This study investigated the quality characteristics of saccharified minor cereal gruel prepared with different grain kojis (rice, buckwheat, sorghum, adlay, and Italian millet koji). The moisture, crude protein, crude lipid, and crude ash contents of raw materials showed ranges of 11.12 - 12.85; 5.81-16.24; 0.56 - 4.36, and 0.28 - 1.93%, respectively. The crude protein, crude lipid, and crude ash contents of the samples showed ranges of 1.64 to 2.44; 0.08 to 0.28, and 0.09 to 0.18%, respectively. The pH, L, a, and b values ranged from 6.11- 6.43; 58.72 - 65.96; 2.92 - 5.76, and 7.81- 9.42, respectively. The viscosities of saccharified minor cereal gruel prepared with different grain kojis were significantly (p<0.05) lower than the unsaccharified minor cereal gruel. After the saccharification, the soluble solids, glucose, and maltose content were significantly (p<0.05) increased, with ranges of $9.58-10.61^{\circ}Brix$; 0.64 - 0.90%, and 0.32 - 0.50%, respectively. A sensory evaluation indicated that minor cereal gruels saccharified using sorghum koji and adlay koji were more acceptable than unsaccharified minor cereal gruel. In conclusion, the cereal kojis could be used as a gruel processing method that would increase the sensory properties and nutritional values of gruels.
Germination conditions were evaluated to obtain barley containing a high content of gamma-aminobutyric acid (GABA), and quality characteristics of saccharified materials manufactured using germinated barley were investigated. Water absorption content of barley increased but pH of the steeping solution decreased with increasing steeping time at all steeping temperatures. The changes in water absorption content and pH were highest at a steeping temperature of $25^{\circ}C$. The highest GABA content was obtained at a steeping condition of $25^{\circ}C$ for 20 h. The highest GABA content was obtained for a germination condition of $5^{\circ}C$ for 36 h after steeping, resulting in an increase of 7.4 times more GABA contents than that in raw material. The pH of saccarified materials decreased but titratable acidity, amino type nitrogen content, and ammonia type nitrogen content increased during the saccharification period. GABA content of saccarified materials increased with increasing saccharification period, resulting in the highest GABA content from saccarified materials containing germinated barley.
Fermentation characteristics of chestnut-added yakju prepared using various proportions of raw materials such as rice koji, rice flour, cornflour koji and cornflour were investigated. The pH of chestnut-added yakju prepared with cornflour koji and saccharified cornflour showed a higher value than that of chestnut-yakju prepared with rice koji and saccharified rice flour. The total acid content of chestnut-added yakju prepared with rice koji and saccharified rice flour was higher than that of chestnut-added yakju prepared using cornflour koji and sacharified cornflour. The reducing of sugar in chestnut-added yakju prepared with saccharified rice flour or saccharified cornflour was rapid at the first brewing stage, decreased dramatically after 2 days, and then decreased slowly after 5 days of fermentation. The value of L and a, the Hunter values, were high in chestnut-added yakju prepared with cornflour koji, and value b was high in chestnut-added yakju with rice koji. The content of iso-amyl alcohol was the highest of seven kinds of fusel oil found in chestnut-added yakju. Ethanol content increased to $17.6{\sim}18.2%$(v/v) after 8 days of fermentation. The content of lactic acid was the highest of all organic acids in the chestnut-added yakju. Sensory test results on chestnut-added yakju prepared with saccharifed corn flour showed that if rice flour is used as a sugar supplement for chestnut, the yakju prepared using koji had better flavor and taste. If cornflour was used in the preparation of chestnut-added yakju, the used of saccharified cornflour offered superior flavor and taste.
Three strains able to efficiently produce ethanol from cellulosic hydrolysates were isolated from soil samples by enrichment culture in liquid saccharified wheat bran medium. The profiles of physiological and biochemical properties of two yeasts KM-09 and KM-402 and a bacterium Hg-225 were almost identical from those of Candida sp. and Klebsiella sp., respectively. Strains KM-09 and HG-225 used xylose and cellobiose as fermentable sugars, and HG-225 had a wide range of sugar utilization for ethanol fermentation. The optimal pH and temperature for growth of KM-09, KM-402 and HG-225 were 5.8, 5.6 and 6.8 and 32t, $30^{\circ}C$~ and $38^{\circ}C$, respectively. During the ethanol fermentation in saccharified wheat bran by the isolated strains, optimal temperature for ethanol production was more or less higher than those for growth, and addition of 0.2% (w/v) $MgSO_4$, into the medium enhanced ethanol productivity. Of the three strains ethanol content of KM-09 was the highest with about 2.3% (v/v), and ethanol production rate of HG-225 was faster than the others and maximum productivity was after 4 days. KM-09 (1.42% v/v) and HG-225 (1.05%, vlv) produced ethanol from 4% (wIv) xylose but growth rate was slower than on glucose. Otherwise KM-402 showed the highest ethanol productivity on glucose, but no ethanol was detected on xylose and cellobiose.
