한반도 중부지방의 벼 뿌리로부터 내생 세균의 분리와 특성 분석 (Isolation and Characterization of Endophytic Bacteria from Rice Root Cultivated in Korea)

  • 한반도 중부지방인 충청남북도 7개 지역에서 재배되고 있는 벼 시료 21점을 채집하여 이들의 뿌리를 표면살균 한 후 내생균을 44주 분리하고 내생성 검정 시스템을 통해 정착력이 상대적으로 우수한 균주를 최종 16주 확보 하였다. 이들의 분리빈도는 뿌리 생체중 1g당 $10^{3-5}$ CFU로 나타났다. 흥미롭게도 이중 7주가 Eurkholderia 속 균으로 동정되어 기존의 다른 벼 내생세균 연구 결과와는 다른 특징을 보였다. 또한 GFP tagging 방법을 이용하여 분리균주 중 하나인 Enterobacter sp. KJ001 균주에 대해 뿌리조직 내 colonization 위치를 확인해본 결과 뿌리 조직 중 관다발 주변에 군락을 이루고 있음을 관찰하였다. Burkholderia 분리주들은 국내 재배 벼에서 높은 빈도로 분리되며 in vitro상에서 광범위한 진균성 식물병원균에 대해 우수한 길항력과 더불어 대부분 질소고정 관련 유전자인 nifH를 가지는 점으로 보아 질소고정에 의해 식물생육에 도움을 줄 수 있을 것으로 예측되며 실제로 오이 유묘의 생장을 30% 이상 촉진하는 효과를 보여 식물병 억제 및 감소와 더불어 작물의 생장 촉진 및 생산성 증대에 활용가치가 높은 내생균으로 사료된다.

비닐하우스 고추재배지의 토양과 근계로부터 분리된 형광성 Pseudomonads의 계통 분류 및 다양성 (Diversity and Phylogenetic Analysis of Fluorescent Pseudomonads Isolated from Soil-Root System of Red Pepper in Greenhouse)

  • 고추 시설 재배지 토양, 근권과 근면등으로부터 형광성 pseudomonads를 분리한 후 이들 중 35균주에 대해 계통 분석을 실시하였다. 계통 분석에 이용된 분류키는 165 rDNA의 일부 염기서열로, 종 수준에서 분류동정을 실시하였다. 고추재배지로부터 분리된 형광성 pseudomonads중 P. putida group으로 분류된 균주는 17균주 였으며, 이들은 주로 비근권 토양으로부터 분리되었다. 이들은 4개의 소그룹 (subgroup I, II, III, IV)으로 분류되었으며, 소그룹 I, IV에 속한 균주는 P putida의 표준균주가 속한 소그룹 II, III의 균주와는 분류학적으로 뚜렸히 구분되는 독특한 균주들이었다. P. fluorescens로 분류된 균주는 15균주였으며, 나머지 3균주는 P. fluorescens와 P. chloraphis의 중간 그룹으로 분류되었다. 근권으로부터 분리된 균주는 대부분 P. fluorescns로 분류되었으며, 이들의 유전적 유연관계는 매우 높게 나타났다. 본보의 결과에 비추어 볼 때, 고추의 근계는 P. fluorescens의 특정 균주에 의해 정착되는 것으로 생각되었다.

Phytophthora drechsleri에 의한 곰취 역병 (Phytophthora Root Rot of Ligularia fishcheri Caused by P. drechsleri)

  • Cultivation of a native wild vegetable, Ligularia fischeri, is hampered by Phytophthora sp. Infected plants showed wilt and eventual death due to rots on the root and basal portion. Eight isolates collected were all identified as P. drechsleri based on their mycological characteristics. The fungi showed relatively strong pathogenicity to L. Fischeri, mild to Aster scaber and Codonoposis lanceolata, and none to Circium setidens and Pimpinella brachycarpa. This is the first report of Phytophthora root rot on wild vegetables in Korea.

Diversity of Root-Associated Paenibacillus spp. in Winter Crops from the Southern Part of Korea

