Preparation of Copper Database of Korean Foods and Copper Nutritional Status of Korean Adults Living in Rural Area Assessed by Dietary Intake and Serum Analysis (한국인 상용 식품의 구리영양가표작성과 식이섭취 및 혈청분석에 의한 한국농촌성인의 구리영양상태 평가)

  • This study was carried out to prepare a copper database of Korean foods which can be used in calculating copper intake from dietary data, and to evaluate the copper nutritional status of Koran adults living in rural areas by dietary intake and serum copper concentrations. A copper database for 1,176 Korean foods was constructed (1) by analysing 112 Korean foods which are frequently consumed by Korean adults living in rural areas, (2) by adapting values form food composition databases from other countries-320 items from the University of Minnesota database, 201 items from the USAD database, and 25 items from U.K. database, and (3) by imputing values from similar foods for 518 food items. Copper intake of 2,034 Korean adults over the age of 30 living in Yeonchongun was Kyunggi province, Korea was estimated by 24-hour recall method. Mean daily copper intake of subjects was 0.98mg. Mean daily intake level of males was 1.11mg which was significantly higher than that of females, 0.88mg. There was a significant difference in the distribution of subjects by the level of copper intake and sex(p<0.05). Mean serum copper concentration was 14.8umol/1 and the percentage of subjects with low, adequate, and high levels of copper concentration were 23.9%, 69.4%, and 6.6%, respectively. The two food groups which contributed most to the dietary copper intake of subjects were cereals and grain products, and vegetables, supplying 46.2% and 12.7% of total copper intake, respectively. Individuallym, rice contributed most, suppling 31% of total copper intake, followed by soybean curd, starch vermicle, barley, etc. Plant foods contributed to 82.1% of the total copper intake. In summary, results of this study show that copper intake of Korean adults living in rural areas is low, and that dietary sources of copper are mainly plant foods. Serum levels of copper in the subjects were relatively normal. The copper database for Korean foods constructed in present study will be a valuable tool for the as-yet limited assessment of copper intake of Koreans. Such studies will contribute to the establishment of a dietary of a dietary allowance of copper and the relationship of copper nutriture and chronic diseases in Koreans.

  • Acorn wine was manufactured experimentally with koji inoculated the strain producing acorn tannin hydrolyzing enzyme in order to apply fungal tannase to food processing. Starch value of several Korean acorns was found to be 72.84 and the acorns were worthy of use as a carbohydrate food. Mixed koji was prepared by combination of rice and acorn powder at a ratio of 50to 50 and inoculation of Aspergillus oryzae producing amylase and Aspergillus sp. AN-11 producing tannase into the mixture in order to hydrolyze efficiently acorn tannin inhibiting alcohol fermentation in the medium, and then the mixed koji was used as a suitable koji to manufacture acorn wine. Acorn wine brewed with medium of the acorn powder treated with water and cooked and the mixed koji prepared was superior about two times to that brewed with medium of untreated acorn powder and general koji with respect to the rate of alcohol production and sugar fermentation during the 1st and 2nd brewing.

  • To select the sntagonistic bacteria against B. cinerea, isolates were screened from the eggplant leaves and rhizosphere soils in the eggplnat fields in the greenhouse. W1 and P99 isolates were selected by the inhibition of mycelial growth of B. cinerea E12 in vitro test. These isolates, W1 and P99, were identified as Bacillus subtilis and Pseudomonas putida, respectively, by the Bergeys manual and API systems, For the formulation of the antagonistic bacteria, the media for the mass production were prepared with biji(soybean curd residues) or soybean flour. B. subtilis W1 or P. putida P99 was mass cultured in biji broth or soybean flour extrect broth and then soybean flour, corn starch flour, rice glutinous flour and biji flour as high molecular substrates were added. These mixtures were dried, grinded and formulated as brofungicides of wettable powder type. The assess the control effect of biofungicides against the infection of B. cinerea, six types of formulations were assayed at the pot culturing with eggplant in the greenhouse. According to the results, there were no significant differences among the formulation methods. However, P99S or PppB formulated with P. putida P99 showed the highest control values as 90.4% and 96.1%, respectively. Then. BSB or BSD formulated whit B. subtilis W1 were 80.8% and 83.0%, respectively. There afforementioned values were more effective than that of chemical fungicide. Ipro W.P which showed as 72.6%.

Preparation of Shikhae with Starch Hydrolysing Enzymes/Malt Mixture in Tea-bag (전분 분해효소 첨가와 종이봉지를 이용한 식혜의 제조 방법)

  • As an improved preparation method of Shikhae, a tea-bag system containing malt and amylolytic enzymes was developed in which extraction of malt enzymes and saccharification occured efficiently. The amylolytic activity of the malt was increased by adding the mixture of ${\alpha}-amylase$, glucoamylase and glucoisomerase. Malt and the mixture of enzymes were placed in tea-bag $(16{\times}20cm)$, extracted in water at $30-40^{\circ}C$ for 1-2 hours and followed by saccharification of the cooked rice at $60-70^{\circ}C$ for 3-4 hours. In the conventional Shikhae, content of maltose was about 50% and that of oligosaccharides larger than trisaccharides was about 40% of total sugar. The content of monosaccharides such as glucose and fructose was about 95% and this improved method would be effective for increasing the sweetness and the monosaccharide contents in the product.

