Comparison of Alcohol Fermentation from Husked Rice and Barley Cooked by Hot Air Puffing and Steaming (열풍팽화 및 자숙한 벼와 보리의 알코올 발효성 비교)

  • The saccharification ratio and efficiency of alcohol fermentation from hot air puffed and steamed flour of husked rice and barley varieties were compared. Enzymatic hydrolysis of husked rice flour was improved by puffing, and that of glutinous rice was higher than that of nonglutinous one. By puffing of husked rice and barley, alcohol production was slightly increased, but fermentation period was reduced comparing with steam cooked husked rice and barley. Fermentation efficiencies of husked rice by puffing and rice by steam cooking were 90.72% and 87.77%, respectively. Residual reducing sugar of barley mash was higher than that of rice mash. The pH of mash was gradually increased during fermentation and was high in case of puffing treatment.

  • To develop high dfficiency-low energy consumption attrition coupled bioreactor for enhanced enzymatic hyerolysis of insoluble biomass, a tumbling drum type bioreactor was installed, and its efficiency was evaluated. The effects of drum structure and poerational conditions were investigated. The optimal saccharification at 3L drum was obtained at 8 baffled drum, drum diameter to baffle height ratio of 1:0.05, 100rpm, and addition of 600g of 3mm glass bead per liter. The consumed power for rolling of drum and energy consumption for half digestion of cellulose were measured, and compared with enhanced rate and yield to predict the economic prospect of the process. The tumbling drum type bioreactor seems to have appropriated structure for industrial scale operation, and further investigation for scale-up need to be conducted.

  • Alakline peroxide pretreatment for the delignification of poplar wood was performed. sinceit is a simple and efficent method for enhancing the enzymatic digestibility of wood residues. Approximately one-half of their lignin and most of the hemicellulose present in poplar wood were removed when the wood sawdust was reacted at 25$^{\circ}C$ for 100 hrs in an alkaline solution (pH 11.5) of 1% peroxide. The rate of decomposition as well as the saccharification efficiency were enhanced up to 350% and 260% respectively in comparision with those of the controll. This enhancement is comparable with that pretreated with 1% sodium hydroxide and 20% peracetic acid successively. The advantages of alkaline peroxide as delignifying agents against other chemicals were also discussed.

  • High-protein rice flour (HPRF) was prepared by an enzymatic process using ${\alpha}-amylase$ and g1ucoamylase without cooking Process and the feasibility of HPRF as infants foods was tested. Rice flour slurry was treated with 0.25% ${\alpha}-amylase$ and 0.5% glucoamylase at $55^{\circ}C$ for 24hrs. After saccharification, the digested rice slurry was centrifuged and the precipitated paste, was then heat-dried to obtain HPRF. The protein content of the HPRF was 20.8%. On the other hand, the supernatant of glucose enriched solution was decolourized, deionized and then isomerised to furctose at $60^{\circ}C$ for 100min by using immobilized glucose isomerase column. The high-fructose solution (HFS) contained 56% glucose, 42% fructose and 2% oligosaccharide. The nutritional quality of the HPRF was compared with milk protein and soybean protein in weight gain, feed efficiency ratio (FER), protein efficiency ratio (PER) and liver weight. HPRF was almost the same in all items with milk and soybean protein, but significantly superior to rice flour group.

  • Seaweed has high growth rate, low land usage, high CO2 absorption and no competition for food resources. Therefore, the use of lignin-free seaweed as a raw material is arising as a third generation biomass for bioethanol production. Various pretreatment techniques have been introduced to enhance the overall hydrolysis yield, and can be categorized into physical, chemical, biological, enzymatic or a combination. Thermal acid hydrolysis pretreatment is one of the most popular methods to attain high sugar yields from seaweed biomass for economic reasons. At thermal acid hydrolysis conditions, the 3,6-anhydro-galactose (AHG) from biomass could be converted to 5-hydroxymethylfurfural (HMF), which might inhibit the cell growth and decrease ethanol production. AHG is prone to decomposition into HMF, due to its acid-labile character, and subsequently into weak acids such as levulinic acid and formic acid. These inhibitors can retard yeast growth and reduce ethanol productivity during fermentation. Thus, the carbohydrates in seaweed require effective treatment methods to obtain a high concentration of monosaccharides and a low concentration of inhibitor HMF for ethanol fermentation. The efficiency of bioethanol production from the seaweed biomass hydrolysate is assessed by separate hydrolysis and fermentation (SHF). To improve the efficiency of the ethanol fermentation of mixed monosaccharides, the adaptation of yeast to high concentration of sugar could make simultaneous utilization of mixed monosaccharides for the production of ethanol from seaweed.

