Housing History of Sakhalin Returnees in Ansan Gohyangmaeul (사할린 영주귀국 동포의 주거생활사 - 안산시 고향마을 거주 강제이주 동포를 중심으로 -)

  • The purpose of this research was to find out the housing history of Sakhalin returnees in Ansan Gohyangmaeul since leaving hometown under the Japanese ruling period, who experienced two international migration for one's life. Face to face interview had been done with 20 returnees in the community center of Ansan Gohyangmaeul during October to December, 2008. The semi-structured questionnaire about housing the respondents lived in major life changes used to guide the individual interview. The results showed that personal life as well as housing histories were differed by the reason to move into Sakhalin, which still influenced the returnee' life up to now. The housing they had lived changed from barracks like a training camp, to Japanese small wooden cottage/row house, and then Russian brick house/ apartment. Housing alteration and addition and rebuilding were common to renew the old existing house. The boundary of residing area was mostly limited to the first residing location under soviet governing system throughout one's life without a long distance move. Housing satisfaction was very high in Gohyangmaeul because of the improvement of housing facilities and residence itself as well as the convenience of housing management, compared to former residence in Sakhalin. Economic and emotional aspects of life satisfaction were also high during about 8 years of living in the apartment. Forced movers still require the compensation on hand to either Korean or Japanese government no matter the amount. Social integration to the Korean community would be one of the main issues to new returnees as well as the already returned. In-depth interviews of case study need to reveal the unique housing experience of the forced mover according to the type of leaving hometown by oneself or by parents, and to returned region and time to motherland.

Teachers' Perceived Dimensions on Elementary School Health Education Curriculum: An Experimental Trial Based on Concept Mapping Approach (초등학교 보건교육 교과영역에 대한 교사들의 인식도 속성: Concept mapping 기법을 활용한 실험적 시도)

  • Background: School is a primary health education setting for children and the continuous support should be provided to renew school health education curriculum correspondent to socio-cultural changes in Korean society. Objectives: This study was conducted to identify the principals' and teachers' needs of health education topics for their students and to analyze their conceptual map for health education curriculum in Korean elementary school. Methods: The sample size of the first survey was 179 and that of the second survey was 691 of elementary school principals and teachers from stratified sampling over Korea. The self-administered mailing survey was conducted to o identify the factor structure of the health education topics and to analyze the conceptual properties with exploratory factor analysis and multidimensional scaling analysis in SPSS 12.0. Results: A total of 21 health education topics were collected from the first survey and 31 topics were, comprehensively, generated for the second survey. The five factors were determined: 'life health promotion behavior,' 'drug control and mental health,' 'bulling and aggression prevention,' 'safety perception and injury prevention,' and 'sex education and sexual harassment.' The educational need scores were the highest in 'safety perception and injury prevention.' The two-dimensional cooperates were generated for the 31 health education topics and the two dimensional properties which divided the conceptual space were 'health-safety' for one and 'public/environmental-individual/personal' for the other. Conclusions: Health education curriculum and textbook should be developed considering teachers' needs and conditions for health education in school fields. The field-based health education programs or textbook would make more possible problem-solving health education for children in real school fields.

In vitro Expansion of Umbilical Cord Blood Derived Mesenchymal Stem Cells (UCB-MSCs) Under Hypoxic Conditions

  • Mesenchymal stem cells (MSCs) have the ability to self-renew and differentiate into multi-lineage cells, thus highlighting the feasibility of using umbilical cord blood-derived MSCs (UCB-MSCs) for cell-therapy and tissueengineering. However, the low numbers of UCB-MSC derived from clinical samples requires that an ex vivo expansion step be implemented. As most stem cells reside in low oxygen tension environments (i.e., hypoxia), we cultured the UCBMSCs under 3% $O_2$ or 21% $O_2$ and the following parameters were examined: proliferation, senescence, differentiation and stem cell specific gene expression. UCB-MSCs cultured under hypoxic conditions expanded to significantly higher levels and showed less senescence compared to UCB-MSCs cultured under normoxic conditions. In regards to differentiation potential, UCB-MSCs cultured under hypoxic and normoxic conditions both underwent similar levels of osteogenesis as determined by ALP and von Kossa assay. Furthermore, UCB-MSCs cultured under hypoxic conditions exhibited higher expression of OCT4, NANOG and SOX2 genes. Moreover, cells expanded under hypoxia maintained a stem cell immnunophenotype as determined by flow cytometry. These results demonstrate that the expansion of human UCB-MSCs under a low oxygen tension microenvironment significantly improved cell proliferation and differentiation. These results demonstrate that hypoxic culture can be rapidly and easily implemented into the clinical-scale expansion process in order to maximize UCB-MSCs yield for application in clinical settings and at the same time reduce culture time while maintaining cell product quality.

