In order to investigate the optimum moisture content of paddy for milling process, a series of tests were conducted by examining the recovery rate and whiteness of milled rice in relation with the various moisture content. Hwabong-byeo and Dongjin-byeo varieties which were major paddies cultivated in Korea were used for the experiment. The test was performed with small experimental milling machines. In order to minimize the unexpected factors, environment conditions were kept in constant during the experiment. As a result, the recovery rate of milled rice were varied as the changes in milling time and degree of whiteness. However, the recovery rate of milled rice increases as its moisture content increases untill a certain point of moisture content and decreases slowly afterward. This certain point can be called optimum moisture content for rice milling. Also, it has a different value depending on the variety. In this experiment, optimum moisture content of Hwabong-byeo and Dongjin-byeo were considered around 14.8% and 15.3%, respectively. It is not sure that these optimum moisture contents for the two varieties would assume the same values irrespective of harvest year and place. However, it could be concluded that the optimum moisture content for rice milling is around 15%(w.b.) for Hwabong-byeo and 15.5%(w.b.) for Dongjin-byeo, respectively.
To study the effects of different nursing methods and transplanting on the growth of rice plant (Oriza sativa L.) in southern region of Korea, Kumo-byeo, Palgong-byeo and Dongjinbyeo were transplanted from April 20 to July 20 at an interval of 15 days with 8 days old seedling (infant seedling) and 25 days old box-seedling for machine transplanting, and 45 days old conventional seedling. Threshold transplanting date in southern region of Korea were June 26 for 8 days old seedling, July 1 for 25 days old seedling and] July 11 for 45 days old seedling for Kumo-byeo, and June 21, June 30, July 10 for Palgong-byeo, June 10, June 24, July 5 for Dongjin-byeo, respectively. Yield has no uniform tendency according to the transplanting date. However, yield were greater in the order of 8 days old seedling >25 days old seedling> 45 days old seedling in Kuma-byeo and 25 days old seedling (equation omitted)8 days old seedling (equation omitted)45 days old seedling in Palgong-byeo, 45 days old seedling(equation omitted)25 days old seedling(equation omitted) 8 days old seedling in Dongjin-byeo. The optimum accumulated air temperature during yield productive stage around heading (40 days from 10 days before heading to 30 days after heading) for high yield were 1,003$^{\circ}C$ for 8 days old seedling, 1,014$^{\circ}C$ for 25 days old seedling and 1,027$^{\circ}C$ for 45 days old seedling in Kumo-byeo. And they were 1,018$^{\circ}C$, 1,015$^{\circ}C$, 1,086$^{\circ}C$ in Palgong-byeo and 998$^{\circ}C$, 984$^{\circ}C$, 949$^{\circ}C$ in Dongjin-byeo, respectively. Earlier transplanting with 8 days old seedling showed higher ratio of broken rice and green kerneled rice in Kuma-byeo, and late transplanting after July 5 showed significant high rate of green kerneled rice. Palgong-byeo and Dongjin-byeo also showed high rate of green kerneled rice at transplanting after July 5.

The effect of genetically engineered rice Bt-9 on the diversity and abundance of plant-dwelling insects and spiders was tested under field conditions. Genetically engineered rice Bt-9, expressing mCry1Ac1 from Bacillus thuringiensis, confers resistance to rice leaf roller (Cnaphalocrocis medinalis) and provides tolerance to the herbicide glufosinate (PPT). The study compared Bt-9 and two non-GM reference varieties, Ilmi-byeo and Dongjin-byeo, at LMO isolated fields in Gunwi (Kyungpook National University) and Jeonju (National Institute Agricultural Sciences) in Southern Korea in 2016 - 2017. A total of 40,817 individuals from 62 families and 11 orders were collected from the two living modified organism (LMO) isolated fields. From the three types of rice fields, a total of 13,982, 14,105, and 12,730 individuals from the Bt-9, Ilmi-byeo and Dongjin-byeo were collected, respectively. Throughout the study, the analysis of variance indicated no significant differences (p < 0.05). Multivariate analysis showed that the abundance and diversity of plant dwelling insects were similar. The data on insect species population densities were subjected to principal component analysis (PCA), which did not distinguish among the three varieties, Bt-9 and the non-GM, reference cultivars, during the cultivation years. However, the results of the PCA analysis were completely divided into four groups based on the yearly survey areas. Therefore, there was no evidence for a negative impact of Bt-9 on the above-ground insects and spiders.
