This study was aimed for determining the biochemical mechanism of cold tolerance in crops and for searching the biochemical genetic marker related with cold tolerance by the analysis of isozyme pattern. We investigated various biochemical changes induced by the long-term cold acclimation in cold sensitive rape (B. napus) and in cold tolerant 'Sandongchae'(B. campestris) seedlings. The cold shock after long-term cold acclimation to B. napus and B. campestris greatly increased the activities of peroxidase 157% and 50% in root fraction and, 201% and 205% in hypocotyl, respectively. Simultaneously, the activity of superoxide dismutase was largely increased in hypocotyl fraction, too. Protein contents of hypocotyl fractions in B. napus and B. campestris were also increased by 11.4% and 57.8%, respectively. The band of pl 6.4 among peroxidase isozymes newly biosynthesized during long-term cold acclimation was emerged in the hypocotyl fraction of cold tolerant B. campestris as well as in the root of both species. From above and previous results, we presented a model of interconversions of molecular oxygen species due to the cold injury and biochemically inferred the mechanism of cold tolerance in crops.
Soybean plants(Glycine max [L.] merr.) inoculated with Bradyrhizobium japonicum MN110 were grown in growth chambers under 400 or $800{\mu}l{\cdot}l^{-1}$ atmospheric $CO_2$ and harvested at 25, 28, 32, and 35 DAT to examine the effect of $CO_2$ enrichment on phosphorus accumulation, uptake, and utilization efficiency during vegetative growth. Phosphorus concentration in leaf was lower in high $CO_2$ plant by 47% at 25 DAT and 34% at 35 DAT than those in the control plant but phosphorus concentrations in stem, root and nodule were not affected by $CO_2$ enrichment. Total phosphorus accumulation increased 3.9-fold in high $CO_2$ plant and 3.2-fold in the control plant between 25 and 35 DAT. Elevated $CO_2$ caused a decrease in the whole plant phosphorus concentration by 35%, which was due almost entirely to a decrease in the phosphorus concentration of leaves. $CO_2$ enrichment increased phosphorus utilization efficiency in the whole plant by 70% during the experimental period. Plants exposed to high $CO_2$ had larger root systems than under ambient $CO_2$, but high $CO_2$ plants had lower P-uptake efficiency. Averaged over four harvests, plants at high $CO_2$ had 38% larger root mass that was more than offset the 20% lower efficiency of P-uptake and accounted for increased phosphorus accumulation by high $CO_2$ plant. These results suggest that the reduced phosphorus concentration in soybean plant under $CO_2$ enrichment may be an acclimation response to high $CO_2$ concentration or enhanced starch accumulation, resulting in the plants to have a lower phosphorus requirement on a unit dry weight basis or a high phosphorus utilization efficiency under these conditions.

Rehmannia glutinosa(Gaertn.) DC. is a herbaceous perennial plant and belonging to the Scrophulariaceae and used as roots for medicinal part and purpose. R. glutinosa is and usually used for fresh rehmannia root or prepared rehmannia root. However, it is very difficult to propagate using the seeds because of lack germination so it is propagated using the vegetative method as the rootstock. Currently, propagation and harvesting using the rootstock of R. glutinosa has difficulties about production of the high quality and quantity in R. glutinosa because of root rot disease. To optimize in vitro cultures and to improve the rootstock and seedling of R. glutinosa after morphological and genetical determination, 5 plant culture media (MS, DJ, LS, QL, and WPM) were used in this study then WPM was selected for better growth, for multiplication condition(WPM + IAA 1.0 mg/L + IBA 0.5 mg/L), and for root enlargement condition(WPM + NAA 0.1 mg/L) of R. glutinosa. Based on these results, in vitro seedlings of R. glutinosa were transferred to soil for acclimation with environment adaptation and shown the positive effects about root enlargement and root formation. Therefore, it can be used for high quality of R. glutinosa production and production of the rootstock based on propagation using in vitro seedlings of R. glutinosa.
The aim of this study was to establish the condition of regeneration for white balloon flower (Platycodon grandiflorum DC. cv. Jangback) and to manage for the raising of seedling with in vitro regenerated plants. It was examined that 0.5 mg/L of NAA and 1.0 mg/L of BA was the best composition for the callus and shoot induction (up to 600%). NAA was better than IBA for the induction of root and it took 16.9 days for the induction of rooting on the MS soild media containing 0.5 mg/L of NAA and the final rooting ratio was up to 75%. Out of 5 different bed soils purchased from local market, "Tosil" was identified to be the best for the acclimation and growth of in vitro regenerated balloon flower. In detail, on 8 weeks after planting of in vitro regenerated plants in pots containing "Tosil" bed soils, the plant hight was increased up to 2-fold (12.8 cm), 3.5-fold (27) for the number of leaf and 1.5-fold (4.5 cm) for the leaf length when compared to the other four bed soils, respectively. Our preliminary results indicate that it is possible to prevent the occurrence of blue balloon flower in the massive cultivated area of white balloon flower by providing the seedlings raised from in vitro regenerated plants.

