피리딜 이미다졸 시리즈의 비펩타이드 안지오텐신 수용체 리간드로 새롭게 개발된 KR-31125에 대한 생체 내활성을 동물모델에서 검증하였다. 척수장애 동물모델에서 KR-31125는 비대칭 농도의존적으로 로자탄보다 18배 이상의 경쟁적인 혈압강하 효과를 나타내었으며, 기타 수용체 촉진제들의 영향을 받지 않았다. 안지오텐신으로 유도된 정상혈압 쥐모델에서는 대조화합물인 로자탄과 비교하여 경구효과는 동등하였으나 더 빠른 초기효과가 관찰되었다. 또한 신성고혈압 쥐모델에서 KR-31125는 로자탄보다 3배 이상의 지속형 혈압강하 효과를 나타내었고, 이뇨제를 투여하여 레닌을 활성화시킨 개실험 모델에서 KR-31125는 로자탄보다 20배 이상의 지속적인 경구혈압강하 효과를 나타내었다. 이러한 KR-31125의 생체 내 활성특징은 대사물질을 통하여 효과를 발휘하는 로자탄과 달리 동일물질의 효과에 의한 것으로 고혈압 및 혈관질환과 깊은 관련이 있는 안지오텐신 조절시스템에 대한 세포영상, 비침투성 진단등의 도구물질로서 가능성이 높을 것으로 판단된다.
Objectives : Yukmijihwang-tang(Wan), a well-known formula for invigorating yin-particular kidney yin, was first recorded in "Xiao er Yao Zheng Zhi Jue", consisting of Radix Rehmanniae Preparata, Fructus Macrocarpii, Rhizoma Dioscoreae Oppositae, Poria, Rhizoma Alismatis and Cortex Moutan Radicis with dose proportion of 8:4:4:3:3:3. Although clinical trials have been lacking, various pharmacological actions for Yukmijihwang-tang has been identified newly using animal models. In addition, it was reported that Yukmijihwang-tang increases structural chromosome aberrations significantly in Chinese hamster lung cells. In this article, it is purposed that new studies for pharmacology and toxicology of Yukmijihwang-tang are reviewed. Insight into new studies of Yukmijihwang-tang at the cellular and animal levels will enhance our understanding of Yukmijihwang-tang against various diseases will provide new tools to diagnose and treat patients. Methods : Recent researches for Yukmijihwang-tang were reviewed and summarized in terms of pharmacological action and toxicity. All sources for review were based on recent studies loaded on data base of web sites such as Science Direct and National Center for Biotechnology Information. Results and Conclusions : Recently, reports showed that YMJ had antiaging effects, antioxidant and free radical scavenging activities, anti-renal hypertension and prevented tumors, and diabetes mellitus. However, there is little information on its safety except general toxicity, acute and sub-chronic oral toxicity, or genotoxicity. In addition, clinical trial for Yukmijihwang-tang was limited even though Yukmijihwang-tang has been used extensively in Korean traditional medicine. Thus, further studies are necessary to focus on safety evaluation and clinical trial for Yukmijihwang-tang.

Present article reviews about clinical pharmacology of mycophenolic acid (MPA), the active form of mycophenolate mofetil (MMF), as widely used component of immunosuppressive regimens in the organ transplantation field. MMF, used alone or concomitantly with cyclosporine or tacrolimus, has approved in reducing the incidence of acute rejection and has gained widespread use in solid organ such as kidney, heart and liver transplantation. The application of MPA and development of MMF has shown a considerable impact on immunosuppressive therapy for organ transplantation as a new immunosuppressive agent with different mechanism of action from other drugs after early 1990s. In particular aspect, use of MMF, a morpholinoethyl ester of MPA, represented a significant advance in the prevention of organ allograft rejection as well as allograft and patient survival. In considering MMF clinical data, it is important to note that there is a strong correlation between high MPA area under curve(AUC) values and a low probability of acute allograft rejection. Individual trials have shown that MMF is generally well tolerated and revealed that MMF decreased the relative risk of developing chronic allograft rejection compared with azathioprine. Recent clinical investigations suggested that improved effectiveness and tolerability will results from the incorporation of MPA therapeutic drug monitoring into routine clinical practice, providing effective MMF dose individualization in renal and heart transplant patients. Therefore, MMF has a selective immunosuppressive effect with minimal toxicity and has shown to be more effective that other agents as next step of immunosuppressive agents and regimens that deliver effective graft protection and immunosuppression along with a more favorable side effect.
