A New Flavonol Glycoside from the Leaves of Boscia senegalensis

  • Detailed chemical investigation of Boscia senegalensis (Per) Lam. ex Poir. led to the isolation of one new flavonol glycoside, rhamnocitrin-3-O-${\beta}$-$\small{D}$-(6"-O-E-feruloyl)-glucopyranoside named bosenegaloside A (1), with seven known compounds, rhamnocitrin-3-O-${\beta}$-$\small{D}$-(6"-O-E-p-coumaroyl)-glucopyranoside (2), rhamnocitrin-3-O-${\beta}$-$\small{D}$-glucopyranoside (3), 3,4,5-trimethoxyphenol-${\beta}$-$\small{D}$-glucopyrinoside (4), lasianthionoside A (5), 3,7-dimethyl-1-octene-3,6,7-triol-6-O-${\beta}$-$\small{D}$-glucopyranoside (6), syringin (7), and austroside B (8). The chemical structures of these compounds were elucidated from spectroscopic data and by comparison of these data with previously published results. The inhibitory activity of the isolated compounds on soluble epoxide hydrolase (sEH) was assessed. Compounds 1-3 potently inhibited sEH activity with $IC_{50}$ values of $12.8{\pm}0.5$, $18.4{\pm}0.2$, and $11.3{\pm}0.9{\mu}M$, respectively.

  • One anthraquinone and four flavonoids were isolated from the extract of leaves and twigs of Rhamnus taquetii. By physicochemical and spectral methods, their structures were identified as physcion (1), keampferol (2), rhamnocitrin (3), quercetin (4), and 3-O-methyl quercetin (5).

  • Various bioactive substances are found in Loranthus tanakae, including quercetin 3-rhamnoside (1), kaempferol 3-rhamnoside (2), rhamnetin 3-rhamnoside (3), and rhamnocitrin 3-rhamnoside (4), which inhibit tyrosinase. These compounds are mainly found in the EtOAc fraction of L. tanakae extract and demonstrate higher rates of tyrosinase inhibition than ascorbic acid, which was used as a control. Our results suggest that L. tanakae extracts can be utilized in skin whitening cosmetics.

Anti-inflammatory Activity of Flavonoids from Populus davidiana

  • An in vitro bioassay-guide revealed that the methanol (MeOH) extract of the stem bark of Populus davidiana showed considerable inhibitory activity against cyclooxygenase (COX-1, COX-2). Continuous phytochemical study of the MeOH extract of this plant led to the isolation of ten flavonoids; sakuranetin (1), rhamnocitrin (2), 7-O-methylaromadendrin (3), naringenin (4), eriodictyol (5), aromadendrin (6), kaempferol (7), neosakuranin (8), sakuranin (9) and sakurenetin-5,4'-di-${\beta}$-D-glucopyranoside (10). Their structures were identified on the basis of their physicochemical and spectroscopic analyses. The isolated compounds, 1-10, were tested for their inhibitory activities against COX-1 and COX-2. Compound 7 was found to have potent inhibitory effect on COX-1 and a moderate effect on COX-2, meanwhile, compounds 1-6 showed moderate inhibition against COX-1 only. Moreover, compounds 5-8 exhibited suppressive effects on xanthine oxidase (XO). These results may explain, in part, the traditional uses of P. davidiana in ethnomedicine.

Isolation of Flavonol Rhamnosides from Loranthus tanakae and Cytotoxic Effect of Them on Human Tumor Cell Lines

  • Loranthus tanakae Fr. et Sav. (Loranthaceae) is a species of mistletoe, a semiparasitic plant growing on the branches of Quercus and Betula species as host trees. In our ongoing search for bioactive compounds from endemic species in Korea, we have investigated to isolate the chemical constituents responsible for the antitumor effect of the MeOH extract of L. tanakae. The ethyl acetate soluble part of the MeOH extract demonstrated a marginal inhibition on the proliferation of the tumor cell lines such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system), and HCT-15 (colon) in vitro. Thus, the activity-guided isolation procedure upon the ethyl acetate soluble part of the extract has been carried out and finally four flavonoid rhamnopyranosides (1-4) were isolated as active principle. The structures of 1-4 were elucidated by the physicochemical and spectral data as rhamnetin 3-O-$\alpha$-L-rhamnoside (1), quercetin 3-O-$\alpha$-L-rhamnoside (2), rhamnocitrin 3-O-$\alpha$rhamnoside (3), and kaempferol 3-O-$\alpha$-L-rhamnoside (4).
