Different kinds of cytokinins and auxins were tested for both shoot induction and rooting in a rare species, Abeliophyllum distichum Nakai. BA in WPM medium was the most effective in shoot induction, whereas zeatin seemed to be the most suitable for shoot elongation. Kinetin, at the concentration of 2.0~5.0 mg/L showed an effect in shoot induction, but the effect was inferior to BA and zeatin. Rapid shoot elongation could be achieved when the cultures were maintained on the diffuse light condition (below 500 lux) regardless of cytokinin treatments. For in vitro rooting, IBA was investigated as the best type of auxin tested when half strength GD medium was incorporated. The frequency of rooting using the plant growth regulator and medium just mentioned above was revealed as approximately 90%. In addition, the survival rate of rooted plantlets was almost 100% in an artificial soil mixture.
This study was undertaken to develop an efficient propagation technique for mature Betula davurica. Using aseptic materials taken from in vitro culture, the effects of media and plant growth regulators on shoot proliferation and rooting were investigated. DKW medium turned out to be the best in shoot proliferation among the media tested. Whereas axillary buds were better culture material than apical buds in proliferation of shoots, apical buds were slightly better than axillary buds on shoot elongation. Neither 1 /2 MS nor WPM medium seemed to be suitable for shoot multiplication or elongation. When the explants were cultured on 1/2 MS medium, shoot elongation was retarded by forming big callus at the base. In the case of WPM, shoots could be formed normally, but they exhibited slow growing. NAA was so effective on in vitro rooting that more than 80% rooting could be achieved on half-strength DKW medium supplemented with 1.0 mg/L NAA after 4 weeks in cultures. Ex vitro rooting using elongated shoot was also applicable to rooting and acclimatization. Rooted plantlets were successfully acclimatized in an artificial soil mixture and grew normally. The results demonstrate that efficient mass propagation of mature B. davurica can be done through tissue culture.

In order to promote in vitro rooting in SOD2-transgenic rose plantlets, which were not well rooted in a rooting medium (MS medium with NAA $0.03mg{\cdot}L^{-1}$), we dipped the plantlets into liquid $Kelpak^{(R)}$ before placing them in the rooting medium. After 4 weeks, $Kelpak^{(R)}$ significantly promoted in vitro rooting in the plantlets. Therefore, $Kelpak^{(R)}$ can be used successfully to aid in the in vitro rooting of rose plantlets with roots that are not well-generated.
This study was conducted to determine the three main factors [rooting time, plant growth regulators (PGRs), rooting medium] of cuttage for mass production of Sageretia thea. The experiment was observed on cuttage tray after 120 days and classified by rooting times, plant growth regulators, rooting medium. Materials were collected from natural habitats of S. thea from the Korean peninsula at March, July and September during 2019. As a result of the experiment, rooting times were showed statistically significant differences. March group's rooting rate and root length were 15 and 200% better than others. In PGRs experiment by concentration, There was no significant correlation between the control and experiment in terms of rooting rate. However, In the case of root length, IBA 500 mg/L, IBA 250 mg/L and IAA 1,000 mg/L groups were lengthened up to 170% compared to the control group, confirming statistically significant differences. As a result, we recommend treating cuttings with IBA 500 mg/L in March with Hardwood cutting.
Effects of growth regulators and medium components were tested for shoot multiplication and callus growth from shoot explants of black locust. During shoot multiplication, callus growth at the cut end of shoot explants proceeded shoot development. The basal callus growth seemed to be a function of both mineral components and cytokinin supplemented in the medium. Maximum callus growth was induced by 0.5${\mu}$M BAP and the callus growth decreased as the level of BAP increased. Positive correlations were found between basal callus growth, and shoot multiplication and growth. Shoot multiplication was greatest on BSM medium (black locust shoot culture medium) supplemented with 1 $\mu$M BAP. With medium containing high nitrogen content, both shoot multiplication and growth were significantly enhanced. A new BRM medium was the most effective for rooting of black locust among three rooting media tested.