  • According to the survey data during the Japanese Occupation Period, the length of South Korea's coastline is about 11,542 km, including the coastlines of land and islands. It will be very expensive and time-consuming to revise/renew accurately this coastline data through site survey, it will cost great money and time. Also, various development projects such as reclamation works on public waters, constructions of ports/harbors, etc. are frequently changing the coastal areas and coastlines, causing many renewal projects on coastal maps. For such reason, appropriate alternatives for site survey are necessary. This paper demonstrate the utilization of RTK-GPS survey data, qualitative analysis and 3D topographic analysis for extracting the change in five coastal areas (Songjeong, Haeundae, Kwanganri, Songdo and Dadaepo). The local experimental areas subjected for this research were limited to five coastal areas near Busan.

  • This study utilized high quality three-dimensional geospatial information produced by high-resolution Digital Aerial Photograph and Airborne LiDAR data in order to analyse landslides and debris flows induced by the heavy rainfall in Chuncheon area. Also, this study analysed correlation between the established landslide hazard map and the landslide factor effect and reviewed the analysis result of debris flows on the area where landslides with debris flows occurred frequently. Finally;the study proposed ways to renew the established landslide hazard map effectively and utilize the high quality three-dimensional Geospatial information on the landslide risk area.

  • Various research studies are underway to explore music classification by genre. Because sound professionals define the criterion of music to categorize differently each other, those classification is not easy to come up clear result. When a new genre is appeared, there is onerousness to renew the criterion of music to categorize. Therefore, music is classified by emotional adjectives, not genre. We classified music by light and shade in precedent study. In this paper, we propose the music classification system that is based on emotional adjectives to suitable search for atmosphere, and the classification criteria is three kinds; light and shade in precedent study, intense and placid, and grandeur and trivial. Variance Considered Machines that is an improved algorithm for Support Vector Machine was used as classification algorithm, and it represented 85% classification accuracy with the result that we tried to classify 525 songs.

No Relevance of NF-${\kappa}B$ in the Transcriptional Regulation of Human Nanog Gene in Embryonic Carcinoma Cells

  • Embryonic stem (ES) cells can self-renew maintaining the undifferentiated state. Self renewal requires many factors such as Oct4, Sox2, FoxD3, and Nanog. NF-${\kappa}B$ is a transcription factor involved in many biological activities. Expression and activity of NF-${\kappa}B$ increase upon differentiation of ES cells. Reportedly, Nanog protein directly binds to NF-${\kappa}B$ protein and inhibits its activity in ES cells. Here, we found a potential binding site of NF-${\kappa}B$ in the human Nanog promoter and postulated that NF-${\kappa}B$ protein may regulate expression of the Nanog gene. We used human embryonic carcinoma (EC) cells as a model system of ES cells and confirmed decrease of Nanog and increase of NF-${\kappa}B$ upon differentiation induced by retinoic acid. Although deletion analysis on the DNA fragment including NF-${\kappa}B$ binding site suggested involvement of NF-${\kappa}B$ in the negative regulation of the promoter, site-directed mutation of NF-${\kappa}B$ binding site had no effect on the Nanog promoter activity. Furthermore, no direct association of NF-${\kappa}B$ with the Nanog promoter was detected during differentiation. Therefore, we conclude that NF-${\kappa}B$ protein may not be involved in transcriptional regulation of Nanog gene expression in EC cells and possibly in ES cells.

