Although sorghum residue production was estimated to be $8{\sim}10Mg\;ha^{-1}$, most sorghum straw was used to be burnt or removed after harvest. This experiment was conducted to evaluate the effect of the incorporation of manured sorghum residues on soil physical properties from 2010 to 2012 in the converted paddy field. Five treatment with 3 replication consisted of control, inorganic fertilizer (NPK), manured residues, sorghum stover and sawdust manure. The incorporation level of organic source was $10Mg\;ha^{-1}$ without inorganic fertilizer NPK. The investigated physical parameters were bulk density (BD), porosity, water stability aggregate (WSA), water infiltration rater (WIR), penetration resistance (PR) and soil water retention characteristics (WRC) with soil depth. The incorporation of manured sorghum residues improved WIR significantly over inorganic fertilizer (NPK), sorghum residues and sawdust manure. The BD by incorporating with manured residues on sorghum was reduced significantly with crop residue over plot of NPK and sawdust. Significant increase in WSA was observed with stubble incorporation alone or manured sorghum residues. Soil WRC were significantly enhanced with manured sorghum residue incorporated without NPK. The average PR at 0~15 cm was 0.54 MPa for manured sorghum residues. For sawdust manure plot it was 0.42 MPa, for Sawdust manure plot 0.39 MPa and for NPK plot 0.54 MPa.

Soil physical and chemical characteristics, soil potential denitrification rates (PDR), community composition and nirK-, nirS- and nosZ-encoding denitrifiers were studied by using MiSeq sequencing, quantitative polymerase chain reaction (qPCR), and terminal restriction fragment polymorphism (T-RFLP) technologies base on short-term (5-year) tillage field experiment. The experiment included four tillage treatments: conventional tillage with crop residue incorporation (CT), rotary tillage with crop residue incorporation (RT), no-tillage with crop residue retention (NT), and rotary tillage with crop residue removed as control (RTO). The results indicated that soil organic carbon, total nitrogen and NH4+-N contents were increased with CT, RT and NT treatments. Compared with RTO treatment, the copies number of nirK, nirS and nosZ in paddy soil with CT, RT and NT treatments were significantly increased. The principal coordinate analysis indicated that tillage management and crop residue returning management were the most and the second important factors for the change of denitrifying bacteria community, respectively. Meanwhile, this study indicated that activity and community composition of denitrifiers with CT, RT and NT treatments were increased, compared with RTO treatment. This result showed that nirK, nirS and nosZ-type denitrifiers communities in crop residue applied soil had higher species diversity compared with crop residue removed soil, and denitrifying bacteria community composition were dominated by Gammaproteobacteria, Deltaproteobacteria, and Betaproteobacteria. Therefore, it is a beneficial practice to increase soil PDR level, abundance and community composition of nitrogen-functional soil microorganism by combined application of tillage with crop residue management.
We describe a method for producing polyclonal antibodies against peptide antigen cytochrome P450 1A2 and 3A4 using a tandem repeat of the epitope region and incorporation of proline residue between the repeated sequences. An ELISA assay revealed more efficient generation of polyclonal antibodies to tandem repeat peptide antigens than mono-epitope peptides. The incorporation of proline residues further stimulated antibody production.
Decomposition of green manure is necessary for the nutrient supply in farm soil. Hairy vetch as a green manure is superior to other winter legumes in terms of wintering ability and N accumulation. This experiment was carried out to investigate the decomposition and N release of hairy vetch and its effect on rice production as the following crop in paddy field. Decomposition of hairy vetch placed by soil depth of 0, 10 and 20cm at transplanting time was investigated by mesh bag method, which was enclosed chopped residue in mesh bags. The fate of $^{15}$ N derived from $^{15}$ N-labeled hairy vetch was investigated at harvest in three levels of N fertilization. Grain yield of the transplanted paddy rice cultured with hairy vetch as starter N were compared with that of applying urea as starter N in the field. Hairy vetch residue decomposed very rapidly both in transplanted and dry-seeded paddy field. In transplanted paddy field, hairy vetch residue lost 72-81 % and 86-90% of its weight after one and five month, respectively, as affected by incorporation depth. The C/N ratio of the decomposing vetch residue increased sharply during the early stages and after then, decreased slowly. The amounts of N and P released from the vetch were about 90% and 97% of initial content after one month, respectively. Recoveries of hairy vetch-$^{15}$ N by rice plant were 30.6, 34.6 and 35.7% in 0, 6 and 12 kg urea-N 10 $a^{-l}$ application, respectively, indicating that N fertilization increased the recovery of hairy vetch. $^{15}$ N. Hairy vetch residue incorporated as starter maintained significant N $H_4$$^{+}$-N concentration in soil water of plow layer until effective tillering stage. Grain yield in the plot applied with hairy vetch was not significantly different from that in the plot with urea. We concluded that hairy vetch incorporation could substitute starter N fertilization and showed possibility to reduce N amount of top-dressing.g.g.
