가미소요산 혈맥약침이 부분 간절제술을 시행한 Rat의 간기능 회복에 미치는 영향 (The Effect of Gamisoyo-san Intravenous Pharmacopuncture on Restoration of Liver Function after Partial Hepatectomy in SD Rat)

  • Objectives: This study researched the effect of Gamisoyo-san intravenous pharmacopuncture on restoration of liver function after partial hepatectomy in SD rat. Methods: Twelve rats were randomly divided into three groups: control group (n=4) underwent partial hepatectomy, saline group (n=4) was injected $1m{\ell}$ saline after partial hepatectomy, Gamisoyo-san group (n=4) was injected with 10mg/kg Gamisoyo-san after partial hepatectomy. Liver function and liver regeneration ratio were measured on the seventh day after partial hepatectomy. Results: The results show that the AST, AST and ALT level in Gamisoyo-san group was significantly lower than those of control group and saline group(P<0.05) Conclusion: In partial hepatectomy model, Gamisoyo-san intravenous pharmacopuncture seems to significantly promote the restoration of liver function.

Analysis of Promoter Elements for Transcriptional Expression of Rat p53 Gene in Regenerating Liver

  • We previously found three transcription factor-binding motifs in the rat p53 promoter. They are two recognition motifs of NF1-like protein (NF1-like element 1: -296 ~ -312, NF1-like element 2: -195 ~ -219) and a bHLH protein binding element (-142 ~ -146). In this study, we investigated the DNA-protein complex formation of the three elements with nuclear extracts from both normal and regenerating liver to find the element involved in the induced transcription of p53. The level of each DNA-protein complex on NF1-like and bHLH motifs was not changed. Instead, a new element located at -264 ~ -284 was detected in the DNase I footprinting assay with regenerating nuclear extract. This element has partial homology to the AP1 consensus motif. However, the competition studies with diverse oligonucleotides suggest that the binding protein is not AP1. An in vitro transcription assay shows that this element is important for the transcriptional activation of the rat p53 promoter. Therefore, for the induced transcription of the rat p53 promoter, the-264 ~ -284 region is required in addition to two NF1-like and one bHLH motif.

흰쥐 부분 간 절제 후 재생 중인 간 조직의 구조와 metallothionein 분포 (Structure and Metallothionein Expression during Rat Liver Regeneration Induced by Partial Hepatectomy)

  • 흰쥐 간의 약 70%정도를 외과적으로 제거한 후, 시간 경과에 따라 간세포들의 증식과 간조직의 재구성 과정에서 나타나는 변화를 관찰하였고, 세포분열과 연관되어 있는 것으로 알려져 있는 metallothionein (MT)의 발현양상 및 세포내 분포 위치를 알아보고자 하였다. 간 절제 후 광학현미경 관찰 결과, 초기부터 소엽의 중심 정맥 주변 세포판이 붕괴되고 군집이 형성되는 모습이 관찰되었으며 간 절제 후 1일째 이미 세포들의 증식이 뚜렷이 증가하여 있는 모습을 관찰할 수 있었다. 이후 활발한 세포분열이 계속되어 재생이 이루어지게 되며 남아 있던 간조직은 약 7일이 경과하면 원 상태의 수준으로 회복되고, 재구성이 완료되는 것으로 조사되었다. 투과전자현미경을 이용하여 증식중인 간세포들의 미세구조 변화를 관찰하였던 결과, 간 절제 후 3시간 경과한 흰쥐 간세포들에서 핵/세포질 비가 비교적 높게 나타났고, 미토콘드리아의 수가 증가하였으며, 유리리보솜이나 폴리솜들의 분포도 증가된 것을 관찰할 수 있었다. 간 절제 후 24시간 전후 실험군에서는 세포질내 지방방울이 극단적으로 증가된 것이 확인되었으며 간 세포들 사이의 세포연접이 사라지거나 약화된 모습을 나타내었다. 7일째 실험군에서는 세포간 연접장치나 세포내 핵과 소기관들의 분포, 담세관의 미세구조 등이 정상대조군과 유사하게 나타났다. MT의 발현 및 위치를 알아보기 위한 면역조직화학결과 정상대조군 간세포에서는 MT에 대한 양성반응이 세포질과 핵에서 약하게 관찰되었으나, 간 절제 후 3시간이 경과한 실험군에서부터 MT에 대한 양성반응이 증가하기 시작하였으며, 일부 세포들에서는 핵에서도 더욱 뚜렷하였다. 시간 경과에 따라 핵에서의 양성반응은 지속되다가 3일이 경과한 실험군에서부터 핵과 세포질에서의 반응이 약하게 나타나며, 7일이 경과한 실험군에서는 정상대조군과 같은 미약한 반응을 나타냈다. 결론적으로, 부분 간 절제 후 재생과정은 간소엽의 체제가 붕괴되고 세포 증식이 이루어지며, 조직의 재구성과정을 거치는 것으로 요약될 수 있다. 또한 MT 단백질은 간세포들의 세포분열에 연관이 있는 것으로 판단된다.

