The expressed sequence tags(ESTs) from immature seed of rice, Oryza sativa cv Milyang 23, were partially sequenced and analyzed by homology. As of 1998, the partial sequences of about 6,600 cDNA clones were analyzed from normal and normalized immature seed cDNA libraries. About 2,200 ESTs were putatively identified by BLASTX deduced amino acid sequence homology analysis. About 20% of them were putatively identified as storage proteins. Also the clones were highly homologous to genes involved particularly in starch biosynthesis, glycolysis, signal transduction and defenses. Compared to 35% of redundancy in the ESTs of normal cDNA library, that from the substracted library was 15%. The Korea Rice Genome Network is maintained to provide the updated information of sequences, their homologies and sequence alignments of ESTs. For the stable expression of transgene in rice, diverse vectors were developed for overexpression, targeting and gene dosage effect with transit peptides (Tp) and matrix attachment region (MAR) sequence from chicken lysozyme locus. The rice calli were transformed via Agrobacterium tumefaciens LBA4404(pSB1) with the triparental mating technique and selected by herbicide resistance. The green fluorescent protein(GFP) gene in expression vector under the control of rbcS promoter-Tp was overexpressed upto 10 % of the total soluble protein. In addition, the Tp-sGFP fusion protein was properly processed during translocation into chloroplast. The expression of sGFP in the presence of MAR sequences was analyzed with Northern and immunoblot analysis. All the lines in which sGFP transgene with MAR sequence, showed position independent and copy number-dependent expression, while the lines without MAR showed the varied level of expression with the integration site. Thus the MAR sequence significantly reduced the variation in transgene expression between independent transformants.
This experiment was conducted to investigate duck-raising system and the optimum nitrogen fertilization level in duck-raising field. Optimum duck rasing density in rice paddy field was 1, 000~1, 500 heads per ha and ducks could be harvested three times in a year starting from 40 days after mature-seedling machine transplanting. When 1, 000~1, 500 heads per ha of ducks was raised once a year, optimum nitrogen application level was 110kg/ha. In the case of three times in a year, it was 8.8kg/ha. The weed control rate of the duck-raised paddy field was superior(93~96%) to control (89%, treated twice with herbicide).
Lactoferrin is an 80-kDa iron-binding glycoprotein known to exert many biological activities, such as facilitating iron absorption and having antimicrobial and anti-inflammatory effects. Rice can be a useful target for edible food plants to introduce human lactoferrin, because it has lower allergenicity and is likely to be safer than microorganisms or transgenic animals. A cDNA fragment encoding human lactoferrin (HLF) driven by the maize polyubiquitin promoter, along with herbicide resistance gene (bar) driven by CaMV 35S promoter, was introduced into rice (Oryza sativa L. cv. Dong Jin) using the Agrobacterium -mediated transformation system. Putative transformants were initially selected on the medium containing bialaphos. The stable integration of the bar and HLF genes into transgenic rice plants was further confirmed through polymerase chain reaction (PCR) and Southern blot analyses. The expression of the full length HLF protein from various tissues such as grains and young leaves of transgenic rice was verified by Western blot analysis. Analysis of progeny also demonstrated that introduced genes were stably inherited to the next generation at the Mendelian fashion.
The loss of green coloration via chlorophyll (Chl) degradation typically occurs during leaf senescence. To date, many Chl catabolic enzymes have been identified and shown to interact with light harvesting complex II to form a Chl degradation complex in senescing chloroplasts; this complex might metabolically channel phototoxic Chl catabolic intermediates to prevent oxidative damage to cells. The Chl catabolic enzyme 7-hydroxymethyl Chl a reductase (HCAR) converts 7-hydroxymethyl Chl a (7-HMC a) to Chl a. The rice (Oryza sativa) genome contains a single HCAR homolog (OsHCAR), but its exact role remains unknown. Here, we show that an oshcar knockout mutant exhibits persistent green leaves during both dark-induced and natural senescence, and accumulates 7-HMC a and pheophorbide a (Pheo a) in green leaf blades. Interestingly, both rice and Arabidopsis hcar mutants exhibit severe cell death at the vegetative stage; this cell death largely occurs in a light intensity-dependent manner. In addition, 7-HMC a treatment led to the generation of singlet oxygen ($^1O_2$) in Arabidopsis and rice protoplasts in the light. Under herbicide-induced oxidative stress conditions, leaf necrosis was more severe in hcar plants than in wild type, and HCAR-overexpressing plants were more tolerant to reactive oxygen species than wild type. Therefore, in addition to functioning in the conversion of 7-HMC a to Chl a in senescent leaves, HCAR may play a critical role in protecting plants from high light-induced damage by preventing the accumulation of 7-HMC a and Pheo a in developing and mature leaves at the vegetative stage.