A series of aqueous extracts and residues from leaves, stems, roots, pods and seeds of Korean black soybean (Glycine max (L.) Merr.) were assayed against alfalfa (Medicago sativa) and barnyard grass (Echinochloa crus-galli) to determine their allelopathic activities through petri-dish and greenhouse experiments, and the results showed highest inhibition in the extracts or residues from the seeds, and followed by pods. The extracts of 40g dry tissue $\textrm{L}^{-1}$ applied on filter paper in petri-dish bioassay significantly inhibited root growth of alfalfa, and especially extracts from seeds and pods reduced root length of alfalfa more than those from leaves, stems, or roots. Plant height, root length, shoot and root dry weights of barnyard grass were reduced significantly by residue incorporation of seeds and pods as the incorporated amount increased. These results suggest that black soybean plants had herbicidal potential, and their activities were exhibited differently depending on plant parts.
Compositae plants are known to contain biologically active substances that are allelopathic to agricultural crops as well as weed species. Aqueous or methanol extracts and plant residues from leaves of Xanthium occidentale were assayed against alfalfa (Medicago sativa) to determine its allelopathic effects, and the results showed that the extracts applied onto filter paper significantly inhibited seed germination as well as root growth of alfalfa. Aqueous leaf extracts of $40g\;L^{-1}$ were completely inhibitory on root growth of alfalfa, while root growths of barley (Hordeum vulgare L.) and soybean (Glycine max L.) were less sensitive. Leaf residue incorporation at $100g\;kg^{-1}$ into soil on seedling growth of barnyard grass (Echinochloa crus-galli Beauv. var. oryzicola Ohwi) inhibited both shoot and root fresh weights of barnyard grass by 94 and $96\%$, respectively. Methanol extracts from BuOn and EtOAc fractions of X. occidentale reduced alfalfa root growth more than did those from hexane and water fractions. The results based on bioassay of extracts and residues show that X. occidentale had potent an allelopathic activity against other plant species.

Background: Some phenolics detected in the soil may inhibit the seed germination and seedling growth of ginseng (Panax ginseng). This study investigated the effect of irrigation and ginseng root residue addition on the soil microbial community and root rot disease in 2-year-old ginseng. Methods and Results: Each $20{\ell}$ pot was filled with soil infected with ginseng root rot pathogens, and irrigated daily with $2{\ell}$ of water for one month. After the irrigation treatment, ginseng fine root powder was mixed with the irrigated soil at a rate of 20 g per pot. In descending order, ${NO_3}^-$, electric conductivity (EC), exchangeable Na (Ex. Na) and K (Ex. K) decreased due to irrigation. In descending order, ${NO_3}^-$, EC, Ex. K, and available $P_2O_5$ increased with the additon of ginseng powder to the soil. The abundance of Trichoderma crassum decreased with irrigation, but increased again with the incorporation of ginseng powder. The abundance of Haematonectria haematococca increased with irrigation, but decreased with the incorporation of ginseng powder. The abundance of Cylindrocarpon spp. and Fusarium spp., which cause ginseng root rot, increased with the incorporation of ginseng powder. The abundance of Arthrobacter oryzae and Streptomyces lavendulae increased with irrigation. The abundance of Streptomyces lavendulae decreased, and that of Arthrobacter spp. increased, with the incorporation of ginseng powder. Aerial growth of ginseng was promoted by irrigation, and ginseng root rot increased with the incorporation of ginseng powder. Conclusions: Ginseng root residues in the soil affected soil nutrients and microorganisms, and promoted ginseng root rot, but did not affect the aerial growth of ginseng.
