Process fault diagnostics using the integrated graph model

  • On-line fault detection and diagnosis has an increasing interest in a chemical process industry, especially for a process control and automation. The chemical process needs an intelligent operation-aided workstation which can do such tasks as process monitoring, fault detection, fault diagnosis and action guidance in semiautomatic mode. These tasks can increase the performance of a process operation and give merits in economics, safety and reliability. Aiming these tasks, series of researches have been done in our lab. Main results from these researches are building appropriate knowledge representation models and a diagnosis mechanism for fault detection and diagnosis in a chemical process. The knowledge representation schemes developed in our previous research, the symptom tree model and the fault-consequence digraph, showed the effectiveness and the usefulness in a real-time application, of the process diagnosis, especially in large and complex plants. However in our previous approach, the diagnosis speed is its demerit in spite of its merits of high resolution, mainly due to using two knowledge models complementarily. In our current study, new knowledge representation scheme is developed which integrates the previous two knowledge models, the symptom tree and the fault-consequence digraph, into one. This new model is constructed using a material balance, energy balance, momentum balance and equipment constraints. Controller related constraints are included in this new model, which possesses merits of the two previous models. This new integrated model will be tested and verified by the real-time application in a BTX process or a crude unit process. The reliability and flexibility will be greatly enhanced compared to the previous model in spite of the low diagnosis speed. Nexpert Object for the expert system shell and SUN4 workstation for the hardware platform are used. TCP/IP for a communication protocol and interfacing to a dynamic simulator, SPEEDUP, for a dynamic data generation are being studied.

Production of $TGF-{\beta}1$ as a Mechanism for Defective Antigen-presenting Cell Function of Macrophages Generated in vitro with M-CSF

  • Macrophages generated in vitro using macrophage-colony stimulating factor (M-CSF) and interleukin (IL)-6 from bone marrow cells (BM-Mp) are defective in antigen presenting cell (APC) function as shown by their ability to induce the proliferation of anti-CD3 mAb-primed syngeneic T cells. However, they do express major histocompatibility (MHC) class I and II molecules. accessory molecules and intracellular adhesion molecules. Here we demonstrate that the defective APC function of macrophages is mainly due to production of $TGF-{\beta}1$ by BM-Mp. Methods: Microarray analysis showed that $TGF-{\beta}1$ was highly expressed in BM-Mp, compared to a macrophage cell line, B6D. which exerted efficient APC function. Production of $TGF-{\beta}1$ by BM-Mp was confirmed by neutralization experiments of $TGF-{\beta}1$ as well as by real time-polymerase chain reaction (PCR). Results: Addition of $anti-TGF-{\beta}1$ monoclonal antibody to cultures of BM-Mp and anti-CD3 mAb-primed syngeneic T cells efficiently induced the proliferation of syngeneic T cells. Conversely, the APC function of B6D cells was almost completely suppressed by addition of $TGF-{\beta}1$. Quantitative real time-PCR analysis also confirmed the enhanced expression of $TGF-{\beta}1$ in BM-Mp. Conclusion: The defective APC function of macrophages generated in vitro with M-CSF and IL-6 was mainly due to the production of $TGF-{\beta}1$ by macrophages.

Implementation of QoS-awared MAC Protocol for Home Networks (홈 네트워크를 위한 QOS 보장형 매체접속제어 프로토콜의 구현)

  • We believe that existing wire solutions such as HomePNA2.0 using phone lines and HomePlug using power line and wireless solution such as HomeRF are the most promising solutions, because of its cost-effectiveness. However, MAC protocols of these solutions provide only Class of Service(CoS) using priority mechanism like HomePNA and HomePlug or consider only voice among real-time traffics like HomeRF. For these reasons, we perceive the needs of the new MAC protocol which is no new wire solution and provides guaranteed Quality of Service (QoS) for not only voice but also video and audio. In light of this, we present the design and software implementation of a new MAC protocol for Home Networks. Our evaluation results of software implementation verify that proposed MAC protocol can provide low delay, low loss, and low jitter to real-time traffic by reserving bandwidth.

