  • This study examined the prevalence of adherence factors, toxin genes, antimicrobial resistance phenotypes, and resistance genes in Escherichia coli (E. coli) isolated from piglets with diarrhea before and after the ban on antibiotic growth promoters (AGPs) in Korea from 2007 to 2018. In this period, pathogenic 474 E. coli isolates were obtained from diarrheic piglets. The virulence factors and antimicrobial resistance genes were assayed using a polymerase chain reaction, and the susceptibility to antibiotics was tested according to the Clinical and Laboratory Standards Institute guidelines. After the ban on AGPs, the frequency of F4 (12.5% to 32.7%) increased significantly, and LT (31.9% to 20.3%) and EAST-I (46.5% to 35.2%) decreased significantly. In addition, the resistance to streptomycin (45.8% to 67.9%), cephalothin (34.0% to 59.4%), and cefazlin (10.4% to 28.8%) increased significantly. Colistin resistance plasmid-mediated genes, mcr-1 and mcr-3, were detected after the ban on AGPs. The results of this study can provide useful data for analyzing the impact of the ban on AGPs on the virulence profiles and antimicrobial resistance of E. coli isolated from piglets with diarrhea in Korea.

  • The optimal quantity of inoculum was determined to screen resistance of garlic cultivars against Sclerotium cepivorum and 30 cultivars was tested. The growth of the pathogen in detached roots, Disease incidence was increased when the inoculum density was raised form 10 to 100 sclerotia. The optimal inoculum density to differentiate resistance or susceptibility of garlic cultivars was seemed to be 50 sclerotia. The cultivars collected from England, Japan, Nepal and Turkey, and cultivars such as common red, PI1356104 and PI135693 were less than the other cultivars in their disease incidence. The growth of S. cepivorum in detached roots varied from 23 to 33 mm according to garlic cultivars. There was no relationship between the disease incidence and the growth in detached roots. The sclerotial germination was increased significantly when root extract was extract was added. The addition of only distilled water resulted in 13% germination, but the addition of 0.25 g of root extract in 100 ml distilled water resulted in more than 85% germination. There was no difference in the stimulation of sclerotial germination among cultivars which showed different resistance.

  • The essential oil fractions from six plant parts including leaf of Zanthoxylum piperitum and flower of Lindera obtusiloba have revealed to possess resistance inhibitory activity on antibiotic resistant Staphylococcus aureus SA2 when combined with ohloramphenicol (Cm). The combination of Cm and essential oil mixtures showed potent resistance inhibition in the level of 10∼20 $\mu\textrm{g}$/ml.

  • In this study, the resistance spot welding process of an aluminum alloy was analyzed through the numerical simulation including the electric contact resistance and the heat generation in the electrode. The finite element model was used to solve the electro-thermal responses in weld cycles. The resistance of the contact area was represented as the contact element modeling, but the thermal resistance between the contact surfaces was neglected. Welding tests of Alclad 2024-T3 aluminum alloy were made not only to get the input data for the numerical simulation, but also to compare the numerical results. The contact resistance was determined initially by the contact resistance tests and assumed to decay exponentially up to the solidus temperature. The temperature distributions and dynamic resistance obtained numerically were in good agreement with the experimental results. Numerical results revealed that nugget growth depends mainly on the heat generated in the workpiece and its contact area. The heat generated in the electrode has, however, only a little effect on the nugget growth, and the heat generation in the electrode-workpiece interface is initially high but decrease repidly.

Enhancing Resistance of Red Pepper to Phytophthora Blight Diseases by Seed Treatment with Plant Growth Promoting Rhizobacteria

  • Plant growth promoting rhizobacteria (PGPR) have been shown to suppress phytopthora blight. This suppression has been related to both microbial antagonism and induced resistance. The PGPR isolates were screened by dual culture plate method and most of the isolates were showed varying levels of antagonism. Among the PGPR isolates pyoverdin, pyochelin and salicylic acid producing strains showed the maximum inhibition of mycelial growth of Phytopkhora capsici and increased plant growth promotion in red pepper. PGPR isolates further analysed for its ability to induce production of defence related enzymes and chemicals. The activities such as Phenyle alanin ammonia Iyase (PAL), Peroxidase (PO), Polyphenol oxidase (PPO) and accumulation of phenolics were observed in PGPR pretreated red pepper plants challenged with Phytopkhora capsici. The present study shows that an addition of direct antagonism and plant growth promotion, induction of defense related enzymes involved to enhance resistance against invasion of P. capsici in red pepper.

Discovery of Performance Traits-Linked Microsatellite Markers in Channel Catfish (Ictalurus punctatus)

  • Genomics research has two ultimate applied goals: to Isolate and clone genes of economic importance for bio-technology and gene-assisted selection (GAS), and to locate and use markers for marker-assisted selection (MAS) in selective breeding programs. To this end, we have identified linked markers for feed conversion efficiency growth rate, and disease resistance to enteric septicemia of catfish (ESC). Three microsatellite markers Ip266, Ip384, and Ip607 were identified to be linked to feed conversion efficiency. Similarly one marker each was identified to be linked to growth rate (Ip607) and disease resistance to ESC (Ip477). Ip607 marker linked to both growth rate and feed conversion efficiency, indicating that the QTL for both growth rate and feed conversion efficiency may either be the same or located in the same chromosomal region in the catfish genome. On phenotypic evaluation, certain traits such as growth rate can be accurately evaluated by body weight evaluation while other traits such as disease resistance can be quite complex. The linked DNA markers will be highly useful for MAS programs and for directing further efforts of genomic mapping for important quantitative traits.

