Effects of Soybean. Cheonggukjang and Doenjang on Serum Cholesterol Level and Weight Reduction in Rats Fed a High-Fat/High-Cholesterol Diet (대두, 청국장 및 된장이 고지방-고콜레스테롤식이를 급여한 흰쥐의 콜레스테롤 함량 및 체중감소에 미치는 영향)

  • This study was conducted to investigate the cholesterol lowering effect and weight reduction of autoclaved soy flour, cheonggulgang and doenjang, which are fermented using Bacillus subtilis DJI, in rats fed a high-fat/high-cholesterol diet. Male Sprague-Dawley rats weighing 250 g, were divided into five groups; a normal diet group (N), a high-fat/high-cholesterol diet group (HFC), a high-fat/high-cholesterol diet with 5% autoclaved soy flour group (HFC-S), a high-fat/high-cholesterol diet with 5% cheonggulgang group (HFC-CJ), and a high-fat/high-cholesterol diet with 5% deonjang group (HFC-DJ). The body weight gain and adipose tissues weights increased in rats fed a high-fat/high-cholesterol diet, but decreased significantly in rats fed autoclaved soy flour, cheonggulgang and doenjang, compared with the HFC group. Food intake was lower in the all HFC groups (HFC, HFC-S.HFC-CJ and HFC-DJ) than that of the N group, However, there was no differences in the food intake between HFC diet groups. Serum triglyceride level increased with a high-fat/high-cholesterol dietgroup whereas HFC-CJ and HFC-DJ groups were markedly decreased serum triglyceride levels compared with N group. Levels of serum total cholesterol and LDL-cholesterol, and level of total cholesterol in the liver were significantly lower in the HFC-S group, the HFC-CJ group and the HFC-DJ group than that of the HFC group, but exerted no significant change between the HFC groups on levels of total lipid and triglyceride in the liver. Fecal total lipid content increased in the HFC-CJ group and the HFC-DJ group than those of the HFC group and HFC-S group. The activities of heparin-releasable lipoprotein lipase (HR-LPL) and total-extractable LPL in adipose tissues decreased in the HFC-S group, the HFC-CJ group and the HFC-DJ group compared with the HFC group. These results suggest that dietary cheonggulgang and doenjang may exert cholesterol-lowering effect and prevent obesity.

  • A novel strain of SFO41 producing a large amount of cholesterol oxidase as an extracellular enzyme isolate from Korean salt fermented foods. The strain was identified as Bacillus megaterium based on morphological, cultural and physiological characteristics. Experiments were carried out to optimized the condition of cholesterol oxidase production using B. megaterium SFO41. B. megaterium SFO41 was shown to give the maximum yield of cholesterol oxidase in the medium containing 2.0% glucose, 0.5% yeast extract. 0.03% $MgSO_4{\cdot}7H_2O,\;0.02%\;K_2HPO_4,\;0.2%\;NH_4NO_3$ and 0.2% cholesterol. The optimum culture conditions, temperature, initial pH and agitation speed were $30^{\circ}C$, 7.0 and 150 rpm, respectively. The enzyme production reached a maximum level at 24 hr of cultivation (2.37 U).

Cholesterol Removal from Whole Egg by Crosslinked β-Cyclodextrin

  • This study was carried out to optimize cholesterol removal in whole egg using crosslinked ${\beta}$-cyclodextrin (${\beta}$-CD) and to recycle the ${\beta}$-CD. Various factors for optimizing conditions were concentration of the ${\beta}$-CD, mixing temperature, mixing time, mixing speed and centrifugal speed. In the result of this study, the optimum conditions of cholesterol removal were 25% crosslinked ${\beta}$-CD, $40^{\circ}C$ mixing temperature, 30 min mixing time, 1,200 rpm mixing speed and $2,810{\times}g$ centrifugal speed. The recycling was repeated five times. The cholesterol removal was 92.76% when treated with the optimum conditions. After determining the optimum conditions, the recyclable yields of the crosslinked ${\beta}$-CD ranged from 86.66% to 87.60% in the recycling and the percentage of cholesterol removal was over 80% until third recycling. However, the cholesterol removal efficiency was decreased when the number of repeated recycling was increased. Based on the result of this study, it was concluded that the crosslinked ${\beta}$-CD was efficient for cholesterol removal in whole egg, and recycling is possible for only limited repeating times due to the interaction of the ${\beta}$-CD and egg protein.

