Effect of Humidification and Hardening Treatment on Seed Germination of Rice

  • To find out the effects of humidification and hardening of rice (Oryza sativa L. cv. Ilpumbyeo) seeds on the germination, both normal and artificially aged seeds with 60% germination rate were humidified at 40, 60, and 80% RHs and $25^{\circ}C$ for five weeks or hydrated for 6, 12, 18, and 24 hours at $25^{\circ}C$ from one to five cycles for hardening. Relative humidity and duration of humidification did not affect the germination rate of normal seeds, while reduced the time to get 50% germination rate ($T_{50}$) by 1.0~1.6 days compared to that of normal seeds at $25^{\circ}C$. Aged seeds humidified at 40 and 60% RHs did not affect the germination rate and $T_{50}$ regardless of relative humidity and duration of treatment, while at 80% RH, the germination rate decreased and $T_{50}$ increased significantly with the duration of humidification. Hardening of normal seeds reduced $T_{50}$ by 0.7~1.1 days without changes in the germination rate. However, the germination rate and $T_{50}$ of aged seeds soaked in water for 6, 12, and 18 hours were similar regardless of soaking/drying cycles, while hardening of artificially aged seeds with 1~4 cycles in 24 hours soaking increased the germination rate by 11-16% and reduced $T_{50}$ by 1.4~2.0 days.

발아찰현미떡의 품질 및 소화특성 (Quality and Digestibility Characteristics of Rice Cake with Germinated Brown Waxy Rice)

  • 백미와 현미, 발아현미 찹쌀떡에 대한 소화율과 품질 특성을 살펴보았다. 또한, 소비자들에게 선호도가 높은 제품의 발아조건을 찾고자 백미와 현미, 발아현미찹쌀떡을 제조하여 색도, 기계적 품질 특성, in vitro 소화율 그리고 관능적 특성을 살펴보았다. 떡의 밝기를 나타내는 명도(L)는 현미보다 발아현미가 낮았으며 발아시간이 길어질수록 감소하였다. 반면 적색도(a)와 황색도(b)는 발아가 진행될수록 증가하여 전체적으로 색이 진해지는 것을 알 수 있다. 현미와 발아현미의 조직감 특성에서 경도, 부착성, 검성, 씹힘성은 현미가 가장 높았고 발아시간이 증가할수록 감소하는 경향을 보였으며 이로써 노화 지연에 도움을 줄 것으로 생각한다. 탄성은 시료 간의 큰 차이를 보이지 않았으며, 응집성은 현미와 발아현미가 백미보다 낮은 값을 보였다. 현미, 발아현미 찹쌀떡의 in vitro 소화율 결과 발아시간이 증가할수록 빠르게 소화되는 전분인 RDS 함량은 증가하고 소화되지 않는 전분인 RS 함량은 감소하는 특성을 나타내어 소화율이 크게 증가함을 알 수 있었다. 관능검사에서는 현미가 색, 부착성, 전체적인 기호도에서 높게 평가되었고, 발아시간이 증가할수록 색, 향에서 높은 점수를 받았지만, 부착성과 전체적인 기호도에서는 낮게 평가되었다. 이상의 결과로 찹쌀떡보다는 높고 찰현미떡과 비슷한 관능적 평가를 받은 10시간 발아한 현미로 만든 발아찰현미떡이 소화조절용 쌀가공식품으로 개발하기에 가장 적당할 것으로 생각된다.

Chitosan/Glutamic acid 처리가 현미의 발아에 미치는 영향 (Effects of Germination in Brown Rice by Addition Chitosan/Glutamic acid)

  • 현미(brown rice)를 이용하여 기능성 성분인 GABA의 고함량 발아조건을 효과적으로 활용한 최적조건을 설정하고자 조단백 함량과 GABA 함량을 분석하였다. 발아온도 $25^{\circ}C$에서 조단백 함량과 GABA 함량은 $260.9\;mg\%$와 425.7 nmole/g으로 나타났으며 발아시간 72시간에서 조단백 함량과 GABA 함량이 $230.6\;mg\%$와 637.0 nmole/g으로 나타났다. chitosan과 glutamic acid 100 ppm을 혼합한 처리구에서 발아시킨 현미의 조단백 함량이 가장 낮게 나타나 이를 토대로 중심합성계획에 의해 발아현미 최적 생성조건을 예측하였다. 발아현미의 조단백 함량에 대한 반응표면은 안장점의 형태로 glutamic acid가 더 큰 영향을 미치며, GABA 함량에 대한 반응표면은 최대점의 형태로 나타났다. 발아현미의 최적 생성조건은 chitosan $105{\sim}160\;ppm$이며 glutamic acid 함량은 $200{\sim}290\;ppm$의 범위로 나타났으며 예측된 범위에서 실험한 결과 예측치 보다 조금 높게 나타났다.

