Influences of Different Planting Times on Harvest Index and Yield Determination Factors in Soybean

  • This experiment was conducted to investigate the changes of harvest index and the relationship between harvest index and yield determination factors by different planting times in the determinate soybean cultivars, Shinpaldal and Danbaeg. Optimum planting were 23 May in 1995 and 1996. Late planting were 13 June in 1995 and 6 June in 1996. Growth period from planting to physiological maturity (R7) was shortened as planting time was delayed in two cultivars due to shortening of reproductive growth period in Shinpaldal, and of vegetative growth period in Danbaeg. Stem weight was distinctly decreased in late planting compared to optimum planting, but seed weight of both cultivars was not different between planting times. Also, seed number per pod and harvest index were significantly increased in late planting and the high correlation was found between two factors. It was suggested that increase of harvest index in late planting would be related with high assimilate use efficiency due to increase of sink capacity. The results of correlation and principal component analysis for yield determination factors showed that main factor on yield determination was pod number per plant at R5 stage associated with dry matter accumulation during early reproductive growth period, seed number per pod and harvest index were the second factor, and one hundred seed weight was the third factor. The result of this experiment indicated that yield determination in soy-bean was dependent mainly on pod number per plant related to dry matter accumulation by early reproductive growth period, and the increase of seed number per pod and harvest index could compensate for yield decrease by shortening of vegetative growth period in late planting. Such result suggests that optimum planting date can be delayed from mid May to early June in improved soybean cultivars in Korea.

  • The Dromedary camel (Camelus dromedaries) is an important species because of its ability to produce good quality meat, milk, and fibers under harsh environmental conditions. Camels are also crucial for transportation, racing, and as draft animals in agriculture. Therefore, dromedary camels play a critical role in the economy for millions of people living in the arid part of the world. The inherent capability of camels to produce meat and milk is highly correlated with their reproductive performance. Compared with other domestic species, the reproductive efficiency in camelids is low. Although recent reproductive technologies such as in vitro fertilization (IVF) and somatic cell nuclear transfer (SCNT) have been successfully applied to camelids and the birth of live offspring following these technologies has been reported; in vitro embryo production (IVP) has lagged in this species. The development of the IVP system for dromedary camels may be a useful tool for the genetic improvement of this species. IVP in farm animals includes three main steps; in vitro maturation (IVM) of an oocyte, IVF of a matured oocyte, and in vitro culture (IVC) of fertilized oocyte up to the blastocyst stage. This review aims to summarize various factors that influence oocyte quality, IVM, and in vitro embryo development in dromedary camel.

냉동절편을 이용한 돼지 자궁내 에스트로겐 수용체의 측정 (A Method for the Determination of Estrogen Receptor Level in Frozen Sections of Porcine Uterus)

  • The present study was designed to develop a new method for the determination of estrogen receptor in porcine uterus using frozen sections. Cryostat sections were incubated with $^3H$-estradiol($^3H$-$E_2$) in the presence or absence of diethylstilbestrol(DES) and the radioactivity of 3H-E2 bound to estrogen receptor(ER) was detected. The level of specific estrogen receptor was determined by Scatchard analysis. The highest ratio of specific binding against total binding was achieved in 3 sec. tions(5mm x 5mm) which was corresponded to lOO${\mu}$/ml protein concentration. Optimal binding was obtained during incubation with $^3H$-$E_2$ for 30 minutes at 23$^{\circ}C$ after treatment of sections with acetone for 20 seconds. Three time-washing of sections was proved to be appropriate for the removal of unbound 3H-E2. 200-fold molar excess of DES was substituted for the binding of $^3H$-$E_2$ to ER sufficiently(binding efficiency of 54.8%). ER was saturated with 4nM of $^3H$-$E_2$ and its dissociation constant was 0.1nM.   • 최근 동결기술이 발달하면서 다양한 목적에 따라 초기 발생단계, 특히 수정 전후의 난자나 수정란의 생명을 연장하는 것이 가능해졌다. 이러한 난자나 수정란의 보존기술은 인간의 수정능력을 배가시키거나 임신조절에서 응용되고 있으며, 동물에서는 우수한 유전자원의 보존과 운영, 저렴한 국제간 운송수단, 그리고 생식보조기술과 유전공학 등의 연구에 필요한 생식세포의 공급하는 데서도 중요하게 활용되고 있다. 최근 개발된 완만동결과 유리화 동결방법은 난자와 수정란을 장기간 동결하여 보존하는데 활용하는 주요 기술이다. 이러한 방법들은 각각 장점과 단점을 가지고 있지만, 상당한 수준의 효율성이 입증되어 실용화되어 있는 실정이다. 무엇보다도 유리화 방법은 완만동결 방법보다 13년이나 늦게 개발되었으나 보다 우수한 기술로 인정을 받고 있다. 비록 유리화 동결은 아직 대한 상반된 의견과 오염문제가 있지만 인간과 동물의 생식보조기술로 활용되는 빈도가 점차 많아지고 있는 실정이다. 따라서 본 원고에서는 먼저 난자와 수정란의 동결보존에 대한 기초적인 기술에 대해서 고찰한 다음, 유리화 동결에 관 한 최근의 연구동향에 대해서 종합적으로 검토하고자 한다.