Effect of Fermented Chub Mackerel Extract on Lipid Metabolism of Rats Fed Diets without Cholesterol

  • The present study was conducted to evaluate the effect of fermented chub mackerel extract (FCME) on lipid metabolism in rats fed diets without cholesterol. Four week-old male rats were divided into three groups of 10 rats with 0, 1% or 2% FCME supplementation to the diets. Purified diets were used in the present study. Feed and water were fed ad libitum. FCME supplementation had no effect on the activities of acetyl-CoA carboxylase, fatty acid synthetase, and the content of free cholesterol, triglyceride and phospholipid in the liver (p>0.05). 1% FCME supplementation significantly increased serum triglyceride (p<0.05) and hepatic 3-hydroxy-3-methylglutaryl-CoA reductase activity (p<0.05) with no effect on serum total cholesterol, free cholesterol and phospholipid concentration. FCME supplementation significantly reduced serum LDL+VLDL-cholesterol (p<0.01) and atherogenic index (p<0.01) with no effect on HDL-cholesterol. The current study showed that FCME inclusion might reduce the risk of atherosclerosis in rats fed diet without cholesterol.

Optimization of β-Cyclodextrin Recycling Process for Cholesterol Removal in Cream

  • This study was designed to find optimum conditions of four different factors (ratio of solvent to cholesterol-$\beta$-cyclodextrin complex, mixing speed, mixing temperature, and mixing time) for cholesterol dissociation in cream. Using the ratio of 6 to 1 (solvent to the complex) showed the highest cholesterol dissociation rate (82.50%) when mixed at 100 rpm at $50^{\circ}C$ for 1 h. Mixing speed did not significantly affect the cholesterol dissociation. Also, mixing time appeared to be insignificant. The optimum mixing temperature was $50^{\circ}C$ and mixing at $40^{\circ}C$ resulted in a significantly lower rate, compared with that at $50^{\circ}C$. In a subsequent experiment, using recycled $\beta$-cyclodextrin only showed 75.07% of cholesterol removal in cream, while the mixture of recycled to unused $\beta$-CD with the ratio of 6 to 4 increased cholesterol removal to 95.59%, which is highly close to that of 100% unused $\beta$-CD.

Effects of Nuruk or Wheat Bran Supplemented Diet on the Serum Levels of Cholesterol and Activities of Hepatic Oxygen Free Radical Metabolizing Enzymes in Rats (누룩 또는 밀기울 첨가식이로 성장시킨 흰쥐의 혈중 Cholesterol 및 간조직 유해산소 대사효소 활성 변동)

  • The effects of nuruk and wheat bran on cholesterol level in serum and activities of free radical metabolizing enzymes were investigated in rats. The rats were fed a diet containing nuruk or wheat bran for one month. Body weight and food intake were measured. Animals were sacrificed after one month. The increased food efficiency ratio throughout whole growth period was observed in the rats fed with either nuruk containing Aspergillus terreus or wheat bran compared with control group on normal diet. In the rats fed with nuruk, hepatic GSH content, glutathione S transferase activity, hepatic cytochrome P 450 content, and aniline hydroxylase activities were generally increased. In the rats fed with nuruk containing other fungi except Aspergillus terreus, xanthine oxidase activity was decreased. The decreased cholesterol level in serum was observed in rats fed with nuruk prepared from Aspergillus terreus and wheat bran. LDL cholesterol level was decreased in rats fed with nuruk prepared with other fungi such as Penicillium sp. and Rhizopus sp. But HDL cholesterol level was increased in all groups fed with nuruk from any fungi and wheat bran. These results suggested that nuruk or wheat bran supplemented diet might exert their effect by decreasing cholesterol level in serum and amount of oxygen free radical level.

  • Demonstration of the highly positive correlation between blood cholestrol levels and heart disease has made consumers wary of the fats in meat, milk and eggs. The egg, as perhaps the single largest common source of cholesterol, has been cited by many members of the medical and scientific world as a food contributing to heart disease. In light of decreasing per capita egg consumption and continuing dietary egg cholesterol controversy, many researchers have focused their efforts on egg nutrition. The results reported, however, are often contradictory. In spite of the disputable scientific evidence, the egg has been labelled (erroneously) as a highly cholesterogenic food. The objective of this presentation is to present a general picture of the problem and discuss our laboratory findings relevant to the problem. An isotope technique was utilized to incorporate $^{14}C$-cholesterol into egg yolk lipoproteins and study the metabolic fate of dietary ovo-cholesterol in rats. Two hundred and fifty micro-curies of 4-$^{14}C$-cholesterol, emulsified in corn oil, were orally administered to five Single Comb White Leghorn laying hens. Eggs were collected, hard-boiled, and the hot dried egg yolk powder (HEY) was prepared. Total radioactivity excreted via feces was determined. The rat groups fed egg yolk powder excreted more than 95% of the ingested ovo-cholesterol, whereas the rat chow group excreted only 47%. No difference was observed between HEY and CEY treatments. Therefore, an unknown lipid factor present in egg folk accelerates cholesterol turnover rate and excretion via feces.