  • The present paper considers the constraint effect on J-R curves under the two-parameter $J-A_2$ controlled crack growth within a certain amount of crack extension. Since the parameter $A_2$ in $J-A_2$ three-term solution is independent of applied loading under fully plasticity or large-scale defor-mation $A_2$ is a proper constraint parameter uring crack extension. Both J and $A_2$ are used to char-acterize the resistance curves of ductile crack growth using J as the loading level and $A_2$ are used to char-acterize the resistance curves of ductile crack growth using J as the loading level and A2 as a con-straint parameter. Approach of the constraint-corrected J-R curve is proposed and a procedure of transferring the J-R curves determined from standard ASTM procedure to non-standard speci-mens or real cracked structures is outlined. The test data(e.g. initiation toughness JIC and tearing modulus $T_R$) of Joyce and Link(Engineer-ing Fracture Mechanics 1997, 57(4) : 431-446) for single-edge notched bend[SENB] specimen with from shallow to deep cracks is employed to demonstrate the efficiency of the present approach. The variation of $J_{IC}$ and $T_R$ with the constraint parameter $A_2$ is obtained and a con-straint-corrected J-R curves is constructed for the test material of HY80 steel. Comparisons show that the predicted J-R curves can very well match with the experimental data for both deep and shallow cracked specimens over a reasonably large amount of crack extension. Finally the present constraint-corrected J-R curve is used to predict the crack growth resistance curves for different fracture specimens. over a reasonably large amount of crack extension.   • As a basic study for the application of the spore-tearing lactic acid bacteria to foods, the effects of the sporulating conditions on the growth and sporogenesis were studied were observed. The results obtained are as follow. 1. All carbohydrates added to sporulation media except dextrin decreased the sporulation rate and the thermal resistance of spores. Dextrin stimulated the growth, however, there in no effect on the thermal resistance. 2. As nitrogen source, the protein hydrolysates such as peptone, casamino acid were effective to obtain were spores of the increased thermal resistance. 3. Ca$\^$++/, Mn$\^$++/ of the metal ions added to casamino acid containing medium validly increased the total growth, sporulation rate and thermal resistance. Its optimum concentration was 40 ppm each. 4. Biotin of vitamines had an effect on the total growth, sporulation and thermal resistance of spores. Its optimum concentration was 30${\gamma}$/ml. 5. The resistant spores required the adequate maturation period, more than 36 hours, sufficient aeration. and optimum temperature, 37∼45$^{\circ}C$.

Aspergillus terreus JF27 Promotes the Growth of Tomato Plants and Induces Resistance against Pseudomonas syringae pv. tomato

  • Certain beneficial microorganisms isolated from rhizosphere soil promote plant growth and induce resistance to a wide variety of plant pathogens. We obtained 49 fungal isolates from the rhizosphere soil of paprika plants, and selected 18 of these isolates that did not inhibit tomato seed germination for further investigation. Based on a seed germination assay, we selected four isolates for further plant tests. Treatment of seeds with isolate JF27 promoted plant growth in pot tests, and suppressed bacterial speck disease caused by Pseudomonas syringae pathovar (pv.) tomato DC3000. Furthermore, expression of the pathogenesis-related 1 (PR1) gene was higher in the leaves of tomato plants grown from seeds treated with JF27; expression remained at a consistently higher level than in the control plants for 12 h after pathogen infection. The phylogenetic analysis of a partial internal transcribed spacer sequence and the b-tubulin gene identified isolate JF27 as Aspergillus terreus. Taken together, these results suggest that A. terreus JF27 has potential as a growth promoter and could be used to control bacterial speck disease by inducing resistance in tomato plants.

  • The ability of P. chlororaphis O6 to induce resistance to Erwinia carotovora subsp. carotovara SCCI and to promote growth in tobacco was demonstrated in microtiter assays on plants pre-inoculated at the root level with the bacteria before challenge with the leaf pathogen. To identify th bacterial determinants involved in induced systemic resistance and plant growth promotion, cell culture of O6 grown in King's medium B was fractionated with organic solvents and purified using various columns. in vivo and in vitro assays with samples from successive fractionation steps of the O6 supernatant led to the conclusion that antibacterial compounds were observed in aqueous layer, and to the isolation of fractions containing metabolites that retained most of the resistance-inducing activity (70:30, methanol:water) and the plant growth promotion (80:20 and 90:10, methanol:water) after ODS column chromatography. Although these molecules remain to be purified further and structurally characterized, its isolation is an addition to the range of determinants from plant growth-promoting rhizobacteria known to stimulate plant defence.