Alfalfa (Medicago sativa L.) plants have been reported to be autotoxic as well as allelopathic. Laboratory and greenhouse experiments through petri-dish and pot test were conducted to determine autotoxic effects of alfalfa leaf and soil extracts on the germination or early seedling growth of alfalfa, and to evaluate allelopathic effects of alfalfa leaf residues on alfalfa, barnyard grass, com, eclipta and soybean. Alfalfa seed germination was delayed depending on aqueous extract concentration, with no difference in final germination after 48 hours. Alfalfa root length was more sensitive to the autotoxic chemicals from leaf extracts than was germination or shoot length. Root growth of alfalfa was significantly inhibited at extract concentration of more than 1 g dry tissue/L (g $\textrm{L}^{-1}$). Hypocotyl growth, however, was not affected by all the concentrations of leaf extracts. Soil extracts from 4-yr-old alfalfa stand significantly reduced alfalfa root length by 66%, while soil extracts from 0,1, and 3yr-old stand stimulated root length up to 14-32% over the control. Residue incorporation with dry matters of alfalfa leaf at 100 g $\textrm{kg}^{-1}$ reduced seedling length of several crop and weed species, ranging from 53 to 87% inhibition. Addition of nutrient solution into alfalfa leaf extracts alleviated alfalfa autotoxic effect. This result indicates alfalfa leaf and soil extracts or residues could exert autotoxic as well as allelopathic substances into soil environments during and after establishment.
Greenhouse and laboratory studies were conducted to determine the allelopathic potentials of extracts or residues from sweet potato (Ipomoea batatas L. (Lam). The extracts applied on filter paper in a Petri dish bioassay significantly inhibited root growth of alfalfa. Aqueous leachates at 40g dry tissue $\textrm{L}^{-1}$ (g $\textrm{L}^{-1}$) from leaves showed the highest inhibition against alfalfa, and followed by stems and roots. Alfalfa root growth was significantly inhibited by methanol extracts of the same plants as the concentration increased. The effect of residue incorporation into soil on seedling growth of com, soybean, barnyard grass and eclipta was examined in the greenhouse, and results showed that the leaf residues at 200g $\textrm{kg}^{-1}$ by plant parts inhibited shoot dry and root dry weights of test plants by 60-80%. By means of HPLC, causative allelopathic substances present in plant parts of sweet potato "Sinyulmi" were identified as coumarin, trans-cinnamic acid, o-coumaric acid, p-coumaric acid, and chlorogenic acid. Total content of these compounds for leaves extracts were detected as the greatest amount in EtOAc fraction, especially trans-cinnamic acid was the greatest component. These results suggest that sweet potato plants have herbicidal potentials, and that their activities exhibit differently depending on plant parts.ant parts.
In contrast to the pyrimidine (6-4)pyrimidone photoproduct [(6-4) adduct], its Dewar valence isomer (Dewar product) is low mutagenic and produces a broad range of mutations with a 42 % replicating error frequency. In order to determine the origin of the mutagenic property of the Dewar product, we used experimental NMR restraints and molecular dynamics to determine the solution structure of a Dewar·lesion DNA decamer duplex, which contains a mismatched base pair between the 3'-T residue and an opposed G residue. The 3'-T of the Dewar lesion forms stable hydrogen bonds with the opposite G residue. The helical bending and unwinding angles of the DW/GA duplex, however, are much higher than those of the DW/AA duplex. The stable hydrogen bonding of the G 15 residue does not increase the thermal stability of the overall helix. It also does not restore the distorted backbone conformation of the DNA helix that is caused by the forming of a Dewar lesion. These structural features implicate that no thermal stability, or conformational benefits of G over A opposite the 3'-T of the Dewar lesion, facilitate the preferential incorporation of an A. This is in accordance with the A rule during translesion replication and leads to the low frequent $3'-T{\rightarrow}C$ mutation at this site.
