EXPRESSION OF BMP-2, -4 DURING DISTRACTION OSTEOGENESIS IN THE RABBIT MANDIBLE (집토끼 하악골에서 신연 골형성술시 BMP-2,-4의 발현)

  • Distraction osteogenesis(DO) is defined as a gradual mechanical process of mechanical stretching two vascularized bone surface apart with a critical rate and rhythm such that new bone forms within the expanding gap, reliably bridges the gap, and ultimately remodels to normal structure. DO has become a mainstay in bone tissue engineering and has significantly improved our armamentarium for reconstructive craniomaxillofacial procedures. But the molecular and biological mechanisms that regulate the formation of new bone during distraction osteogenesis are not completely understood. BMPs are potent osteoinductive agents. Our hypothesis was that BMPs, especially BMP-2 and BMP-4, might play an importent role in the signaling pathways that link the mechanical forces created by distraction to biological responses and in promting new bone formation. Using a rabbit's mandible, we investigated the expression of BMP-2, -4 at different time points during distraction osteogenesis. The purpose of this study is to research the pattern of expression of BMP-2, -4 in new bone formation during distraction osteogenesis of the rabbit mandible. The experimental group was applied gradual distraction (0.7mm a day by twice a day, 4.9mm in total, for 7 days) and the control group was carried out osteotomy alone. They were examined clinically, histologically, and by RT-PCR analysis. On 3 days after osteotomy, the high level of expression of BMP-2, -4 was detected. But, the expression of BMP-4 was decreased during latency period. As distraction was started, its expression was increased and maintained till postoperative 28days. In control group, the expression of BMP-4was remarkably decreased till postoperative 14 days. On the other hand, the expression of BMP-2 was no difference between experimental group and control group. The expression of BMP-4 was maintanined at high level during the entire experimental period in both group. These findings suggested that excellent bone formation during distraction osteogenesis is associated with enhanced expression of BMP-4 genes by mechanical tension stress.

Spaciotemporal Plasticity of Intergeniculate Leaflet Using Genetically Modified Pseudorabies Virus Recombinant (유전자 조작된 Pseudorabies Virus 변종을 이용한 무릎사이작은핵의 시.공간적 가소성)

  • This study was carried out to investigate the spaciotemporal plasticity of intergeniculate leaflet in postnatal mongolian gerbil using genetically modified pseudorabies virus recombinant, which was a kind of excellent neurotracer with the ability to transpass the neuronal synaptic cleft. In addition, we tried to evaluate the special role of intergeniculate leaflet as a signal controler of circardian rhythm by expression of various nourotransmitters in suprachiasrnatic nucleus. The PRV-BaBlu, a genetically modified strain of PRV-Bartha with lac-Z gene, was injected into vitreous body of postnatal mongolian gerbil, and immunostained. The PRV-Bablu infected the neurons in intergeniculate leaflet of postnatal mongolian gerbil, and the degree of viral infection in postnatal period of experimental animals had tendency to increase with time consuming. This results showed that the mutant PRV-Bar-tha strain with lac-Z gene, PRV-BaBlu, was a very excellent neurotracer to localize the retinogeniculate tract with infection of neurons in intergeniculate leaflet specially.

Diversity and Function of Retinal-binding Protein in Photosynthetic Microbes

  • Photosynthetic microbes possess a wealth of photoactive proteins including chlorophyll-based pigments, phototropin-related blue light receptors, phytochromes, and cryptochromes. Surprisingly, recent genome sequencing projects discovered additional photoactive proteins, retinal-based rhodopsins, in cyanobacterial and algal genera. Most of these newly found rhodopsin genes and retinal synthase have not been expressed and their functions are unknown. Analysis of the Anabaena and Chlamyrhodopsin with retinal synthase revealed that they have sensory functions, which, based on our work with haloarchaeal rhodopsins, may use a variety of signaling mechanisms. Anabaena rhodopsin is believed to be sensory, shown to interact with a soluble transducer and the putative function is either chromatic adaptation or circadian rhythm. Chlamydomonas rhodopsins are involved in phototaxis and photophobic responses based on electrical measurements by RNAi experiment. In order to analyze the protein, we developed a sensory rhodopsin expression system in E. coli. The opsin in E. coil bound endogenous all-trans retinal to form a pigment and can be observed on the plate. Using this system we could identify retinal synthase in Anabaena PCC 7120. We conclude that Anabaena D475 dioxygenase functions as a retinal synthase to the Anabaena rhodopsin in the cell.