  • Some properties of six kinds of waxy starches isolated from Olchal and Hankang(both of waxy rice), Chalborie (waxy barley), Yullmoo (waxy Job' tears), Chajoe (waxy foxtail millet), Chalsusu (waxy great millet) and Chalocsusu(waxy corn) were investigated. The average diameters of their starch granules were $4.2{\sim}19.0$ microns. X-ray diffraction study showed that these starches had A pattern. Blue values of iodine complexes of these waxy starches were $0.02{\sim}0.08$. The number of glucose units per segment of amylopectin molecule were $21{\sim}23$. Their raising powers were $180{\sim}280$.

Isolation and Identification of Burkholderia pyrrocinia CH-67 to Control Tomato Leaf Mold and Damping-off on Crisphead Lettuce and Tomato

  • A bacterial strain CH-67 which exhibits antagonism towards several plant pathogenic fungi such as Botrytis cinerea, Fulvia fulva, Rhizoctonia solani, Sclerotinia sclerotiorum, Colletotrichum sp. and Phytophthora sp. was isolated from forest soil by a chitin-baiting method. This strain was identified as Burkholderia cepacia complex (Bcc) and belonging to genomovar IX (Burkholderia pyrrocinia) by colony morphology, biochemical traits and molecular method like 16S rRNA and recA gene analysis. This strain was used to develop a bio-fungicide for the control of tomato leaf mold caused by Fulvia fulva. Various formulations of B. pyrrocinia CH-67 were prepared using fermentation cultures of the bacterium in rice oil medium. The result of pot experiments led to selection of the wettable powder formulation CH67-C containing modified starch as the best formulation for the control of tomato leaf mold. CH67-C, at 100-fold dilution, showed a control value of 85% against tomato leaf mold. Its disease control efficacy was not significantly different from that of the chemical fungicide triflumidazole. B. pyrrocinia CH-67 was also effective in controlling damping-off caused by Rhizoctonia solani PY-1 in crisphead lettuce and tomato plants. CH67-C formulation was recognized as a cell-free formulation since B. pyrrocinia CH-67 was all lethal during formulation process. This study provides an effective biocontrol formulation of biofungicide using B. pyrrocinia CH-67 to control tomato leaf mold and damping-off crisphead lettuce and tomato.

Effects of Methanol Extract of Prosomillet on Cholesterol and Fatty Acid Metabolism in Rat

  • To study effects of methanol extract of prosomillet on lopid metabolism , five groups of male Sprang-Dawley rats weighing 116$\pm$9 g were fed test diets for four weeks. The five diets consisted of one low fat(5% w/w) diet containing starch as carbohydrate source(normal) and four high fat diets(15% w/w) containing 40.5%(w/w)sucrose(control) and additional 80% nethanol extractof prosomillet at the levels of 0.3% and 1%(w/w) or prosomillet powder at the level of 20%(w/w). Serum level of total cholesterol was a little higher but that of triglyceride was 41% lower in 20% (w/w) prosomillet powder group than in the control group. The cholesterol levels of two Liver cholesterol levels were lower and phospolipid levels higher in all three prosomillet powder group . Fecal excretionof bile acid was most increased in the prosomillet powder group among all five test groups. Acitivity of liver microsomal 3-hydroxy-3-methylglutaryl coenzyme A (HMG CoA) reductase was significantly lower in 0.3% methanol extract fed group than the control and also appeared to be reduced in 1% extract fed one, wherease those of 20 cholesterol 7$\alpha$-hydroxylase were not different among the five groups. Activities of liver cytosilic glucose-6-phosphate dehydrogenase(G6PDH) and malic enzyme were decreased in 0.3% prosomillet methanol extract and 20% powder groups. The results indicate that in addition to fiber, certain active components in prosomillet have potential to exert hypolipidemic effects via regulating hepatic cholesterogenesis and lipogenesis.

