Characterization, detection and identification of transgenic chili pepper harboring coat protein gene that enhances resistance to cucumber mosaic virus

  • Previously, two events (H15 and B20) of transgenic pepper (Capsicum annuum L.) that enhanced resistance to Cucumber mosaic virus (CMV) by the introduction of CMV coat protein (CP) gene were constructed. Presently, a single copy number of the CP gene was revealed in H15 and B20 by Southern blot. To predict possible unintended effects due to transgene insertion in an endogenous gene, we carried out sequencing of the 5'-flanking region of the CP gene and a Blastbased search. The results revealed that insertion of the transgene into genes encoding putative proteins may occur in the H15 and B20 transgenic event. Mutiplex polymerase chain reaction (PCR) for simultaneous detection and identification of transgenic pepper was conducted with a set of nine primers. Both transgenic event were differentiated from non-transgenic event by the presence of 267 bp and 430 bp PCR products indicative of CP gene specific primer pairs and primer pairs targeting the CP gene and 35S promoter. H15 and B20 uniquely possessed a 390 bp and 596 bp PCR product, respectively. The presence of a 1115 bp product corresponding to intrinsic pepper actin gene confirmed the use of pepper DNA as the PCR template. The primer set and PCR conditions used presently may allow the accurate and simple identification of CMV resistant transgenic pepper.

  • Objectives: By the end of the 2000s, the economic situation in many European countries started to deteriorate, generating financial uncertainty, social insecurity and worse health status. The aim of the present study was to investigate how the recent financial crisis has affected the lifestyle health determinants and behaviours of older adults living in the Mediterranean islands. Methods: From 2005 to 2015, a population-based, multi-stage convenience sampling method was used to voluntarily enrol 2749 older adults (50% men) from 20 Mediterranean islands and the rural area of the Mani peninsula. Lifestyle status was evaluated as the cumulative score of four components (range, 0 to 6), that is, smoking habits, diet quality (MedDietScore), depression status (Geriatric Depression Scale) and physical activity. Results: Older Mediterranean people enrolled in the study from 2009 onwards showed social isolation and increased smoking, were more prone to depressive symptoms, and adopted less healthy dietary habits, as compared to their counterparts participating earlier in the study (p<0.05), irrespective of age, gender, several clinical characteristics, or socioeconomic status of the participants (an almost 50% adjusted increase in the lifestyle score from before 2009 to after 2009, p<0.001). Conclusions: A shift towards less healthy behaviours was noticeable after the economic crisis had commenced. Public health interventions should focus on older adults, particularly of lower socioeconomic levels, in order to effectively reduce the burden of cardiometabolic disease at the population level.

Determination of Optimal Unit Hydrographs and Infultration Rate Functions from Single Rainfall-Runoff Event (단순 강우-유출 사상으로부터 최적단위도와 침투율의 결정)

  • This paper is to present the determination of the optimal Joss rate parameters and urnt bydrographs from the observed single rainfall-runoff event using optimization models coupled with a stochastic technique for the global solution. Two kinds of the linear program models are formulated to derive the optimal unit hydrographs and loss rate parameters for gaged basins; one mimmizes the summation of the absolute residual between predlCted and observed runoff ordinates and the other, the maximum absolute residuaL Multistart algorithm which is one or stochastic techniques for the global optimum is adopted to perturb the parameters of the loss rate equations. Multistart efficiently searches the feasIble region to identify the global optimlUll for loss rate parameters, which yields the optimal loss rate parameters and unit hydrograph for Kostiakov's, Plulip's, and Horton's equation. The unique unit hydrograph ordinates for a gIven rainfall-runoff event iS exclusrvely obtained WIth $\Phi$ index, but unit hydrograph ordinates depend upon the parameters [or each loss rate equations. The parameters of Green-Ampt's are determined through a trial and error method. In this paper the single rainfall-nmoff event observed from a watershed is considered to test the proposed method. The optimal unit hydrograph herein found has smaller deviations than the ones reported previously by other researchers.

Actor's Role and Networks in the Environmentally Friendly Farming in Busan Metropolitan Agricultural Region (부산 김해평야 농업지역 친환경농업의 행위자-연결망 연구)

  • This study is to analyze the adoption of environmentally friendly fanning(EFF) by Actor-Network Theory(ANT). ANT declares that the world is full of hybrid entities containing both human and non-human elements and maintains that adoption of an innovation comes as a consequence of the action of everyone in the chain of actors who has anything to do with it. In this study, adoption of EFF will be analysed through the role of actors and networks. And this paper try to identify the intermediaries and obligatory passage point(OPP) in each networks. In study area, 5 actors, -nature, governmental institute, food processors, consumers and farmers-, have each roles in their networks, But only 18 farmers adopted EFF. This study revealed that three OPPs were not overcome in each network. The one is nature, such as water and soil pollution. Another is shortage of reliability between farmers and governmental institute. The other is shortage of information about agricultural commodity trade. And through this application of ANT to the EFF, we contend that ANT can be useful for studies of diffusion of EFF and sustainability of rural systems in situations where interactions of the social, technological and political are regarded as particularly important.

