Plants exposed to environmental stress for long durations often can adapt to stress conditions with improved tolerance. Moreover this acquired tolerance to stress can be retained even after reverting to destressed growth conditions, which is known to stress memory. In these adaptation and stress memory processes, epigenetic regulation, such as DNA methylation and histone modifications play a key role. Here, we showed that regenerated rice plants from embryogenic callus exposed to gradually increasing NaCl concentrations (up to 120 mM NaCl) acquired salt tolerance and their enhanced tolerance are inherited to subsequent generations. The rice plants (R0) regenerated from rice callus under saline conditions were transplanted into normal paddy field and R1 seeds were harvested. These R1 seeds displayed higher germination rate on MS medium containing 100mM NaCl than wild-type. The callus derived from R1 seeds showed better growth than control callus on high salinity medium. And the salt-adapted R1 plants exhibited higher chlorophyll contents and also higher $K^+/Na^+$ ratio than wild-type rice under saline conditions. The results indicated that rice plants successfully adapted to saline growth conditions during regeneration on high salt medium and moreover this acquired tolerance to salt stress was inherited subsequent generation.
The aim of this study was to develop rice cookies based on 'Goami 2' rice, and made with sesame (white and black) and perilla seeds. Furthermore, we studied the physicochemical and sensory characteristics of the cookies as well as the alternative effects of the sesames or perilla seeds for butter, where we substituted the sesames or perilla for 30% of the total butter. The pH values of the cookie doughs made with the sesames or perilla seeds were significantly different (p<0.001), whereas the spread factors of the doughs were not significantly different. The Hunter color L-, a-, and b- values were significantly different for the cookies made with the added sesames or perilla seeds (p<0.001). The brittleness of the Goami 2 rice cookies (control) was significantly higher than that of the cookies made with the sesames or perilla (p<0.001). In a descriptive analysis performed by 8 trained panelists, the following sensory characteristics: yellow, black, sweet taste, nutty taste, nutty odor, oily flavor, and off flavor, were significantly different among the cookie samples. In a consumer test by 50 women students, the Goami 2 rice cookies were preferred in the following order: Goami 2 rice cookies with no additions > black sesame cookies > white sesame cookies > perilla cookies. In conclusion, some physicochemical and sensory characteristics of the cookies were different, in relation to being made with or without the sesame or perilla seeds. However, because of the small difference in acceptance among the cookies, the sesame and perilla rice cookies show strong potentials for consumer acceptance. Moreover, the black sesame cookies were more acceptable in terms of butter substitution for the cookie preparation.

The experiment was conducted at the growth chamber to determine germination, seedling growth and fresh weight between the untreated control and iron-coated seeds in germination test of the rice seeds. There was of 95% germination in rice seeds regardless of treatments and days for seedling emergence was relatively 13-14days. Seedling height was 4.9cm of the iron-coated seeds but 0.4cm of the untreated control and there was 6.S roots of seedling in iron-coated seeds and 5.5 roots of seedling in the untreated control, respectively. The fresh weight of seedling was 107mg of the iron-coated seeds and 66mg of the untreated control at 35days after sowing.
To find out the effects of humidification and hardening of rice (Oryza sativa L. cv. Ilpumbyeo) seeds on the germination, both normal and artificially aged seeds with 60% germination rate were humidified at 40, 60, and 80% RHs and $25^{\circ}C$ for five weeks or hydrated for 6, 12, 18, and 24 hours at $25^{\circ}C$ from one to five cycles for hardening. Relative humidity and duration of humidification did not affect the germination rate of normal seeds, while reduced the time to get 50% germination rate ($T_{50}$) by 1.0~1.6 days compared to that of normal seeds at $25^{\circ}C$. Aged seeds humidified at 40 and 60% RHs did not affect the germination rate and $T_{50}$ regardless of relative humidity and duration of treatment, while at 80% RH, the germination rate decreased and $T_{50}$ increased significantly with the duration of humidification. Hardening of normal seeds reduced $T_{50}$ by 0.7~1.1 days without changes in the germination rate. However, the germination rate and $T_{50}$ of aged seeds soaked in water for 6, 12, and 18 hours were similar regardless of soaking/drying cycles, while hardening of artificially aged seeds with 1~4 cycles in 24 hours soaking increased the germination rate by 11-16% and reduced $T_{50}$ by 1.4~2.0 days.