A Time to Fast, a Time to Feast: The Crosstalk between Metabolism and the Circadian Clock

  • The cyclic environmental conditions brought about by the 24 h rotation of the earth have allowed the evolution of endogenous circadian clocks that control the temporal alignment of behaviour and physiology, including the uptake and processing of nutrients. Both metabolic and circadian regulatory systems are built upon a complex feedback network connecting centres of the central nervous system and different peripheral tissues. Emerging evidence suggests that circadian clock function is closely linked to metabolic homeostasis and that rhythm disruption can contribute to the development of metabolic disease. At the same time, metabolic processes feed back into the circadian clock, affecting clock gene expression and timing of behaviour. In this review, we summarize the experimental evidence for this bimodal interaction, with a focus on the molecular mechanisms mediating this exchange, and outline the implications for clock-based and metabolic diseases.

  • The aims of this article is to argue that the effect of catharsis through satire and humor in Korean music, Saseolnanongga. This song is a cheerful song of four beats and the representative rap music of Korean traditional music. This song is a folk song that reveals satire and humor such as curse to the person who left me and compassion and self-absorption. The lyrics harmonize with cheerful rhythm and create laughter and the lyrics of the song itself alone cause music therapy. The contents of the lyrics of the song are already refreshing. It means that you will be able to satisfy your surrogacy through the explicit lyrics. In this way, author thinks emotional expression through singing lyrics is an alternative solution. There has been a cathartic element in many twentieth-century approaches to psychological healing. The author thinks developmental criticism leads society to health, and satirical criticism reflects and heals individuals through catharsis.

A study of Movement on Modern Fashion (현대 의상에 나타난 움직임의 표현성)

  • The purposes of this research are firstly to define the characteristics of movement on the costume, secondly to demonstrate that fine art, expecially in relation to kinetic art, has become a good motif in modern costume de-sign, and thirdly to examin the characteristics of movement as shown in modern costume de-sign. For this study, we selected and analyzed some costume designs among those introduced in fashion magazines, which are Collections, Fashion-Preview, Fashion Show, Collezioni, and L'Officiel that have been issued since 1988. As this research is carried out only through the analysis of pictures, it is not ac-companied by a study on material itself. The result of this study is that the ex-pression of movement on modern fashion can be shown in two ways, one is the inclusive and subordinate movement expressed on the cloth-ing according to the motion of body when they are worn, and the other is the movement caused by the very designs of clothing. And the latter can be also divided in two ways, the illusionistic movement and the actual move-ment. The expression of actual movement is made through the emphasis of direction, rhythm, and dimension of the costume. And the illusionistic movement is expressed by the de-sign to which the textile pattern or reflection of light is applied so that the movement is larger than the actual movement. A sense of direction expressed on the cos-tume by the actual movement creates a light rhythm and adds interest to the costume. This reflects a part of psychology of modern men who are not accustomed to the statics. In the rapidly changing society of information our modern men seek after movement even in the statics. It is also true in the clothing and they seem to get a psychological rest through the immediated visual change as shown in fashion design. The simple and cheerful rhythm expressed by the illusionistic movement on the costume is attempted in order to overcome the static tediousness caused by simple design. These elements of movement cause the en-largement of visible range and create another enlarged shape other than the real one, so that they come to remove the dullness and monot-ony in costume design and provoke interest in costume, as if kinetic art reveals the various aspects that transcend the closed area of fine art upon the motif of 'movement'. finally, as this paper deals with the ex-pression of movement as shown in the costume design which prevailed after 1988, it is desir-able hereafter to study more about the fashion since 1950 when kinetic art began in earnest, along with the comparative study on the ex-pression of movement in our Korean designer's and foreign designer's works.

Gut Microbial Metabolites Induce Changes in Circadian Oscillation of Clock Gene Expression in the Mouse Embryonic Fibroblasts

  • Circadian rhythm is an endogenous oscillation of about 24-h period in many physiological processes and behaviors. This daily oscillation is maintained by the molecular clock machinery with transcriptional-translational feedback loops mediated by clock genes including Period2 (Per2) and Bmal1. Recently, it was revealed that gut microbiome exerts a significant impact on the circadian physiology and behavior of its host; however, the mechanism through which it regulates the molecular clock has remained elusive. 3-(4-hydroxyphenyl)propionic acid (4-OH-PPA) and 3-phenylpropionic acid (PPA) are major metabolites exclusively produced by Clostridium sporogenes and may function as unique chemical messengers communicating with its host. In the present study, we examined if two C. sporogenes-derived metabolites can modulate the oscillation of mammalian molecular clock. Interestingly, 4-OH-PPA and PPA increased the amplitude of both PER2 and Bmal1 oscillation in a dose-dependent manner following their administration immediately after the nadir or the peak of their rhythm. The phase of PER2 oscillation responded differently depending on the mode of administration of the metabolites. In addition, using an organotypic slice culture ex vivo, treatment with 4-OH-PPA increased the amplitude and lengthened the period of PER2 oscillation in the suprachiasmatic nucleus and other tissues. In summary, two C. sporogenes-derived metabolites are involved in the regulation of circadian oscillation of Per2 and Bmal1 clock genes in the host's peripheral and central clock machineries.

