Effect of Medium Composition on in vitro Plant Root Regeneration from Axillary Buds of Cassava (Manihot esculenta Crantz) (카사바 액아배양 시 배지조성이 기내 식물체 발근에 미치는 영향)

  • The Cassava (Manihot esculenta Crantz) is one of the major food crops in the tropical or subtropical regions. Recently, clean planting materials of improved cassava cultivars are in high demand. Problems in the propagation of cassava are virus vulnerable and low rates of seed germination. Thus, the study was undertaken to develop an efficient in vitro mass propagation protocol of Manihot esculenta Crantz. So we tried to optimize protocols for mass production from axillary buds of Cassava. Young and actively growing stem segments were excised from adult plants of cassava. Samples were cut into a 3~4 cm nodal segments with axillary buds, and cultivated in the different medium supplemented with various plant growth regulators for 4 weeks. For shoot multiplication, axillary buds approximately 1 cm in length were taken from in vitro derived shoots and subcultured. After 4~6 weeks, the shoot generation rate showed 55.6%. The shoot number and its length was 1.0/explant and 2.3 cm in the most favorable medium composition. The auxin β-indolebutyric acid(IBA) 0~2.0 mg/L was proved to be effective on root development. Plantlets with fibrous roots easily generated tuberous roots in vitro. The tuberous roots were induced only when both kinetin and IBA were used in combination. after 8 weeks, the root generation rate showed 100%. The root number and its length was 17.2/explant and 2.2 cm in the most promising medium composition. Our experiments confirmed that in vitro growth and multiplication of plantlets could depend on its reaction to the different medium composition, and this micropropagation techniques could be a useful system for healthy and vigorous plant production.

Changes in the Growth of Young Rice Seedlings in the Root Extension Stage under Different Growth Conditions (벼 착근기 생육 환경에 따른 어린모의 생육변화 분석)

  • Root extension is the most important growth change that occurs during cultivation. We analyzed growth changes according to young seedling age, temperature, and the degree of root cutting in order to identify factors affecting rooting after transplanting. Root cutting did not affect plant height growth rate, root growth rate was increased in plants that experienced root cutting, and 14-day-old seedlings exhibited a higher growth rate than 7-day-old seedlings. Growth temperature experiments revealed that elongation was high at 25℃ and 28℃, but tended to be low at 18℃, and root elongation was high at 22-28℃ for 7-day-old seedlings and 22-25℃ for 14-day-old seedlings. Nitrogen absorption decreased in the following growth temperature order: 25, 28, 22, 18℃, and differences in nitrogen absorption under different growth temperatures tended to be lower in 7-day-old seedlings. The amount of nitrogen taken up by roots did not differ significantly between the short root treatment and the control, and 7-day-old seedlings tended to start nitrogen absorption faster than 14-day-old seedlings. Root vitality was highest in short-rooted 7-day-old seedlings with 3 cm of root remaining, and vitality also tended to be high in short-rooted 14-day-old seedlings.

The Effect of Activated Ion Calcium for Production of Panax ginseng Seedlings in Paddy Field (논 인삼 우량묘 생산을 위한 활성이온칼슘 처리효과)

  • When ginseng seedlings are cultured in paddy fields, quality degradation and yield reduction are induced by severe plant loss with chlorosis on leaves occurred physiological disorder by excessive salt and poor drainage, rusty-root occurrence, and root rot etc. Accordingly, in order to solve these problems, this study was performed to investigate the treatment method, concentrations and time of activated ion calcium as environment-friendly agricultural materials. Activated ion calcium is an enriched and purified water-soluble mineral calcium component for absorbing quickly into plant as a highly functional calcium and it is an alkaline calcium of 37% (370 $m{\ell}$/1 ${\ell}$) concentration with pH 13. Treatment method was that ginseng seeds were sown after removing water in the shade after seed immersion for 1 minute with active ion calcium of 20-fold diluted solution, and then irrigated $4{\ell}$ per 3.3 $m^2$ with 200-fold, 400-fold, and 600-fold diluted solution before emergence on late March, and supplied 1 time on leaves with 500-fold diluted solution in June and July respectively. The disease rate by treatment of activated ion calcium was that on the treatment of soil irrigated with 200-fold diluted solution compared to non-treated soil, damping-off was 33%, anthracnose was 100% reduced and the occurrence rate of rusty-root was 30% reduced. In addition, when active ion calcium of 200-fold diluted solution were soil irrigated, first and second grade ginseng were respectively 26% and 22% produced more, compared with control.

Effects of Non-drainage Hydroponic Culture on Growth, Yield, Quality and Root Environments of Muskmelon (Cucumis melo L.) (멜론 수경재배 시 배액제로화가 근권환경 및 수량에 미치는 영향)

  • This study was conducted to figure out the possibility of non-drainage in muskmelon (Cucumis melo L.) hydroponics culture. Plants were grown under 3 different levels of drainage, standard (20~40%, SD), minimum (5~10%, MD), and non-drainage (ND). Throughout cultivation periods, constant water content and electrolyte conductivity changes in root zone were observed in SD in the range of 60~70% and $1.5{\sim}2.5dS{\cdot}m^{-1}$, respectively. ND treatment caused the fluctuation in water content and electrolyte conductivity of root zone and its change ranges were 30~50% in water content and $2{\sim}6dS{\cdot}m^{-1}$ in electrolyte conductivity, but ND treatment did not decrease fruit quality. Even if fruit fresh weight was slightly lower in ND with 1,863 g, than in SD with 1,990 g, the fruit weight in ND meets standard market size, 1,800~2,000 g. Higher soluble solids content was observed in fruit in ND than in SD and MD. Total amount of drainage per plant was 27,718, 15,769 and 2,346 mL in SD, MD and ND, respectively. SD showed $83.2m^3$ drainage, 34.5% drainage of irrigation amount whereas required total irrigation amount in ND was very low with $7m^3$.

