Measuring elastic modulus of bacterial biofilms in a liquid phase using atomic force microscopy

  • With the increasing interest in using bacterial biofilms in geo-engineering practices, such as soil improvement, sealing leakage in earth structures, and hydraulic barrier installation, understanding of the contribution of bacterial biofilm formation to mechanical and hydraulic behavior of soils is important. While mechanical properties of soft gel-like biofilms need to be identified for appropriate modeling and prediction of behaviors of biofilm-associated soils, elastic properties of biofilms remain poorly understood. Therefore, this study investigated the microscale Young's modulus of biofilms produced by Shewanella oneidensis MR-1 in a liquid phase. The indentation test was performed on a biofilm sample using the atomic force microscopy (AFM) with a spherical indentor, and the force-indentation responses were obtained during approach and retraction traces. Young's modulus of biofilms was estimated to be ~33-38 kPa from these force-indentation curves and Hertzian contact theory. It appears that the AFM indentation result captures the microscale local characteristics of biofilms and its stiffness is relatively large compared to the other methods, including rheometer and hydrodynamic shear tests, which reflect the average macro-scale behaviors. While modeling of mechanical behaviors of biofilm-associated soils requires the properties of each component, the obtained results provide information on the mechanical properties of biofilms that can be considered as cementing, gluing, or filling materials in soils.

Dispersive Activity and Physical Properties of Exopolysaccharide (Cystan) produced by Cystobacter sp. MSL-9

  • The dispersing activity and physical properties of exopolysaccharide (Cystan) produced by Cystobacter sp. MSL-9 was studied. Dispersing activity of Cystan was measured by using bentonite, calcium carbonate, and kaolin. Rheological properties of Cystan were investigated by using a rheometer MCR300. It was found that dispersing activity of Cystan was maintained under various pH and temperature conditions. A particle size analyzer BI-9000AT was used for the particle size measurement of Cystan in aqueous solution. The high dispersing activity and stability under various pH and temperature conditions showed the feasibility of Cystan as a industrial biodispersant.

  • A new, commercially available polishing process called magnetorheological finishing is used to polish and figure precision optics. To understand and model this process correctly it is important to determine the mechanical properties of the fluid under the influence of the magnetic field. Magnetorheological (MR) fluids are commonly modeled as Bingham fluids, so one of the essential properties to measure is the yield stress. Since MR fluids are inherently anisotropic, the yield stress will depend on the mutual orientation of the magnetic field and the direction of deformation. The relative orientation of the field and deformation in polishing does not coincide with common rheological setups, so a new rheometer has been designed and tested. This new magnetorheometer design has been shown to give correct stresses during calibration experiments using Newtonian fluids with a known viscosity. The measured stress has also been shown to have a magnitude consistent with published finite element approximations for magnetic fluids. The design of the instrument was complicated because of the requirements imposed upon the magnetic field, and the difficulty in satisfying the no slip boundary condition. Our results show the importance of having a homogeneous field in the test region during measurements. The solutions to these problems and discussion of the measurements on nonmagnetic and magnetic fluids are given.

A Study on the Storage Stability of Waste Vinyl-Modified Asphalt (폐비닐로 개질된 아스팔트의 저장안정성에 관한 연구)

  • It is said that polymer modified asphalt using polyethylene as modifier would show phase separation due to density difference and incompatibility between asphalt and polyethylene. In this study, to prevent coalescence of polyethylene in asphalt, we employed peroxides as phase separation inhibitor. On microscope, peroxides (dicumyl peroxide, lauroyl peroxide) with waste vinyl (comprising low density polyethylene) did not show phase separation, however, rheometer test showed phase separation at molecular level, i.e., polyethylene and asphalt are immiscible ultimately. Mechanical properties (tensile strength, Marshall stability, dynamic stability) showed waste vinyl-modified asphalts are highly resistant to plastic deformation and these properties are even better than those of Superphalt.

Study of WMA Additive's Compaction Characteristics in Terms of Temperature Change by Using DSR (DSR을 이용한 온도변화에 따른 중온화 첨가제의 다짐특성 연구)

  • PURPOSES : This study is to develop a method to evaluate the compaction effects of asphalt binders using WMA additives and compare their compaction effects on two types of WMA additives, two types of testing temperatures, and three types of asphalt film thicknesses. METHODS : This study is based on laboratory experiments and rheological analysis of the experimental results. Testing materials are aggregate disks, asphalt, and WMA additives. The main testing method is the stress sweep test by using dynamic shear rheometer (DSR). In addition, the testing parameters obtained from the stress sweep results to evaluate lubrication effects are complex modulus and LVE-Limit. RESULTS : At both the first compaction condition ($110^{\circ}C$, 0.3mm) and second compaction condition ($80^{\circ}C$, 0.2mm) assumed, LEADCAP showed better compaction effects than Sasobit. CONCLUSIONS : The temperature $30^{\circ}C$ lower than general compaction temperatures can provide a better sensitivity for the evaluation of compaction effects. If a testing temperature and film thickness are grouped for the proper compaction conditions in the testing results, the compaction performance of each WMA additive can be more clearly discriminated in the grouped testing results matched with the grouped conditions.

