Differential capacity of the parthenogenetic embryonic stem cells (PESCs) is still under controversy and the mechanisms of its neural induction are yet poorly understood. Here we demonstrated neural lineage induction of PESCs by addition of insulin-like growth factor-2 (Igf2), which is an important factor for embryo organ development and a paternally expressed imprinting gene. Murine PESCs were aggregated to embryoid bodies (EBs) by suspension culture under the leukemia inhibitory factor-free condition for 4 days. To test the effect of exogenous Igf2, 30 ng/ml of Igf2 was supplemented to EBs induction medium. Then neural induction was carried out with serum-free medium containing insulin, transferrin, selenium, and fibronectin complex (ITSFn) for 12 days. Normal murine embryonic stem cells derived from fertilized embryos (ESCs) were used as the control group. Neural potential of differentiated PESCs and ESCs were analyzed by immunofluorescent labeling and real-time PCR assay (Nestin, neural progenitor marker; Tuj1, neuronal cell marker; GFAP, glial cell marker). The differentiated cells from both ESC and PESC showed heterogeneous population of Nestin, Tuj1, and GFAP positive cells. In terms of the level of gene expression, PESC showed 4 times higher level of GFAP expression than ESCs. After exposure to Igf2, the expression level of GFAP decreased both in derivatives of PESCs and ESCs. Interestingly, the expression level of $Tuj1$ increased only in ESCs, not in PESCs. The results show that IGF2 is a positive effector for suppressing over-expressed glial differentiation during neural induction of PESCs and for promoting neuronal differentiation of ESCs, while exogenous Igf2 could not accelerate the neuronal differentiation of PESCs. Although exogenous Igf2 promotes neuronal differentiation of normal ESCs, expression of endogenous $Igf2$ may be critical for initiating neuronal differentiation of pluripotent stem cells. The findings may contribute to understanding of the relationship between imprinting mechanism and neural differentiation and its application to neural tissue repair in the future.
Purpose : These studies were undertaken to evaluate the effects of the Cynomorii Herba (CH) on the spermatogenic abilities such as the concentration, motility and morphological normality of sperm from the testis, and the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. Materials and Methods : We used the 2-month-old mice and administered 0.2ml extract solution of CH in the 0.1mg/ml, 1mg/ml, 10mg/ml and 100mg/ml once a day for 60days. The control group was administered the distilled water in the same way. After the administration of extract solution, we examined the number of total, motile and normal sperm from the cauda epididymis, the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. We observed the histological changes of isolated testis and compared to the testicular tissue especially seminiferous tubules between control and CH groups by histochemical method. Results : The concentration of total sperm and the motility of spermatozoa were significantly increased in the 1mg/ml, 10mg/ml and 100mg/ml CH groups, especially in 10mg/ml group, compared to the control group. The significant differences were observed in the normality of spermatozoa of the CH groups compared to the control group. In the histolocal analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the CH groups compared to the control group. Also, the activity of hyaluronidase was significantly increased in the CH groups compared to the control group. In the antioxidant activity analysis, the activities of testicular peroxidase and testicular catalase were significantly increased in the CH groups compared to the control group, respectively. Conclusion : This study shows that CH has the beneficial effect on the concentration, morphology and motility of sperm, the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. We can suggest that CH extract solution be useful for the treatment of male sexual dysfunctions and infertility.

