Physicochemical Properties of Organic Liquid Fertilizer with Oil Cake and Rice Bran as Affected by Microorganism and the Ratio of Molasses

  • The study was conducted to investigate changes in the characteristics of inorganic components during fermenting process of organic liquid fertilizers according to the type and ratio of microorganism and the amount of molasses when producing organic liquid fertilizers using sesame oil cake and rice bran. To select appropriate microorganisms, liquid fertilizers were produced through a 90-day fermentation process by adding mag-ggeo-li, yogurt, dried yeast, and leaf mold. The pH in liquid fertilizer was decreased, and then increased in all microorganism samples except the mag-ggeo-li sample. The EC was rapidly increased in all samples until the $10^{th}$ days after production, and showed no changes after the $60^{th}$ days in dried yeast and after the $30^{th}$ days in the other samples. The concentration of $NH_4$-N was generally increased with time. The concentration of $P_2O_5$ was rapidly increased until the $10^{th}$ days after production and was maintained at about 1% regardless of the type of microorganism. In terms of the characteristics of liquid fertilizers according to the ratio of selected dried yeast, the pH was decreased until the $30^{th}$ days after producing the liquid fertilizers, and then was increased regardless of the ratio of dried yeast. The EC was increased with time and showed no differences depending on the amount of dried yeast. The concentration of $NH_4$-N was increased with time and in proportion to the amount of dried yeast. In terms of the characteristics of liquid fertilizers according to the ratio of molasses, the pH was decreased with increasing the molasses. The EC and concentration of $P_2O_5$ were no differences according to the amount of molasses. When 3% molasses was added, the content of $NH_4$-N was 2.6 mg $L^{-1}$ at the beginning and was at 3,025 mg $L^{-1}$ on the $90^{th}$ days.

Sterol Composition of Rice Bran Oil (미강유중(米糠油中)의 Sterol조성(組成))

  • The unsaponifiable from rice bran oil was fractionated into 4-desmethyl-'4-monomethyl- and 4, 4-dimethylsterol (triterpene alcohol) fraction by thin layer chromatography (TLC), and sterol composition of the each fraction was analyzed by gas liquid chromatography(GLC). The sterol peaks not well separated by GLC were further fractionated by $AgNO_3-TLC$, then analyzed using GLC. Each components in the three sterol fractions were identified by GLC and gas chromatography-maps spectrometry. As the results, ten sterols were confirmed as 4-desmethylsterol, nine as 4-monomethylsterol and four as 4, 4-dimethylsterol. Such uncommon phytosterols in higher plants as fucosterol, 24-ethyllophenol, 4${\alpha}$-methylstigmasta-7, 25-dienol and 28-isocitrostadienol were detected in rice bran oil and the biosynthetic pathways of the phytosterols were deduced with all the identified sterols.

Characteristics of Submerged and Solid-State Fermentations for Production of Arachidonic Acid Mortierella alpina (Arachidonic Acid 생산을 위한 Mortierella alpina 곰팡이의 심부 및 고체 발효 특성 연구)

  • The objective of this work was to evaluate a solid-state fermentation process for the practical production of arachidonic acid(AA) by Mortierella alpina ATCC 32222. In the present investigation, batch culture kinetics for both submerged- and solid-state fermentations was carried out at $25^{\circ}C$ to identify the relationship between growth and arachidonic acid (AA) production. Glucose and yeast extract were used in submerged fermentations by using flasks, while rice bran was used as a sole raw material in the other type of fermentations by using a series of Petri dishes. It was evident that a mixed-growth associated pattern existed between the two variables, irrespective of modes of fermentations. The effect of carbon to nitrogen (CfN) ratio on AA production in solid-state fermentation was studied in the range of 6.5 - 20. As a result, an optimum condition was found to be 6.5. Supplementary carbon source was not necessary to meet the optimum C/N ratio. Unlike the Previous results obtained by other researchers, a supplement of sodium glutamate up to $4\%$ (w/w) to the rice bran medium did not have a positive effect on the AA productivity. However, an increase in AA productivity was obtained with the rice bran medium supplemented with sesame oil.

Suppression of β-catenin and Cyclooxygenase-2 Expression and Cell Proliferation in Azoxymethane-Induced Colonic Cancer in Rats by Rice Bran Phytic Acid (PA)