Mapping quantitative trait loci controlling low-temperature germinability in rice

  • Low-temperature germination is one of the major determinants for stable stand establishment in the rice direct seeding method in temperate regions and at high altitude areas. Quantitative trait loci (QTL) controlling low-temperature germinability in rice were identified using 96 introgression lines (ILs) derived from a cross between Oryza rufipogon and the Korean japonica cultivar, 'Hwaseongbyeo'. The germination rate at $15^{\circ}C$ was measured to represent low-temperature germination and used for QTL analysis. The germination rate at $15^{\circ}C$ for 7 days of Oryza rufipogon and Hwaseongbyeo was 93.3 and 28.7%, respectively, and that of progenies ranged from 0 to 48%. A linkage map was constructed using 135 simple sequence repeat (SSR) markers. Five putative QTLs associated with low-temperature germination were detected on chromosomes 1, 3, 4, 10 and 11. The QTL, qltg10 on chromosome 10 accounted for 19.2% of the total phenotypic variation for low-temperature germinability. Four additional QTL, accounted for 10.4 - 15.1% of the total phenotypic variation. The O. rufipogon alleles in all detected QTLs loci increased the low-temperature germination rate. No QTL associated with low temperature germinability has been detected near the qltg10 QTL in this study suggesting that qltg10 is a new QTL. The locus, qltg10 is of particular interest because of its independence from undesirable height and maturity effects. The DNA markers linked to the QTL for low temperature germinability would be useful in selecting lines with enhanced low temperature germinability in rice breeding program.

벼 저온발아성의 효율적 검정 조건 (Effective Screening Condition for Low-Temperature Germinability of Rice)

  • 수도 저온발아성이 양호한 품종의 육성을 위한 효율적인 저온발아성 검정 조건을 모삭하기 위해 최근 육성된 신품종과 도입품종을 합하여 50품종을 공시, 1994년 영남농업시험장에서 13 , 16 , 25 에서 20일간 발아시험을 실시한 바, 그 결과를 요약하면 다음과 같다. 1. 13 , 16 , 25 에서 발아시킨 50품종들의 발아율 기초통계량 분석 결과, 13 11일 치상일에서 평균발아율이 43.8%, 발아율의 절위가 92, 그리고 분산이 748로 가장 높아 품종간 저온발아성의 차가 가장 현저한 조건이었다. 2. 13 11일 치상일의 발아율과 평균발아일수, 발아계수와의 상관에서 고도의 상관이 검증되어 13 11일 치상일의 발아율로써 저온발아성을 나타내는 지표로 시용 가능할 것으로 여겨진다. 3. 13 11일 치상일의 발아율을 근거로 공시품종들의 저온발아성을 구분해본 바, 동진벼 등 11품종이 저온발아성이 양호하였고 상주벼 등 17품종이 저온발아성이 매우 불량하였다. 4. 공시품종의 저온발아성과 몇가지 미질관련형질들과의 상관에서 출수 소요일수 및 아밀로스함량은 고도의 정의 유의상관이, 입폭과는 부의 유의상관이 인정되었다.

Some Physiological Activities Observed from Ethanol Extracts of Germinated Specialty Rough Rice

  • This study analyzed the some of the physiological activities of 70% ethanol extracts of specialty rough rice ('Goami2', 'Keunnunbyeo', and 'Heugkwangbyeo' rough rice cultivars) before and after germination. The physiological activities of the extracts were analyzed for their inhibition of linoleic acid peroxidation, hydrogen peroxide radical scavenging activity, superoxide dismutase-like activity, xanthine oxidase inhibitory activity, and nitrite scavenging effects. The most active rough rice cultivar was 'Heugkwangbyeo' and germinated 'Heugkwangbyeo' rough rice was more effective than ungerminated rough rice, having higher inhibition activity of linoleic acid peroxidation (1.2-fold), hydrogen peroxide radical scavenging activity (1.4-fold), superoxide dismutase-like activity (1.2-fold), and xanthine oxidase inhibitory activity (1.1-fold). The nitrite scavenging effect was increased after germination (2.1-fold) and decreased as pH increased. According to the results obtained in this study, 70% ethanol extracts from germinated 'Heugkwangbyeo' rough rice can be used for industrial applications such as the functional food materials.

