seawater interaction with wall rock at depth during deep circulation of seawater. Base on the relationship between major ions and temperature, saline geothermal waters are diluted and are cooled by mixing of groundwaters during ascent. Isotope study and reaction path modeling of the overall geochemical system are required in order to better quantify the evolution and origin of geothermal waters in the Busan area.

Determination of Amisulbrom Residues in Agricultural Commodities Using HPLC-UVD/MS (HPLC-UVD/MS를 이용한 농산물 중 Amisulbrom의 잔류분석)

  • This experiment was conducted to establish an analytical method for residues of amisulbrom, as recently developed an oomycete-specific fungicide showing inhibition of fungal respiration, in crops using HPLC-UVD/MS. Amisulbrom residue was extracted with acetonitrile from representative samples of five raw products which comprised apple, green pepper, kimchi cabbage, potato and hulled rice. The extract was diluted with 50 mL of saline water and directly partitioned into dichloromethane to remove polar co-extractives in the aqueous phase. For the hulled rice sample, n-hexane/acetonitrile partition was additionally employed to remove non-polar lipids. The extract was finally purified by optimized Florisil column chromatography. On an octadecylsilyl column in HPLC, amisulbrom was successfully separated from sample co-extractives and sensitively quantitated by ultraviolet absorption at 255 nm with no interference. Accuracy and precision of the proposed method was validated by the recovery test on every crop samples fortified with amisulbrom at 3 concentration levels per crop in each triplication. Mean recoveries ranged from 85.3% to 105.6% in five representative agricultural commodities. The coefficients of variation were all less than 10%, irrespective of sample types and fortification levels. Limit of quantitation (LOQ) of amisulbrom was 0.04 mg/kg as verified by the recovery experiment. A confirmatory method using LC/MS with selected-ion monitoring technique was also provided to clearly identify the suspected residue. The proposed method was sensitive, reproducible and easy-to-operate enough to routinely determine the residue of amisulbrom in agricultural commodities.

  • Chemical burns onto oral mucosa which are infrequent, may result from contact with a wide variety of chemical agents. The degree of injury depends on the chemical, its concentration, duration of contact, and the natural penetrability and resistance of the tissues involved. Chemicals do not usually "burn" in that they do not cause destruction by hyperthermic activity. Rather, they damage tissue by causing coagulation of protein by one of several processes, reduction, oxidation, desiccation, corrosion, or vesication. Paraquat(Gramoxon) is the most frequently agricultural chemicals that induce the severe toxic reactions onto the organs of human body in Korea. The toxic reaction are composed of pulmonary edema and fibrosis, formation of hyaline membrane, inflammatory reaction and bleeding tendency, owing to the cell damage by the production of superoxide radicals. The contents of essential treatment in paraquat intoxication are commonly airway and breathing maintenance, gastric lavage, much hydration and diuresis, hemoperfusion and medications for the removal of the chemicals and the prevention of various complications. The sedative oral dressings, such as, orabase ointment application, warm saline gargling, lidocaine viscous gargling and oral gargling by the mixed solutions(tetracycline, prednisolone and 10% dextrose water) are important for the improvement of chemical oral mucositis and the comfortable feeding of diet. The authors managed properly two cases of oral chemical mucositis that were occurred by the incorrect use of agricultural chemicals(paraquat) and report the cases with the review of literatures about care of the chemical intoxication and oral mucositis.

Suppressive Effect of Zinc on the Formation of Colonic Preneoplastic Lesions in the Mouse Fed High Levels of Dietary Iron

  • We investigated the effect of zinc on the formation of colonic aberrant crypt foci induced by azoxymethane (AOM) followed by dextran sodium sulfate (DSS) in mice with high iron diet (HFe; 450 ppm iron). Sixweek old ICR mice were fed on high iron diets with combination of three different levels of zinc in diets, low-zinc (LZn; 0.01 ppm), medium-zinc (MZn; 0.1 ppm), and high-zinc (HZn; 1 ppm) for 12 weeks. Animals were received weekly intraperitoneal injections of AOM (10 mg/kg B.W. in saline) for 3 weeks followed by 2% DSS (molecular weight 36,000~50,000) in the drinking water for a week. To confirm the iron storage in the body, the hepatic iron concentration has been determine chemically and compared with histological assessment visualized by Prussian blue reaction. Aberrant crypt (AC) and aberrant crypt foci (ACF) were analyzed in the colonic mucosa of mouse fed high dietary iron. Superoxide dismutase (SOD) activity and thiobarbituric acid-reactive substances (TBARS) level were also investigated. Apoptosis in the preneoplastic lesion was determined by terminal deoxynucleotidyl transferase-mediated dUTP nickend labeling (TUNEL). In addition, immunohistochemistry of ${\beta}$-catenin was also performed on the mucous membrane of colon. The number of large ACF (${\geq}4$ AC/ACF), which possess greater tumorigenic potential, was significantly lower in MZn and HZn groups compared with LZn group. Cytosolic SOD activity in the liver was significantly higher in HZn group compared with LZn group. Hepatic MDA level was decreased significantly in HZn group compared with MZn and LZn groups. Apoptotic index was significantly higher in HZn group. Taken together, these findings indicate that dietary zinc might exert a protective effect against colonic preneoplastic lesion induced by AOM/DSS in ICR mice with high iron status, and suggest that dietary supplement of zinc might play a role in suppressing colon carcinogenesis in mice.