Efficiency of laser-assisted intracytoplasmic sperm injection in a human assisted reproductive techniques program

  • Objective: Laser-assisted intracytoplasmic sperm injection (LA-ICSI), also known as micro-opening or thinning of the zona pellucida (ZP) prior to ICSI, may help to reduce mechanical damage to the oocyte during the procedure. The aim of the present study was to evaluate and analyze the efficacy of our institutional LA-ICSI program, which features laser-assisted ZP thinning prior to ICSI, in comparison with conventional ICSI (C-ICSI), performed on patients with different clinical characteristics. Methods: Patients undergoing a total of 212 ICSI cycles were randomly divided into an LA-ICSI group (106 cycles) and a conventional ICSI group (106 cycles). To reduce tissue damage, we thinned the ZP by approximately 70%, using a laser, before ICSI. Patients thus treated formed the LAICSI group. Comparisons included the morphological quality of transferred embryos, blastocyst development of the remaining embryos, and clinical pregnancy, in terms of ICSI method and patient characteristics. Results: Fertilization, development of remaining embryos, and pregnancy rate were significantly higher in the LA-ICSI group compared with the C-ICSI group. Fertilization, embryonic development, and the pregnancy rate were all improved in younger patients (<38 years of age) and in those who underwent a low number of IVF-ET attempts (<3 trials). In addition, the pregnancy rate was increased in older patients. Conclusion: LA-ICSI may be useful in improving the chance of pregnancy in all ICSI patients.

Fluorescence in Situ Hybridization 시행을 위한 인간정자 탈응축법의 적정화 (The Optimization of Human Sperm Decondensation Procedure for Fluorescence in Situ Hybridization)

  • Studies were conducted to determine the efficiency of decondensation protocols. Sperm obtained from seven normal donors was immediately washed after liquefaction and then decondensed using the method of West et al. (1989) and my original protocol. My optimized protocol entailed mixing 1 ml aliquots of semen with 4 ml phosphate buffered saline (PBS). Following centrifugation, pellets were resuspended in 1 ml PBS containing 6 mM EDTA. After centrifugation, pellets were resuspended in 1 ml PBS containing 2 mM dithiothreitol at $37^{\circ}C$ for 45 min. Following mixing with 2 ml PBS and centrifugation, pellets were resuspended by vortexing. While vortexing, 5 ml of fixative were gently added. Slide preparation was accomplished using the smear method and it was stored at $4^{\circ}C$. When comparing these protocols, the degree of sperm decondensation and head swelling was monitored by measuring nuclear length, area, perimeter, and degree of roundness using FISH analysis software. Apparent copy number for chromosome 1 and, separately, for the sex chromosomes was determined by FISH using satellite DNA probes for loci DIZ1, DXZ1 and DYZ3. Sperm treated by my decondensation protocol showed significant increases (p<0.05) in length, area, perimeter, and degree of roundness. There was a significant decrease (p<0.05) in the frequency of nuclei displaying no signal but no change in the frequency of nuclei with two signals in samples decondensed by my protocol. My data suggested that decondensation using my original protocol may lower the frequency of cells with spurious "nullisomy" due to hybridization failure without inducing spurious "disomy" resulting from increased distances between split signals.