A study on the children's oral health behavior according to the general characteristics of parents (부모의 일반적인 특성에 따른 자녀의 구강보건 행태에 관한 연구)

  • Objective : The purpose of this study was to examine the relationship of the general characteristics of parents to their children's oral health care. The subjects in this study were 117 parents and their children who resided in rural communities in the region of K. Methods : A survey was conducted with the consent of the parents from July 1 to 30, 2010. As for the survey on the children, the children who were in the lower grades were interviewed, and the upper graders filled out the questionnaires in person. A frequency analysis was carried out to find out the general characteristics of the parents and children, and x2-test was utilized to grasp the links between the general characteristics of the parents and the children's oral health care. A SPSS WIN 12.0 program was employed to analyze all the collected data. The findings of the study were as follows: Result : 1. As a result of investigating the relationship of the occupation of the parents to the children's regular dental checkup, the 71.8 percent of the entire children didn't get a dental checkup on a regular basis. The biggest percentage of the children of the self-employed parents(100%) got a regular dental checkup, and 90.9 percent of the children of the government workers did that(p<.05). 2. As a result of checking the relationship of the occupation of the parents to the children's dental-caries experiences, 39.3 percent of the whole children had no such experiences. The largest percentage of the children of the company employees(51.9%) had dental-caries experiences(p<.05). 3. Concerning the links between the academic credential of the parents and the children's toothbrushing method, the biggest percentage of the children(40.2%) brushed their teeth up and down. The largest rate of the children whose parents were high-school graduates(41.5%) brushed their teeth in that way(p<.05). 4. Regarding the links between the toothbrushing time of the parents and the children's view, 43.6 percent found their parents to take good care of their teeth to make them clean. 60 percent whose parents brushed their teeth after having breakfast and dinner took the best view(p<.001). 5. As for the oral-health education experiences of the parents and a time for the children's change of toothbrushes, 29.1 percent changed their toothbrushes every two months, and 29.1 percent did that not on a regular basis but when the bristles of their toothbrushes got bent(p<.05). Conclusions : The above-mentioned findings suggest that in order to promote children's oral health in consideration of the characteristics of their parents, children should be urged to get a regular dental checkup, and the development of oral-health education programs in which parents and children can participate together is urgently required.

Habitat Connectivity Assessment of Tits Using a Statistical Modeling: Focused on Biotop Map of Seoul, South Korea (통계모형을 활용한 박새류의 서식지 연결성 평가: 서울시 도시생태현황도 자료를 중심으로)

  • Species distribution modeling is one of the most effective habitat analysis methods for wildlife conservation. This study was for evaluating the suitability of species distribution to distance between forest patches in Seoul city using tits. We analyzed the distribution of the four species of tits: varied tit (Parus varius), marsh tit (P. palustris), great tit (P. major) and coal tit (P. ater), using the landscape indexes and connectivity indexes, and compared the resulting suitability indexes from 100m to 1,000m. As factors affecting to the distribution of tits, we calculated landscape indices by separating them into intra-patch indices (i.e. logged patch area (PA), area-weighted mean patch shape index (PSI), tree rate (TR)) and inter-patch indices (i.e. patch degree (PD), patch betweenness (PB), difference probability of connectivity (DPC)), to analyze the internal properties of the patches and their connectivity by tits occurrence data using logistic regression modeling. The models were evaluated by AICc (Akaike Information Criteria with a correction for finite sample sizes) and AUC (Area Under Curve of ROC). The results of AICc and AUC showed DPC, PA, PSI, and TR were important factors of the habitat models for great tit and marsh tit at the level of distance 500~800m. In contrast, habitat models for coal tit and varied tit, which are known as forest interior species, reflected PA, PSI, and TR as intra-patch indices rather than connectivity. These mean that coal tit and varied tit are more likely to find a large circular forest patch than a small and long-shaped forest patch, which are higher rate of forest. Therefore, different strategies are required in order to enhance the habitats of the forest birds, tits, in a region that has fragmented forest patches such as Seoul city. It is important to manage forest interior areas for coal tit and varied tit, which are known as forest interior species and to manage not only forest interior areas but also connectivity of the forest patches in the threshold distance for great tit and marsh tit as adapted species to the urban ecosystem for sustainable ecosystem management.