Analytical Method for Flusulfamide as Benzenesulfonamide Fungicide, Residues in Major Agricultural Commodities (주요 농산물 중 Bezenesulfonamide계 살균제 Flusulfamide의 잔류 분석법)

  • BACKGROUND: An analytical method was developed using HPLC-UVD/MS to precisely determine the residue of flusulfamide, a benzenesulfonamide fungicide used to inhibit spore germination. METHODS AND RESULTS: Flusulfamide residue was extracted with acetone from representative samples of five raw products which comprised apple, green pepper, Kimchi cabbage, hulled rice, and soybean. The extract was diluted with large volume of saline water and directly partitioned into dichloromethane to remove polar co-extractives in the aqueous phase. For the hulled rice and soybean samples, n-hexane/acetonitrile partition was additionally employed to remove non-polar lipids. The extract was finally purified by optimized Florisil column chromatography. On an octadecylsilyl column in HPLC, flusulfamide was successfully separated from co-extractives of sample, and sensitively quantitated by ultraviolet absorption at 280 nm with no interference. Accuracy and precision of the proposed method was validated by the recovery experiment on every crop sample fortified with flusulfamide at 3 concentration levels per crop in each triplication. CONCLUSION: Mean recoveries ranged from 82.3 to 98.2% in five representative agricultural commodities. The coefficients of variation were all less than 10%, irrespective of sample types and fortification levels. Limit of quantitation (LOQ) of flusulfamide was 0.02 mg/kg as verified by the recovery experiment. A confirmatory method using LC/MS with selected-ion monitoring technique was also provided to clearly identify the suspected residue.

Hepatoprotective effects of the aqueous extract from Taraxacum officinale (Dandelion) against Thioacetamide-induced hepatotoxicity in rats (Thioacetamide로 유발된 간 손상에 대한 Taraxacum officinale (Dandelion) 추출물의 효과)

  • The genus Taraxacum, known by the common name Dandelion, is a medicinal herb in the family Asteraceae. It has been traditionally used as a folk medicine for the treatment or prevention of various diseases due to its anti-inflammatory and anti-oxidative properties. In this study, we attempted to evaluate protective effects of Dandelion related with anti-oxidative activity to Thioacetamide (TAA)-induced liver damage. 36 rats were randomly assigned to six experimental groups : Control, Dandelion water extract (DWE), TAA, TAA&DWE 300, TAA&DWE 600, TAA&DWE 1,200 groups. Rats in DWE and TAA&DWE groups were pretreated with DWE (300, 600 or 1,200 mg/kg BW) by gavage for 7 days. All rats were treated intraperitoneally with TAA (200 mg/kg BW) or normal saline at 12 hours after last oral administration and sacrificed at 12 hours after last treatment. Levels of WBC and Neutrophil count were significantly decresed in TAA&DWE 1,200 group compared with that in TAA group (P<0.05). In TAA&DWE 600 and TAA&DWE 1,200 groups, serum AST, ALT, GGT levels were lower than TAA group (P<0.05). The serum TG level was significantly elevated in TAA&DWE groups compared with those in TAA group. Liver tissues from TAA group showed extensive histopathological changes, characterized by moderate or severe hepatocytes degeneration, inflammatory cell infiltration, and congestion. In the TAA&DWE group, The severity of histopathological lesions were decreased compared to those in the TAA group. The MDA concentration was significantly decreased and GSH content was significantly increased in the TAA&DWE 1,200 group compared to those in the TAA group. GR, CAT and GST activities in the TAA&DWE 1,200 group were significantly increased compared to those in the TAA group.