Ginseng is a popular medicinal plant highly valued throughout the world. Asian ginseng is one of the most common species of ginseng. It has long occupied a significant position in oriental medicine and has been justified its name as the 'king herb'. As a nutritional supplement, ginseng is an extremely common and popular herbal medicine in the United States and Canada in recent decades. The multiple constituents of ginseng possess equally multifaceted pharmacological actions as demonstrated by numerous studies. Ginseng root and its constituents influenced the central nervous system, endocrine, cardiovascular, gastrointestinal system, sexual, renal organ and immune system, etc. One important action is its anti-hyperglycemic effect. Previous studies on ginseng demonstrate that only the root of ginseng has been used in the treatment of diabetes, while the other parts of ginseng plant were always neglected. Recently, we analyzed the constituents of ginseng berry, leaf and discovered that ginseng berry, leaf extracts and its total ginsenosides have the ability to reduce hyperglycemia and body weight and increase the peripheral glucose utilization in obese or diabetic ob/ob or db/db mice. Our data suggest that all parts of ginseng plant, including root, berry, leaf and stem exhibit potent anti-hyperglycemic and anti-obese effects and may provide an opportunity to develop a novel class of anti-diabetic agents.
Mizoribine (MZR) has been shown to possess immunosuppressive activity that selectively inhibits the proliferation of lymphocytes by interfering with inosine monophosphate dehydrogenase. The efficacy of MZR is not only in patients who have had renal transplantation, but also in patients with rheumatoid arthritis (RA), lupus nephritis, and primary nephritic syndrome. Because the exact mechanism of its immunosuppressive action is not clear, the object of this study was to examine the ability of MZR to regulate the antigen presenting cells (APCs), dendritic cells (DCs). In this work, we tested whether MZR ($1{\sim}10\;{\mu}g/mL$) could inhibit the cross-presentation of DCs. DC2.4 cells ($H-2K^{b}$) or bone marrow-derived DCs (BM-DCs) generated from BM cells of C57BL/6 mouse ($H-2K^{b}$) were cultured in the presence of MZR with OVA-microspheres, and the amount of OVA peptide-class I MHC complexes was measured by a T cell hybridoma, B3Z, that recognizes OVA (257-264 : SIINFEKL)-$H-2K^{b}$ complex and expresses-galactosidase. MZR profoundly inhibited the expression of SIINFEKL-$H-2K^{b}$ complexes. This inhibitory activity of MZR appeared to affect the phagocytic activity of DCs. MZR also decreased IL-2 production when we examined the effects of MZR on $CD4^{+}$ T cells. These results provide an understanding of the mechanism of immunosuppressive activity of MZR on the inhibition of MHC-restricted antigen presentation and phagocytic activity in relation to their actions on APCs.
비펩타이드성 안지오텐신 수용체 길항제로 새롭게 개발된 KR-31081에 대한 생체 내 활성을 세가지 동물모델에서 검증하였다. 척수장애 동물모델에서 KR-31081은 로사탄보다 40배 이상의 경쟁적인 혈압강하 효과를 나타내었으며, 신성고혈압쥐 모델에서 KR-31081은 로사탄보다 10배 가량의 지속형 효과를 나타내었다. 또한 개실험에서 구강 투여한 KR-31081은 로사탄보다 20배 이상의 지속적인 혈압강하 효과를 나타내었다. 실험에 사용된 동물 모델 시스템에서 다른 혈관조절물질들과 상호작용을 하지 않는 것으로 나타난 KR-31081은 향후 고혈압 및 혈관질환에 대한 연구 및 진단에 활용될 수 있을 것이라고 판단된다.