Identification and Safety Assessment of Cucumber Mosaic Virus Coat Protein in Genetically Modified Pepper (Capsicum annuum)

  • The great economic losses caused by Cucumber mosaic virus (CMV) infection of peppers has led to the development of genetically modified (GM) CMV-resistant peppers. We developed virus-resistant pepper plants using Agrobacterium tumefaciens -mediated transformation. The expressed recombinant protein was purified using nickel-nitrilotriacetic acid resin and immunoaffinity chromatography, and purity was assessed by sodium dodecyl sulfate polyacrylamide gel electrophoresis. Immunoblot analysis revealed the purified CMV coat protein (CMV-CP) had a molecular mass of 25 kDa. After in-gel digestion and desalting, the internal peptide fragments of CMV-CP were sequenced by matrix-assisted laser desorption/ionization-time of flight. Most GM pepper and Escherichia coli BL21 internal peptides had identical peptide sequences and contained 137 of 183 whole peptides in CMV-CP. A quantitative enzyme-linked immunosorbent assay was performed to detect CMV-resistant GM peppers. We also provide basic information about the expressed protein in GM peppers for further safety assessment. The contents of soluble protein and CMV-CP were measured in GM and control peppers cultivated in three different areas of Korea. Statistical significance in terms of cultivation areas, harvest times, generations, and plant tissue origin were determined based on a P value of 0.05. The highest amount of CMV-CP was detected at the seedling stage from plant grown in each region. T3 and T5 showed significantly different levels of CMV-CP from T4 in leaves in the whorl stage. No statistical differences were observed among GM peppers at different stages of maturity in any cultivation area. The results from this study contribute to the safety evaluation of newly designed CMV-resistant GM peppers and provide a standard against which to compare other virus-resistant GM peppers.

  • In order to create a successful regional development model, there should be an effort to foster a strategic industry which boosts a geographical features of rural areas. Because, It must be clear that the strategic industry chosen between choice and concentration of the development of the local industry development which boosts the local economy revitalization. The local government should have continual concern and roles to fulfil political subjects and keep estimating about the possibility that there could be the sustainable development by Task Ahead of the local industry. This study's indicators were established for the adequacy evaluation of the industrial policy by focusing on domestic regional development's success example. Next, Conjoint analysis was conducted on the Experts. On the basis of this analysis, the weights of indicators are derived. And this study suggest the reasonable evaluation system to take Sustainable industry plan of regional strategic. Consequently, 'Regional development' sector separate 3 categories which are 'Local economy', 'Human Resource Development', 'local marketing' and 9 indicators were derived. 'Industrial development' sector derived 9 indicators about 'Construction of infrastructure', 'Technical development' and 'business support'. Looking through the implications of this study. First, the problem needed to be supplemented based on the Diversity and concreteness of Evaluation system when government assesses regional industry development policy. Second, local government have to strategically consider the possibility of convergence between industries and easiness of network in a broad region. Finally, Foundation and support of the professional manpower and companies for the regional industries must be established.

  • Sens of insecutity of the public and professionals about harmful effects of radiation is increasing in an accident at the Chernobyl and Fkushima nuclear power plant.Anxiety was amplified to lack of information about radiation majority of people. To target the middle and high school in the region of Daegu and Gyeongsangbuk-do, to investigate the radiation recognition of the next generation, it is intended to present a model of education for the safe use of radiation. The High School of the six metropolitan cities, city, town through the questionnaire and needs to be educational experience of radiation and use knowledge level of radiation, experience in daily life, understanding of man-made radiation and natural radiation, information channel on radiation, the radiation Distribute the total 800 parts of, to recover the 629 unit, was analyzed for 155 females 474 males. Many people 75.36% of the people, to 24.64% female subjects of this investigation, was constant, respectively from 13 to 18 years age. It is a large number and 30.37% of the respondents as "normal" level of knowledge of radiation, for the type of radiation, most knew. You have answer for risk experience of the medical radiation was higher, touching a lot of information via the broadcast medium in general, and the accuracy is low. I thought we wanted to be educated three or more twice a year, as an educator,about 71.37% and radiation-related understanding of knowledge and background in accordance with the diversification of information channels, the regional differences between urban and rural areas. But I considered the difference age (grade) for each is displayed, intended for junior high school students, the target surface and use the occurrence of radiation, high school students, the need for education about risk and application of radiation through this study.

Water Scarcity Assessment Using Green and Blue Water Concepts (그린워터 및 블루워터를 이용한 물부족 평가)

  • With climate change and population growth, there are significant increases in water scarcity. There have been water security assessments to abate the gap between water demand and availability to support water resource management. However, most of the assessments are focusing on the water that flows through either on or below the land surface, failing to consider water that infiltrates and can be used by vegetation. This study presents water scarcity assessment accounting for Blue and Green water concept, and applied the method to Boryung region. Monthly streamflow, evapotranspiration, and soil moisture were estimated by SWAT modeling, and each of them was used to analyze Blue and Green water scarcity. Blue and Green water scarcity had different aspect, and the result indicated the time when water scarcity is more likely to happen. The water scarcity assessment framework presented in this paper provides novel assessment method integrating hydrologic and ecosystem aspects, thereby improving the understanding of how water resources should be managed.