Characterization of Exolytic GH50A β-Agarase and GH117A α-NABH Involved in Agarose Saccharification of Cellvibrio sp. KY-GH-1 and Possible Application to Mass Production of NA2 and L-AHG (Cellvibrio sp. KY-GH-1의 아가로오스 당화 관련 엑소형 GH50A β-아가레이즈와 GH117A α-NABH의 특성 및 NA2와 L-AHG 양산에의 적용 가능성)

  • Recently, we sequenced the entire genome of a freshwater agar-degrading bacterium Cellvibrio sp. KY-GH-1 (KCTC13629BP) to explore genetic information encoding agarases that hydrolyze agarose into monomers 3,6-anhydro-L-galactose (L-AHG) and D-galactose. The KY-GH-1 strain appeared to possess nine β-agarase genes and two α-neoagarobiose hydrolase (α-NABH) genes in a 77-kb agarase gene cluster. Based on these genetic information, the KY-GH-1 strain-caused agarose degradation into L-AHG and D-galactose was predicted to be initiated by both endolytic GH16 and GH86 β-agarases to generate NAOS (NA4/NA6/NA8), and further processed by exolytic GH50 β-agarases to generate NA2, and then terminated by GH117 α-NABHs which degrade NA2 into L-AHG and D-galactose. More recently, by employing E. coli expression system with pET-30a vector we obtained three recombinant His-tagged GH50 family β-agarases (GH50A, GH50B, and GH50C) derived from Cellvibrio sp. KY-GH-1 to compare their enzymatic properties. GH50A β-agarase turned out to have the highest exolytic β-agarase activity among the three GH50 isozymes, catalyzing efficient NA2 production from the substrate (agarose, NAOS or AOS). Additionally, we determined that GH117A α-NABH, but not GH117B α-NABH, could potently degrade NA2 into L-AHG and D-galactose. Sequentially, we examined the enzymatic characteristics of GH50A β-agarase and GH117A α-NABH, and assessed their efficiency for NA2 production from agarose and for production of L-AHG and D-galactose from NA2, respectively. In this review, we describe the benefits of recombinant GH50A β-agarase and GH117A α-NABH originated from Cellvibrio sp. KY-GH-1, which may be useful for the enzymatic hydrolysis of agarose for mass production of L-AHG and D-galactose.

Ethanol Fermentation of Fusant between Heterologous Transformant of Saccharomyces cerevisiae and Candida tropicalis in Mini-jar Fermentor Scale (Mini-jar fermentor Scale에서의 Fusant의 Ethanol 발효)

  • The optimum conditions for ethanol fermentation and ethanol productivity of the fusant ESC-14-15 were examined in a mini-jar formentor scale (working volume : 2.5 liters) to assess the possibility of practical application. Addition of yeast extract to fermentation broth greatly enhanced the ethanol productivity and shortened the period of fermentation. The pH 4.2 was more favorable than pH 5.5 with respect to ethanol productivity and fermentation speed. The optimum concentration of liquefied potato starch for ethanol fermentation of FSC-14-15 was 15%(w/v) and the corresponding productivity was 8.7%(v/v) of ethanol with an efficiency of 80.6% to the theoretical maximum. When the fresh fermentation broth containing 20% of liquefied potato starch was inoculated with love(v/v) of inoculum, the fusant FSC-14-75 produced 11.0%(v/v) of ethanol in 4 days, which is considered comparable to that from an industrial process. From the liquefied cassava starch or the equal mixture of liquefied barley and sweet potato starch prepared according to the same method as in the industrial process except saccharification step, the fusnnt FSC-14-75 produced 8.5%(v/v) or 7.6%(v/v) of ethanol in 4 days, respectively.

Development of "Miscanthus" the Promising Bioenergy Crop (유망 바이오에너지작물 "억새" 개발)

  • In order to suggest correct direction of researches on Miscanthus spp. which are promising bioenergy crop, authors had reviewed and summarized various literature about botanical taxonomy, morphology and present condition of breeding, cultivation and utilization of miscanthus. Among the genus of Miscanthus which are known 17 species, the most important species are M. sinensis and M. sacchariflorus which origin are East Asia including Korea, and M. x giganteus which is inter-specific hybrid of tetraploid M. sacchariflorus and diploid M. sinensis. Miscanthus is superior to other energy crops in resistance to poor environments including cold, saline and damp soil, nitrogen utilization efficiency, budget of input energy and carbon which are required for producing biomass and output which are stored in biomass. The major species for production of energy and industrial products including construction material in Europe, USA and Canada is M. x giganteus which was introduced from Japan in 1930s. In present, many breeding programs are conducted to supplement demerits of present varieties and to develop "Miscanes" which is hybrid of miscanthus and sugar cane. In Korea, the researches on breeding and cultivation of miscanthus were initiated in 2007 by collecting germplasms, and developed "Goedae-Uksae 1" which is high biomass yield and "mass propagation method of miscanthus" which can improve propagation efficiency in 2009. In order to develop "Korean miscanthus industry" in future, the superior varieties available not only domestic but also foreign market should be developed by new breeding method including molecular markers. Researches on production process of cellulosic bio-ethanol including pre-treatment and saccharification of miscanthus biomass also should be strengthen.