Methods : We used the 8-week-old ICR mice and administered 0.3mg/g extract solution of Cynomorii Herba once a day for 30, 60, 90 and 120 days. The control group was administered the normal saline in the same way and duration. We examined the number of total, motile and normal sperm from the cauda epididymis. And we compared the testicular tissue especially seminiferous tubules between control and treated groups by histochemical methods. At the end we observed the difference of sperm hyaluronidase activities between control and treated groups. Results : The significant differences were observed in the concentration of total sperm, the motility and normality of spermatozoa of the Cynomorii Herba extract solution administered groups compared to the control group in 60 and 90 days groups. In the histological analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the Cynomorii Herba extract solution administered groups compared to the control group, respectively. Also, the activity of hyaluronidase was significantly increased in the Cynomorii Herba extract solution administered groups compared to the control group. The control group was administered the normal saline in the same way and duration. We examined the number of total, motile and normal sperm from the cauda epididymis, the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. Also we observed changes of isolated testis before and after administration of Morindae officinalis Radix extract solutions the mice. And we compared to the testicular tissue especially seminiferous tubules between control and treated group by histochemical methods. Results : The significant dose-dependent differences were observed in the concentration of total sperm, the motility and normality of spermatozoa of Morindae officinalis Radix extract solution administered groups compared to the control group, respectively. In the histological analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the Morindae officinalis Radix extract solution administered groups compared to the control group, respectively. Also, the activity of hyaluronidase was significantly increased in the Morindae officinalis Radix extract solution administered groups compared to the control group. The aim of present experiment was to examine hatching rate as in vitro indicator of viability of porcine embryos before early stage embryo transfer such as zygotes or 2-cell stage embryos. Cumulus-oocyte complexes (COCs) collected from ovaries were matured in North Carolina State University 23 (NCSU-23) containing 10% porcine follicular fluid (pFF), 10 ng/ml epidermal growth factor (EGF), $10{\mu}g/ml$ follicle stimulating hormone (FSH), $35{\mu}g/ml$ luteinizing hormone (LH), and 1mg/ml cysteine. After 24 hours, the COCs were transferred to the same medium without hormones. After 65h of maturation, oocytes were exposed to phosphate buffered saline (PBS) with 7% ethanol (v/v) for 7 minutes, and then the oocytes were washed and cultured in tissue culture medium (TCM) 199 containing 5 ug/ml cytochalasin B for 5h at $38.5^{\circ}C$ in an atmosphere of 5% $CO_2$ and 95% air with high humidity. After cytochalasin B treatment, the presumptive parthenotes were cultured in porcine zygote medium (PZM)-5 and cleavage of the parthenotes was assessed at 72h of activation, Normally cleaved parthenotes were cultured for an additional 8 days to evaluate their ability to develop to blastocyst and hatching stages. The fetal bovine serum (FBS) were added at Day 4 or 5 with concentrations of 2.5, 5 or 10%. The blastocyst rates were ranged within $39.1{\sim}70%$ in each treatment. However hatching rate was dramatically decreased in non-addition group. In this experiment, embryo viability in female reproductive tract may be estimated before embryo transfer with in vitro culture adding FBS by hatching ability.
본 연구에서는 우단일엽(Pyrrosia linearifolia)의 전엽체 및 포자체의 생활환을 이용한 주년 대량번식체계의 기초를 제공하기 위하여 전엽체 증식 및 포자체를 이용한 식물체 재생에 미치는 기내 환경요소를 구명하였다. 전엽체 증식은 2MS배지에서 가장 왕성하였다. 질소급원 또한 첨가량이 많아질수록 증식율과 생육이 우수하였으며, 120mM 첨가구에서 생육이 가장 왕성하였다. 전엽체 증식에 가장 적합한 sucrose의 첨가농도는 3%였다. IAA, NAA, BA 및 kinetin을 $5{\sim}20{\mu}M$정도 첨가하는 것은 전엽체의 생육을 촉진하였으나, 2iP는 전엽체의 생육을 억제하였다. 액체배지에서는 전엽체의 생육이 억제되었으나, 진탕배양했을 때에는 고체배지와 증식량이 유사하였으며, 생식기관 또한 정상적으로 형성되었다. 포자체의 엽신과 근경의 절편을 다져서 배양한 결과, 근경 절편에서만 포자체가 재생되었다. 곱게 다진 근경의 절편은 1/8MS배지에서 포자체 재생이 가장 왕성하였으나, 재생된 포자체의 생육은 1/2MS배지에서 가장 우수하였다. 1/8MS배지에서 포자체 재생 및 생육을 촉진시킬 수 있는 적정 sucrose의 첨가 농도는 1%이며, $NaH_2PO_4$를 $50{\sim}100mg{\cdot}mL^{-1}$ 첨가하는 것은 균질배양한 근경의 절편에서 포자체가 형성되는 것을 촉진하였다.