  • Background: Phytic acid (PA) is a polyphosphorylated carbohydrate that can be found in high amounts in most cereals, legumes, nut oil, seeds and soy beans. It has been suggested to play a significant role in inhibition of colorectal cancer. This study was conducted to investigate expression changes of ${\beta}$-catenin and cyclooxygenase-2 (COX-2) and cell proliferation in the adenoma-carcinoma sequence after treatment with rice bran PA by immunocytochemistry. Materials and Methods: Seventy-two male Sprague-Dawley rats were divided into 6 equal groups with 12 rats in each group. For cancer induction two intraperitoneal injections of azoxymethane (AOM) were given at 15 mg/kg bodyweight over a 2-weeks period. During the post initiation phase, two different concentrations of PA, 0.2% (w/v) and 0.5% (w/v) were administered in the diet. Results: Results of ${\beta}$-catenin, COX-2 expressions and cell proliferation of Ki-67 showed a significant contribution in colonic cancer progression. For ${\beta}$-catenin and COX-2 expression, there was a significant difference between groups at p<0.05. With Ki-67, there was a statistically significant lowering the proliferating index as compared to AOM alone (p<0.05). A significant positive correlation (p=0.01) was noted between COX-2 expression and proliferation. Total ${\beta}$-catenin also demonstrated a significant positive linear relationship with total COX-2 (p=0.044). Conclusions: This study indicated potential value of PA extracted from rice bran in reducing colonic cancer risk in rats.

Effects of $^{60}Co-gamma$ Radiation on Ricebran Oil (미강유에 대한 코발트 60-감마선의 영향에 관한 연구)

  • The accumulation of peroxides, acid values, and carbonyl values during irradiation and post-irradiation storage of the ricebran oil has been studied. The rice bran oils were irradiated two doses of 2 and 7 megarads (300 rads/sec) at $23^{\circ}C$ atmospheric circumstance. The acid values, peroxide values and carbonyl values were measured at regular intervals of one week during the storage at $5^{\circ}C$ and $25^{\circ}C$. 1) During the storage, the acid values of the irradiated rice bran oils increased or decreased insignificantly regardless of the addition of antioxidants and storage temperature. 2) The peroxide values were not increased continuously but increased zigzag. The result was indicated that the composition and decomposition of peroxides occurred continuously throughout the storage. 3) As the peroxide values increased, carbonyl values decreased and changed quite differently, but, especially in 7th week, they were constant or insignificant. 4) Dibutylhydroxytoluene is more effective than caffeic acid in retarding the formation of peroxides during irradiation of rice bran oils and post-irradiation storage. The effect of antioxidant is more efficient at 2 megarads than at 7 megarads irradiation. When we store the rice bran oil, the addition of antioxidants of post-irradiation is more desirable than that of preirradiation. 5) In spite of changing conditions such as storage temperature and addition of antioxidants, the peroxide values of rice bran oils irradiated at 2 megarads were always greater than those at 7 megarads during the storage. Peroxide values of samples at high temperature $(25^{\circ}C)$ storage increased as twice as those of low temperature $(5^{\circ}C)$ storage samples. At low temperature, peroxide values in the first week increased twice during the period of 8th weeks storage, but those did from three to four times at higher temperature in the same period Therefore, the low temperature storage is recommandable too.

Identification of Quantitative Trait Loci for Fatty Acid Content in Brown Rice (Oryza sativa L.)

  • The rice bran oil contained in brown rice is composed of highly valued ingredient. Improving the content of unsaturated fatty acids in rice seed, such as oleic acid, linoleic acid, and ${\alpha}$-linolenic acid, would provide more benefit to human health. Fatty acid content is quantitative trait controlled by multiple genes. We have utilized high-density SNP data from highly advanced breeding populations to identify QTLs for fatty acid contents in brown rice. Here, we identified 51 major QTLs (M-QTLs) and 25 epistatic QTLs (EpQTLs) related to eleven fatty acid contents. Eight and four M-QTLs were pleiotropically associated with the content of different fatty acids in MT-RILs and DT-RILs, respectively. Total effect of M-QTLs for palmitic acid (16:0), oleic acid (18:1), and linoleic acid (18:2), could explain phenotypic variations of 36.7%, 63.7%, and 41% in MT-RILs, respectively. Alpha-linolenic acid which is important for a human's health could be explained phenotypic variation of 15.7% by six M-QTLs. These QTLs identified in this study can be used to improve nutritious content in rice breeding programs.

Nitrogen mineralization of oil cakes according to changes in temperature, moisture, soil depth and soil texture (유박의 온도, 수분, 토심, 및 토성에 따른 질소의 무기화)

  • To investigate the characteristics of the nitrogen mineralization of oil cakes according to changes in temperature(10, 20, $30^{\circ}C$) moisture(40, 50, 60, 70% of field capacity in loam and 50, 60, 70, 80% of field capacity in sandy loam), mineral nitrogen was measured in soil after incubation for 30 days. In addition, the mineralization in sandy loam and loam which had different soil texture were compared. According to incubating the castor seed, soybean, and rice bran cakes with soil, the higher the temperature and moisture content were, the higher the content of mineral nitrogen was observed. The content of mineral nitrogen was higher in sandy loam than loam. The content of mineral nitrogen was decreased with soil depth increasing and was also higher in sandy loam than loam.