  • The genus Paenibacillus is a new group of bacilli separated from the genus Bacillus, and most of species have been isolated from soil. In the present study, we collected 450 spore-forming bacilli from the roots of winter crops, such as barley, wheat, onion, green onion, and Chinese cabbage, which were cultivated in the southern part of Korea. Among these 450 isolates, 104 Paenibacillus-like isolates were selected, based on their colony shape, odor, color, and endospore morphology, and 41 isolates were then finally identified as Paenibacillus spp. by 16S rDNA sequencing. Among the 41 Paenibacillus isolates, 23 were classified as P. polymyxa, a type species of the genus Paenibacillus, based on comparison of the 16S rDNA sequences with those of 32 type strains of the genus Paenibacillus from the GenBank database. Thirty-five isolates among the 41 Paenibacillus isolates exhibited antagonistic activity towards plant fungal and bacterial pathogens, whereas 24 isolates had a significant growth-enhancing effect on cucumber seedlings, when applied to the seeds. An assessment of the root-colonization capacity under gnotobiotic conditions revealed that all 41 isolates were able to colonize cucumber roots without any significant difference. Twenty-one of the Paenibacillus isolates were shown to contain the nifH gene, which is an indicator of $N_{2}$ fixation. However, the other 20 isolates, including the reference strain E681, did not incorporate the nifH gene. To investigate the diversity of the isolates, a BOX-PCR was performed, and the resulting electrophoresis patterns allowed the 41 Paenibacillus isolates to be divided into three groups (Groups A, B, and C). One group included Paenibacillus strains isolated mainly from barley or wheat, whereas the other two groups contained strains isolated from diverse plant samples. Accordingly, the present results showed that the Paenibacillus isolates collected from the rhizosphere of winter crops were diverse in their biological and genetic characteristics, and they are good candidates for further application studies.

한국에 있어서의 'Kresek'에 관한 연구 III. 품종저항성 및 균주에 따른 도체내에서의 균증식과 발병과의 관계 ('Kresek' Disease in Korea III. Varietal Resistance, and Relationship among Pathotype, Multiplication in the Tissue and Disease Development)

  • 수도 흰빛마름병 이병엽과 Kresek 발병주로부터 분리한 I균군(각5균주), II균군(각4균주)의 18균주를 "밀양23호"및 "유신" 품종 5엽기의 전개엽에 침접종한후 수도체내에서의 균증식과 병반장율 및 Kresek 발병율을 비교조사하여 균주간차이를 비교하였고, 밀양23호외28품종에 대해 II, IV균군을 사용하여 침접종및 근단접종법으로 품종간 저항성을 조사한 결과 1. 각균주간 수도체내에서의 증식은 큰 차이를 볼수 없었으나 "Kresek"발병주로부터 분리된 균주가 병반장률이 약간 높은 경향을 나타내었다. 2. Kresek 발병은 흰빛잎마름병 이병주에서 분리한 균주와 "Kresek"발병주로부터 분리한 균주간에는 아무런차이를 인정할수 없었고, 모두 발병을 볼수있었다. 3. Kresek에 대한 품종저항성에서는 흰빛잎마름병의 저항성반응과 비슷한경향을 나타내고 있으나 Wase Aikoku 품종군에서도 발병을 볼수있었다. 4. 황옥품종군인 "IR20"은 Kresek에라해 고도의 저항성반응을 나타내었다. 5. 접종반응으로는 근단접종법에서 침접종법에 비해 더욱 많은 발병율을 볼수있었다.

MicroTom - A Model Plant System to Study Bacterial Wilt by Ralstonia solanacearum

  • MicroTom is a miniature tomato plants with various properties that make it as a model system for experiments in plant molecular biology. To extend its utility as a model plant to study a plant - bacterial wilt system, we investigated the potential of the MicroTom as a host plant of bacterial wilt caused by Ralstonia solanacearum. We compared the disease progress on standard tomato and MicroTom by two inoculation methods, root dipping and soil drenching, using a race 1 strain GMI1000. Both methods caused the severe wilting on MicroTom comparable to commercial tomato plant, although initial disease development was faster in root dipping. From the diseased MicroTom plants, the same bacteria were successfully reisolated using semiselective media to fulfill Koch's postulates. Race specific and isolate specific virulence were investigated by root dipping with 10 isolates of R. solanacearum isolated from tomato and potato plants. All of the tested isolates caused the typical wilt symptom on MicroTom. Disease severities by isolates of race 3 was below 50 % until 15 days after inoculation, while those by isolates of race 1 reached over 50% to death until 15 days. This result suggested that MicroTom can be a model host plant to study R. solanacearum - plant interaction.