Basis for differential response of rice and E. crus-galli to cyhalofop-butyl ester (R-butyl 2-(4-(4-cyano-2-fluoro-phenoxy)phenoxy)propionate) was anatomically compared. Cell division in the growing point of E. crus-galli was severely inhibited by the herbicide, leading to malformation and irregular arrangement of cells. The inhibitory symptom of rice by cyhalofop-butyl ester appeared to be very similiar to that of E. crus-galli although its injury was negligible in rice when applied at the rate of 180ppm.

This study was conducted to investigate the herbicidal efficacy of up granule formula of benzobicyclon mixtures, benzobicyclon + azimsulfuron + oxaziclomefone up granule (BAO UG) and benzobicyclon + bensulfuron-methyl + fentrazamide up granule (BBF UG) to control annual and perennial weed species in a rice paddy field. Four annual weed species including Echinochloa crus-galli L. and three perennial weeds were dominated in the experimental field. Application of BAO UG and BBF UG was highly effective to control both annual and perennial weed species. No phytotoxic effect was observed in the rice based on the plant height, the number of tillers, and culm and panicle length. Finally, rice yield in the BAO UG and BBF UG treatment was as much as that in the hand weeding. This study indicates that self-dispersible floating formula of benzobicyclon mixtures can be applied to control both annual and perennial weed species in rice paddy fields and contribute to save labor costs.
The leaching behaviour of quinclorac was elucidated using soil columns. On top of each glass column packed with a rice paddy soil up to the 30 cm height were applied three different treatments of [$^{14}C$]quinclorac: quincloiac only (T-1), quinclorac adsorbed onto active carbon (T-2), and quinclorac adsorbed onto a mixture of active carbon and $Ca(OH)_{2}$ (T-3). Half of the columns were planted with rice plants for 17 weeks and half of them unplanted for comparison. Average amounts of $^{14}C$-activity percolated from tile soil columns without rice plants in T-1, T-2, and T-3 were 81.1%, 27.8% and 48.0%, respectively, of tile originally applied $^{14}C$, whereas those with rice plants grown were 36.8%, 9.6% and 11.0%, respectively, indicating that the leaching of [$^{14}C$]quinclorac was significantly affected by vegetation and by treatment with the adsorbents. The bioavailability of the herbicide to rice plants in T-1, T-2, and T-3 were 13.6%, 11.0% and 13.9%, respectively. The residue levels of quinclorac in the edible part of rice grains would be far less than the maximum residue limit (MRL, 0.5 ppm). After the leaching, the amounts of $^{14}C$ remaining in soil in with rice planting T-1, T-2, and T-3 were 36.3%, 73.7%, and 61.8%, whereas those without rice planting were 19.7%, 71.1%, and 52.3%, respectively. The balance sheets indicate that [$^{14}C$]quinclorac translocated to rice shoots would be lost by volatilization and/or in other ways in T-1 and T-3. The $^{14}C$-activity partitioned into the aqueous phase of the leachates collected from all treatments was less than 7% of the total, but it increased gradually with time in the case of rice growing, suggesting tile formation of some polar degradation products.

The effect of different cultural patterns on the growth and herbicidal response of rice(Oryza sativa L.) and barnyardgrass(Echinochloa crus-galli var. oryzicola) was studied tinder direct-seeded rice and early rice seedling conditions. In dry-seeded rice condition, the growth of rice and barnyardgrass was the greatest in 3cm-seeding depth and 40% soil moisture, respectively. At 10 days after herbicide application, plant height of rice was severely inhibited in 1cm-seeding depth and 50% soil moisture, and the degree of inhibition was severer at application of pendi+molinate EC as compared to that of butachlor EC. On the other hand, plant heigth of barnyardgrass by treatment of herbicides was severely reduced regardless of seeding depth and soil moisture. The lengths of barnyardgrass mesocotyl and rice coleoptile were increased with increasing seeding depth and were severely inhibited by applied herbicides in 1cm-seeding depth under dry-seeded rice condition. The dry weight of rice at different leaching times after herbicides application showed slightly decreasing trend as the leaching time was delayed, but the growth of barnyardgrass was very severely inhibited regardless of the leaching time. In addition, the phytotoxicity of rice showed decreasing trend with the increase in transplanting depth and delay of application time under early rice seedling condition.