  • It is generally agreed that strong linkages exist between rubber chains and reinforcing filler particles. The purpose of this study is to examine the effect of rubber-filler attachments on the various mechanical properties of the rubber. In particular, the modulus and strength will be altered by these attachments. For this study, the curing properties are examined by means of ODR (Oscillating Disk Rheometer), the physical properties by means of Instrong. The results of this study can be summarized as follows. In the ODR test, the carbon black filled stock has shorter scorch time than gum stock and, the silica and clay filled stock has longer scorch time than gum stock. In the modulus, ten sile and swelling properties, the vulcanizates filled with carbon black had higher those values than inorganic filler loaded NR vulcanizates, but the rebound rate showed that the silica filled NR vulcanizates was lower than other inorganic filler contained NR vulcanizates and the ISAF filled NR vulcanizates was lower than other carbon block contained NR vulcanizates.

  • The purpose of this study is to investigate the reinforcement of inorganic filler silica, treated by MDI about SBR vulcanizate. The characteristics of vulcanization, physical properties, surface properties and dynamic properties were investigated after mixing those silica with SBR and unmodified silica with SBR. In this experiment only the quantity of silica was variable. In the vulcanization characteristics tested by rheometer, S-series showed the fastest scorch $time(t_{10})$ and optimum cure $time(t_{90})$. And in test or tensile characteristics hardness, tensile strength, 100%, 300% modulus and elongation were all appeared in the order of M>S-series. The characteristic bonding of urea between unmodified silica and MDI could be confirmed in IR spectrum. The shapes of silicas treated chemically were observed by SEM. And the dispersion of the filler in the SBR composite was uniform. In the dynamic characteristics by the RDS, the order of elastic modulus G' values was as follows : M>S-series, and also the order of damping values was as follows : M>S-series.

  • Mechanical properties of 9-30% starch gels from naked and covered barley were investigated with rheometer or rotation viscosimeter. The compression-penetration curves of 20 and 30% gels were characterized by deformations containing elastic, plastic and fracture regions under the load of 0-2kg. The compressive stress relaxation test showed that the viscoelastic properties of 20% gels may be represented by four element Maxwell model consisting of two Maxwell element in parallel. Also, stress-decay under the steady shear of 9% covered starch gel was able to be interpreted by linear viscoelastic model and stress-decay process was suggested to be effective to investigate the effect of temperature or additives on gel structure.

Mechanical Properties of Styrene-Butadiene Rubber Reinforced with Hybrids of Chitosan and Bamboo Charcoal/Silica

  • Chitosan-polyvinyl alcohol (PVA) -bamboo charcoal/silica (CS-PVA-BC/SI) hybrid fillers with compatibilized styrene-butadiene rubber (SBR) composites were fabricated by the interpenetrating polymer network (IPN) method. The structure and composition of the composite samples were characterized by scanning electron microscope (SEM) and Fourier transform infrared spectroscopy (FT-IR). The viscoelastic behaviors of the rubber composites and their vulcanizates were explored using a rubber processing analyzer (RPA) in the rheometer, strain sweep and temperature sweep modes. The storage and loss moduli of SBR increased significantly with the incorporation of different hybrid fillers, which was attributed to the formation of an interphase between the hybrid fillers and rubber matrix, and the effective dispersion of the hybrid fillers. The mechanical properties (hardness, tensile strength, oxygen transmission rate, and swelling rate) of the composite samples were characterized in detail. From the results of the mechanical test, it was found that BC-CS-PVA0SBR had the best mechanical properties. Therefore, the BC-CS-PVA hybrid filler provided the best reinforcement effects for the SBR latex in this research.

Chemical Compositions & Texture of Allium senescens (두메부추의 화학적 조성 및 조직감 특성)

  • Chemical compositions and texture of Allium senescens that is one of Korean indigenous culinary herbs were analyzed. The contents of carbohydrate and calorie were higher than those of Chinese chive. Proline, glutamine, valine, leucine and threonine were the major total amino acids in white and green portions of Allium senescens leaves. Methionine and cysteine were identified in white and green portions of Allium senescens leaves. The values of sodium and iron in Allium senescens were lower than in Chinese chive. Trace amount of selenium was also detected in Allium senescens leaves. The C$_{20:5}$ and C$_{20:6}$ were identified in leaves of Allium senescens. As a result of Rheometer analysis and sensory evaluation, texture of Allium senescens was more tender than that of green portion of Chinese chive leaves.