랩틴은 지방세포의 비만유전자에서 분비되는 포식인자로써 음식섭취, 에너지대사, 체중, 번식생리 및 신경호르몬 분비를 조절하는 역할을 한다. 본 연구는 antisense 비만유전자를 발현하는 형질전환 생쥐를 생산하기 위하여 실시하였다. 먼저 랩틴을 분비하는 지방세포에서 RNA 를 추출한 후 역전사 PCR을 실시하여 303 bp의 anti I과 635 bp의 anti II cDNA 들을 합성하였다. 이러한 cDNA 들을 지방세포 특이적 발현 프로모터인 aP2 프로모터와 SV40 poly(A) 사이에 역방향으로 결합하여 미세주입용 유전자를 구축하였다. 생쥐의 수정란전핵에 antisense 비만유전자를 미세 주입하여 14 마리의 형질전환 생쥐를 생산하였으며, anti I 을 지닌 4 마리의 형질전환 생쥐와 anti II를 지닌 5마리의 형질전환 생쥐계통을 확립하였다. 그리고 형질전환 생쥐의 지방세포를 추출하여 RT -PCR을 실시한 결과 antisense 비만유전자 mRNA발현을 확인하였다. 따라서, 본 연구에서 생산된 형질전환생쥐는 생체 랩틴저하에 의해 비만을 일으키는 질환모텔동물로써의 사용가능성을 나타내었다.
알킬화제로 잘 알려진 ethane 1,2-dimethane sulfonate(EDS)는 여러 종에서 선택적인 Leydig 세포(LC) 독성과 정소 기능장애에 연구 모델로 널리 사용된다. EDS 투여에 의해 유도된 LC 녹아웃 흰쥐의 경우, 부정소와 저정낭과 같은 테스토스테론 의존성 부속 생식기관들의 급격한 무게 감소가 초래됨이 이전의 연구들에게 보고되었다. 본 연구는 EDS투여가 흰쥐 부속 생식기관에 미치는 영향에 대해 조사한 것이다. 생체 수컷 흰쥐에 EDS를 1회 주사(75 mg/kg, i.p.)한 후 0, 1, 2, 3, 4, 5, 6, 7주 후에 각각 희생시겼다. 정소와 부정소, 저정낭 그리고 전립선의 무게를 각각 측정하였다. 조직의 변화는 hematoxylin & eosin 염색과 광학현미경을 통하여 분석하였다. 생식소와 그 부속기관의 무게는 EDS 처리 후 3주 까지는 점진적으로 감소하였고, 이후 서서히 정상으로 회복되었다. 부정소의 무게는 정상 수준의 60%까지 회복되는 양상을 보였다. 조직학적 결과에서 부정소의 상피세포층은 주사 후 1주에서 3주까지 비대해지는 것이 관찰되었다. 저정낭에서는 주사 후 1주부터 3주까지 상피세포층의 두께가 감소하였다. 전립선 역시 주사 후 1주에서 3주까지 상피세포층의 두께가 감소되고, 4주부터 회복되었다. 본 연구는 EDS 주사에 의해 일시적으로 안드로겐 결핍이 일어난 흰쥐에서 부속 생식기관에 뚜렷한 조직학적 변화가 일어남을 보였다. EDS 모델을 사용하여 이들 안드로겐 의존적인 부속 생식기관의 생리적, 분자적인 조절 기작에 대해 심층 연구할 경우 이들 조직의 정상 및 병리학적 발생과 분화를 이해하는데 도움이 될 것이다.
