Effect of Polyphenol Oxidase Activity and Total Phenolic Content on Browning and Quality of Dried-Persimmon According to Maturity Degree of Astringent Persimmon

  • Hunter's color value "a" in dried-persimmon of table and full ripe fruit was higher than that in unripe fruit. In case of soluble solid content, full ripe fruit was $50^{\circ}Brix$, the highest degree, while unripe fruit was $40^{\circ}Brix$, the lowest degree. PPO activation of unripe fruit was 4.7, which was higher than table-ripe fruit (0.7) and full ripe (1.0). Polyphenol oxidase activation remained even while drying, but there was no difference in PPO activation degree as drying period increased. Total phenol content of unripe fruit was 101.4, which was higher than table-ripe fruit (57.5) and full ripe fruit (67.4). Total phenol content level increased as drying period increased, which was based on fresh weight. Hardness of unripe and table ripe fruit continued to decrease until three weeks during softening. After that, hardness was high and it started drying. However, in full ripe fruit, hardness increased after two weeks and softening was fast during the drying period, and its weight reduction rate was lower than that of unripe and table ripe fruit.

Differential Induction of PepTLP Expression via Complex Regulatory System against Fungal Infection, Wound, and Jasmonic Acid Treatment during Pre-and Post-Ripening of Nonclimacteric Pepper Fruit

  • Ripe fruit of pepper (Capsicum annuum) showed resistance to Colletotrichum gloeoporioides, but unripe fruit was susceptible. We previously isolated the PepTLP gene that induced in both unripe and ripe fruit by fungal infection and wound, and only in ripe fruit by jasmonic acid (JA) treatment. To examine further regulation of PepTLP, the action of specific agonist and antagonists of known signaling effector on the .PepTLP expression by fungal infection, wound, and JA was investigated. A similar dephosphorylation event negatively activated all the PepTLP expression in the ripe fruit by fungal infection, wound, and JA. The induction of PepTLP expression by wound is differentially regulated via phosphorylation and dephosphorylation step during pre- and post-ripening, respectively. In addition, the induction of PepTLP expression in the ripe fruit by wound and JA is differentially regulated via dephosphorylation and phosphorylation step, respectively. Only both wound and JA treatment has synergistic effect on the PepTLP expression in the unripe fruit. Both SA and JA treatments on the unripe fruit, and both wound or JA and SA on the ripe fruit could not do any effect on the expression of PepTLP. These results suggest that the induction of PepTLP expression is differentially regulated via complex regulatory system against fungal infection, wound, and JA treatment during pre- and post-ripening of pepper fruit.

Transport Distance Affects the Quality of Both Mature and Ripe Tomato Fruits (수송거리에 따른 녹숙 및 적숙 토마토의 품질변화)

  • Tomato fruits(Lycopersicon esculentum $M_{ILL}$.) picked at mature (from mature green to breaker) and ripe (from pink to red) stage were packed into two layers in corrugated fiberbox and transported 200km and 400km from the harvested site. Loss (abrasion + cracking) in marketability was lowest (11.0%) from mature + 200km transport, followed by mature + 400km transport(13.5%), ripe + 200km transport(28.5%), and ripe + 400km transport (45.5%). When data were pooled by fruit maturity, firmness of mature fruit was 1.55 times higher than ripe fruit. However, vitamin C content was 1.25 times higher in ripe fruit than in mature fruit. Fruit transported longer distance and packed in bottom layer within a box exhibited lower firmness and vitamin C content in both maturity. It was concluded that it is desirable to transport relatively shorter distance for ripe tomato and longer distance for less ripe fruit.