재생중인 흰쥐간의 Lipid Peroxidation과 약물대사효소의 상관관계에 관한 연구 (Studies on the Relationship of Lipid Peroxidation and Drug Metabolizing Enzyme in Regenerating Rat Liver)

  • Wistar계 흰쥐 간의 약 70%를 절제한 후 재생중인 나머지 간의 microsome에서 lipid peroxidation과 약물 대사효소중 특징적인 aminopyrine demethylase의 활성도를 측정하고 NADPH-cytochrome c reductase, glutathione peroxidase의 활성도와 cytochrome P-450의 함량을 측정하였다. Aminopyrine demethylase의 활성도는 재생 제1일에 최저치를 나타냈으며 재생이 진행될수록 정상인 수준으로 증가하였고 lipid peroxidation, NADPH-cytochrome c reductase, cytochrome P-450의 함량 역시 재생초기에 급히 감소하여 재생2일에 가장 낮은 값을 나타내며 점차 정상인 수준으로 증가하여 7일 이후에는 정상치에 도달하였다. Glutathione peroxidase의 활성도는 재생중인 간이나 정상인 개체의 간에서 별다른 차이가 없었다. 이것은 재생중인 간에서 재생초기에 cytochrome P-450의 함량과 NADPH-cytochrome c reductase의 활성도 감소가 lipid peroxidation과 약물 대사효소의 활성도 감소를 일으키며 이러한 현상은 부분 간 절제 후 7일 이후에는 거의 나타나지 않았다.

생약복합제제(生藥複合製劑)의 약효연구(藥效硏究) 제7보(第7報) -생간건비탕(生肝健脾湯)의 간조직부활(肝組織復活)에 대(對)한 작용(作用)- (Studies on the Efficacy of the Combined Preparation of Crude Drugs (VII) -Effect of 'Saengkankunbi-Tang' on the Liver Tissue Recovery-)

  • To investigate the effect of 'Saengkankunbi-Tang' on the liver tissue recovery in about 70% cutted rat liver, the observation on the photographing by the Gamma-camera and the electromicroscopic section against the rabbit liver induced by $CCl_4$, these studies were conducted. The results were summarized as follows; Observed on the liver tissue recovery, function regeneration and body weight increase in about 70% cutted rat liver, the recovery of liver tissue and body weight were increased with the passage of the time and also they were showed to the high increase rate in proportion to administration amount. The glucose content in the serum was found in increasing tide as compared with the control group. The alkaline phosphate activity in the serum showed the significant difference in the 10th day and 13th day as compared with the control group. The ammonia content in the blood showed the significant difference only in the 10th day. Observed on the photographing by the Gamma-camera against the rabbit liver induced by $CCl_4$, sample groups were shown to be significant recovery as compared with the control group. Observed on the electromicroscopic section, sample groups showed the control action of the nuclear destruction against the rabbit liver induced by $CCl_4$ and also they showed the control action of the expansion of glycogen and granular endothelial reticulum in cytoplasm. According to the above finding, it is presume that 'Saengkankunbi-Tang' can be applicable to the extensive treatment of liver disease.

