Formation of Genetic Tumor and Characteristics of Teratoma Shoot from Tobacco Interspecific Reciprocal Hybrids (연초종간 상호교잡에 의한 Genetic Tumor의 유도 및 Teratoma Shoot의 특성)

  • Reciprocal interspecific hybrids between N. glauca(2n=24) and N. langsdorffii(2n=18) were obtained by intercrossing. One hundred percent of F$_1$ seeds was produced from intercrossing of N. glauca $\times$ N. langsdorffii, whereas the frequency of F$_1$ hybrid seed formation from N. langsdorffii $\times$ N. glauca was very low. However, all the hybrid seeds were germinated well and then grown to normal plantlets. All the plants of F$_1$ hybrids have chromosome number of interspecific hybrids (2n=21). From observation of morphological characteristic, the structure of petrol, leaf, flower, and the morphology of pollen have characteristics of F1 hybrid. Spontaneous tumors (genetic tumor) were formed from each F$_1$ hybrid; the genetic tumor arose at the reproductive phase when the maternal type of F$_1$ hybrid came from N. glauca, while the genetic tumor arose only after reproductive phase when the maternal type of F$_1$ hybrid came from N. langsdorffii. The genetic tumor actively proliferated on hormone-free medium and produced numerous teratoma shoots. In addition, normal leaf or stem explants of F$_1$ hybrid produced calli on hormone-free medium after 15 days of culture, the calli produced new numerous teratoma shoots after 30 days. The frequency of teratoma shoot formation from rnterspecific hybrid was higher in the N. glauca $\times$ N. langsdorffii than in the N. langsdorffii $\times$ N. glauca. Root development from the teratoma shoots was hardly obtained. Teratoma shoots without roots in vitro can form genetic tumor at the vegetative growth phase after tissue culture.

Suppressive Effect of Organic Farming Materials on the Development of Tomato Gray Mold (토마토 잿빛곰팡이병에 대한 유기농업자재의 억제효과)

  • Botrytis cinerea infects stems, leaves and fruits of greenhouse tomato and can cause serious economic losses. This study was conducted to develop organic farming control method against tomato gray mold. Twenty two organic farming materials including mineral and plant extracts were screened for the suppressive activity against Botrytis cinerea, in vitro and in vivo. Among the organic farming materials, sulfur, copper, Chinese twinleaf extract and rhubarb extract decreased by 51.7-90% of the spore germination of Botrytis cinerea. Also, gray mold incidence was reduced more than 90% on tomato stems by treating sulfur, seaweed extracts, rhubarb root extracts and Chinese twinleaf extract. After the selected four organic farming materials were applied on tomato cultivated in greenhouse, their control effects against the tomato gray mold were tested. When the water soluble sulfur was foliar-sprayed on the tomato leaves infected by artificial inoculation with spore suspension of Botrytis cinerea, it showed 87.9% of control value. Also, control activity of the water soluble sulfur was paralleled with chemical fungicide, diethofencarb+carbendazim. The above mentioned results indicate the sulfur formulation can be used as chemical fungicide alternatives for controlling tomato gray mold in the greenhouse.

Geographical Distribution and Ecology in Microhabitats of the Rare Species, Pulsatilla tongkangensis Y.N. Lee et T.C. Lee (희귀종인 동강할미꽃(Pulsatilla tongkangensis Y.N. Lee et T.C. Lee)의 지리적 분포와 미소생육지의 생태)

  • A perennial plant species of Ranunculaceae, Pulsatilla tongkangensis is a very rare species only found in limestone vegetation of South Korea. It is not only rare but also has great value as a horticultural resource, thus resulting in serious problems of overexploitation and habitat disturbance. For the conservation of the rare species and its development as a resource, the study was conducted with the following three aims: first, to investigate any new geographical distribution of the species; second, to identify the site condition and vegetation structure of its habitat; and third, to compare the ecology of P. tongkangensis in other microhabitats such as rock, slope and ridge area. A new distribution was found in limestone vegetation at Danyang-gun, Chungcheongbuk-do. It was, therefore, confirmed that the distribution of P. tongkangensis exactly coincides with the area of distribution of limestone, indicating a possibility that the species might be a calcicole. 87% of the microhabitats found in rocks. Compared to P. tongkangensis, at ridges or slopes, number of P. tongkangensis plants were few in rocks. It is possibly due to the lack of microhabitats that could settle in rocky areas. Once established on rocks, however, plants grew large enough to flower with higher number of leaves and flowers per plant, resulting in higher flowering rate on rocky areas. Cracks in/between rocks are likely to be safe places for the plants with favorable light conditions, abundant fine particles to root down and reduced competition for resources. Yet, further research is necessary in terms of determining whether the species is indeed a calcicole, whether other limiting factors work in its establishment stage, and genetic diversity exists in the population.

