Cloning and Sequence Analysis of a Levansucrase Gene from Rahnella aquatilis ATCC15552

  • An intracellular levansucrase gene, lscR from Rahnella aquatilis ATCC 15552, was cloned and its nucleotide sequence was determined. Nucleotide sequence analysis of this gene revealed a 1,238 bp open reading frame coding for a protein of 415 amino acids. The levansucrase was expressed by using a T7 promoter in Escherichia coli BL21 (DE3) and the enzyme activity was detected in the cytoplasmic fraction. The optimum pH and temperature of this enzyme for levan formation was pH 6 and $30^{\circ}C$, respectively. The deduced amino acid sequence of the lscR gene showed a high sequence similarity (59-89%) with Gram-negative levansucrses, while the level of similarity with Gram-positive enzymes was less than 42%. Multiple alignments of levansucrase sequences reported from Gram-negative and Gram-positive bacteria revealed seven conserved regions. A comparison of the catalytic properties and deduced amino acid sequence of lscR with those of other bacterial levansucrases strongly suggest that Gram-negative and Gram-positive levansucrases have an overall different structure, but they have a similar structure at the active site.

An Experimental Study of Heat Transfer Characteristics on the Electronic Module Arrangement (전자모듈의 배열에 따른 열전달특성의 실험적 연구)

  • Heat transfer from three-dimensional heat-generating modules was investigated. A simulated electronic module in an array configured with dummy module elements was used to measure the average heat transfer coefficients. Various module arrangements were tested using module spacings of 0.85 and 1.15 cm for six Reynolds numbers ranging from 500 to 975. The results show that a module placed in-line with and upstream of a heated module results in the heat transfer enhancement due to a high level in turbulence prompted by upstream modules. The highest enhancement occurs when the separation distance between modules is close to the module length in the flow direction. Flow visualization reveals laminar flow on the front of the first module, slow recirculation regions on the sides parallel to the air stream, and turbulence on the back side. It appears that the first module serves to trip the air stream and produce a high level of turbulence, which enhances the heat transfer rate downstream.

  • In this paper, we propose a novel tone mapping method that implements histogram modification framework on two local regions that are classified using K-means clustering algorithm. In addition, we propose automatic parameter tuning method for histogram modification. The proposed method enhances local details better than the global histogram method. Moreover, the proposed method is fully automatic in the sense that it does not require intervention from human to tune parameters that are involved for computing tone mapping functions. In simulations and experimental studies, the proposed method showed better performance than existing histogram modification method.

  • Carbon nanotubes exhibit strong fluorescence emissions in the region of near infrared regions where most biomolecules are transparent. Such signals are highly sensitive to environment variations as well as adsorption of specific biomolecules. In this research, single walled carbon nanotubes(SWNTs) are assembled with different types of DNAs and used to target specific types of DNAs. Dot blot investigations and corresponding raman spectroscopy observations demonstrated excellent selectivity of carbon nanotube-DNA bioconjugates. The results show possibility of using SWNT as generic nano-bio markers for precise detection of different kinds of genes.

Characterization and Genetic Diversity of Benzimidazole-resistant and -sensitive Monilinia fructicola Isolates in Korea

  • To characterize benzimidazole-resistant and -sensitive Monilinia fructicola populations, the fungal isolates were obtained from peach plants showing brown rot and bloosom blight. Benzimidazole-sensitive isolates did not grow on potato dextrose agar(PDA) amended with $\geq1.0{\mu}g$ active ingredient(a.i.)/ml of the fungicides. However, benzimidazole-resistant isolates grew on PDA regardless of the tested concentrations of fungicides. Benzimidazole-resistant isolates did not grow on diethofencarb-PDA, but sensitive isolates grew on the same PDA. In the nucleotide sequences of $\beta$-tubulin gene, only codon 198(GAG: glutamic acid), a target site for benzimidazole, was replaced with GCG(alanine) in all of the resistant isolates, and this substitution seems to play an important role in the development of resistance. Other interesting codons such as 165(GCT), 200(TTC), and 241(GCT) were not changed among the isolates. Benzimidazole-resistant and -sensitive isolates were clustered clearly in random amplified polymerphic DNA analyses and the results revealed that low levels of genetic diversity between benzimidazole-sensitive and -resistant isolates of M. fructicola in the investigated regions.

A Study on Improvement of the Fisheries Census on Fisheries Households (어가실태조사를 통한 농림어업총조사의 개선방안 연구)

  • The purpose of this research is to define the problems of the Fisheries Census of Korea and to make better policies for improvement. In order to complete the purpose, the research team has firstly conducted a survey in the six regions to reinvestigate the yes or no for accuracy of the primary data related to the Fisheries Census. Moreover, the team has defined the problems of the Fisheries Census through conducting a survey for the experts and users of the fisheries statistics. Also, the team has derived the short/long term improvement of Fisheries Census through out this survey. This study has proposed that the name of census should be changed from the Fisheries Census to the Fishing Industry Census, the owners of distribution & processing companies, and their employees should be included in order to figure out the total size of the fishing industry and to plan right policies. In addition, the right information can evaluate the long-term planning for the fishing industry. The proposals from this study require a lot of changes on the Fisheries Census; so, it is impossible to adopt the proposals right from the Fisheries Census of 2015. Therefore, the changes should be adopted from the next Fisheries Census.