  • This paper presents a multiple face detector based on a robust pupil detection technique. The pupil detector uses active illumination that exploits the retro-reflectivity property of eyes to facilitate detection. The detection range of this method is appropriate for interactive desktop and kiosk applications. Once the location of the pupil candidates are computed, the candidates are filtered and grouped into pairs that correspond to faces using heuristic rules. To demonstrate the robustness of the face detection technique, a dual mode face tracker was developed, which is initialized with the most salient detected face. Recursive estimators are used to guarantee the stability of the process and combine the measurements from the multi-face detector and a feature correlation tracker. The estimated position of the face is used to control a pan-tilt servo mechanism in real-time, that moves the camera to keep the tracked face always centered in the image.

Cytoprotective Effect of Lespedeza Cuneata Extract on Glucose Toxicity (야관문(夜關門)의 포도당 독성에 대한 세포 보호 효과)

  • Objective: Production of ROS from glucose toxicity results in injury of pancreatic $\beta$-cells in diabetes models. This study was undertaken to examine the influence of Lespedeza Cuneata extract (LCE) on cytoprotective effects on glucose toxicity, insulin secretion and gene expression in RIN-m5F cells. Methods: First, we measured LCE's antioxidant activity by DPPH free radical-scavenging activity and SOD activity. After the various concentrations of LCE were added to the RIN-m5F cells, we measured cell viability with glucose stimulation by MTT assay and glucose-stimulated insulin secretion. We analyzed gene expression with Agilent whole mouse genome 44K oligo DNA microarray and searched for related pathways in KEGG (Kyoto Encyclopedia of Genes and Genomes). Lastly we measured INS-1, INS-2, INS-R, IRS-1, IRS-2, IRS-3, GLP-1R, and GLP-2R mRNA expression by real time RT-PCR. Results: Free radical-scavenging activity, SOD activity and insulin secretion increased dependent on LCE concentration, but LCE did not show considerable cytoprotective effect on RIN-m5F cells. More than twice expressed gene was 6362 in Oligo DNA chip. In KEGG, the most related pathway was the metabolic pathway. In the insulin signaling pathway, up expressed genes were Irs1, Mapk8, Akt1, and Lipe and down expressed genes were Rhoq, Fbp2, Prkar2b, Gck, and Prkag1. In real time RT-PCR, IRS-2, and IRS-3 expression increased significantly compared to the control group on LCE $12{\mu}g/m{\ell}$ concentration and GCK expression decreased significantly compared to the control group. Conclusions: These results show that LCE encourages insulin secretion and insulin metabolism by complicated gene mechanisms. Further mechanism study and clinical study seem to be necessary about Lespedeza Cuneata.

Tributyltin increases the expression of apoptosis- and adipogenesis-related genes in rat ovaries

  • Objective: Tributyltin (TBT), an endocrine disrupting chemical, has been reported to decrease ovarian function by causing apoptosis in the ovary, but the mechanism is not fully understood. Therefore, we examined whether TBT increases the expression of adipogenesis-related genes in the ovary and the increased expression of these genes is associated with apoptosis induction. Methods: Three-week-old Sprague-Dawley rats were orally administered TBT (1 or 10 mg/kg body weight) or sesame oil as a control for 7 days. The ovaries were obtained and weighed on day 8, and then they were fixed for terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) or frozen for RNA extraction. Using the total RNA of the ovaries, adipogenesis- and apoptosis-related genes were analyzed by real-time polymerase chain reaction (PCR). Results: The ovarian weight was significantly decreased in rats administered 10 mg/kg TBT compared to that in control rats. As determined by the TUNEL assay, the number of apoptotic follicles in ovary was significantly increased in rats administered 10 mg/kg TBT. The real-time PCR results showed that the expression of adipogenesis-related genes such as $PPAR{\gamma}$, ${\alpha}P2$, CD36, and PEPCK was increased after TBT administration. In addition, apoptosis-related genes such as $TNF{\alpha}$ and TNFR1 were expressed more in the TBT-administered rats compared with the control rats. Conclusion: The present study demonstrates that TBT induces the expression of adipogenesis- and apoptosis-related genes in the ovary leading to apoptosis in the ovarian follicles. These results suggest that the increased expression of adipogenesis-related genes in the ovary by TBT exposure might induce apoptosis resulting in a loss of ovarian function.