Sex-linked Dwarf Gene for Broiler Production in Hot-humid Climates

  • This review has been done to examine sex-linked dwarf gene in broiler production in hot-humid climates. Introduction of sex-linked dwarf gene especially in hot harsh tropical environments brings a great advantage for broiler production. The heavy broiler parent suffers due to the stress of these adverse climates. Sex-linked dwarf genes reduce body weight, egg weight, but are superior for adaptability under harsh tropical environments, with a lower requirement for housing and feed, better survivability and reproductive fitness giving fewer defective eggs, more hatching eggs, better fertility, hatchability, feed conversion efficiency and resistance to disease. Overall the cost of chick production from dwarf hens is lower than from their normal siblings. Market weights of broilers from sexlinked dwarf dams is almost similar to those of broilers from normal dams with normal sires. But the net benefit of broiler production from sex-linked dwarf dams is found to be greater than that of broilers from normal dams. This will be the most important to the rural communities in Bangladesh and in other countries where the similar environment and socio-economic conditions exist. Therefore, sexlinked dwarf hens might be used in broiler breeding plan as well as broiler production in the tropics.

Triploid hybridization as a reproductive containment method of genetically modified fish, exemplified by fast-growing transgenic mud loach

  • Transgenic triploid hybrid between fast-growingtransgenic mud loach (Misgurnus mizolepis) males and cyprinid loach (M. anguillicaudatus) females were generated and their performance on growth, feed conversion ability and reproduction were evaluated. Although the growth accelerations of diploid and triploid transgenic hybrids were not as much as those of original transgenic mud loaches, they still represented persistent growth stimulation ranging 11 to 28 fold when compared to their non-transgenic counterparts, with significantly improved feed conversion efficiency up to 2-fold (compared to non-transgenic hybrid) and 1.5-fold (compared to non-transgenic mud loach) in maximum. The gonad development of diploid hybrids was fertile in histological views regardless of transgenic genotypes but the extent of developmentin hybrid fish were less than mud loach diploids at the same age. On the other hands, very stringent sterility was obtained in both sexes of the triploid hybrid transgenics: ovary and testis from transgenic triploid hybrids were significantly depressed and any notable sign for maturation to ovum or spermatids was not detected. No viable embryo was obtained in a fertilization trial using the suspension prepared from the minced testes of transgenic triploid hybrids. This study may indicate the potential usefulness of triploid hybridization as a mean for reproductive containment of transgenic mud loach.

Comparison of physiological responses soybean [Glycine max (L.) Merill] of different irrigation Periods

  • The water in the crop cultivation shows difference according to the variety of crop, cultivations period and climatic condition. The growth and development, quantity and fruit enlargements are affected by soil water conditions. In previous study, leaf area and photosynthesis are decreased by lower soil moisture. Other research reported that excess moisture condition at vegetative and reproductive growth period in cultivation of soybean caused highest reduction in crop growth rate (CGR) and dry weights of plant parts. In particular, the damage was bigger during vegetative growth stage than reproductive growth period. Soybean (Glycine max (L.) Merill) is useful and popular crop throughout the world. It is very popular crop in Korea, China, Japan and other Asian countries. Soybeans used in various way including soybean sprouts, paste, soymilk, oil and tofu. Two soybean cultivars grown in four different irrigation conditions were determined for physiological responses. In this study, we examined leaf area (LA), leaf dry weight (LDW), specific leaf area (SLA), root dry weight (RDW) and shoot height (SH) in different water conditions. 50mL/9day irrigation periods showed the lowest contents in LA, LDW, RDW, SH. Water deficit caused increase of leaf Water saturation deficits (WSD), Cheongjakong 3 and Taekwangkong showed increase of leaf water saturation deficits (WSD) in drought conditions and leaf water potential and stomatal conductance were decreased. Photochemical efficiency was decreased in 50mL/1day irrigation condition while, there was decrease of growth and development in 50mL/9day with drought.

  • Milk progesterone concentrations were measured in 111 dairy cows for confirming the estrus observation and for the early pregnancy diagnosis. Of the 56 cows inseminated, 52 cows(92.8%) were an ovulatory estrus, 2 cows(3.6%) were an unovulatory estrus, and 2 cows(3.6%) were the error of estrus observation, respectively. Milk progesterone concentrations at 21 and 24 days after artificial insemination were significantly higher in 23 pregnant cows compared with those in 5 non-pregnant cows(P<0.05). The accuracy rate for early pregnancy diagnosis in 27 cows achieved when the discriminatory concentration at 21 days after artificial insemination was placed at 2.0 ng/ml skim milk, was 91.3% for positive diagnosis and 100% for negative diagnosis, respectively. These results indicated that milk progesterone determination at 0, 6 and 21 days after artificial insemination can be utilized for confirming the estrus observation and for early pregnancy diagnosis. In conclusion, milk progesterone determination is useful diagnostic tool for monitoring the reproductive performance.