Soil salinity due to accumulation of salts particularly sodium chloride affects agricultural lands and their vegetation. Generally, rice is a moderately sensitive plant with some cultivars with varying tolerance to salinity. Though there are physiological differences between salt-sensitive and salt-tolerant rice cultivars, both are still affected especially during high salinity and prolonged exposure. This also ultimately affects their indigenous bacterial endophytes particularly those that inhabit the rice seed endosphere. This study investigates the dynamic structure of seed bacterial endophytes of salt-sensitive and tolerant rice cultivars grown in different levels of soil salinity. Endophytic bacterial diversity was studied Terminal-Restriction Fragment Length Polymorphism (T-RFLP) analysis. Results revealed a very interesting pattern of diversity and shifts in community structure of bacterial endophytes in the rice seeds. There is a general decrease in diversity for the salt-sensitive rice cultivar, IR29 as soil salinity increases. For the salt-tolerant cultivars, IC32 and IC37, diversity interestingly increased at moderate salinity then decreased at high soil salinity. The patterns of community structure is also strikingly different for the salt-sensitive and salt-tolerant rice cultivars. IR29 has a more even distribution of abundance, but under soil salinity, the community shifted where Curtobacterium, Pantoea, Flavobacterium and Microbacterium become the more dominant bacterial communities. For IC32 and IC37, the dominant bacterial groups under normal stress conditions were also the dominant bacterial groups during salt stress conditions. Their seed bacterial community is dominated by endophytes belonging to Microbacterium, Flavobacterium, Pantoea, Kosakonia and Enterobacter. Stenotrophomonas and Xanthomonas have not changed in terms of abundance under different salinity stress level in the salt-sensitive and salt-tolerant rice cultivars. This study showed that soil salinity greatly influenced the seed bacterial communities of rice seeds irrespective of their physiological tolerance to salinity.
Experiments were conducted to investigate seed persistence of Chinese milk vetch(CMV) in naturally reseeded rice field in 2007~2009. The seed and pods with seeds were buried in rice field at 0, 5 and 10 cm depths and retrieved from the field at one to three month intervals from July to the following year March and determined change of seed dormancy and viability. In the second experiment, persistence of the CMV seeds in the naturally reseeded rice field at different tillage methods and soil depths were also investigated after rice harvest in autumn. Burial depths and durations affected recovery rate, dormancy and viability of CMV seed. The viability loss was faster and greater in the seed than the pod with seeds and on the soil surface than the 5 or 10 cm burial depths. The recovery rate of CMV seed was decreased starting from one month as seed burial and it was significantly decreased to 52~65% for the seed in September. However, unlike the seed burial, the nearly 100% CMV seeds were recovered for burial as pod with seeds even after four months burial in both 0 and 5 cm depths. However, the recovery rate was sharply declined to below 30% at October in 2007 in both seed and pods with seeds and in the 2008/2009 experiment. the 15~47% of CMV seeds still remained even after October. The CMV had high seed dormancy of 95%, showing only 4~5% germination at the beginning in June but the seed germination increased to 25 to 35% in seed and 55 to 61% in pod with seeds in September due to breakage of hard seed dormancy. The viability loss was faster in the seed than in the pod with seeds regardless of depths of placement in the soil base on decayed seeds. Also the seed placed on the soil surface lost viability faster than the 5~10 burial depths. On the other hand, field observation in the naturally reseeded CMV rice field showed that as many as 917~2,185 CMV seeds $m^2$ were from the 0~15 cm soil depth in the rotary tillage and 250~10,105 CMV seeds in minimum tillage treatmints. The recovered seed germinated 25~33%, 23~43% but still had high percentage of hard seed having 64~72% and 51~77% even after rice harvest in autumn. These results indicate that freshly harvested CMV seeds had high level of primary dormancy and the dormancy was gradually broken in soil with time during rice cultivation periods and appreciable number of CMV seeds remained even 4 month after burial in soil. CMV plant regenerated naturally from the remained seed bank at rice harvest time in autumn. The CMV seedling still emerged even after 2 years of continuous destructive killing of emerged CMV plant by rotary tillage in naturally reseeded CMV plant in rice field, indicating that CMV seeds do persistent as least two years in soil.
