Hybridization between Marine Medaka Oryzias dancena and Javanese Medaka Oryzias javanicus (바다송사리 Oryzias dancena와 자바송사리 Oryzias javanicus 간 잡종 유도)

  • Inductions of hybrids and reciprocal hybrids between Oryzias dancena and O. javanicus (ODJ and OJD) were conducted and backcross hybrids between female O. dancena and male ODJ were also produced for biological and cytogenetic analysis. Embryonic development of ODJ and OJD were compared with those of their parents. Developmental time was fastest in O. dancena and ODJ, followed by O. javanicus and OJD. Oryzias dancena hatched 11 days (d) after fertilization, ODJ at 13 d, O. javanicus at 14 d and OJD at 15 d. The abnormality of external morphology rate in ODJ was 10.6%; however, OJD showed a high degree of abnormality, over 90%. The proportion of males was 90.0% and 31.3% for ODJ and OJD, respectively. Cytogenetic analysis was conducted to obtain basic information for genetic identification of O. dancena, O. javanicus and their hybrids. The karyotypes of all experimental groups showed 2n=48 chromosomes and the fundamental number (FN) was 48. The first pair carried secondary constrictions near the centromeric regions. Erythrocyte area and volume were $9.8\;{\pm}\;0.5\;{\mu}m^2$ and $18.2\;{\pm}\;1.0\;{\mu}m^3$, respectively, for O. dancena, $8.3\;{\pm}\;0.5\;{\mu}m^2$ and $15.8\;{\pm}\;1.5\;{\mu}m^3$ in O. javanicus, and $18.3\;{\pm}\;0.5\;{\mu}m^2$ and $15.7\;{\pm}\;1.3\;{\mu}m^3$ in ODJ. Erythrocyte area and volume in ODJ were similar to those of O. javanicus. In backcross hybrids between female O. dancena and male ODJ, all embryos failed to develop and died in the late gastrula stage.

Improving Accuracy of Chapter-level Lecture Video Recommendation System using Keyword Cluster-based Graph Neural Networks

  • In this paper, we propose a system for recommending lecture videos at the chapter level, addressing the balance between accuracy and processing speed in chapter-level video recommendations. Specifically, it has been observed that enhancing recommendation accuracy reduces processing speed, while increasing processing speed decreases accuracy. To mitigate this trade-off, a hybrid approach is proposed, utilizing techniques such as TF-IDF, k-means++ clustering, and Graph Neural Networks (GNN). The approach involves pre-constructing clusters based on chapter similarity to reduce computational load during recommendations, thereby improving processing speed, and applying GNN to the graph of clusters as nodes to enhance recommendation accuracy. Experimental results indicate that the use of GNN resulted in an approximate 19.7% increase in recommendation accuracy, as measured by the Mean Reciprocal Rank (MRR) metric, and an approximate 27.7% increase in precision defined by similarities. These findings are expected to contribute to the development of a learning system that recommends more suitable video chapters in response to learners' queries.

  • Artificial interspecific matings between Matsumuraeses phaseoli and M. falcana (Lepidoptera: Tortricidae) were conducted to know the possibility of hybridization of the two sympatric species. Reciprocal crossings successfully produced F1 hybrids. Most of F2 crosses yielded progenies except all mating trials with females of F1 hybrid obtained from M. phaseoli female. Inbreedings of the F2 hybrids produced F3 progenies. In backcrossings between F1 hybrids and parent lines, all the two mating trials with females of F1 hybrid obtained from M. phaseoli females did not produce any progeny, while other 6 backcrosses produced the next generations. Inbreedings of the backcross lines also produced subsequent progenies. These results indicated that F1 females produced from hybridization between M. phaseoli females and M. falcana males were sterile. Conclusively, it suggested that a partial reproductive isolation at a postzygotic stage can occur between the two sympatric species.

Relationship Between Heterosis and Genetic Divergence in Tongil-type Rice(Oryza sativa L.) (벼에서 RAPDs 분석에 의한 양친의 유전적 유사성과 잡종강세의 상관)

  • In this study we evaluated genetic divergence among six Tongil-type rice varieties and assessed the relationship between genetic distance and hybrid performance in all possible non-reciprocal crosses between them. The 15 F1 hybrids along with the six parents were evaluated for eight traits of agronomic importance including yield in a replicated field trial. The six parents were examined for DNA polymorphism using 120 random decamer oligonucleotide primers. A total of 168 polymorphic variants were generated and based on the polymorphism data, genetic distances (GDs) ranged from 0.157 to 0.383. Heterosis was observed in hybrids for most of the traits, and yield exhibited the highest heterosis among the eight traits examined. The correlation values of GDs with F1 performance were not significant. Also the correlations of GDs with midparent and better-parent heterosis were not significant except for spikelets per panicle. Our results have indicated that GDs based on the RAPDs markers may not be useful for predicting heterotic combinations in Tongil-type rice and are supportive of the idea that the level of correlations between hybrid performance and genetic divergence is dependent on the germlplasm employed.