This study was designed to investigate the effects of a new herbal medicine named CWS-A on the changes in regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats, and further to determine the mechanism of action. Also, this study was designed to investigate the effects of CWS-A on the changes in rCBF in cerebral ischemic rats and the safety examination of CWS-A in rats regared to body weight change, renal and liver function test, CBC, $LD_{50}$ and histological observation in rats. The rCBF was significantly and stably increased and MABP was significantly decreased by CWS-A during the period of cerebral reperfusion in normal rats. However, there were no here were no significant changes of rCBF in ischemic rats. In additional, there were no significant changes on the safety examination. In conclusion, these results suggest that medication of CWS-A is effective for the improve the cerebral blood flow and CWS-A can be prescribed as vertigo(眩暈) symptoms treatment.
Morphine을 비롯한 opioid peptide가 말초 또는 중추에 투여시 혈압하강과 심박동수감소를 보이며 opiate 수용체 길항제인 naloxone에 의해 길항됨이 관찰되었던바 근래 몇몇 보고들은 중추신경내에서 adrenergic및 opioidergic system이 서로 관련되어 있음을 시사하고 있다. 이에 본 실험에서 고혈압 연구에 널리 사용되고 있는 2-kidney, 1-clip (2K1C) 방법으로 실험적 고혈압을 유발시킨 백서의 측뇌실내 clonidine또는 morphine의 심맥관계에 대한 효과와 각각의 차단제에 의한 영향 그리고 정상 및 고혈압상태 백서의 뇌내 ${\beta}-endorphin$의 함량과 specific opiate receptor binding을 정량하여 고혈압 유발에 따른 뇌내 opiate system의 변동을 관찰하였다. 2K1C 고혈압 또는 sham-operated대조백서에서 측뇌실내 clonidine $(3-30\;{\mu}g/kg)$은 용량에 비례하여 혈압하강과 심박동수감소를 일으켰으며 clonidine의 혈압강하 효과는 2K1C고혈압 백서에서 더욱 현저하였다. clonidine의 혈압강하효과는 고혈압 백서에서 측뇌실내 yohimbine 또는 naloxone 전처리에 의해 약화되었고 대조군에서는 yohimbine ($30\;{\mu}g/kg$, i.v.t.)에 의해 억제되었으나 naloxone ($50\;{\mu}g/kg$, i.v.t.)에 의해서는 영향 받지 않았다. clonidine과 마찬가지로 측뇌실내 morphine $(10-100\;{\mu}g/kg)$은 2K1C 고혈압 또는 sham-operated 대조백서에서 용량에 비례하여 혈압하강과 심박동수감소를 일으켰으며, morphine의 혈압강하효과는 2K1C 고혈압백서에서 더욱 현저하였다. 대조군과 고혈압군에서 morphine의 혈압강하효과는 naloxone 전처리에 의해 현저히 약화되었으나 yohimbine에 의해서는 영향 받지 않았다. 2K1C 시술익일부터 투여한 clonidine은 2K1C 시술에 의한 혈압 상승을 억제하였으며 naloxone (2 mg/kg, i.p.)에 의해 반전되었다. 2K1C 시술에 의해 고혈압이 유발된 백서의 뇌내 ${\beta}-endorphin$ 함량은 sham-operated 군에 비하여 유의하게 감소되어 있었고 (3H)-naloxone의 specific binding의 Bmax는 증가되었으나 Kd치는 변동되지 않았다. 이상의 실험 성적은 뇌내 opiate계가 혈압조절에 중요한 역할을 담당하고 있으며 2K1C 고혈압백서의 고혈압상태 유지에 뇌내 opiate계의 기능저하가 일부관여하고 있음을 강력히 시사한다.