Appropriate Each Irrigation Quantity in Irrigation System Controlled by Drainage Level Sensor for Perlite Bag Culture of Tomato (배액전극제어법을 이용한 토마토 펄라이트 자루재배시 일회급액량 구명)

  • This research was conducted to investigate the effects of irrigation quantity in irrigation management system controlled by drainage level sensors for perlite bag culture on the growth and yields of tomatoes during different growth stages. Tomato plants were irrigated with four selected methods; supplying small quantity (~70 mL) during entire growth (S-S), large quantity (~145 mL) during entire growth (L-L), small quantity before harvesting the first cluster fruits and large quantity after harvesting (S-L), and large quantity until harvesting the first cluster fruits and small quantity after harvesting (L-S). The irrigation quantity supplied in each time was gradually adjusted along with the ratios as the tomato crop grew during different growth stages. The growth of the tomato plants was unstable and slow during the entire cropping period when the plant was irrigated by small or large quantities (S-S or L-L). In L-S treatment, the growth phase of the tomato was changed from vegetative to generative growth on the basis of the plant development index when each irrigation quantity was changed. The L-S treatment exhibited the largest root volume and yields with stable drainage ratios. Therefore, the optimum irrigation quantity was determined as 145 mL before harvesting the first cluster fruits and 70 mL after harvesting.

Distribution and Diversity of Saprophytic, Mycorrhizal and Parasitic Higher Fungi in Kwangnung Experimental Forest in Korea (광릉시험림(光陵試驗林)의 부생성(腐生性), 균근성(菌根性) 및 기생성(寄生性) 고등균류(高等菌類)의 분포(分布)와 다양성(多樣性)에 관(關)한 연구(硏究))

  • Higher fungi (Basidiomycetes) were collected from forest stands of Kwangnung Experimental Forest, Kwangnung, Kyonggido, during the summer and fall seasons of 1976, 1984, 1985 and 1986. A total of 257 species and varieties in 104 genera were identified. Saprophytic fungi accounted for 135 species in 75 genera while mycorrhizal fungi accounted for 120 species in 28 genera. Two parasitic species were collected and they were root parasites. Among the mycorrhizal species Amanita, Russula, Lactarius, and species in the Boletaceae were the dominant taxa. The mycorrhizal fungi appear to be an important component in the essential elements needed for forest management in Korea. A reevaluation of the higher fungi previously described in Korea was made and we report 77 higher fungi previously undescribed from Korea. This is from a single experimental forest and suggests that there are many unreported higher fungi which play an important role in Korean forests.

Characteristics of Seed Germination and Seedling Growth of Native Hydrangea serrata for. acuminata (자생 산수국의 종자 발아와 유묘 생육 특성)

  • This work aims to obtain basic information for seed propagation of Hydrangea serrata for. acuminata. The germination percentage of the seeds taken on 15 November, 30 December, and 23 January was $90.0{\pm}4.16%$, $84.4{\pm}5.52%$, and $88.9{\pm}2.40%$, respectively. This suggest that seeds of Hydrangea serrata for. acuminata are non-dormant seeds. The optimum temperature for germination was $25^{\circ}C$ and light was necessary. Most of the growth parameters (shoot and leaf length, stem diameter, root length, no. of roots, T/R ratio, and fresh and dry wts.) were significantly greater at $25/20^{\circ}C$ and $25^{\circ}C$ than at the other temperatures. Low T/R ratio at relatively cool temperatures (15 and $20^{\circ}C$) was caused by suppressed top growth. In light quality treatment, red light (RL) significantly enhanced stem elongation. The greatest photosynthetic pigments (total chl, chl a/b, and carotenoid) were observed in seedlings grown in blue light (BL), followed by seedlings grown in RL+BL. When blue light was added, higher pigment contents were found. Effect of plug cell size (50, 72, 128, 162 and 200 cells) on the growth of seedlings was investigated. The highest top growth was observed in seedlings grown in 50 cell trays, followed by seedlings grown in 72, 128, 162, and 200 cell trays. However, there was no significant differences between 162 and 200 cell trays. Especially, smaller size leaves were observed in seedlings grown in smaller cell trays (lower volume and high plant density).