A Novel Phototransduction Pathway in the Pineal Gland and Retina

  • Light is a major environmental signal for entrainment of the circadian clock, but little is known about the phototransduction pathway triggered by light-activation of photoreceptive molecule(s) responsible for the phase shift of the clock in vertebrates. The chicken pineal gland and retina contain the autonomous circadian oscillators together with the photic entrainment pathway, and hence they provide useful experimental model for the clock system. We previously demonstrated the expression and light-dependent activation of rod-type transducin $\alpha$-subunit (Gtl$\alpha$) in the chicken pineal gland. It is unlikely, however, that the pineal Gt$_1$$\alpha$ plays a major role in the photic entrainment, because the light-induced phase shift is unaffected by bloking the signaling function of Gt$_1$$\alpha$. Here, we show the expression of G 11 $\alpha$, an $\alpha$-subunit of another heterotrimeric G-protein, in the chicken pineal gland and retina by cDNA cloning, Northern blot and Western blot analyses. GIl$\alpha$-immunoreactivity was colocalized with pinopsin in the chicken pineal cells and it was found predominantly at the outer segments of photoreceptor cells in the retinal sections, suggesting functional coupling of G11 $\alpha$ with opsins in the both the tissues. By coimmunoprecipitation experiments using the retina, we showed the light- and GTP-dependent interaction between rhodopsin and G11 $\alpha$. Upon ectopic expression of a Gq/ 11-coupled receptor in cultured pineal cells, pharmacological (non-photic) activation of endogenous G11 induced phase-dependent phase shifts of the melatonin rhythm in a manner very similar to the effect of light. These results suggested opsin-G11 pathway contributing to the photic entrainment of the circadian clock.

  • The techno fashion presents the new formative beauty of fashion with a stream of light and dynamic phenomenon on human body. Also it opens the futurism arising from the combination of technology with fashion. The purpose of this paper was to investigate the internal trend and external form that techno-fashion aims at, and to analyze the expressive characteristics in design. The results were as follows. ${\cdot}$ The techno fashion created the new formative artifact through the dismantlement and reorganization of form. ${\cdot}$ The techno fashion used the composition of the geometrical abstraction as an expression of beauty symbolizing the modernity. ${\cdot}$ The techno fashion created the diverse colors by using the light along with colors of gold, silver, metal, intense fundamental colors, and artificial rotor. ${\cdot}$ The techno fashion expressed the dynamism, brilliance, youthfulness, futuristic image by using the sense of dazzling brightness by means of metal. ${\cdot}$ The techho fashion along with the development of science and technology made the atmosphere of silhouette of clothes different by expanding the category of materials more widely, together with the possibility of continual emergence of new material. ${\cdot}$ The techno fashion expressed the dynamic movement on clothes and gave the sense of periodical rhythm by the reflective action of light when they were clad. ${\cdot}$ The techno fashion grafted wearable technology into the fashion and made the information instruments recognized as a concept of clothes. ${\cdot}$ The techno fashion pursued the values of not only the beauty of the simple geometrical design, but also the values of functionality and expressed the image of high quality of life through the harmony of technology with human.

A Study on Metal Molding Using Line Transformation and Repetition - Using phosphor bronze wire (선의 변형과 반복을 활용한 금속조형 연구 - 인청동선을 활용하여)

  • This study, which was begun in order to create a metal molding by using lines, a basic element of design, examined various characteristics and concept of line and the beauty of lines based on previous research on the subject. The result showed that formative interpretation of lines largely depends on intentional expression by using thickness and rhythm of lines. In order to obtain a metal molding that provides both visual freedom and beauty, rather than the conventional formative expression of lines, I used red copper plate and Phosphor bronze line for creating a pattern based on transformation and repetition of lines and an artwork based on the pattern. The result was not only a creation of a plane but the senses of three-dimension, speed, and unity in the overall shape. Further research will need to be conducted regarding formative interpretation of lines and emotional and intrinsic beauty through expression of lines.
