한국인 급성진행성 및 성인성 치주염의 원인균인 Bacteroides gingivalis에 대한 미생물 및 면역학적 연구 (Microbiological and Immunological Investigation on the Bacteroides gingivalis in Rapidly Progressive and Adult Periodontitis in Korean)

  • For the investigation of microbiological and immunological specificity of Bacteroides gingivalis, Bacteroides gingivalis were isolated, enumerated and characterized from 13 Korean rapidly progressive periodontitis and 7 healthy control by anaerobic culture technique. The total proportion of black-pigmented Bacteroides from Korean R.P.P. patients and healthy control were 8.78% and 0.92%, respectively, among total isolated black-pigmented Bacteroides. In antibiotic susceptibility test, Bacteroides gingivalis isolated from R.P.P. patients were sensitive to Ampicillin and Tetracycline, and resistant to Gentamicin and Erythromycin in disc diffusion method. In antibiotic broth dilution method, the minimum inhibitory concentration(MIC) to Bacteroides gingivalis was 2 unit/ml of Penicillin and $0.25{\sim}1{\mu}g/ml$ of Tetracycline, respectively. The concentration of serum IgG in rapidly progressive periodontitis patients were sigificantly higher than that of healthy control, and concentration of diluted gingival crevicular IgG has not any significant differences between two groups. Serum and gingival crevicular IgG antibody to Bacteroides gingivalis were significantly higher titer in rapidly progressive periodontitis patients to compare with healthy control. The lipopolysaccharide profiles of 2 Korean B. gingivalis in silver stained sodium dodecyl sulfate-polyacrylamide gel electrophoresis were similar to type strains of B. gingivalis and typical LPS band were appeared around the 24-Kd molecular weight. Immunodiffusion test and immunoelectrophoresis of the L.P.S. extracted from 2 Korean B. gingivalis and 2 kinds of type strains of B. gingivalis showed that B. gingivalis Korean-1 was reacted identically to B. gingivalis ATCC 33277. In trypsin and ${\alpha}$-glucosidase activity test of 2 Korean B. gingivalis, both of them revealed positive trypsin and negative ${\alpha}$-glucosidase activity, respectively. These investigation suggested that B. gingivalis is important pathogenic plaque bacteria for the pathogenesis of periodontitis and further study is needed to purify and characterize of the species-specific antigens of this organisms to develop monoclonal antibody and potential diagnostic reagents.

느타리 신품종 '솔타리'의 육성 및 자실체 특성 (Characteristics and breeding of a new cultivar of Pleurotus ostreatus 'Soltari')

  • 느타리의 새로운 품종을 개발하기 위해 갓이 짙은 흑회색이며 대가 굵고 길어 고품질인 느타리 신품종을 육성하였다. 2014년에 경기도농업기술원으로부터 'ASI 0665(흑타리)'를 분양받아 포자를 분리하여 'ASI 2504(수한)'의 이핵균주와 교잡하여 꺾쇠연결체(clamp connection)를 형성한 교잡주 25점을 선발하였다. 그 후 미토콘드리아 마커 MtPo1을 이용하여 모균주 'ASI 0665'와 같은 밴드 양상을 보이는 교잡주를 선발하여 반복 재배실험을 수행하였다. 병 당 수량이 대조구인 'ASI 2504'보다 높고 자실체의 갓과 대의 형태가 더 우수한 'Po2013-538'을 최종 우량계통으로 선발하여 특성검정, 생산력검정시험을 통해 2015년 농작물 직무육성 품종 심의회에서 '솔타리'로 명명되었다. 주요 특성은 균사 생장 적온이 $25{\sim}30^{\circ}C$이며 버섯 원기형성 및 발생온도가 $13{\sim}20^{\circ}C$로 중고온성 계통이다. 자실체의 갓 색깔은 진한 흑회색이며 갓의 형태는 깊은 깔때기형이다. 대길이는 75.24 mm, 대굵기는 19.69 mm로 대조구 'ASI 2504'와 비슷한 값을 가지나 대의 육질이 탄탄하고 색깔이 밝아 고품질로 선호된다. 자실체 수량은 병 당(1100 mL) 158.8 g으로 'ASI 2504'의 수량지수를 100으로 놓았을 때 솔타리는 103으로 수량이 약 3% 증가하였다. 가변특성으로는 감자한천배지(PDA)와 버섯완전배지(MCM)에서 균사를 배양한 결과 감자한천배지에서 생장이 양호하였고 대조구 또한 같은 결과를 보였다. 2종류의 primer(UPF 2, UPF 5)를 이용하여 새로운 품종 '솔타리'와 모균주에 대한 DNA profile을 분석한 결과 솔타리가 양친의 주요 DNA 밴드를 갖고 있으며 대조구인 'ASI 2504'와는 구별되는 것을 볼 수 있었다. 신품종 느타리 '솔타리'는 소비자들이 선호하는 짙은 흑회색의 갓과 굵으면서 색이 하얀 대의 특징을 갖고 있어 느타리 재배 농가에서 널리 보급될 것으로 기대된다.