벼 수발아의 품종간 차이와 왕겨 추출물질의 발아억제효과 (Varietal Difference of Viviparity and Germination - Inhibition of Rice Hull Extracts)

  • 수발아가 벼의 품질 및 정현 비율 감소에 미치는 영향, 수발아성의 품종간 차이 및 왕겨로부터 발아 억제물질을 추출하여 수발아 방제를 위한 기초자료를 얻고자 실시한 결과는 다음과 같다. 1. 현미중은 무처리에 비하여 수발아 처리일수가 길어질수록 감소되는 경향으로 6일 처리시 약 20% 감소되었다. 2. 수발아 처리일수에 따른 완전립 비율은 무처리가 99.5%인데 반해 6일 처리시 41.3%로 현저히 저하되었다. 3. 인디카품종은 대부분 난수발아성이었으나 자포니카(9.2∼79.7%) 및 통일형 품종(2.1∼63.7%)은 품종간 차이가 현저히 나타났다. 4. 난수발아성인 IR36, 신운봉벼 및 중원벼의 왕겨를 메탄올을 이용하여 추출한 물질이 발아억제에 효과가 있었다.

왕겨 바이오차 및 음식물쓰레기 바이오차가 밭 사양토에서 상추발아 및 수용성 유기탄소 용출에 미치는 영향 평가 (Evaluating germination of lettuce and soluble organic carbon leachability in upland sandy loam soil applied with rice husk and food waste biochar)

  • This study was carried out to evaluate the effect of rice husk (RHB) and food waste biochar (FWB) on upland soil with sandy loam texture, in terms of physico-chemical analysis, lettuce seed germination test, and orgainc carbon leaching experiment. RHB and FWB had different physico-chemical properties each other. Carbon to nitrogen ratio (C/N ratio) of RHB was 32, showing two times higher than that of FWB. FWB had high salt and heavy metal content, compared to RHB. This is probably due to different ingredients and production processing between two biochars each other. Results of germination test with Lettuce showed lower germination rate when FWB was applied because of higher salt concentration compared to control and RHB. Organic carbon leaching test using saturated soil column (${\Phi}75{\times}h75mm$) with $10MT\;ha^{-1}$ biochar application rate, showed higher saturated hydraulic conductivity in rice husk biochar treatment column, compared to control and food waste biochar treatment. The highest total organic carbon concentration in column effluent was lower than those in both of rice husk biochar and food waste biochar, whereas the differences was negligible after 9 pore volumes of effluent. Consequently, biochars from byproducts such as rice husk and food waste in sandy loam textured upland soil could enhance a buffer function such as reduction of leaching from soil, but the harmful ingredient to crops such as high salt and heavy metals could limit the agricultural use of biochars.

Changes of Germination Rate of Rice Seeds with Variable Status after 10-years of Storage Period

  • The seeds of rice with variable status were examined the germination rate after 10 years of long-term storage (-18℃) conservation. For seeds of wild relatives, 2 accessions were examined and germination rate of all accessions showed no changed after freezing conservation. For seeds of native germplasm, 1,259 accessions were examined and germination rate of 696 accessions was increased or showed no change after 10 years of long-term storage. Germination rate of 432 accessions was decreased with below 15% of initial germination rate. For 79 accessions of native germplasm, germination rate of seeds was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. For seeds of developed varieties, 873 accessions were examined and germination rate of 486 accessions of developed varieties was increased or showed no change after 10 years of long-term storage. Germination rate of 251 accessions was decreased with below 15% of initial germination rate. For 25 accessions of developed varieties, germination rate of seeds was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. For seeds of developed line, 2,131 accessions were examined and germination rate of 1,245 accessions of developed line was increased or showed no change after 10 years of long-term storage. Germination rate of 588 accessions was decreased with below 15% of initial germination rate. For 96 accessions, germination rate of seeds was decreased with above 15% of initial germination rate, which is needed to be rejuvenated.

Antioxidative and antiproliferative activities of ethanol extracts from pigmented giant embryo rice (Oryza sativa L. cv. Keunnunjami) before and after germination

  • BACKGROUND/OBJECTIVES Oxidative stress is a major cause of cancer. This study investigated the effects of the ethanol extracts from germinated and non-germinated Keunnunjami rice, a blackish-purple pigmented cultivar with giant embryo, on selected human cancer cell lines and on the antioxidant defense system of mice fed with a high-fat diet. MATERIALS/METHODS: High fat-fed mice were orally administered with either distilled water (HF) or extracts (0.25%, w/w) from brown (B), germinated brown (GB), Keunnunjami (K), and germinated Keunnunjami (GK) rice. RESULTS: In comparison with the brown rice extract, Keunnunjami extract showed higher anticancer effect against cervical and gastric cell lines but lower anticancer activity on liver and colon cancer cells. Mice from the HF group showed significantly higher lipid peroxidation and lower antioxidant enzyme activities than the control group. However, the oxidative stress induced by high-fat diet markedly decreased in B, GB, K, and GK groups as compared with the HF group. CONCLUSIONS: Germination may be an effective method for improving the anticancer and antioxidative properties of Keunnunjami rice and extracts from germinated Keunnunjami rice may serve as a therapeutic agent against cervical and gastric cancers and oxidative damage.