다낭성 난소증후군(polycystic ovary syndrome, PCOS)은 내분비 및 대사장애로 호르몬 불균형 상태를 야기하여 가임기 여성의 불임 및 여러 합병증을 유발한다. 폐경기 여성의 호르몬대체 치료에 사용되는 estradiol valerate (EV)는 과다 투여 시 PCOS를 유발하는 것으로 알려졌다. 신경성장인자(nerve growth factor, NGF)는 발생 중인 신경세포의 생존과 성숙을 조절하는 인자로 난소에서도 합성되고, EV 처리에 의해 유도된 다낭성난소(polycystic ovary, PCO)에서 발현이 크게 증가하는 것으로 보고됐다. 따라서 본 연구에서는 난소유래 세포인 CHO 세포주와 다낭성 난소를 이용해 호르몬 불균형 상태인 PCOS 진단 시 NGF가 생물학적 표지인자로서 사용될 수 있는 가능성을 규명하고자 하였다. CHO 세포에 EV 2 mg과 3 mg 처리는 초기에 세포증식을 억제했으나 1 mg 처리는 영향을 미치지 않음으로써 실험 시 최적농도로 사용하였다. 하지만 농도별 EV 처리에 따른 CHO 세포의 형태학적 차이는 관찰되지 않았다. CHO 세포에 EV 처리 후 NGF mRNA 및 단백질 발현은 대조군에 비해 30분 후 크게 증가하였고, 다수의 난포낭이 관찰된 PCOS 조직에서도 정상군에 비해 크게 증가하였다. 따라서 이상의 결과들을 종합하면 EV 처리 후 난소에서 유도되는 NGF 과발현은 비정상적 난포발달을 유도하는 인자로 작용할 것으로 보이며, 다낭성난소 진단 시 표지인자로서 사용될 수 있을 것으로 생각된다.
본 실험은 돼지체외수정란을 생산하는데 적합한 배양액과 산소조건을 구명하기 위하여 NCSU(North Carolina State University)-23, PZM(Porcine Zygotes Medium)-3, PZM-4 및 TCM(Tissue Culture Medium)-199 배양액과 5% 산소조건(39, 5% $O_2$, 5% $CO_2$ , 90% $N_2$ 및 포화습도) 및 20% 산소조건(39, 5% $CO_2$ 및 포화습도)에서 돼지 미성숙난포란의 체외성숙과 체외배발달을 실시하였다. 본 연구에서 얻어진 결과를 요약하면 다음과 같다. 1) 돼지 난포란을 5 및 20% 산소조건하에서 NCSU-23, PZM-3, PZM-4 및 TCM-199 배양액에 44시간 동안 성숙을 유도한 결과 난핵포붕괴율과 핵성숙률에 있어서 산소조건 및 배양액간 유의적(p>0.05)차이는 없었다. 2) 체외수정을 실시한 결과 정자 침투율, 자ㆍ웅전핵형성률, 다정자 침입률 및 평균정자수에 있어서 산소조건 및 배양액간에 유의적인 차이가 없었다. 3) 체외수정을 실시한 후, 5 및 20% 산소조건의 NCSU-23, PZM-3, PZM-4 및 TCM-199 배양액에서 배양하여 배발달에 미치는 영향을 조사한 결과, 난할률에 있어서는 유의적(p<0.05)인 차이가 없었으며. 배발달 7일째 배반포기 발달률에 있어서 5% 산소조건에서는 PZM-3 배양액이, 20% 산소조건에서는 NCSU-23 배양액이 유의적(p<0.05)으로 높은 결과를 나타냈다. 4) 배발달 7일째 배반포기 배의 세포수를 조사한 결과, 내부세포괴세포수와 총세포수는 산소조건과 배양액간에 통계적인 유의성이 인정되지는 않았으나, 5% 산소조건에서 PZM-3 배양액(36.8$\pm$6.5개)이 다소 높은 총세포수를 나타냈다. 위의 결과를 종합하면 돼지체외수정란의 생산에는 5% 산소조건에서 PZM-3 배양액에서 배발달을 유도하는 것이 배반포기 발달률과 세포수에 있어 효과적이라고 판단된다.

회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