팽이버섯(Flammulina velutipes) 교잡분리집단의 형태학적 특성 분석 (Analysis of phenotypic characterization of segregation population developed by crossing in Flammulina velutipes)

  • 팽이버섯의 야생 갈색 균주에서 유래한 단핵 계통 ASI 4019-20과 상용 품질에서 유래한 백색 단핵균주 M3를 교잡하여 분리집단을 만들고 그 특성을 조사하여 육종의 기초 자료를 작성하였다. 4019-$20{\times}M3$ 교잡으로 육성된 $F_1$인 M3-Sn 계통으로부터 단핵균주 94개를 분리하고 다시 교배 모본의 하나인 M3와 교잡하여 $BC_1F_1$인 M3-n 집단을 육성하여 생장특성을 조사하였다. 분리된 단핵균주는 형태상으로는 균사의 생장 형태가 불규칙한 것이 있었으며 톱밥 생장이 늦은 것이 많았다. 그 반면 이핵균주인 $BC_1F_1$(M3-n) 계통의 균사는 생장형태가 대부분 균일하며 톱밥에서 생장이 양호하였다. 갈색 야생 균주와 백색 상용 품종 유래의 단핵균주간 교잡으로 육성된 $F_1$의 자실체 갓색은 연갈색으로 나타나 유색이 백색에 우성이었으며, 백색 회보친을 교잡하여 육성한 $BC_1F_1$ 분리집단에서 색깔이 분포는 진갈색 7%, 갈색 25%, 연한갈색 27%, 아이보리 16%, 흰색 25%로 나타나 전체 유색에 백색의 비율은 3:1이었다.

가잠의 품종별 세포질다각체 바이러스에 대한 저항성 및 이형 바이러스 첨식에 의한 유발에 관한 연구 (Interstrain Difference in the Resistance of the Silkworm, Bombyx mori,. to Peroral Infection and Induction with the Two Strains of Cytoplasmic Polyhedrosis Virus)

  • 세포질다각체 바이러스에 대한 가잠의 품종별 저항성과 이형 세포질다각체 바이러스 첨식에 의한 유발에 관한 시험을 한 결과는 다음과 같다. 1. 품종간의 저항성 1) 일본종 및 중국종 사이에 뚜렷한 차이가 없었다. 2) 장잠기(4령)가 추잠기(2령) 보다 저항성이 강했다. 3) 육각형 세포질다각체 바이러스가 사각형 세포질다각체 바이러스보다 병원성이 높았다. 4) 육각형 세포질다각체 바이러스의 LC$_{50}$의 평균치는 원종에서는 2령기잠 처리에서 6.12$\times$$10^{6}$/$m\ell$, 4령기잠 처리에서 1.57$\times$$10^{7}$ /$m\ell$ 이었고 교잡종에서는 2령기잠 처리에서 1.28$\times$$10^{6}$/$m\ell$, 4령기잠 처리에서 4.99$\times$$10^{6}$/$m\ell$이었다. 5) 사각형 세포질다각체 바이러스의 LC$_{50}$의 평균치는 원종에서는 2령기잠 처리에서 2.06$\times$$10^{7}$ /$m\ell$, 4령기나 처리에서 5.67$\times$$10^{7}$ /$m\ell$ 이었고 교잡종에서는 2령기잠 처리에서 9.84$\times$$10^{6}$/$m\ell$, 사령기잠 처리에서 3.86$\times$$10^{7}$ /$m\ell$이었다. 2. 이형 세포질다각체 바이러스 첨식에 의한 유발사각형 세포질다각체 바이러스 첨식에 의한 육각형 세포질다각체 바이러스의 유발은 1) 영기별로 볼 때 2령기잠 처리에서가 4영기잠 처리에서보다 월등히 높은 유발률을 보였다. 2) 유형별로 볼 때 대부분이 육각형과 사각형의 혼합감염 형태로 나타났다. 3) 농도별로 볼 때 대체로 1.3$\times$$10^{6}$/$m\ell$ 농도를 중심한 중간농도에서 높은 유발률을 나타냈다.타냈다.