  • Morphological characteristic during formation of tobacco crown gall tumor and genetic tumor, and their differential response to growth regulator were investigated in in vitro culture. Crown gall tumor was induced from tumor tissue transformed by infecting Agrobacterium tumefaciens C58. Genetic tumor was induced from tumor tissue which was induced spontaneously from reciprocal interspecific hybrids between Nicotiana glauca (2n=24) and Nicotiana langsdorffii (2n=18). Morphological characteristic of crown gall tumor, genetic tumor, and teratoma shoot was very similar, and they were actively proliferated on hormone-free medium. Typical tumor callus and teratoma shoot formed from crown gall tumor on the hormone-free medium. On the contrary, tumor callus derived from genetic tumor formed as a crown gall tumor callus on the medium supplemented with 0.5 mg/L of 2,4-D, and lots of teratoma shoots without any root formed on the hormone-free medium. Root development from the teratoma shoots was hardly obtained on the medium with IAA, GA and active carbon. However, teratoma shoots with roots, as normal shoots, were initiated occasionally on the hormone-free medium. These shoots also formed new genetic tumor on the stem, which leaves formed lots of teratoma shoot on the hormone-free medium in in vitro culture.

Plant Regeneration via in Vitro Culture of Ovule Obtain by Intergeneric Crossing Between Citrus junos Sieb. et Tanaka and Poncirus trifoliata Raf. (유자와 탱자의 속간교잡후 배주배양에 의한 식물체 유기)

  • As a basic research for breeding new varieties, reciprocal -intergeneric crosses between Citrus junos and P.trifoliata were made. F$_1$ hybrid production using in vitro ovule culture, gametogenesis, and fertilization phenomena were investigated. Frequency of fruit set resulting from crossing of Citrus junos and Poncirus Trifoliata was 16.6% while that of Poncirus Trifoliata and Citrus junos was 11.7%. Callus formation occurred well when ovules at the 6th week after pollination were cultured on MT (Murashige and Tucker) medium supplemented with zeatin 0.5 mg/L and NAA 1.0 or 3.0 mg/L. Immature ovules developed into mature embryos of the MT medium supplemented with 2,4-D 0.1 or 3.0 mg/L. Immature ovules developed into mature embryos of the MT medium supplemented with 2,4 D 0.1 or 0.5 mg/L. The invitro germination rates of 20-week-old ovules set C. junos $\times$ P. Trifoliata and P. Trifoliata $\times$ C. junos were 54.5% and 48.6%, respectively. The emergence ratios of trifoliate hybrids obtained by C. junos $\times$ P. Trifoliata and P. Trifoliata $\times$ C. junos were 56.7% and 100%, respectively. The chromosome number of C. junos and P. Trifoliata was n = 9 or 2n = 18, and the sizes of their pollen grain were 33.75 $\mu$ and 25.0 $\mu$. The length and width of embryo sac in C. junos and P. Trifoliata were 69.38~79.23 $\mu$ and 27.50~38.56 $\mu$, and those of egg cells were 17.50~41.50 $\mu$ and 6.25~8.12$\mu$. Fertilization of C. junos and P. trifoliata terminated 72 h after pollination.

Cytoplasmic Inheritance of High Tillering and Earing Characters of a Korean Local Maize Line(MET) (다수.다얼성 옥수수(NET)의 세포질적 유전)

  • A Korean local maize line, MET, which has multi-ears and tillers has been proved as a potential source for silage production. However, no fundamental genetic nature for the line has been investigated. Therefore, this study was done to find genetic information on the multi-earing and -tillering habits of MET line. MET line and a hybrid. (Mo 17 ${\times}$ B68), with monoculm and single ear per plant were used for production of F$_1$(F$\_$1-12/ and F$\_$1-21/), F$\_$2-12/, F$\_$2-21/, BC$\_$1-12/ and BC$\_$1-21/ generations. From the comparison of reciprocal crosses, it was found that the tillering and earing habits of the MET line are controlled by cytoplasmic factors. The tiller and ear numbers, and barren ears were all characters associated with the MET cytoplasm. The cytoplasmic effect of MET on tiller and ear numb en was not evident in F$_1$ generation, probably because of suppressing effect of heterosis on appearance of tillers or ears. Genetic parameters for the gene action for both tiller and ear number also indicated a lack of mono- or digenic-chromosomal gene effects. The heritability (broad) was very low for both characters. Therefore, it is strongly concluded that the tillering and earing characters of MET line are due to cytoplasmic reasons.