Regulation of Pluripotency-related Genes and Differentiation in Mouse Embryonic Stem Cells by Direct Delivery of Cell-penetrating Peptide-conjugated CARM1 Recombinant Protein

  • Coactivator-associated arginine methyltransferase 1 (CARM1) is included in the protein arginine methyltransferase (PRMT) family, which methylates histone arginine residues through posttranslational modification. It has been proposed that CARM1 may up-regulate the expression of pluripotency-related genes through the alteration of the chromatin structure. Mouse embryonic stem cells (mESCs) are pluripotent and have the ability to self-renew. The cells are mainly used to study the genetic function of novel genes, because the cells facilitate the transmission of the manipulated genes into target mice. Since the up-regulated methylation levels of histone arginine residue lead to the maintenance of pluripotency in embryos and stem cells, it may be suggested that CARM1 overexpressing mESCs elevate the expression of pluripotency-related genes in reconstituted embryos for transgenic mice and may resist the differentiation into trophectoderm (TE). We constructed a fusion protein by connecting CARM1 and 7X-arginine (R7). As a cell-penetrating peptide (CPP), can translocate CARM1 protein into mESCs. CPP-CARM1 protein was detected in the nuclei of the mESCs after a treatment of 24 hours. Accordingly, the expression of pluripotency-related genes was up-regulated in CPP-CARM1-treated mESCs. In addition, CPP-CARM1-treated mESC-derived embryoid bodies (EBs) showed an elevated expression of pluripotency-related genes and delayed spontaneous differentiation. This result suggests that the treatment of recombinant CPP-CARM1 protein elevates the expression of pluripotency-related genes of mESCs by epigenetic modification, and this protein-delivery system could be used to modify embryonic fate in reconstituted embryos with mESCs.

Incidence of Viral Infection of Main Garlic Growing Areas of Gangwon Province in Korea (강원도 마늘 주산지 재배마늘의 바이러스 감염 실태)

  • This research was conducted to investigate the incidence of viral infection in Samcheok-shi and Jeongseon-gun, main garlic growing areas of Gangwon province in Korea. Four primers sets (GLV, LYSV, GCLV, OYDV) were used for RT-PCR to test of 100 samples collected from each location. Infection rates of GLV, LYSV, GCLV and OYDV of garlic samples from Samcheok-shi and Jeongseon-gun were 95, 95, 92 and 33%, respectively. All garlic samples tested in this research were infected at least one virus. Coinfection of 3 kinds of virus (GLV, LYSV and GCLV) and 4 kinds of virus (GLV, LYSV, GCLV and OYDV) were 60 and 25%, respectively. While infections of GLV, LYSV and GCLV were evenly found in both two regions, OYDV infection of garlic samples collected from Jeongseon-gun was higher than Samcheok-shi. Viral infections at garlic fields of whole country and Gangwon province are very serious now. Therefore, it is very necessary to develop technic for virus-free bulb and renew seed bulb of garlic.

Chemical Composition of Acacia Flower(Robinia pseudo-acacia) (아카시아(Robinia pseudo-acacia) 꽃의 화학성분 조성)

  • Chemical composition was determined to renew interest in acacia flower as food. The moisture content was 86.60%. The chemical composition showed 24.55% of protein, 8.51% of ash, 40.97% of total sugar and 160.44mg% of ascorbic acid on dry matter basis, respectively. Free sugar was mainly composed of fructose, sucrose and glucose. In fatty acid composition, the ratio of saturated and unsaturated fatty acids was 1.7 : 1. The unsaturated acids were primarily composed of polyenoic acid by more than 90%. The amino acid was distributed with a ratio 0.32 of essential to total amino acids. Important elements of acacia flower were K, Mg, Ca, Fe, and Na. Flavor components such as 24.19% of octadecanoic acid, 9.41% of benzyl alcohol, 7.05% of linalool, 5.43% of heptacosane and 4.28% of geraniol were identified as major volatile compounds of acacia flower.