재생 쥐간에서 분리한 DNA topoisomerase II에 결합된 protein kinase 활성 (The Identification of Type II DNA Topoisomerase-Associated Protein Kinase Activity from Regenerating Rat Liver)

  • 재생쥐간에서 분리한 topoisomerase II에서 protein kinase 활성이 발견되었다. ,topo II 활성 및 kinase 활성은 hydroxyapatite, phosphocellulose, double strand DNA cellulose chromatography 등의 순수 분리 과정 중에도 서로 분리되지 않았으며 glycerol gradient sedimentation 분석에서도 같은 분획에서 활성이 존재하였다. Kinase는 topo II 저해제인 N-ethylmaleimide와 novobiocin 등에 의해 그 활성이 저해되었다. 그러나 이러한 증거들 만으로 kinase 활성이 topo II가 아닌 다른 polypeptide에 의한 것일 가능성을 완전히 배제 할 수는 없다. Topo II와 결합된 kinase 활성에는 Mg++가 절대적으로 필요하였으며 다른 일가 또는 이가 이온으로는 그 효과가 대체되지 않았다. Histone H1은 kinase 활성을 증가 시키며 또 kinase에 의해 강하게 인산화된다. 이러한 효과는 다른 histone 류 및 casein 등에 의해 대체되지 않았다.

부분 간절제술을 시행한 랫드에서 간재생에 관한 이과두주 급성투여에 따른 영향 (Effects of acute LeeKwaDoo administration on liver regeneration induced by partial hepatectomy in rats)

  • Alcoholism and alcohol abuse are major public health concerns. This is linked to the injury of many organs, especially liver. Experiments were peformed to know the acute effects of LeeKwaDoo (LKD) induced by two-third partial hepatectomy (PH) in rats. In liver samples, regeneration parameters and histological assessment were performed. For the blood biochemical study, the blood were assayed with AST, ALT. The portal branch of liver lobes was ligated in the male Sprague-Dowley rats, two-thirds partial hepatectomies were also performed. It was estimated bodyweight and relative liver weight for the index of liver mass. For the marker of blood chemistry, we investigate the serum sample of rats and demonstrated the level of AST, ALT. Remaining tissues of liver developed as microscopic structures. Resection of the lobes in PH+LKD group resulted in a marked change of liver weight, blood chemistry and histological changes. The initiation of the proliferative response in PH group stimulated as well as reduction of the liver mass. On the other hands, the Initiation of the proliferative response in PH+LKD group delayed. Eventually, both PH group and PH+LKD group was restored relative liver weigh after 7 day. In conclusion, the acute adminstration of LKD seems to inhibit the initial response of liver regeneration through alcohol effects.

머위(Petasites japonicus maxim)를 급여한 rat와 mouse에 대한 병리학적 관찰 I. 육안적 및 병리조직학적 관찰 (Pathological changes on rats and mice fed with Petasites japonicus Maxim I. Macroscopical and histopathological observations)

  • In order to know the toxic effect and carcinogenic activity in rats and mice fed with juice of Korean native Petasites japonicus Maxim of its pellet(4% or 8%) which were dried, milled and mixed with basal diet, the investigations were carried out by macroscopy and histopathology. Macroscopically, although remarkable changes were not observed in the liver of mice, there were slight to moderate swelling of rat livers in the whole groups at 12 to 14 weeks after feeding and milky spots in rats fed with its juice and 8% pelleted Petasites japonicus Maxim diet and a normal diet for 1 week alternatively for 14 weeks. Moreover, moderate to severe swelling and milk spots were recognized in livers of all rats fed with its juice and 8% pellet or 8% pelleted Petasites japonicus Maxim for 16 weeks. But, in cases of rats fed with its juice and 4% pellet or 4% pelleted Petasites japonicus Maxim, only swelling of livers was recognized moderately or severely. Histopathologically, major lesions were found in livers of both rats and mice. There were congestion, hemorrhage, fatty change, focal necrosis, megalocytosis and hyperplasia of endothelial cell in livers of mice and rats, the additional lesions such as proliferation of bile duct and nodular regeneration with diffuse regenerating cells were seen in livers of rats. In addition, preneoplastic lesions, the areas of milky spots macroscopically, were observed in livers of rats fed with Petasites japonicus Maxim for 14 to 16 weeks. In a few cases, haemangioendothelial sarcoma in livers was detected in rats fed with Petasites japonicus Maxim for 16 weeks. Petasites japonicus maxim growing naturally in Korea seem to exhibit toxic effect especially in liver and it contained a causative agent of primary liver tumors.