Changes in flavor-relevant compounds during vine ripening of tomato fruit and their relationship with ethylene production

  • Flavor quality is import for determining consumer perception and acceptance of tomato products. In this study, 'Fendou' tomato fruit were harvested at six ripening stages and sampled to investigate the development of flavor-relevant compounds during vine ripening. Results showed that upon the initiation of ripening there was an increase in respiration rate and concomitant ethylene evolution that was associated with increased membrane permeability. In accordance with these physiological changes, flavor-relevant compounds demonstrated different expression patterns as fruit ripened, which contributed to 'red-ripe' flavor characteristics of red-ripe fruit. Based on correlation analysis between ethylene evolution and the flavor-relevant compounds during 'Fendou' tomato ripening and the other researchers' reports, the activation of System 2-dependent autocatalytic ethylene production plays an important role in the development of most flavor-relevant compounds during tomato vine ripening. Overall, our results suggested that most flavor-relevant compounds that accumulated the most during tomato fruit ripening at red stage could be under ethylene regulation and were among the most important contributors to the 'red-ripe' flavor. Due to the development of these compounds, the flavor quality at late ripening stages is different from that of fruit at early ripening stages.

Quantitative Determination of the Triterpenoids and Total Tannin in Korean Rubus species by HPLC

  • The triterpenoids contained in four Rubus species (Rosaceae) were quantitatively analyzed using HPLC to select plants with large quantities of niga-ichigoside $F_1$. Unripe fruits, ripe fruits, and leaves were extracted to estimate the quantity of niga-ichigoside $F_1$, together with Rubus-specific $19{\alpha}$-hydroxyursane-type triterpenoids, euscaphic acid, tormentic acid, and kaji-ichigoside $F_1$, and a dimeric triterpenoid coreanoside $F_1$. Niga-ichigoside $F_1$ was most abundant in the leaves of R. crataegifolius (23.4 mg/g dry weight). The amount of triterpenoid in the black, ripe fruits of R. coreanus was lower than the unripe fruits of the same plant. On the other hand, the ripe fruits of three plants, R. crataegifolius, R. parvifolius and R. pungens var. oldhami, which are reddish, contained higher or similar level of triterpenoids than their unripe fruits. In addition, the concentration of niga-ichigoside $F_1$ in the ripe fruit of R. crataegifolius was 20.5 mg/g, suggesting that the fruits could be used as a functional food. Methyl gallate and ellagic acid were used as quantitative indices of total tannin. Methyl gallate levels were higher in ripe fruits than unripe fruits in R. crataegifolius, R. pungens var. oldhami, and R. parvifolius. In R. crataegifolius, the quantity of methyl gallate was 30.5 mg/g in ripe fruit, but 1.19 mg/g in unripe fruit.

  • Objectives : The purpose of this study is to determine new source of Jisil(枳實). Methods : Find out the source of Jisil in the history of herbal medicine. Results : 1. The source of jisil(枳實) is known as the immature fruit of Poncirus trifoliata Rafinesqul(Rutaceae) in the Korean Pharmacopoeia Eight Edition, the dried young fruit of Citrus aurantium L. and its cultivars or Citrus sinensis Osbeck(Fam. Rutaceae) in Pharmacopeia of the people's republic of china(English edition 2000). 2. Until Song dynasty, Jisil is the pericarp of the ripe fruit of Poncirus trifoliata 3. From Myeong dynasty the source of jisil(枳實) turn to the immature fruit of C. wilsonii, C. junos, C. aurantiun var. amara Conclusions : The source of Jisil(枳實) is the ripe fruit of Poncirus trifoliata.

Diagnosis and Integrated Management of Major Fungal Fruit Rots on Kiwifruit in Korea

  • Ripe rot, Botrytis storage rot and Sclerotinia rot are major fungal diseases that lead to deterioration of fruit quality in Korea. Ripe rot, which is caused by Botryosphaeria dothidea, affects harvested fruits during post-storage ripening, while Botrytis storage rot, caused by Botrytis cinerea, affects harvested fruits during cold storage, and Sclerotinia rot, caused by Sclerotinia sclerotiorum, mainly affects immature fruits on the trees. Major fungal fruit rots tend to affect yellow- and red-fleshed cultivars of kiwifruit more severely because of the sharp increase in their cultivation acreage in recent years in Korea. In this review, we summarize symptoms and epidemiological characteristics of the major fungal fruit rots and propose integrated management methods of the diseases that can be practically utilized at the farmers' orchards in order to prevent the diseases based on our research works and field experiences and the research works of others conducted during the last three decades worldwide.

