  • Objectives : This study was to investigate the effects of GCP treatment on the monosodium iodoacetate-induced osteoarthritis in rats. Methods : Arthritis was induced by injection of Monosodium lodoacetate(0.5mg) into knee joints of rats. Arthritic rats were divided into control(n=8) and treated(n=8) group. Control group was taken distilled water for 20 days. Treated group was taken extracts of GCP by oraly for same duration. Normal group(n=8) was injected with normal saline and was taken distilled water for 20 days. Body weight was measured at 0, 5, 10, 15, 20 days after injection. Macroscopic examination and histopathological study on articular cartilage of knee joint were operated at 20 days after injection. Proteoglycan(PG) content of articular cartilages of knee joint was represented by safranine O staining, was measured at 20 days injection. Tumor necrosis $factor-{\alpha}$, $Interleukin-1{\beta}$, Interleukin-6 in synovial fluid were measured with ELISA kit at 20 days after injection. Immunohistochemical staining of COX-2, iNOS in knee joints were observed at 20 days after injection. Results : 1. Body weight of the treated group increased compare with control group at 20 days after injection. 2. Macroscopically, degree of osteoarthritis in the treated group were evaluated compared with the control group. 3. PG content in articular cartilage of the treated group was significantly increased compared with the control group. 4. Histopathologically, osteoarthritic scores of the treated group was significantly decreased compared with the control group. 5. $TNF-{\alpha}$ content in synovial fluid of the treated group was decreased compared with the control group. 6. $IL-1{\beta}$ content in synovial fluid of the treated group was significantly decreased compared with the control group. 7. Positive reaction of COX-2 in chondrocytes and synovial membrane of the treated group was faint compared with the control group. Conclusions : On the basis of these results, we concluded that GCP has inhibiting effects on the $IL-1{\beta}$ and COX-2 secretion of chondrocytes and synovial membrane in Monosodium lodoacetate-Induced osteoarthritis model of rats.

  • Objectives: This study was performed to investigate the effects of Gagamsosokmyeong-tang(Jiajianxiaoxuming-tang) on the monosodium iodoacetate(MIA) induced early state osteoarthritis in rats. Methods: Osteoarthritis was induced by injection of MIA(0.25 mg) into knee joints of rats. Osteoarthritis rats were divided into control(n=8) and treated=8 group respectively, Control group was taken distilled water and treated group was taken extracts of Gagamsosokmyeong-tang(Jiajianxiaoxuming-tang) by orally for 20 days. Body weight was measured at 0, 5, 10, 15, 20 days after MIA injection. At the end of experiment, gross and histopathological examination on the articular cartilages of the knee joints were performed. Proteoglycan(PG) content of articular cartilages were analysed by safranine O staining method. The content of tumor necrosis $factor-{\alpha}(TNF-{\alpha})$, $interleukin-1{\beta}(IL-1{\beta})$ in synovial fluids were analysed by enzyme-inked inmunosorbent assay(ELISA) method. And also cycloxygenase-2(COX-2), matrix metalloproteinase 3(MMP-3), inducible nitric oxide synthase(iNOS), calpain immunochistochemical examination on the knee joints were performed. Results: PG content in articular cartilages of the treated group was significantly increased compared with control group. Histopathological osteoarthritic score of the treated group was significantly decreased compared with control group. $TNF-{\alpha}$ content in synovial fluids, expression of iNOS and calpain in synovial membrane of the treated group were significantly decreased compared with control group. But body weight, $1L-1{\beta}$ content in synovial fluids, expression of iNOS and MMP-3 of the treated group were not significantly changed compared with control group. Conclusions: On the basis of these results, we conclude that Gagamsosokmyeong-tang(Jiajianxiaoxuming-tang) has anti-arthritic effects on the MIA induced early stage osteoarthritis in rats.

  • Objectives : This study was carried out to investigate the effects of Samgi-eum($S{\bar{a}}nq{\grave{i}}-y{\check{i}}n$) on the monosodium iodoacetate-induced osteoarthritis in rats. Methods : Osteoarthritis was induced by injection of monosodium iodoacetate intraarticularly in both knee joints. Arthritic rats were divided into control and treated group. Control group were taken distilled water for 20days. Treated group were taken extracts of Samgi-eum($S{\bar{a}}nq{\grave{i}}-y{\check{i}}n$) by orally for the same duration. Normal group were injected normal saline and taken distilled water. Body weights were measured at 0, 5th, 10th, 15th, 20th day after injection. At the end of the experiment, gross and histopathological examination on the articular cartilages of the knee joints were performed. Proteoglycan contents of articular cartilages were analyzed by safranine O staining method. The contents of $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6 in synovial fluids were analyzed by ELISA method. Results : 1. Body weights of the treated group were significantly increased compared with control at 20days after injection. 2. Grossly, the severity of osteoarthritis in the treated group were alleviated compared with control. 3. • We investigated the effects of oyster shell extract (OSE) on papain-induced osteoarthritis in C57BL/6J mice. Osteoarthritis was induced in mice by a papain injection into the knee joint. The mice were divided into a total of five groups (n=8). The normal group was untreated, whereas the papain group (negative control) was induced with osteoarthritis and treated with water daily. The papain+DS group (positive control) was treated with diclofenac sodium. Papain+OSE groups were treated with OSE concentrations of 100 and 200 mg/kg/bw for 20 days. Proteoglycan content in articular cartilage was analyzed through safranine-O fast green staining and H&E staining. The histopathological changes in cartilage were measured by the Rudolphi score approach. The contents of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin (IL)-$1{\beta}$, and IL-6 in plasma were analyzed by the ELISA method. After experiments, body weights of the treated groups were not significantly different compared with the normal group. Cartilage loss and joint instability significantly improved in a dose-dependent manner in the OSE-treated group compared with the papain group (P<0.05). Proteoglycan content was significantly higher in the OSE-treated group than the papain group (P<0.05). Osteoarthritis scores of the OSE-treated group were significantly decreased compared with the papain group (P<0.05). TNF-${\alpha}$, IL-$1{\beta}$, and IL-6 content in the plasma of the papain+OSE treated groups significantly decreased in a dose-dependent manner compared with the papain group (P<0.05). These results suggest that OSE treatment might have anti-arthritic effects on papain-induced osteoarthritis in C57BL/6J mice.