Controlled release of nerve growth factor from heparin-conjugated fibrin gel within the nerve growth factor-delivering implant

  • Objectives: Although nerve growth factor (NGF) could promote the functional regeneration of an injured peripheral nerve, it is very difficult for NGF to sustain the therapeutic dose in the defect due to its short half-life. In this study, we loaded the NGF-bound heparin-conjugated fibrin (HCF) gel in the NGF-delivering implants and analyzed the time-dependent release of NGF and its bioactivity to evaluate the clinical effectiveness. Materials and Methods: NGF solution was made of 1.0 mg of NGF and 1.0 mL of phosphate buffered saline (PBS). Experimental group A consisted of three implants, in which $0.25{\mu}L$ of NGF solution, $0.75{\mu}L$ of HCF, $1.0{\mu}L$ of fibrinogen and $2.0{\mu}L$ of thrombin was injected via apex hole with micropipette and gelated, were put into the centrifuge tube. Three implants of experimental group B were prepared with the mixture of $0.5{\mu}L$ of NGF solution, $0.5{\mu}L$ HCF, $1.0{\mu}L$ of fibrinogen and $2.0{\mu}L$ of thrombin. These six centrifuge tubes were filled with 1.0 mL of PBS and stirred in the water-filled beaker at 50 rpm. At 1, 3, 5, 7, 10, and 14 days, 1.0 mL of solution in each tubes was collected and preserved at $-20^{\circ}C$ with adding same amount of fresh PBS. Enzyme-linked immunosorbent assay (ELISA) was done to determine in vitro release profile of NGF and its bioactivity was evaluated with neural differentiation of pheochromocytoma (PC12) cells. Results: The average concentration of released NGF in the group A and B increased for the first 5 days and then gradually decreased. Almost all of NGF was released during 10 days. Released NGF from two groups could promote neural differentiation and neurite outgrowth of PC12 cells and these bioactivity was maintained over 14 days. Conclusion: Controlled release system using NGF-HCF gel via NGF-delivering implant could be an another vehicle of delivering NGF to promote the nerve regeneration of dental implant related nerve damage.

Antitumor and Immuno-modulatory Effect Against Mouse Sarcoma 180 of Crude Polysaccharides Extracted from Fruiting Body of Armillaria tabescens (뽕나무버섯부치(Armillaris tabescens)의 자실체에서 추출한 조다당류의 생쥐 Sarcoma 180에 대한 항암 및 면역증강 효과)

  • Armillaria tabescens, one of edible and medicinal mushrooms belonging to Agaricales of Basidiomycota, has been known to have outstanding curative effects on chronic hepatitis and cholecystitis and inhibitory effects on the sarcoma 180 and Erhrlich carcinoma of mice. Neutral saline soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr, NaCl, Fr. HW and Fr, MeOH, respectively) were extracted from fruiting body of the mushroom. In vitro cytotoxicity tests showed that crude polysaccharides were not cytotoxic against cancer cell lines such as NIH3T3 and Sarcoma 180 at the concentration of $2000\;{\mu}g/ml$. Intraperitoneal injection with crude polysaccharides exhibited life prolongation effect of $28.8{\sim}46.5%$ in mice inoculated with Sarcoma 180, respectively. Fr. NaCl improved the immunopotentiation activity of B lymphocyte by increasing the alkaline phosphatase activity by $1.8{\sim}2.1$ folds, respectively. In case of Fr, NaCl, the numbers of peritoneal exudate cells and circulating leukocytes were increased by 9 and 1.9 folds, respectively.

Studies on Immuno-Modulatory and Antitumor Effects of Crude Polysaccharides Extracted from Fruiting Body of Oudemansiella radicata (민긴뿌리버섯(Oudemansiella radicata)의 자실체로부터 추출한 조다당류의 항암 및 면역 활성 효과에 관한 연구)

  • Oudemansiella radicata, edible and medicinal mushroom belonging to Agaricales of Basidiomycota, has been known to exhibit outstanding curative effects on the fungal infection and hypertension caused by high blood pressure. Neutral saline soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr, NaCl, Fr. HW and Fr. MeOH, respectively) were extracted from fruiting body of the mushroom. In vitro cytotoxicity test showed that Fr. NaCl was not cytotoxic against NIH3T3 and Sarcoma 180 at the concentration of $10{\sim}1,000\;{\mu}g/ml$. Intraperitoneal injection with Fr. NaCl exhibited antitumor activity with life prolongation effect of $42.9{\sim}66.7%$ in mice inoculated with Sarcoma 180. Fr. NaCl improved the immunopotentiating activity of B lymphocyte by increasing the alkaline phosphatase activity by $1.4{\sim}3$ folds compared with controlled and LPS groups, respectively. Intraperitoneal injection with Fr. MeOH increased the numbers of peritoneal exudate cells and circulating leukocytes by 3.5 and 2.5 folds, respectively. Therefore, the antitumor effect exhibited on mouse Sarcoma 180 cells was likely due to immune-modulating activity of crude polysaccharides extracted from fruiting body of O. radicata.