The physicochemical and gelatinization properties of rice flours, which were prepared by polishing nonwaxy brown rice (Dongjin byeo), depending on polishing degrees were investigated. The contents of protein, ash, lipid and fiber decreased significantly as the degrees of polishing increased and water uptake of rice grains decreased during hydration. When the surface of the soaked rice grains was examined by the SEM, extensive crack formation was observed. The water binding capacities, swelling power and solubility of rice flour increased as the polishing degree increased. By the amylogram, the initial gelatinization temperatures of rice flour decreased as the polishing degree increased and the peak viscosities, the breakdown and consistency of rice flour increased as the polishing degree increased.

This study was conducted to develop environmental risk assessments and biosafety guides for insect-resistant genetically modified rice in an LMO (Living Modified Organism) isolation field. In the LMO quarantine area of Kyungpook National University, the species diversities and population densities of non-target insects found on insect-resistant genetically modified rice (Bt-T), rice resistant to Cnaphalocrocis medinalis, and non-GM rice (Dongjin-byeo and Ilmi-byeo) were investigated. The Bt-T plants were, therefore, evaluated under field conditions to detect possible impacts on above ground insects and spiders. In 2016 and 2017, the study compared transgenic rice and two non-GM reference rice, namely Dongjin-byeo and Ilmi-byeo, at Gunwi. A total of 9,552 individuals from 51 families and 11 orders were collected from the LMO isolation field. From the three types of rice fields, a total of 3,042; 3,212; and 3,297 individuals from the Bt-T, Dongjin-byeo, and Ilmi-byeo were collected, respectively. There was no difference between the population densities of the non-target insect pests, natural enemies, and other insects on the Bt-T compared to non-GM rice. The data on insect species population densities were subjected to principal component analysis (PCA) without distinguishing between the three varieties, namely GM, non-GM, and reference cultivar, in all cultivation years. However, the PCA clearly separated the samples based on the cultivation years. These results suggest that insect species diversities and population densities during plant cultivation are determined by environmental factors (growing condition and seasons) rather than by genetic factors.
To study the effects of different nursing methods and transplanting on the growth of the rice plant(Oriza sativa L.hn southern region of Korea, Kumo-byeo, Palgong-byeo and Dong-jinbyeo were transplanted from April 20 to July 20 with 15 days intervals as 8 days seedling (infant rice), 25 days box seedling for machine transplanting and 45 days conventional seedling, respectively. No. of leaves at transplanting were in order of 45 days seedling>25 days seedling>8 days seedling. In 25 days seedling, they were increased as transplanting dates were later from April 20 to June 5, and in 45 days seedling, it showed same tendency until June 5 with Dongjinbyeo, June 20 with Palgong-byeo and Kumo-byeo. Root activity according to the $\alpha$-naphthylamine oxidation activity and rooting activity were higher in order of 8 days seedling>25 days seedling>45days seedling. Panicle formation initiated after maximum tillering stage as transplanting at April 20 with 8 days seedling and 25 days seedling, but it intiated before maximum tillering stage as transplanting at July 20 in Kumo-byeo. and in Palgong-byeo intiations of panicle formation were shown after maximum tillering stage as transplanting until May 20, but June 5 in Dongjin-byeo. However, there was no significant tendency with 45 days seedling. Missing hill ratio were less than 5% as transplanting with 8 days seedling and 25 days seedlingregardless of transplanting periods or varieties.
Physicochemical properties of nonwaxy (Dongjin byeo) and waxy (Sin-seunchal byeo) brown rice flour were investigated. The proximate composition of nonwaxy brown rice flour was 7.02% of protein, 1.14% of ash, 3.2% of lipid and that of wfny brown rice was 7.74% of protein, 1.48% of ash,4.83% of lipid. X-ray dif- fraction patterns were A types, and relatives crystallinity of nonwaxy brown rice flour was higher than that of waxy brown rice flour. Swelling power and solubility increased with the increase of temperature, waxy brown rice flour showed higher than nonwaxy rice flour until 70$^{\circ}C$, but lower after 80$^{\circ}C$. The gelatinization temperature, breakdown and consistency of nonwaxy brown rice flour by amylogram were higher than those of the waxy brown rice flour but setback was same. The initial gelatinization temperature and the enthalpy of nonwaxy brown rice flour by DSC were higher than those of the waxy brown rice flour.