Weedy rice (Oryza sativa) is a semi-wild rice grown in paddy fields and is more adapted to the environment than cultivated rice. Therefore, it is expected that the seeds of weedy rice might contain some chemicals related to its environmental adaptation, such as antioxidants, which may be useful for nutritional and medicinal purposes. This study was carried out to investigate the antioxidant activities of weedy rice compared to a Korean-bred rice cultivar, Sindongjin (SDJ), and to screen lines that show higher antioxidant activity in 199 accessions of weedy rice germplasm collected in Korea. DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity ranged from 31.0 to 91.7%, with an average of 82.5%, and 19 accessions that showed more than 91% antioxidant activity were selected. The 19 accessions were re-screened against non-germinated brown rice (BR) and germinated brown rice (GBR) using four assays, total phenol determination, DPPH radical scavenging, ABTS radical scavenging, and reducing power activities. The results showed that accession 'WD3' had the highest antioxidant capacity in both BR and GBR, suggesting that WD3 is a promising potential source of antioxidants and could be developed as a potentially functional substance material.
To investigate the effect of seeding date, seeding rate, and sprouted seeds on seedling establishment, yield and its components of rice in barley /rice reley cropping as a minimum tillage, Gancheokbyeo rice seeds sowed by hand broadcast in various overlapped days befer barley harvest. The highest rice seedling stand was at sowed on the day just before barley harvest and l00kg per hectare of seeding rate with sprouted seeds. Also number of tiller and panicle showed the similar tendency. Delayed rice heading dates were found 2 days at l0-day overlap, 2 days at 5-day overlap, and 4 days at 0-day overlap before barley harvest compared with machine transplanting rice at just after barley harvest using l0-days old seedling. The culm length of broadcast rice shortened 2∼4cm compared with the transplanting rice. The rice plants lodged slightly at seeding rate l00kg per hectare and at sowed on the day of barley harvest. The rice panicle number increased with later date sowing and higher seeding rate. Number of spikelets per panicle and ripened grains decreased at higher seeding rates. Sprouted seeds and higher seeding rates showed higher rice yield at later sowing dates.
Physical and cultural properties were investigated on the rice-seed pellets made by the pelleting machine(Yu, 2003) as the changes with mixing ratios of soil to rice seed of 6 : 1, 7 : 1, and 8 : I, and rotating speeds of forming rolls of 7, 10, and 13 rpm. Average weight, average diameter, and average sphericity of the pellets were 1.70 g, 12.0 ㎜, and 99.1 %, respectively. Average number of seeds per pellet was more than 3, and almost all pellets had more than 3 seeds in the cases of mixing ratios of 6 : 1, and 7 : 1 at the forming rolls' speed of 7 rpm. Gradual drying was needed because rapid drying caused cracks on surface of the pellets. Compression strength of the pellets dried in shady room was in the range of 132 ∼ 152 N, which was enough for handling. Comparing with the previous pellets(Park, 2002), average number of seeds per pellet, ratio of pellets including more than 3 seeds, and compression strength increased due to the effects of pressure feed of pellet materials, and improvements of the forming rolls. Emergence ratio of the pellets made at the mixing ratio of 6 : 1 and the forming rolls' speed of 7 rpm, was 100 % on dry paddy and was 97 % on flooded paddy surface. Good growth characteristics, and yield except number of seedling stand and ratio of missing plant were shown in planting of the pellets made at the mixing ratio of 7 : 1 and the forming rolls' speed of 7 rpm on flooded paddy surface field. Considering the cultural results, the mixing ratio of 6 : 1, and the forming rolls' speed of 7 rpm seems to be optimum operating condition for the improved pelleting machine.
To make certain of the white tip infection on rice, nematodes isolated from the rice seeds grown at Chongdo-goon Isu-myun and Kimje-goon Chooksan-myun were artificially inoculated and their pathway was studied. Also, studies were made for the hot water seed treatment as a measure of white tip control at different levels of water temperature and their germinablity was checked. The results are summarized as follows; 1. Causal nematodes survive in the paddy soil and cause white tip year after year. 2. Causal nematodes can survive in the rice seeds as long as the seeds loose their germinability. 3. Causal nematodes can disseminate throught the irrigation water. 4. Causal nematodes can disseminate through the soil move from a place to other place. 5. Causal nematodes can infect all of the parts of the rice and they can disseminate through any of the parts of rice. 6. The hot water treatment at 5$0^{\circ}C$ for 60 minutes were not effective controlling causal nematodes. But, either treatment of at 55$^{\circ}C$ for 15-20 minutes or at 6$0^{\circ}C$ for 10-15 minutes were effective and germination of seeds was not affected.