Spatial Distribution and Casual Causes of Shallow Landslides in Jinbu Area of Korea

  • In temperate monsoon regions, extensive shallow landslides triggered by heavy rainfall are recurrent phenomena in mountainous areas. 1,357 landslides over Jinbu area, Korea that totaled 127 km2 were identified from aerial photographs and field survey. We examined characteristics of rainfall-induced shallow landslides and casual factors affecting landslide distribution with respect to topographic and forest settings, and land use. Most landslides occurred in the study area were the results of a complex combination of precondition, preparatory factors and triggering factors. Cumulative rainfall and high intensity rainfall during short period of time made the study area very sensitive to landslides and played as catalysts to enable other factors including topographic and forest settings, and land use to act more effectively. In addition, some landslides at lower elevation involved channel incision or bank erosion influenced by land use changes such as deforestation and intensification of agriculture surrounding riparian forests or hillslopes. The results suggest that most of landslide were triggered by heavy rainstorms while topographic, forest settings, and land use affected landslide distribution occurred in the study area.

Extended Adaptively Sampled Distance Fields Method for Rendering Implicit Surfaces with Sharp Features (음함수 곡면의 날카로운 형상 가시화를 위한 확장 Adaptively Sampled Distance Fields 방법)

  • Implicit surfaces are geometric shapes which are defined by implicit functions and exist in three-dimensional space. Recently, implicit surfaces have received much attention in solid modeling applications because they are easy to represent the location of points and to use boolean operations. However, it is difficult to chart points on implicit surfaces for rendering. As efficient rendering method of implicit surfaces, the original Adaptively Sampled Distance Fields (ADFs) $method^{[1]}$ is to use sampled distance fields which subdivide the three dimensional space of implicit surfaces into many cells with high sampling rates in regions where the distance field contains fine detail and low sampling rates where the field varies smoothly. In this paper, in order to maintain the sharp features efficiently with small number of cells, an extended ADFs method is proposed, applying the Dual/Primal mesh optimization $method^{[2]}$ to the original ADFs method. The Dual/Primal mesh optimization method maintains sharp features, moving the vertices to tangent plane of implicit surfaces and reconstructing the vertices by applying a curvature-weighted factor. The proposed extended ADFs method is applied to several examples of implicit surfaces to evaluate the efficiency of the rendering performance.

Recent Advances in the Seborrheic Dermatitis (지루성 피부염의 최신 지견)

  • Seborrheic dermatitis is characterized by the appearance of red, flacking, greasy lesions in regions rich in sebaceous glands. There are various opinions about the relation between seborrheic dermatitis and dandruff. Considering all available data, we consider seborrheic dermatitis and dandruff to be differing severity manifestations of similar origin. The known etiologies of seborrheic dermatitis are the abnormality of lipid and hormnes, Malassezia species, immune response. We recently focus on the relation between seborrheic dermatitis and HIV positive and AIDS patients, seborrheic dermatitis and neurological disease. The treatments of seborrheic dermatitis include topical treatments, oral treatments. There are non-specific agents, antifungal medication, corticosteroids in the topical treatments. Narrow band UVB and oral medications are known as the prophylaxis of seborrheic dermatitis but we need further study. In traditional korean medicine, the literature studies and case reports have been published. But we don't know what herbal medicine has antifungal effect against Malassezia species. So we should discriminate herbal medicines which have antifungal effect against Malassezia specieses. Then we could treat seborreic dermatitis by traditional korean medicine effectively.

Incidence of Severe Crown Gall Disease on Tetraploid Cultivars of Grape in Korea

  • The main grape producing regions in Korea were surveyed for the occurrence and relative incidence of crown gall disease on grapevine. The results of the survey showed that the disease tended to affect tetraploid cultivars, which produce a large and sweet fruit but are very weak to cold weather. Incidence of crown gall disease was high on the tetraploid cultivars, Kyoho (Gerbong), Daebong, and Black Olympia while it was low on the diploid cultivars, Cambell Early and Sheridan. The disease incidence was very high on Anseong, Cheonan, and Chincheon, where the major growing areas of tetraploid cultivars and grapevines were burried in the winter to protect from freeze injury, whereas it was low in Yungdong, Kimcheon, and Nonsan. Crown gall disease did not increased with grapevine age on both Cambell Early and Kyoho. It remained low in Cambell Early, but high for all ages on Kyoho, Galls were found on the crown region and mid part of the trunk, but more galls were on small branches and canes on Kyoho grapevines. More than one third of ZKyoho grapevines inspected had galls on multiple locations on grapevines. On Kyoho, 56.3% of the galls covered more than 50% of the crown gall disease affects severely on the cultivar.