Effects of Incorporation of Green Manure Crops on Growth and Quality in Cynanchum wilfordii Hemsley (녹비작물 토양환원이 백수오 생육 및 품질에 미치는 영향)

  • Background: The study aimed to obtain data on the effects of cultivation and soil reduction of green manure crop on the quantity and quality of organically cultivated Cynanchum wilfordii Hemsley. Methods and Results: The experiment comprised four treatments: control, hairy vetch, barley, and hairy vetch + barley (3 : 2). The plant height in the hairy vetch treatment (86.3 cm) was significantly different from that in the other treatments, whereas the stem diameter leaf area, and special product analysis division (SPAD) value did not differ across the treatments. The largest soil reduction of green manure crop was recorded in the barley treatment (440 kg/10 a), whereas the smallest was recorded in the single treatment with hairy vetch (80 kg/10 a). The hairy vetch + barley (60 : 40) treatment showed 63% more soil microorganisms than control. Radical scavenging activity estimation revealed that the total polyphenol content was highest (1,740 mg/kg), and the 2,2-diphenyl-1-picrylhydrazyl (DPPH) was 92.6% in the barley treatment. The 2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) activation was highest in the control (51.1%), and the root yield was the highest in the barley treatment (310 kg/10 a). Conclusions: The root yield, total polyphenol content, and antioxidant activity of Cynanchum wilfordii (Maxim.) Hemsley increased in presence of the green manure crop barley.

Cytotoxicity of Methanol Extracts from Cudrania tricuspidata Bureau (꾸지뽕나무 메탄올 추출물의 세포독성)

  • In order to develop as a natural source of anticancer materials of Cudrania tricuspidata, the cytotoxicity of methanol extracts by harvesting parts and times against 8 cell lines including 293 (normal kidney cells) and A-431 (epidermoid carcinoma cells) were investigated using MTT assay. All harvesting parts had hardly cytotoxicity against 293. And methanol extracts of stem bark and root bark showed very high cytotoxicities against 7 cancer cell lines. The cytotoxicity was the highest against HeLa (cervix adenocarcinoma cells) and followed by MCF-7 (breast adenocarcinoma cells), AGS (stomach adenocarcinoma cells), HT-29 (colon adenocarcinoma cells), HepG2 (hepatoblastoma cells), A549 (lung carcinoma cells) and A-431. By the way, leaf extract had a cytotoxicity against only AGS and ripe fruit extract had no cytotoxicity. Among harvesting times, the cytotoxicity of root bark were high from April to September but that of stem bark showed a little difference. These results showed that anticancer activities of Cudrania tricuspidata extracts were eventful changes by harvesting parts and times.

Inhibition Effect of Achyranthes japonica N. Root Extract on Cathepsin B (우슬뿌리 추출물의 Cathepsin B에 대한 저해효과)

  • This study was carried out to investigate the cathepsin B inhibition effect by Achyranthes japonica N. root extract in vitro. The methanol/$H_{2}O$(4:1, v/v) extract was fractionated by ethyl acetate(F1), chloroform(F2), chloroform/methanol(3:1, v/v)(F3) and methanol(F4). The yield of F4 in Achyranthes japonica N. root was $8.27\%$. As an index material of Achyranthes japonica N. root, 20-hydroxy ecdysone was detected by TLC, and HPLC and it's content was $0.33\%$. Three isolates(F1, F3, F4) showed the cathepsin B inhibition activity, and F4 showed the highest inhibition activity among them. In the inhibition activity on cathepsin B, leupeptin, 20-hydroxy ecdysone and F4(at the same concentration of 20-hydroxy ecdysone.) were 92, 88 and $97\%$ on BANA($N{\alpha}$-benzoyl-DL-arginine ${\beta}$-naphthylamide) substrate, and 62, 36 and $67\%$ on CLN($N{\alpha}$-CBZ(carbobenzlyoxy)-L-lysine p-nitrophenyl ester HCI) substrate, respectively.

Effect of Grafting Methods on Plug Seedling Quality, Growth after Transplanting and Yield of Oriental Melon (Cucumis melo L. var. makuwa Mak.) (참외 접목방법이 플러그 묘 소질과 정식 후 생육 및 수량에 미치는 영향)

  • This study was conducted to investigate the effect of grafting methods on plug seedling quality, growth after transplanting, and yield of oriental melon that those seedlings were raised in the 32 hole plug tray for protected cultivation. Number of leaf and leaf area in approach grafting were higher until 15 days after grafting but lower on 20 days after grafting. Plant height, stem diameter and content of chlorophyll were nearly the same of each grafting methods on 20 days after grafting. Growth of top part, that is, plant height, leaf number, and leaf area on root removed single cotyledon ordinary splice grafting was the highest among the grafting methods. In the underground part, dry weight of root was high and T/R ratio was the lowest. Seedling quality of root removed single cotyledon ordinary splice grafting was the highest among grafting methods before transplanting. Plant height, number of leaf, leaf area and dry weight of root removed single cotyledon ordinary splice grafting in the growth of field were the highest on 30 days after grafting among grafting methods and days required for first flowering was also shorter, 38.4 days among grafting methods. But Fruit weight, content of soluble solids, fruit hardness, and color characteristics in fruit quality were insignificant among each grafting methods. Consequently, grafting methods influenced on the seedling quality, of oriental melon early growth after transplanting and yields. It was concluded that root removed single cotyledon ordinary splice grafting was the best methods in present study. It will be needed to convert grafting methods. Thus new grafting method should be applied.