We analyzed the growth characteristics of each cell line and acclimating substrate of Larix kaempferi plantlets regenerated from somatic embryos, with the goal of increasing the survival rate during the acclimation phase. Somatic embryos from three embryogenic cell lines (L14-66, L16-18, and L17-B4) were used, and the acclimating substrates were commercial soils for Larix species (Larix-Soil) and horticultural corps (Hort-Soil), Elle-pot, and Peat-plug. The average initial shoot and root length was shortest in L14-66 and longest in L17-B4. The average survival rate by cell line was highest (87.0%) in L17-B4 and lowest (64.3%) for L14-66. Survival rates by substrate were highest in Elle-pot (88.5%) and Peat-plug (88.9%). The initial shoot length of the L14-66 plantlets was highly correlated with survival rates in the Larix-Soil (r = 0.852), Hort-Soil (r = 0.692), and Elle-pot (r = 0.867) substrates, but not in Peat-plug with high total nitrogen content. The initial shoot length of the L17-B4 plantlets was not correlated with the survival rate in any of the substrates. The initial root length of the L14-66 plantlets was highly related to survival rates in the Larix-Soil (r = 0.986), Elle-pot (r = 0.846), and Peat-plug (r = 0.802) substrates, and the survival rate of the plantlets was higher as the initial root length was longer. The initial root length of the L17-B4 plantlets was related to survival rate only in the Larix-Soil (r = 0.896) and Elle-pot (r = 0.696) substrates. In conclusion, to increase the survival rate of plantlets, root length should be considered over shoot length, and it is recommended to use substrates with high nitrogen content, such as Peat-plug, or to add fertilizer, during the acclimating process. In addition, in order to increase the survival rate, lines with high initial growth should be developed.

An osmotin-like protein (CAOSMl) gene was isolated from pepper leaves infected with the avirulent strain Bv5-4a of Xmthomonas campestris pv. vesicatoria. The cDNA encodes a polypeptide of 250 amino acids with a molecular mass of 27, 361 Da. Its amino acid sequence is highly homologous to various osmotin-like proteins from other plant species. The CAOSMl gene expression was organ- and tissue-specifically regulated In pepper plants. The CAOSMl mRNA was intensely localized in the endodermis area of root tissue and in the phloem cells of vascular bundles of red fruit tissue, but not in leaf, stem, and green fruit tissues of healthy pepper plants. Infection by X. c. pv vesintoria, Colletotrichum coccodes, or Phytopkhora capsici iinduced CAOSMl transcription in the leaf or stem tissues. Expression of the CAOSMl gene was somewhat higher in the incompatible than the compatible interactions of pathogens with pepper. The CAOSMl mRNA was prevalently localized in the phloem cells of the vascular bundle of leaf tissues infected by C. coccodes. The CAOSMl gene was activated in leaf tissues by treatment with ethylene, methyl jasmonate, high salinity, cold acclimation and mechanical wounding, but not by abscisic acid (ABA) and drought. These results indicate that the pepper CAOSMl protein functions in response to Pathogens and some abiotic stresses in pepper plants
An efficient regeneration system was developed using leaf, petiole, and internode explants. Highly embryogenic callus was obtained following cultivation on MS basal nutrient supplemented with 2 $mg/{\ell}$ 2,4-D. Globular, heart, torpedo and cotyledon shaped somatic embryo were produced from the surface of embryogenic callus. Direct shoot regeneration without intermediate callus formation has been achieved on MS medium supplemented NAA and BAP. The percentage of response varies with different concentration of auxin and cytokinin treated individually or in combination. The best shoot regeneration response (54.28%) and number of shoot per explant (12.67) were achieved on the medium supplemented with 0.1 $mg/{\ell}$ NAA and 1 $mg/{\ell}$ BAP. The regenerated shoot transformed into young plant when cultured into elongation and root induction medium. More than 90% of in vitro propagated plants could survive when transferred to the greenhouse for acclimation. This optimized regeneration system can be used for rapid shoot proliferation and genetic transformation.
Plant Regeneration via organogenesis from leaf disk of Korean dandelion was investigated. Leaf disk cultured on MS medium with various combinations of BA (0-4 mg/L) and 2,4-D (0-1 mg/L). Shoot regeneration from leaf explant was observed after 3 weeks of culture. The highest shoot regeneration frequency from leaf disk was obtained with 2 mg/L BA. To analyze the effect of leaf age along shoot formation, we measured number of shoots per explant, shooting rate, fresh and dry weight of leaf explant. The highest number of shoots (11.5) per explant were obtained leaf from 7 weeks old plantlets after seed germination. The regenerated shoots were transferred in 1/2 MS medium with 0.5 mg/L NAA for root formation. Regeneratied plantlets thought organogenesis were growing to whole plants in the pots with acclimation.

The experiment was carried out to study the effect of Korean wild ginseng adventitious root supplementation on the laying performance, egg quality, cytokine expression, ginsenoside concentration, and microflora quantity of Institut de selection Animale (ISA) brown laying hens at 24 weeks old. A total of 90 laying hens were subjected to a completely randomized design at three treatments, five repetitions and six laying hens per replicate. The experiments were divided by diets into the basic feed (CON), basic feed + 0.1% wild ginseng (WG1), and basic feed + 0.5% wild ginseng (WG2). The feeding trial was carried out over a duration of 12 weeks after an initial acclimation period of 2 weeks. Feeds and water were administered ad libitum in mash form, and light was available for 16 hours per day. At the end of study, henday egg production (HDEP), average egg weight (AEW), and egg mass (EM) were increased (p <0.05) in WG2 at week 12. Feed conversion ratio (FCR) was decreased (p < 0.05) in WG2 at week 12. The ginsenoside content in egg yolk was increased (p <0.05) in laying hens in the WG2 treatment at week 12. Relative expression of tumor necrosis factor alpha (TNF-α) was reduced (p < 0.05) in the WG supplemented diets at week 12. The fecal microflora quantity of Lactobacillus was increased (p < 0.05) in WG2 at week 8 to week 12, and Escherichia coli (E. coli) was significantly decreased (p < 0.05) in the WG2 at week 12. We concluded that the result observed in the HDEP, AEW, EM and FCR was due to an increase in ginsenoside content, leading to an improvement in the TNF-α, and fecal microflora quantity such as Lactobacillus and E. coli in the WG2 supplemented diets. We therefore recommend the use of WG at application level 0.5% per basal diet for optimum laying performance in layer hens.