The aim of this study attempted to verify the possibility of bioethanol production using wasted medium density fiberboard (wMDF). In order to produce bioethanol from wood cellulosic materials must be carried out the process of pretreatment, saccharification, fermentation and distillation. First, the wMDF was pretreated using sodium chlorite and pretreated wMDF was prepared to 8% slurry and then slurry was saccharified with the commercial enzyme (Cellic CTec3). The fermentable sugar and pH of saccharified substrate were about 5.5% glucose and 4.4, respectively. Herein we compared the results of ethanol yield according to the nutrients added or without addition to increase ethanol yield. Ethanol fermentation was finished in about 24 hours, but it was delayed in experimental group without nutrients. Ethanol content and fermentation ratio of the final fermented mash prepared by utilizing jar fermenter was 25.40 g/L and 86.64%, respectively. At this time, the maximum ethanol productivity was confirmed as 1.78 g/Lh (ethanol content 21.38 g/L, 12 h), and the overall ethanol productivity was 1.05 g/Lh (ethanol content 25.27 g/L, 24 h). Using fermented liquid we could produced bioethanol 95.37% by continuous distillator packed with copper element in laboratory scale. These results show that wMDF has a potential valuable for bioethanol production.

To investigate the usefulness of freshwater alga Water-net (Hydrodictyon reticulatum, HR) as resources for production of fermentable sugars, the easiness of enzymatic saccharification was evaluated at first. When 6 plant materials (HR, Spirulina, Chlorella, Scenedesmus, Cladophora, Corn stover) were enzymatically hydrolyzed with 2% solid loading at the same condition, HR showed the highest ratio of saccharification based on glucose production. No milled HR was also completely saccharified at the amounts of optimal enzyme mixture. Glucose yield was not changed though the citrate buffer strength for saccharification was decreased from 0.1 M to 0.1 mM. Only about 10% yield reduction was observed compared to that of $120^{\circ}C$ treatment when HR was enzymatically hydrolyzed at room temperature. The saccharification was normally occurred at $37^{\circ}C$ and pH 6.5 which is general growth condition of fermentable microrganisms, suggesting that HR have a biomass characteristics applicable for the simultaneous saccharification and fermentation. The saccharification was occurred by more than 70~80% of one of the best condition although the supplied enzyme amounts was reduced to 1/10 volume. And the glucose yield by enzymatic hydrolysis was not decreased by 10% HR solid loading and began to decrease at more than 15% solid contents. Above these results show that HR is an interesting algal biomass which is relatively easy to be saccharified by hydrolyzing enzymes. In addition, HR is a flilamentous alga and very easy to be collected. Therefore, HR seems to be an useful and valuable resources in the economical production of fermentable sugars for manufacture of bio-chemical products.
In order to brew foxtail millet wine, a folk wine of Cheju, properties of raw materials, optimum brewing conditions were inveatigated. Carbohydrate and crude fat content of glutinuous foxtail millet are 71.27% and 3.47%, respectively. Since the ratio of water to steamed millet and ethanol concentration of wine showed negative correlation, less than 250% water had to added to steamed millet to maintain ethanol concentration in wine above 13%, Sugar consumption and ethanol production increased rapidly for the first 2 days, and main fermentation was done in 4 days. Ethanol concentrations were $13.0{\sim}13.4%$ when foxtail millet was used, and they were $14.0{\sim}14.3%$ for the mixture substrates of 90% millet and 10% rice or barley. Organic acids in millet wine were lactic acid, malic acid and succinic acid. The residual carbohydrates after fermentation were mainly xylose and oligosaccharides. A trace of methanol was detected in millet wine. The content of fusel oil was low, while the concnetration of organic acids was high. Optimum conditions for millet wine-making were as follows. Glutinuous foxtail millet with 10% rice as fermentation source need to be soaked in water and steamed for enough time. Water was added to steamed millet with the ratio of 2 : 1. The resulting mixture was stmnultaneously saccharified and fermented by Aspergillus orzae and Saccharomyces cerevisiae IAM 4274 at $20^{\circ}C$ for a week. Millet wine was prepared after filtering fermented broth while pressing for a week.