This study was conducted to investigate the inhibition of protox activity and the PPIX accumulation of the oxyfluorfen-tolerant and-susceptible rice cultivars with barnyardgrass, a typical susceptible weed in accordance by oxyfluorfen treatment. The susceptible rice cultivars and barnyardgrass showed more inhibition of protox activity due to the treatment of oxyfluorfen than the tolerant rice cultivars. Especially in the concentration at $10^{-6}$M treatment, protox activity of the susceptible rice cultivars and barnyardgrass were the completely inhibited but the tolerant rice cultivars kept 32~59% of activity compared to the control. As the treatment concentration increased, the content of PPIX accumulation increased and it increased untill four hours of light exposure but it tended to decrease these after. The content of PPIX accumulation by the treatment of oxyfluorfen was more pronounced in the light condition than in the dark. Under the light and dark conditions, the susceptible rice cultivars and barnyardgrass showed more PPIX accumulation than the tolerant rice cultivate. Especialiy the susceptible barnyardgrass had more than the rice. With the treatment of GC and DA, tetrapyrrole biosynthesis inhibitor, the herbicidal activity by oxyfluorfen was inhibited, and the susceptible rice cultivars and barnyardgrass tended to have less effective than the tolerant rice cultivars and the content of chlorophyll or PPS accumulation tended to be similar.

Sulfonylurea (SU)-resistant weeds include seven annual weeds such as Monochoria vaginalis, Scirpus juncoides and Cyperus difformis, etc., and three perennial weeds of Scirpus planiculmis, Sagittaria pigmaea and Eleocharis acicularis as of 2010 since identification Monochoria korsakowii in the reclaimed rice field in 1998. The Echinochloa oryzoides resistant to acetyl CoA carboxylase (ACCase) and acetolactate synthase (ALS) inhibitors has been confirmed in wet-direct seeding rice field of the southern province, Korea in 2009. In the beginning of occurrence of SU-resistant weeds the M. vaginalis, S. juncoides and C. difformis were rapidly and individually spreaded in different fields, however, theses resistant weeds have been occurring simultaneously in the same filed as time goes by. The resistant biotype by weed species demonstrated about 10- to 1,000-fold resistance, base on $GR_{50}$ (50% growth reduction) values of the SU herbicides tested. And the resistant biotype of E. oryzoides to cyhalofop-butyl, pyriminobac-methyl, and penoxsulam was about 14, 8, and 11 times more resistant than the susceptible biotype base on $GR_{50}$ values. In history of paddy herbicides in Korea, the introduction of SU herbicides including besulfuron-metyl and pyrazosulfuron-ethyl that control many troublesome weeds at low use rates and provide excellent crop safety gave farmers and many workers for herbicide business refreshing jolt. The products and applied area of SU-included herbicides have been rapidly increased, and have accounted for about 69% and 96%, respectively, in Korea. The top ten herbicides by applied area were composed of all SU-included herbicides by 2003. The concentrated and successive treatment of ACCase and ALS inhibitors for control of barnyardgrass in direct-seeded rice led up to the resistance of E. oryzoides. Also, SU-herbicides like pyrazosulfuron-ethyl and imazosulfuron which are effective to barnyardgrass can be bound up with the resistance of E. oryzoides. The ALS activity isolated from the resistant biotype of M. korsakowii to SU-herbicides tested was less sensitive than that of susceptible biotype. The concentration of herbicide required for 50% inhibition of ALS activity ($I_{50}$) of the SU-resistant M. korsakowii was 14- to 76-fold higher as compared to the susceptible biotype. No differences were observed in the rates of [$^{14}C$]bensulfuron uptake and translocation. ALS genes from M. vaginalis resistant and susceptible biotypes against SU-herbicides revealed a single amino acid substitution of proline (CCT), at 197th position based on the M. korsakowii ALS sequence numbering, to serin (TCT) in conserved domain A of the gene. Carfentrazone-ethyl and pyrazolate were used mainly to control SU-resistant M. vaginalis by 2006, the early period, in Korea. However, the alternative herbicides such as benzobicyclone, to be possible to control simultaneously the several resistant weeds, have been developing and using broadly because the several resistant weeds have been occurring simultaneously in the same filed. The top ten herbicides by applied area in Korea have been occupied by products of 3-way mixture type including herbicides with alternative mode of action for the herbicide resistant weeds. Mefenacet, fentrazamide and cafenstrole had excellent controlling effects on the ACCase and ALS inhibitors resistant when they were applied within 2 leaf stage.