Genetic Distance and Heterosis Degree Among the Developed Waxy Corn Lines (찰옥수수 육성계통간 유전적 거리와 잡종강세 정도)

  • This study was carried out to find relationships of genetic distance and heterosis expression degree in the developed CNU waxy corn lines. The used material were developed in Corn Breeding Laboratory, Coll. of Agri. & Life Sci., CNU. Total 10 primers used for SSR maker analysis. In genetic distance, the used lines divided into four groups; A group has two lines, B group has one line, C group has ten lines and D group has eleven lines, respectively. Among used lines, CNU427 and CNU588 were very closed as a 74, while CNU451 and CNU429 were remote as a 40 inbred coefficient, respectively. While heterosis degree were very variable not only hybrids but also cross parents. Average heterosis of most cross-parent was high in plant height, ear height, flowering day and ear length. Especially, CNU H09-23 hybrid was high as 79.4% than other hybrids. Here, we gained the fact has closely relationship between genetic distance and heterosis.

The Test of Combining Ability and Heterosis on the Silkworm(Bombyx mori) Breeding (누에 견.사질에 관한 잡종강세 및 조합능력검정)

  • The study was conducted to obtain the genetic information on heterosis and combining ability of the quantitative characters for F1 hybrid breeding in silkworms. Six parental varieties and each set of 30 diallel crosses in F1's were used as materials, and bred on the randomized complete block design with three replications. Fourteen characters were observed with the twenty samples in each tray. The data were analyzed for (1) heterosis and combining ability in F1 hybrid. The heterosis in the weight and the length of cocoon showed positively high at 24.51%, and 23.4%, respectively and the weight of the whole cocoon as well as the weight of the whole cocoon layer showed a siginificant heterosis ranging from 15.56% to 15.71% and from 17.14% to 19.01%, but the fifth and the total instar period showed negative heterosis. It was found that the combination between, C70XRomogua and N9 X Romogua showed highly a negative heterosis on the fifth instar period and for the cocoon weight. The female of N9+Sansuian and the male of Romogua X Sansurian have a high heterosis effect, on the cocoon shell weight, and Sansurian X Romogua(reciprocal) on the length and the weight of cocoon filament with no regard to sexuality. The significant maternal and cytoplasmic effect on heterosis of the cocoon weight and the cocoon shell weight were observed with the combinations, N9 X C5, N63 X C70 and on the length of the cocoon filament with the combinations, Sansurian X N63, Sansurian X C5, Sansurian X C70 and N9 X C70, N63 X C70 on the weight of cocoon filament. As mean squared of GCA, SCA and RCA were significant with these combining ability for all characters resulted from additive and non-additive altogether and there is a significant difference between reciprocals. Sansurian showed a negative GCA effect on the fifth and total larval duration, but the higher positive GCA effects took places with varieties N9 and C5 on the length, width, weight of cocoon, cocoon shell weight, percentage of cocoon shell weight, length and weight of cocoon filament, percentage of raw-silk with no regard to both generations and silkworm sexuality. The values of SCA between the cross combinations varied generation-wise and sex-wise. It was shown that SCA value for the fifth instar period was highly negative for Sansurian X C70, Romogua X C70, Sansurian X C5, Romogua X C5, but it was positive effect on the cocoon weight, cocoon shell weight with N9 X C5, and C70 X Sansurian, on the length of cocoon filament with N9 X C5, Romogua X Sansurian on the weight of cocoon filament between Romogua and N63 and on the percentage of raw-silk between the combination of Sansurian X Romoga.

Enhanced Resistance to Sulfur Dioxide Gas in Transgenic Petunia by Stacking both SOD2 and NDPK2 Genes (SOD2와 NDPK2 유전자 집적에 의한 페튜니아의 아황산가스 저항성 증진)

  • The aim of this study was to develop a transgenic petunia with enhanced resistance to sulfur dioxide ($SO_2$) gas by stacking two genes, SOD2 and NDPK2, which are both known to confer resistance to abiotic stresses. The first-generation hybrids ($TF_1$) were obtained through reciprocal crosses between an SOD2-transgenic line SOD2-2-1-1-35($T_4$)[S($T_4$)] and an NDPK2-transgenic line NDPK2-7-1($T_2$)[N7-1($T_2$)]. Approximately 32.1-73.0% of the first-generation hybrids ($TF_1$) carried both SOD2 and NDPK2 genes. These hybrids showed 2.6 and 5.1 times less damage than hybrids carrying only SOD2 or NDPK2 genes, respectively, when they were treated with $SO_2$ gas at 30 ppm. This confirmed that the heterozygous hybrids were more resistant to $SO_2$ than the hybrids carrying either one of the resistance genes. Second-generation hybrids ($TF_2$) were obtained by selfing the $TF_1$ individuals. We confirmed the expression of the stacked genes in the $TF_2$ hybrids by phenotypic observation of their response to $SO_2$ gas at 30 ppm as well as using RT-qPCR and photosynthetic efficiency.