Characterization of Growth-supporting Factors Produced by Geobacillus toebii for the Commensal Thermophile Symbiobacterium toebii

  • Symbiobacterium toebii is a commensal symbiotic thermophile that cannot grow without support from a partner bacterium. We investigated the properties of Symbiobacterium growth-supporting factors (SGSFs) produced by the partner bacterium Geobacillus toebii. SGSFs occurred in both the cell-free extract (CFE) and culture supernatant of G. toebii and might comprise multifarious materials because of their different biological properties. The heavy SGSF contained in the cytosolic component exhibited heat- and proteinase-sensitive proteinaceous properties and had a molecular mass of >50 kDa. In contrast, the light SGSF contained in the extracellular component exhibited heat-stable, proteinase-resistant, nonprotein properties and had a molecular mass of <10 kDa. Under morphological examination using light microscopy, S. toebii cultured with the culture supernatant of G. toebii was filamentous, whereas S. toebii cultured with the CFE of G. toebii was rod-shaped. These results strongly suggest that the SGSFs produced by G. toebii comprise two or more types that differ in their growth-supporting mechanisms, although all support the growth of S. toebii. Upon the examination of the distribution of SGSFs in other bacteria, both cytosolic and extracellular components of Geobacillus kaustophilus, Escherichia coli, and Bacillus subtilis had detectable growth-supporting effects for S. toebii, indicating that common SGSF materials are widely present in various bacterial strains.

Pathogenesis of Hantaan Virus Infection in Suckling Mice -Clinical, Virologic and Serologic Observations-

  • Hemorrhagic fever with renal syndrome (HFRS) is a debilitating disease of humans caused by Hantaan virus (HV), the prototype member of a newly proposed genus of Bunyaviridae. Studies of HV pathogenesis have been limited by the absence of a well defined model for a virus-induced disease state. In an attempt to devise a model for HV pathogenesis in laboratory rodents, newborn outbred suckling ICR mice were shown to be uniformly susceptible to lethal infection with non- mouse adapted HV by intracerebral (IC), intraperitoneal (IP), intramuscular (IM), and subcutaneous (SC) inoculation routes. Clinical coures, mean time to death, and fatal outcome were age-dependent. With an inoculum of 10 $LD_{50}$, mortality was 100% in mice infected within 72h of birth, but declined to 50% by 7 days. By 2-2.5 weeks, animals developed complete resistance to clinical disease. Virus was consistently detected in serum by day 6 post-infection in IC- and IP- inoculated animals, and reached peak levels of $10^5\;PFU/ml$ by day 8 Mice infected IM and SC showed delays in onset of viremia, but achieved similar titers. Immunofluorescent antibody appeared by 17-18 days, and neutralizing antibody by 15 days, in all experimental groups. Two of 8 inbred mouse strains were identified as resistant to clinical disease : SJL/J and A/J. Manipulation of this model will allow investigation of natural rodent pathogenesis with HV, as well as offer insight into disease mechanisms and therapy of HFRS.

Comparison of Chemical Compositions and Antimicrobial Activities of Essential Oils from Three Conifer Trees; Pinus densiflora, Cryptomeria japonica, and Chamaecyparis obtusa

  • The chemical compositions, and antibacterial and antifungal effects of essential oils extracted from three coniferous species, Pinus densiflora, Cryptomeria japonica, and Chamaecyparis obtusa, were investigated. Gas chromatography mass analysis of the essential oils revealed that the major components and the percentage of each essential oil were 16.66% $\beta$-phellandrene and 14.85% $\alpha$-pinene in P. densiflora; 31.45% kaur-16-ene and 11.06% sabinene in C. japonica; and 18.75% bicyclo [2,2,1] heptan-2-ol and 17.41% 2-carene in Ch. obtusa. The antimicrobial assay by agar disc diffusion method showed that $2.2{\mu}g$ of Ch. obtusa oil inhibited most effectively the growth of Escherichia coli ATCC 33312 and Klebsiella oxytoca ATCC 10031, whereas the C. japonica oil gave weak antimicrobial activity. The minimal inhibitory concentration(MIC) values for bacterial strains were in the range of 5.45-21.8 mg/ml depending on essential oils, but most Gram-negative bacteria were resistant even at 21.8 mg oil/ml. P. densiflora oil showed the most effective antifungal activity and the MIC values for Cryptococcus neoformans B42419 and Candida glabrata YFCC 062CCM 11658 were as low as 0.545 and 2.18 mg/ml, respectively. Cryp. neoformans B42419 was the most sensitive to all essential oils in the range of 0.545-2.18 mg/ml. Our data clearly showed that the essential oils from the three conifers had effective antimicrobial activity, especially against fungi.