Botrysphaeria dothidea, the Causal Organism of Ripe Rot of Kiwifruit (Actinidia deliciosa) in Korea

  • Ripe rot was commonly found in overripe kiwifruits (Actinidia deliciosa) during a disease survey conducted in Jeju and Jeonnam provinces in Korea in 1999. While the disease did not manifest any clear external symptoms on kiwifruits, it caused portion of the fruit surface to collapse. Watersoaked flesh tissue could be seen on the sunken part when the skin of the collapsed portion was peeled off. The milky internal symptom with dark green margin developed concentrically as the fruit ripened. A species of Botryosphaeria was consistently isolated from lesions showing typical symptoms of ripe rot on kiwifruit, and its pathogenicity was confirmed by artificial inoculation test on healthy kiwifruits. The fungus was also pathogenic to apple and pear. The mycological characteristics of the fungus were consistent with those of Botryosphaeria dothidea. This is the first report on the detailed mycological characteristics of the causal organism of post-harvest ripe rot of kiwifruit in Korea.

Growth and Fruit Characteristics of Blueberry 'Northland' Cultivar as Influenced by Open Field and Rain Shelter House Cultivation (노지와 비가림 하우스 재배에 따른 블루베리 'Northland' 품종의 생육및 과실 특성 분석)

  • This study was conducted to find the effects of microclimates such as air and soil temperature, photosynthetic photon flux density (PPFD) on the berry development and physiological property between cultivation conditions (open field and rain shelter house) in 'Northland' blueberries (Vaccinium corymbosum). The rate of berry growth and development was stimulated in plants grown in rain shelter house, thus, berry reached to the ripe stage about one week earlier than those in open field. Berry weight and size at ripe stage seemed not affected by microclimates. However, total soluble solids content was higher in berries from open field whereas the titratable acidity was significantly higher in berries grown in rain shelter house. Berry firmness at ripe stage was little affected by growing condition. Total anthocyanin content of ripe berries was higher in berries harvested from rain shelter house. Total phenolics content and anti-oxidation activity of berries were higher in open field than those of rain shelter house during berry development but no differences were found at ripe berries.

Suppression of Ripe Rot on 'Zesy002' Kiwifruit with Commercial Agrochemicals

  • Ripe rot caused by Botryosphaeria dothidea is one of the serious diseases of postharvest kiwifruit. In order to control ripe rot on Actinidia chinensis cultivar 'Zesy002', several commercial agrofungicides were selected by an antifungal test on an artificial medium. Furthermore, disease suppression by the selected fungicides was evaluated on the kiwifruit by inoculation with a conidial suspension of B. dothidea. On the artificial media containing boscalid + fludioxonil was shown to be the most effective antifungal activity. However, in the bio-test pyraclostrobin + boscalid and iminoctadinetris were the most effective agrochemicals on the fruit. On the other hand, the infection structures of B. dothidea on kiwifruit treated with pyraclostrobin + boscalid were observed with a fluorescent microscope. Most of the fungal conidia had not germinated on the kiwifruit treated with the agrochemicals whereas on the untreated fruit the fungal conidia had mostly germinated. Electron microscopy of the fine structures showed morphological changes to the conidia and branch of hyphae on the kiwifruit pre-treated with pyraclostrobin + boscalid, indicating its suppression effect on fungal growth. Based on this observation, it is suggested that ripe rot by B. dothidea may be suppressed through the inhibition of conidial germination on the kiwifruit treated with the agrochemicals.