Production Medium Optimization for Monascus Biomass Containing High Content of Monacolin-K by Using Soybean Flour Substrates (기능성 원료를 기질로 이용하는 Monacolin-K 고함유 모나스커스 균주의 생산배지 최적화)

  • During the last decade, monacolin-K biosynthesized by fermentation of red yeast rice (Monascus strains) was proved to have an efficient cholesterol lowering capability, leading to rapid increase in the market demand for the functional red yeast rice. In this study, the production medium composition and components were optimized on a shake flask scale for monacolin-K production by Monascus pilosus (KCCM 60160). The effect of three different soybean flours on the monacolin-K production were studied in order to replace the nitrogen sources of basic production medium (yeast extract, malt extract and beef extract). Among the several experiments, the production medium with dietary soybean flour to replace a half of yeast extract was very good for monacolin-K production. Plackett-Burman experimental design was used to determine the key factors which are critical to produce the biological products in the fermentation. According to the result of Plackett-Burman experimental design, a second order response surface design was applied using yeast extract, beef extract and $(NH_4)_2SO_4$ as factors. Applying this model, the optimum concentration of the three variables was obtained. The maximum monacolin-K production (369.6 mg/L) predicted by model agrees well with the experimental value (418 mg/L) obtained from the experimental verification at the optimal medium. The yield of monacolin-K was increased by 67% as compared to that obtained with basic production medium in shake flasks.

Physicochemical and Sensory Properties of Kakdugi Added with Various Thickening Agents During Fermentation (점증제 첨가 깍두기의 이화학적.관능적 특성)

  • Effect of various thickening agents on kakdugi fermentation was investigated by measuring physicochemical and sensory properties during fermentation at 2$0^{\circ}C$. Paste of seven kinds of thickening agents (wheat flour (WF), waxy rice flour (WR), corn starch (CS), acid modified starch (AM), aretylated distarch adipate (AA), hydroxypropyl distarch phosphate (PP) and xanthan gum (XG) ) at 0.25% concentration was added to kakdugi. Total acidity during fermentation was not different among thickening agents, but slightly lower in XG than control at the 7th day of fermentation. At 0 day of fermentation, free sugar amount were higher in thickening agent addition groups than control, but rapidly decreased to below control at the 7th day of fermentation, except XG. Glucose and fructose which were the major free sugars, decreased rapidly during fermentation, whereas mannitol increased in all samples. Viscosity of kakdugi liquid was much higher in thickening agent addition groups than control at 0 day of fermentation, but rapidly decreased from 1 day of fermentation. However, initial viscosity was maintained only in XG. Hardness at the 7th day of fermentation was higher in WR, PP, XG than control. The result of sensory evaluation shows that there were no significant difference in sour odor, sour taste and savory taste among samples. Moldy odor was higher in WR, WF and AM, but was not significantly different in XG, PP, AA compared to control. Viscosity of XG and PP, and starchy taste of XG were higher than those of control. Overall preference of XG, AM, PP were not significantly different from that of control. Xanthan gum was considered to be a good thickening agent for kakdugi but it is necessary to find a minimum concentration for kakdugi since starch taste of xanthan gum.

Studies on the Epidemiology and Control of Bacterial Leaf Blight of Rice in Korea (한국에 있어서의 벼흰빛잎마름병의 발생생태와 방제에 관한 연구)

  • The study has been carried out to investigate the occurrence, damage, characteristics of the pathogen, environmental conditions affecting the disease outbreak, varietal resistance, forecasting, and chemical control of bacterial leaf blight of rice in Korea since 1964. Bacterial leaf blight of rice became a major disease in Korea since 1960. A correlation was found between the annual increase of epidemics and increase of cultivation area of susceptible varieties, Jinheung, Keumnampung etc. Areal damage within the country showed that the more was at southern province, Jeonnam, Gyeongnam and western coast, and at flooded rice paddy. Yield reduction directly related with the amount of infection on upper leaves at heading stage. Fifty per cent of reduction resulted when the lesion area was more than 60 per cent. Less than 20 per cent of lesion area, however, was not affected so much on yield loss One hundred and six isolates collected from all over the country were classified as 8 strains by using 4 different bacteriophages in 1973. It was, however, only two in 1965. There were some specificities on varietal distributions among the strains such as that the Jinheung attacked mainly by strain A, B, C and I, those attack Kimmaze were A, B, H and I. Most strains were found from Tongil except D and E, whereas Akibare was only variety that attacked by strain E. Low temperature, high humidity, heavy rainfall and insutficient daylight favored the disease epidemics. Especially, typhoon and flooding at heading stage were critical factors. The earlier transplanting the more disease was resulted, and more nitrogen fertilizer application accerelated the diseased development in general. The resistance to the disease varied by growing stage of the sane plants. All of recommended varieties in Korea were susceptible to the disease except Norm No. 6 and Sirogane which moderately resistant. The pathogen, Xanthomonas oryzae, was detectable from extract of healthy seedlings that were grown in the field with an heavy infection previous year. The more bacteriophage in irigation water resulted the more disease outbreak, and the existence of more than 50 bacteriophages in 1ml. of irrigation water were necessary to initiate the disease out break. The curves representing occurrence of bacteriophages and disease outbreak were similar with 15 days interval. The survey of bacteriophage occurrence can be utilized in forecasting of the disease two weeks ahead of disease outbreak. Three applications of chemicals, Phenazin and Sangkel, in weekly intervals at the early satage of out-break depressed the symptom development, and increased yield by 20per cent. Proper period for the chemical application was just before the number of bacteriophage reaches 50 in 1ml. of irrigation water.