100% root formation in in vitro cultures was observed regardless of kind and levels of auxin used and explant source. The number of roots/explant was increased in 0.5~1.0 mg/L IAA treatment. Thicker roots were observed with the addition of 9% sucrose compared with medium containing lower sucrose concentrations. Paclobutrazol and chlormequat had no effect on tuberization of formed roots but slightly increased the number of root. In in vivo rooting, soaking of regenerated shoot cuttings to 100 mg/L IBA for 15 to 60 minutes was found effective. Treatment of 0.1% IBA rooting powder and planting in rooting medium composred of vermiculite(1) : perlite(1) gave 100% rooting and survival.
This study was conducted to determine the optimum medium and humidification period for the strawberry (Fragaria × ananassa Duch. cv. Maehyang) cutting seedling production. The cuttings were planted in coir (CO), rockwool (RW), phenolic foam (PF), and organic foam medium (OFM) with fogging treatment in 0, 3, 6, 9, or 12 days on February, 2019. And, as the field verification examination research, cuttings were planted in CO, RW, PF, and OFM, with misting treatment in 0, 6, 9, 12, or 15 days on June, 2019. In fogging treatments, rooting and survival rates of seedlings tended to increase with longer fogging periods, and rooting and survival rates were showed significantly higher in CO medium with more than 9 days of fogging periods. In misting treatments, rooting rate was significantly higher in CO and RW medium with more than 9 days of misting periods, and survival rate was significantly higher with more than 12 days of misting periods. There was no significantly difference by growth medium and humidification period in shoot and root growth. Therefore, when considering the rooting and survival rate the CO medium with 9 days of humidification period could be beneficial for the production of the 'Maehyang' strawberry cutting seedlings.
This study was carried out to investigate the optimum composition and concentration of medium for the acclimation of the shoots derived from bioreactor culture of Scrophularia buergeriana. Rooting and growth of the shoots cultured in bioreactor for 4 weeks were better on liquid full strength MS medium while one-tenth of MS medium was good for rooting and growth of the shoots cultured in bioreactor for 6 weeks. Leaf expansion was more effective in the shoots cultured for 6 weeks on solid MS medium than in those cultured for 4 weeks. No leaf expanded in liquid medium. Vitrification rate was higher in full strength solid MS medium than in half strength MS medium. It is concluded that acclimation of the shoots is more effective on the half strength solid MS medium containing 1.2% agar.
An efficient in vitro plant regeneration system was developed through shoot proliferation from axillary buds of Tectona grandis (L.f), APNBV-1 (Andhra Pradesh North Badrachalam Venkatapuram-1) clone. Multiple shoots of high quality were produced in vitro from axillary bud explants. An average of 4.39 shoots/explant were obtained on Murashige and Skoog's (MS) medium supplemented with plant growth regulators (PGRs) benzyl amino purine (BA), kinetin (KN), indole acetic acid (IAA), gibberillic acid ($GA_3$), growth adjuvants casein hydrolysate (CH), adenine sulphate (Ads) and antioxidants ascorbic acid, polyvinyl pyrrollidine (PVP). Eighty five percent of rooting was observed in ex vitro rooting media containing IBA and vermiculite. In ex vitro rooting, single shoots with 2 to 3 nodes were subjected to IBA of different concentrations at different periods of time intervals. Direct rooting in vermiculite at 500 ppm concentration of IBA resulted in 4.3 number of roots with 2 cm length. Minimum response of rooting and length of roots were recorded at 100 ppm concentration of IBA. Planlets were transferred to plastic bags for short acclimatization stage in green house where they survived at 95%.
Optimum conditions for rooting of hardwood cuttings of two mulberry varieties have been determined with respect to the concentration of IBA applied to the base of cutting, dipping depth into IBA solution, season of cutting collection and rooting medium in the bottom heat bins. 1. Optimum range of IBA concentration was 2,000 p.p.m.∼4,000 p.p.m. in Sariasi and Ichinose. 2. Most optimum dipping depth of cutting was 2.5 cm from the base of cutting 3. Rooting occurred most readily during December and March. 4. Perlite was the best medium to produce hardwood cuttings in the bottom heat bins.