Physicochemical properties of native and defatted nonwaxy(Dongjin byeo) and waxy(Sinseunchalbyeo) rice starches were investigated. The granule shapes of rice starches were polygonal and X-ray diffraction patterns were A types, but relative crystallinity was decreased by defatting. The amylose content of defatted starches slightly increased, but water binding capacity of defatted starches decreased. Swelling power and solubility of starches increased with the increase of temperature, at each temperature increased by defatting. Transmittance of Dongjinbyeo and Sinseunchalbyeo starch suspensions showed a rapid increase at $60{\sim}65^{\circ}C$, $55{\sim}60^{\circ}C$ respectively. The initial pasting temperature by amylograph of Dongjinbyeo and Sinseunchalbyeo starches were $66^{\circ}C$ and $64^{\circ}C$, respectively. The gelatinization temperature of defatted starches was higher than that of the nondefatted starches. Dongjinbyeo starch decreased peak viscosity and breakdown by defatting, but Sinseunchalbyeo starch unchanged.
Rice (Dongjin-byeo) added with black rices (Chindo and Suwon-415) was cooked and the changes of texture and color of this cooked rice were investigated by texture analyser and color & color difference meter. The hydration time to come to the equilibrium condition was at least 11 hr in black rices. The optimum ratio of water to cooked rice added with black rice was 1.6 (ratio of water to rice) and the hardness of cooked rice added with 5% Chindo black rice was $5.66\;kg_f$. Regardless of ratio of water to rice, Hunter a value increased as the ratio of black rice addition to rice increased, while L value decreased. The color elution rate of Suwon 415 was 4 times greater than that of Chindo black rice. The pH of the steep water of Suwon 415 at $20^{\circ}C$ during 120 min decreased from pH 6.40 to pH 6.16 as the steeping time increased. The optimum heating time by microwave oven of cooked rice added with black rice was between $90{\sim}120\;sec$ to recover the original texture after cold storage treatment of 7 days.

Transposon-mediated insertional mutagenesis provides one of the most powerful tools for functional studies of genes in higher plants. This project has been performed to develop a large population of insertional mutations, and to construct databases of molecular information on Ds insertion sites in rice. Ultimate goals are to supply genetic materials and information to analyze gene function and to identify and utilize agronomically important genes for breeding purpose. Two strategies have been employed to generate the large scale of transposon population in a Japonica type rice, Dongjin Byeo; 1) genetic crosses between Ac and Ds lines and 2) plant regeneration from seeds carrying Ac and Ds. Our study showed that over 70% of regenerated plants generally carried independent Ds elements and high activity of transposition was detected only during regeneration period. Ds-flanking DNA amplified from leaf tissues of F2 and T1 (or T2) plants have been amplified via TAIL-PCR and directly sequenced. So far, over 65,000 Ds lines have been generated and over 9,500 Ds loci have been mapped on chromosomes by sequence analysis. Database of molecular information on Ds insertion sites has been constructed, and has been opened to the public and will be updated soon at http://www.niab.go.kr. Detailed functional analysis of more than 30 rice mutants has been performed. Several Ds-tagged rice genes that have been selected for functional analysis will be briefly introduced. We expect that a great deal of information and genetic resources of Ds lines would be obtained during the course of this project, which will be shared with domestic and international rice researchers. In addition to the Japonica rice, we have established the tagging system in an rice line of indica genetic background, MGRI079. MGRI079 (Indica/Japonica) was transformed with Agrobacteria carrying Ac and Ds T-DNA vectors. Among transgenic lines, we successfully identified single-copy Ds and Ac lines in MGR1079. These lines were served as ‘starter lines’ to mutagenize Indica genetic background. To achieve rapid, large scale generation of Ds transposant lines, MGR1079 transformants carrying homozygous Ac were crossed with ones with homozygous Ds, and $F_2$seeds were used for plant regeneration. In this year, over 2,000 regeneration plants were grown in the field. We are able to evaluate the tagging efficiency in the Indica genetic background in the fall.