이온운반계는 생체의 각기 다른 세포의 성장을 조절하는 성장조절인자들의 효과를 매개하는데 깊은 관련이 있는 것으로 보고되고 있다. 신장 근위세뇨관에서 솔변 연 $Na^+/H^+$ 상호운반계는 사구체에서 여과된 나트륨의 재흡수와 수소이온의 분비를 조절하는 중요한 기능을 수행한다. 이 연구는 초대배양된 신장 근위세뇨관세포의 나트륨 운반을 Insulin-like Growth Factor-I(IGF-I)이 어떤 경로를 통하여 조절하는지를 알아보고자 실시하였다. 결과는 아래와 같다. 1. 초대배양된 신장 근위세뇨관세포에서 $Na^+$ uptake는 시간의존적으로 증가되었으며, 30분동안 $Na^+$ uptake를 실시한 결과 세포외 NaCl 농도의존적으로 $Na^+$ uptake를 유의성있게 감소시켰다(대조군; $40.11{\pm}1.76$, 140mM군; $17.82{\pm}0.94pmole\;Na^+/mg\;protein/min$). 2. $Na^+$ uptake는 iodoacetic acid(IAA, $1{\times}10^{-4}M$) 또는 valinomycin($5{\times}10^{-6}M$)처리시 대조군에 비해 각각 $50.51{\pm}4.4%$와 $57.65{\pm}2.27%$ 억제되었으며, ouabain($5{\times}10^{-5}M$)을 처리한 경우는 $140.23{\pm}3.37%$ 증가되었다. IGF-I($1{\times}10^{-5}M$)으로 배양한 세포를 actinomycin D($1{\times}10^{-7}M$)와 cycloheximide($4{\times}10^{-5}M$)로 처리시 $Na^+$ uptake는 대조군에 비해 각각 $90.21{\pm}2.39%$와 $89.64{\pm}3.69%$로 감소되었다. 3. IGF-I으로 배양한 세포에서 세포외 cAMP는 농도의존적($10^{-8}-10^{-4}M$)으로 $Na^+$ uptake를 유의성있게 감소시켰고, 3-isobutyl-1-methyl-xanthine(IBMX, $5{\times}10^{-5}M$)도 억제시켰다. Pertussis toxin(PTX, 50pg/ml)이나 cholera toxin(CTX, $1{\mu}g/ml$)의 처리시에도 $Na^+$ uptake는 억제되었다. 세포외 phorbol 12-myristate 13 acetate(PMA) 또한 농도의존적(1-100ng/ml)으로 $Na^+$ uptake를 감소시켰다. 그러나 staurosporine($1{\times}10^{-7}M$)은 $Na^+$ uptake에 영향을 미치지 않았으며 PMA와 stauiosporine을 동시에 처리했을 때도 $Na^+$ uptake는 억제되지 않았다. 결론적으로 초대배양된 토끼 신장 근위세뇨관세포에서 $Na^+$ uptake는 막전위와 세포내 에너지 의존적이며 IGF-I은 부분적으로 단백질 및 RNA 합성을 통해서 그리고 세포내 cAMP나 PKC 경로를 통해서 $Na^+$ uptake를 조절하는 것으로 생각된다.
본 연구에서는 호두박 분획층의 항산화 활성을 검색하기 위해 MeOH, $CH_2Cl_2$, EtOAc, BuOH 그리고 $H_2O$ 분획으로 추출하여 수행하였고, 각 분획층의 산화적 세포손상과 지질 과산화의 방지 효과를 in vitro에서 확인하였다. 그 결과 호두박의 free radical (DPP보 radical) 소거 활성은 EtOAc층에서 가장높게 나타났고, 활성산소종 억제율과 peroxynitrite $(ONOO^-)$ 소거 활성은 $CH_2Cl_2$층을 제외한 모든 분획층에서 높게 나타났다. 신피질 절편에 t-BHP의 처리 시 LDH의 방출과 지질과 산화를 증가시켰고 이러한 변화는 호두의 MeOH, EtOAc, n-BuOH 분획에 의해서 완전하게 방지되었다. 이러한 결과는 호두 추출물이 t-BHP에 의한 신장 세포 손상에 효과적이고 이러한 효과는 항산화력에 의한 것으로 추측된다. 또한 이러한 결과는 호두 추출물에서 많은 질병의 원인이 되고 있는 산화적 스트레스를 방지할 수 있는 효과적인 약물 개발의 가능성을 시사한다

제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