  • The landing gear is a component that requires a high degree of safety to protect the lives of rotary-wing aircraft and boarding personnel, absorbing the impact on transfer/landing and supporting the fuselage during taxiing and mooring on the ground. In particular, the wheel landing gear supporting the aircraft fuselage absorbs most of the shock from the ground through the shock absorber and tires. This ensures the safety of the pilot on board the aircraft and satisfies the operational capability of the soldiers between missions. During the operation of a rotary-wing aircraft, a number of piston pins, which are a component of the right main wheel landing gear, were found to be broken. Therefore, this study examined the root cause of the piston pin crack phenomenon found in the main wheel landing gear. For this purpose, various causes were identified from fracture surface analysis of a flight test. In particular, the possibility of cracking was analyzed based on the influence on the fastening torque with the drag beam component applied to the piston pin at the time of development. This ensures the fatigue life and structural integrity.

Investigation of Rhizome Enlargement Stage and Harvest Time in Rehmannia glutinosa (Gaertn.) Libosch. ex Steud. (지황의 비대시기와 수확시기 구명 연구)

  • Background: There have been no studies to date on rhizome development and optimal harvest timing for Rehmannia glutinosa. We therefore, undertook this investigation. Methods and Results: R. glutinosa 'Jihwang 1' was sown in early May and harvested in early November. Growth investigations were carried out at intervals of 10 days between 90 and 180 days after sowing (DAS). Leaf length, leaf width, and number of leaves increased until 150 DAS but decreased after 160 DAS. Rhizome length increased until 120 DAS subsequently, rhizome diameter increased rapidly between 130 and 150 DAS. Thus, the key period for rhizome enlargement in R. glutinosa is thought to be 130 to 150 DAS. Fresh root yield increased sharply from 916 kg/10a to 1,914 kg/10a between 4 and 5 months after sowing (MAS). Dry matter ratio increased gradually from 19.2% at 4 MAS to 24.4% at 6 MAS. Finally, the level of catalpol, a key active ingredient, increased sharply from 0.41% to 4.21% between 5 and 6 MAS. Given the dry matter ratio, catalpol content and yield measured, we suggest that optimal R. glutinosa harvest time is 6 MAS. Conclusions: Based on our results, the key period for rhizome enlargement is 130 to 150 DAS and optimal harvest timing is 6 MAS. We anticipate that these and other results of this study can be used to inform cultivation of R. glutinosa.

Physicochemical and Sensory Characteristics of Moju Sold at Restaurants Located in Jeonju (전주지역 음식점에서 판매되는 모주의 이화학적, 관능적 특성)

  • Moju is a low-alcoholic beverage made by filtering after boiling a mixture of makgeolli, ginseng, arrowroot, licorice root, cinnamon, jujube, ginger, and raw sugar. It is known to alleviate hangovers. To provide information for use in the development of palatable Moju, this study evaluated the physicochemical and sensory properties of 22 kinds of Moju being sold at different restaurants in Jeonju city. The mean values of obtained in the physicochemical analysis were as follows: water content, 91.28%; alcohol content, 1.09%; pH, 4.25; total acidity, 0.27%; reducing sugar content, 40.68 mg/mL; soluble solid content, 13.75 $^{\circ}Brix$; and viscosity 11.19 cP. The Lactic-, malic-, and citric- acid contents were higher than the contents of other organic acids. The sucrose content was higher than the contents of other free sugars. The mean value of the free amino acids was 175.3 mg%, lower than that of Takju, the main ingredient of Moju. It was considered that the free amino acids in Takju can be used as a substrate for the browning reaction in the process of Moju manufacture. In the sensory evaluation, the Moju with 0.15~0.25% total acidity, 10.6~13.4 $^{\circ}Brix$, and 5.73~9.57 cP was preferred.

Effect of Organic Fertilizer Application on the Growth and Yield of Leaf Lettuce and Garland Crysanthemum (유기질비료(有機質肥料)(Bio-com)시용(施用)이 상치와 쑥갓의 생육(生育) 및 수량(收量)에 미치는 영향)

  • Effect of microbiological product manure (Bio-com) on the growth and yield of vegetables and on physico-chemical properties of soil. Leaf lettuce and Garland Chrysanthemum were taken for test plants. A Jungdong (Series) silty loam soil was used for pot experiments. The results are summarized as following: 1. Treatment with Bio-com stimulated plant growth that were increased in the number of leaf, leaf area, plant height and root length both for Leaf lettuce and Garland Cyrysanthemum. 2. Yield increase of 123-1170% was obtained from Bio-com treatments compared to N.P.K.+manure treatment. Application of 2250 kg/10a of Bio-com gave the highest yield. 3. Fertilizer effect of Bio-com appeared much earlier than that of manure. 4. Plant content of $K_2O$ was highest when treated with Bio-com. Plants absorbed considerably more N, $P_2O_5$ and MgO. 5. Content of N, $P_2O_5$ in Garland Chrysanthemum were higher when treated with Bio-com. 6. Content of $P_2O_5$ in soil was at the highest level. Soil pH, content of exchangeable soil Ca and Mg were also high with Bio-com fertilization.