Characterisation of Phenotypic and Genotypic Antibiotic Resistance Profile of Enterococci from Cheeses in Turkey

  • The aim of this study was to determine the prevalence of enterococci in cheese samples and to characterize their antimicrobial resistance profiles as well as the associated resistance genes. A total of 139 enterococci were isolated from 99 cheese samples, the isolates were identified as E. faecalis (61.2%), E. faecium (15.1%), E. gallinarum (12.9%), E. durans (5.0%), E. casseliflavis (2.9%) and E. avium (2.9%). The most frequent antimicrobial resistance observed in enterococci isolates was to lincomycin (88.5%), followed by kanamycin (84.2%), gentamycin (low level, 51.1%), rifampin (46.8%) and tetracycline (33.8%). Among the isolates, the frequencies of high level gentamycin and streptomycin resistant enterococci strains were 2.2% and 5.8%, respectively. Apart from the mentioned antibiotics, low levels of resistance to ciprofloxacin, erythromycin and chloramphenicol were found. Moreover no resistance was observed against penicillin and ampicillin. The antimicrobial resistance genes including tetM, tetL, ermB, cat, aph(3’)-IIIa, ant(6)-Ia and aac(6’)-Ieaph(2”)-Ia were found in enterococci from Turkish cheese samples. In the current study, we provided data for antibiotic resistance and the occurrence of resistance genes among enterococci. Regulatory and quality control programs for milk and other dairy products from farms to retail outlets has to be established and strengthened to monitor trends in antimicrobial resistance among emerging food borne pathogens in Turkey.

전남지역(全南地域) 유우유방염(乳牛乳房炎)의 역학적(疫學的) 조사연구(調査硏究) 1. 원유중(原乳中)의 세균수(細菌數) 및 유방염검진(乳房炎檢診) (Studies on Epidemiological Investigations of Bovine Mastitis in Jeonnam District 1. Total Bacterial Count of Raw Milk and Survey of Bovine Mastitis)

  • A total of 119 raw milk samples from ten dairy farms were examined for total bacterial count, and 739 quarter milk samples of 118 dairy cows of 14 herds were examined for mastitis. The results obtained were as follows: 1. The mean of total bacterial counts of the 119 raw milk samples was 132,000 per ml. The total bacterial counts of 81 samples (68.1%) were under the standard of 100,000 per ml and those of the 38 samples (31. 9%) were over the standard. The number of bacteria showed a tendency to increase in summer. 2. One hundred and ninety five quarters (26.4%) of 98 cows (52.7%) were proved to be infected with mastitis. Clinical mastitis was found at 7 qtarters (3.5%) of 5 cows (5.0%). 3. Staphylococcus (44.9%) and Streptococcus (26.7%) were two main causative organisms of mastitis. Coliform bacteria (4.6%), Pseuedomonas spp. (4.6%), yeasts (1. 3%) and corynebacterium sp. (0.7%) were also isolated from the infected quarters. 4. The isolates were more sensitive to chloramphenicol ((96.1%), leukomycin (78.8%), streptomycin (75.5%) and tetracycline (72.4%). On the other hand, they were less sensitive to colistin (11.0%), oreandomycin (18.1%), sulfisoxazole (24.6%), penicilline (27.6%), kanamycin (43.3%) and erythromycin (49.7%). Especially the strains of Pseudomonas spp. isolated from the infected quarters were resistant to almost all the drugs examined.

  • The purpose of study was to confirm theory about the effectiveness of routine mental care on the reduction of catheter-associated urinary tract infection. The study was carried out at a university hospital from September 1,1987 to April 17, 1989 : 32 Patients with a foley -catheter were studied. The study compared the urinary tract infection rate of an experimental group with that of a control group and tested the antibiotic susceptibility of the isolated bacteria. The experimental group(16 patients) was given daily meatal care with 10% Betadine for periods ranging from 4 to 21 days. The control group(16 patients) was not given that care. The results obtained were as follows : 1. The urinary tract infection rate of the experimental group was 50.0 %, and that of the control group 43.8%. There was no significant difference between the groups. 2. Organisms isolated in the control group were bacteria 100%, and in the experimental group bacteria 50% and fungus 50%. The most common organisms of the 15 strains isolated in the total group were Staphylococcus coagulase negative (3 patients), and E-coli (3 patients). 3. Most of bacteria isolated in this study were sensitive to Norfloxacillin, but resistant to Ampicillin, Chloramphenicol, Kanamycin, Tetracycline, and Erythromycin. Hence the importance of controling catheter-associated urinary tract infections. Findings suggest the need to search for other sources of infection, further experimentation controling various sources of urinary tract infection and larger group of subjects.