Real-Time Moving Object Tracking System using Advanced Block Based Image Processing (개선된 블록기반 영상처리기법에 의한 실시간 이동물체 추적시스템)

  • In this paper, we propose a real tine moving object tracking system based on block-based image processing technique and human visual processing. The system has two nun features. First, to take advantage of the merit of the biological mechanism of human retina, the system has two cameras, a CCD(Charge-Coupled Device) camera equipped with wide angle lens for more wide scope vision and a Pan-Tilt-Zoon tamers. Second, the system divides the input image into a numbers of blocks and processes coarsely to reduce the rate of tracking error and the processing time. Tn an experiment, the system showed satisfactory performances coping with almost every noisy image, detecting moving objects very int and controlling the Pan-Tilt-Zoom camera precisely.

Whitening Effect of Banana Leaf Extract (바나나잎 추출물의 미백 개선 효과)

  • This research was carried out to identify the whitening effect of Banana leaf extract. B16F10 cells were used to measure cell viability, mRNA expression, and tyrosinase activity inhibition assay from B16F10 cell. We also carried out clinical test of the cream product containing banana leaf extract. In this study, we elucidated the effects of banana leaf extract on TRP1 / TRP2 / Tyr mRNA expression and tyrosinase activity inhibition. Quantitative real-time PCR showed that banana leaf extract decreased mRNA level of TRP1, TRP2 and Tyr gene and tyrosinase activity inhibition assay also revealed that banana leaf extract 65% decreased melanin production in B16F10 cell. Banana leaf extract cream can whiten the skin darkness induced by ultraviolet. Therefore, we successfully identified the whitening effect of banana leaf extract, and this finding suggested the banana leaf extract is a considerable potent cosmetic ingredient for skin whitening. Based on this, we anticipated further researches about banana leaf extract for mechanism to develop not only cosmetics but healthcare food or medicines.

  • Red tide(harmful algae) in the Korean Coastal Waters has a given a great damage to the fishery every you. However, the aim of our study understands the influence of meteorological factors (air and water temperature, precipitation, sunshine, solar radiation, winds) relating to the mechanism of red tide occurrence and monitors red tide by satellite remote sensing, and analyzes the potential area for red tide occurrence by GIS. The meteorological factors have directly influenced on red tide formation. Thus, We want to predict and apply to red tide formation from statistical analyses on the relationships between red tide formation and meteorological factors. In future, it should be realized the near real time monitoring for red tide by the development of remote sensing technique and the construction of integrated model by the red tide information management system (the data base of red tide - meteorological informations). Finally our purpose is support to the prediction information for the possible red tide occurrence by coastal meteorological information and contribute to reduce the red tide disaster by the prediction technique for red tide.

The Effect of Morphine on REST Expression in Human Neuroblastoma NMB Cells

  • The mu opioid receptor (MOR) has been regarded as the main site of interaction with analgesics in major clinical use, particularly morphine. The repressor element-1 silencing transcription factor (REST) functions as a transcriptional repressor of neuronal genes in non-neuronal cells. However, it is expressed in certain mature neurons, suggesting that it may have complex and novel roles. In addition, the interactions between MOR and REST and their functions remain unclear. In this study, we examined the effects of morphine on the expression of REST mRNA and protein in human neuroblastoma NMB cells to investigate the roles of REST induced by MOR activation in neuronal cells. To determine the effects of morphine on REST expression, we performed RT-PCR, real-time quantitative RT-PCR, western blot analysis and radioligand binding assays in NMB cells. By RTPCR and real-time quantitative RT-PCR, the expression of REST was found to be unchanged by either the MOR agonist morphine or the MOR specific antagonist CTOP. By western blot, morphine was shown to significantly inhibit the expression of REST, but this suppression was completely blocked by treatment with CTOP. In the radioligand binding assay, the overexpression of REST led to an increased opioid ligand binding activity of endogenous MOR in the NMB cells. These results together suggest that morphine inhibits the expression of REST in human neuroblastoma cells through a post-transcriptional regulatory mechanism mediated through MOR.