Changes in Fatty Acid and Cholesterol Composition of Koran Styled Beef Broths (Gom-Guk) during Cooking (쇠고기 곰국의 조리중 지방산 및 Cholesterol 조성변화)

  • The content of total lipid, cholesterol and fatty acid in Korean styled various beef broths was examined before and during toiling to determine the influence of cooking time. The values obtained from the broths of brisket, ox-tail and small-intestine were determined by using G. C. The highest total lipid value among small-intestine, brisket and ok-tail was in the small-intestine. The content of total lipid in the boiled meat decreased during cooking with the exception of the brisket. Total lipid content found in the broth of brisket meat increased until 6hr and thell decreased at cooking time of 8hr and 10 hr. The highest total lipid contents in the broths of brisket, ox-tail and small-intestine were found when the cooking time were 6 hr, 10 hr and 4 hr, respectively. The main fatty acids found in the broths of brisket, ok-tail and small-intestine were $C_{14}$, $C_{16}$, $C_{18}$, $C_{18:1}$, and $C_{18:1}$. The content of $C_{18:1}$ and $C_{18:2}$ from the brisket meat increased until 4hr's cooking, especially $C_{18:2}$ increased significantly until 6 hr cooking. The highest ratios of unsaturated to saturated fatty acid (UNS/S) in the meat of brisket and ox-tail were found in the 2hr and 4hr cooking, respectively. The ratios of UNS/S in the broths of both of brisket and small-intestine, and ok-tail were highest in the 4 hr and 2 hr cooking, respectively. The content of total cholesterol (TC) was the highest in the small-intestine and the highest percentage of ester cholesterol was 5.4 in the brisket. The content of TC in small-intestine, brisket and ox-tail was decreased significantly after 2 hr cooking. Precentage of the ester in the brisket was the highest when the cooking time was 8 hr. Only trace amount of total cholesterol was found in the broth.

Preventive Effects of Nokyongdaebotang on Hyperlipidemia Rats (녹용대보탕(鹿茸大補湯) 투여에 의한 흰쥐의 고지혈증(高脂血證) 예방(豫防)에 관한 실험 연구)

  • In order to study the preventive effects of hyperlipidemia depending on endogenous and exogenous methods of induction, after observing what happens when Nokyongdaebotang, a strengthening up treatment is orally administrated into the ways that cause hyperlipidemia either by the exogenous hyperlipidemia condition model method, which is the way where you orally administrate the cholesterol that was dissolved in olive oil, or the endogenous hyperlipidemia model method, where it uses the injecting the Triton WR-1339 vein method, or to the already inducted white rats, the following conclusions could be drawn. 1. The endogenous induction method, cholesterol diet, helps preventing Total Cholesterol, TG, and LDL-Cholesterol, Free Fatty Acid, Phospholipid's augmentation within the blood in the white rats that is being induced or just inducted with hyperlipidemia 2. The exogenous induction method, Triton WR injection, helps preventing Total Cholesterol, TG, and LDL-Cholesterol, Free Fatty Acid, Phospholipid's augmentation within the blood in the white rats that is being induced or just inducted with hyperlipidemia 3. The HDL-Cholesterol did not increase in regard. This is considered to be because when the geological features the HDL-Cholesterol increases proportionally. In deference to the above results, Nokyongdaebotang, which strengthens the vitality, showed that it helps prevent white rats that is being induced or just inducted with hyperlipidemia no matter whether it is endogenous or exogenous.

Analysis on the Difference of Dietary Intake Behavior in Subjects with/without Various Types of Dyslipidemia from the Seventh (2016) Korea National Health and Nutrition Examination Survey (KNHANES) (2016년 제7차 국민건강영양조사를 이용한 이상지질혈증 유무 및 형태에 따른 식품섭취행태 차이 분석)

  • This study examined the difference of dietary intake behavior between groups of adults aged >20 years with or without dyslipidemia, and comparing the various types of dyslipidemia, based on the Seventh (2016) KNHANES. Men showed higher rate for triglyceridemia and LDL-cholesterolemia than women and subjects in the age group 50-59 years exhibited the highest incidence of triglyceridemia, total cholesterolemia and HDL-cholesterolemia. The ratio of obese people was higher in most types of dyslipidemias. The top five foods in each group of cereal & grain, meat, vegetable, and fruit were selected for analysis, based on the food frequency. The dyslipidemia group showed higher intake frequencies for mixed grains, cabbage kimchi and leafy vegetable than the no-dyslipidemia group, and the group with high blood total cholesterol for mixed grains, apple, and cutlassfish-croaker. Group with high blood triglyceride showed higher intake frequencies of rice and mackerel-mackerel pike than the other groups of high blood triglyceride. No food showed significant difference in the frequencies between groups of LDL-cholesterol. Group with high blood HDL-cholesterol showed higher intake frequency of bibim-fried rice, deep fried chicken, stir fried chicken, apple, tangerine, banana and butter-margarine. These results indicate that people with high levels of blood total cholesterol are more concerned with food having health benefits than those with abnormal levels of other blood lipid. More information on dietary benefits need to be provided to patients having high levels of triglyceride, LDL-cholesterol and HDL-cholesterol.