충북지방의 뿌리혹병 감염 포도나무 뿌리에서 분리한 Agrobacterium속 균의 특성 (Characterization of Agrobacterium spp. Isolated from Roots of the Crown Gall-infected Grapevine in Chungbuk)

  • 최근 혹병이 심하게 발생한 충북지방의 포도나무의 뿌리로부터 선택배지를 이용하여 Agrobacterium속 세균을 분리하고 동정하였다. 분리균의 지방산 분석을 통한 MIDI에 의한 동정, 16S rDNA 염기서열, 생화학적 특성, 종 특이적 primer을 이용한 PCR 결과로 13개 분리균은 모두 A. tumefaciens로 동정 되었으며, 포도나무 혹병균인 A. vitis는 분리되지 않았다. 모든 분리균은 토마토와 포도나무의 줄기와 뿌리에 병원성을 나타내지 않았다. Rep-PCR 결과 분리균은 A. tumefaciens와 일부 유사성이 있지만 전체적으로 병원균인 A. tumefaciens와 A. vitis와는 유사성이 낮았다. 이상의 결과는 충북 일부 지방의 MBA와 거봉에서 발생한 혹병을 일으키는 병원균은 토양이나 뿌리로부터 전반되지 않고 묘목을 통해서 전반되었을 가능성을 시사하고 있다.

Occurrence of Dry Rot on Cymbidium Orchids Caused by Fusarium spry. in Korea

  • Cymbidium orchids with blight and rot symptoms were collected, and a total of 63 isolates of Fusarium app. was obtained from pseudobulbs, roots, and leaves of the diseased plants. The isolates were identified based on their morphological characteristics. Out of the 63 isolates of Fusatium sup., 51 isolates were identified as F. oxysporum, 10 isolates as F. solani, and the rest as F. proliferatum. F. oxysporum was isolated from all the Cymbidium spp., while F. solani and F. proliferatum were isolated only from Cymbidium ensifolium and C. ginatum, respectively. Isolates of the three Fusarium spp. were tested for pathogenicity to their hosts by artificial inoculation. The strongly pathogenic isolates of Fusarium spp. induced severe dry rot of pseudobulbs and roots of the host plants. The symptoms progressed up to the basal part of the leaves, which later caused blight of the entire plant. The dry root symptoms induced on the plants by artificial inoculation with the isolates of Fusarium app. were similar to those observed in the growers'greenhouses. This is the first report of dry rot of Cymbidium spp. caused by F. oxysporum, F. solani, and F. proliferatum in Korea.

Fusarium oxysporum Protects Douglas-fir (Pseudotsuga menziesii) Seedlings from Root Disease Caused by Fusarium commune

  • Fusarium root disease can be a serious problem in forest and conservation nurseries in the western United States. Fusarium inoculum is commonly found in most container and bareroot nurseries on healthy and diseased seedlings, in nursery soils, and on conifer seeds. Fusarium spp. within the F. oxysporum species complex have been recognized as pathogens for more than a century, but attempts to distinguish virulence by correlating morphological characteristics with results of pathogenicity tests were unsuccessful. Recent molecular characterization and pathogenicity tests, however, revealed that selected isolates of F. oxysporum are benign on Douglas-fir (Pseudotsuga menziesii) seedlings. Other morphologically indistinguishable isolates, which can be virulent, were identified as F. commune, a recently described species. In a replicated greenhouse study, inoculating Douglas-fir seedlings with one isolate of F. oxysporum prevented expression of disease caused by a virulent isolate of F. commune. Moreover, seedling survival and growth was unaffected by the presence of the F. oxysporum isolate, and this isolate yielded better biological control than a commercial formulation of Bacillus subtilis. These results demonstrate that an isolate of nonpathogenic F. oxysporum can effectively reduce Fusarium root disease of Douglas-fir caused by F. commune under nursery settings, and this biological control approach has potential for further development.

Occurrence of Clubroot Caused by Plasmodiophora brassicae in Baecheongchae

  • Clubroot symptoms were frequently observed on the roots of Baecheongchae plants grown in vinyl greenhouses of a farmer located in Yangpyeong area of Korea during a disease survey in June 2019. The incidence of diseased Baecheongchae plants ranged from 30 to 90% in the vinyl greenhouses investigated. Many resting spores were found in the tissue of root galls collected. The resting spores were hyaline and spherical and measured 2.5-4.2 ㎛ in diameter. Three inoculum suspensions of resting spores prepared from the root galls were inoculated to the roots of healthy Baecheongchae plants. All the inoculum suspensions caused clubroot symptoms to appear on the roots of the inoculated Baecheongchae plants. The symptoms on the roots induced by artificial inoculation were similar to those observed in the plants of the vinyl greenhouses during the disease survey. Resting spores of the pathogen were recovered from the root galls of the inoculated plants. Three root gall isolates obtained from the inoculated plants were used for molecular identification. Comparing the isolates to the Plasmodiophora brassicae strains in GenBank, the amplification products demonstrated 100% similarity with the internal transcribed spacer (ITS2) sequences. The clubroot pathogen was identified as P. brassicae according to its morphological, pathological, and molecular characteristics. This is the first report of P. brassicae causing clubroot in Baecheongchae.
