PREPROCESSING EFFECTS ON ON-LINE SSC MEASUREMENT OF FUJI APPLE BY NIR SPECTROSCOPY

  • The aims of this research were to investigate the preprocessing effect of spectrum data on prediction performance and to develop a robust model to predict SSC in intact apple. Spectrum data of 320 Fuji apples were measured with the on-line transmittance measurement system at the wavelength range of 550∼1100nm. Preprocess methods adopted for the tests were Savitzky Golay, MSC, SNV, first derivative and OSC. Several combinations of those methods were applied to the raw spectrum data set to investigate the relative effect of each method on the performance of the calibration model. PLS method was used to regress the preprocessed data set and the SSCs of samples, and the cross-validation was to select the optimal number of PLS factors. Smoothing and scattering corection were essential in increasing the prediction performance of PLS regression model and the OSC contributed to reduction of the number of PLS factors. The first derivative resulted in unfavorable effect on the prediction performance. MSC and SNV showed similar effect. A robust calibration model could be developed by the preprocessing combination of Savitzky Golay smoothing, MSC and OSC, which resulted in SEP= 0.507, bias=0.032 and R$^2$=0.8823.

  • The method of measuring and classifying the energy category of neutrons directly using raw data acquired through a CZT detector is not satisfactory, in terms of accuracy and efficiency, because of its poor energy resolution and low measurement efficiency. Moreover, this method of measuring and analyzing the characteristics of low-energy or low-activity gamma-ray sources might be not accurate and efficient in the case of neutrons because of various factors, such as the noise of the CZT detector itself and the influence of environmental radiation. We have therefore developed an efficient method of analyzing radiation characteristics using a neutron and gamma-ray analysis algorithm for the rapid and clear identification of the type, energy, and radioactivity of gamma-ray sources as well as the detection and classification of the energy category (fast or thermal neutrons) of neutron sources, employing raw data acquired through a CZT detector. The neutron analysis algorithm is based on the fact that in the energy-spectrum channel of 558.6 keV emitted in the nuclear reaction 113Cd + 1n → 114Cd + in the CZT detector, there is a notable difference in detection information between a CZT detector without a PE modulator and a CZT detector with a PE modulator, but there is no significant difference between the two detectors in other energy-spectrum channels. In addition, the gamma-ray analysis algorithm uses the difference in the detection information of the CZT detector between the unique characteristic energy-spectrum channel of a gamma-ray source and other channels. This efficient method of analyzing radiation characteristics is expected to be useful for the rapid radiation detection and accurate information collection on radiation sources, which are required to minimize radiation damage and manage accidents in national disaster situations, such as large-scale radioactivity leak accidents at nuclear power plants or nuclear material handling facilities.

  • To develop the somatic cell counting NIR Spectrum method within a range of 400-2500 nm, eosin-Y, methyl red, methylene blue, resazurin and amido black 10B were tested at 0.01% in raw milk. The PLS (Partial Least Square) results are summarized as follows: Correlation coefficients of the calibration model measurements by NIR spectroscopy in raw milk for eosin-Y, methyl red, methylene blue, resazurin and amido black 10B were 0.78, 0.65, 0.63, 0.65, 0.98 and 0.99, respectively. The correlation coefficients of the prediction model measurements by NIR spectroscopy in raw milk for eosin-Y, methyl red, methylene blue, resazurin and amido black 10B were 0.49, 0.21, 0.36, 0.47, 0.95 and 0.98 respectively. Based on these results, amido black 10B was the best additive for the NIRS somatic cell count method.

Development of Nondestructive Moisture Sensor for Agricultural Products Using Hydrogen NMR : Feasibility Experiments (수소자기공명원리를 이용한 비피괴적 농산물 함수율 측정장치 개발을 위한 기초실험)

  • Nuclear magnetic resonance(NMR) was used to study a feasibility test for the possibility to develop a moisture sensor applicable for agricultural products such as raw and powdered grain. Samples of wheat flour and brown rice were tested with a 200MHz hydrogen NMR. The samples were loaded into 5mm NMR glass tubes and $30^{\circ}$ pulses were supplied to get resonance signals. Hydrogen spectrum intensities of the samples were compared to moisture content measured by a drying oven method. High linear correlations were obtained between the hydrogen spectrum intensity and moisture content. Therefore, moisure contents of agricultural products could be measured nondestructively utilizing the NMR principle. A low-cost design concept of the moisture sensor with the NMR principle was proposed for a practical use.

  • HRV(heart rate variability) is the time series data of R-R interval time duration based on ECGs. Power spectral analysis of HRV has recently been used to define the activity of ANS(autonomic nervous system). In this study, 14 rats were divided into two groups, sympathectomy and vagotomy. During the experiments, ECGs of rats were collected three times at each experimental conditions or the duration of 5 minutes, where sampling frequency was set at 2KHz. After the application of the Berger's Serires algorithm to ECG raw data, power spectrum of HRV was obtained via FFT. Results showed that HF/LF were increased or the sympathectomy group and decreased or the vagotomy group. It implies that the variations in HF/LF components could be used or the ANS function classification.

A Study on the Physical and Chemical Properties of Teat Cup Liners (유두컵 라이너의 물리화학적 특성 조사)

  • This study was carried out to investigate the physical and chemical properties of teat cup liners. The hardness of the liners was $50\~67$, and their fatigue to failure 38-1,185 cycles. The elongation and tensile strength of these liners were about $134 kgf/cm^2\;and\;473\%$, respectively. The infrared spectrum and the gas chromatogram revealed that the liner A was consisted of NR, SBR, and BR, with a composition ratio of 60:20:20 (part per hundred rubber). The raw rubber materials used for liners B to G, on the other hand, were NBR only. However, the liner H was made of silicon rubber. The thermogravimetric analysis showed that the liners tested in this study contained raw rubber material, carbon black, organic compounds and metallic compounds.

  • To confirm the feasibility as raw materials for cosmetic formulation, ethanol, n-hexane, ethylacetate and normal butyl alcohol were extracted from 1kg of Ginko leaves. Ultimately, investigated chemical structure and physicochemical properties of ginkgetin and isoginkgetin. The purity of those two components have been evaluated higher than 99%. They are isomers with molecular weight as 566 and a molecular formular, $C_{32}H_{22}O_{10}$. Flavonoid's frame and NMR spectrum exhibited the existence of aromatic proton and the proper ratio of the carbon, which were expected structures exactly corresponded to the reported references.

Chemical Composition of Glycoprotein from Urechis unicinctus (개불(Urechis unicinctus) 당단백질의 성분조성과 특성)

  • To confirm the food quality of Urechis unicinctus which have been favored as a special raw seafood in southern area in Korea, the chemical composition of Urechis unicinctus and those glycoprotein were studied. Freeze dried Urechis unicinctus was composed of more than 70% of crude protein and 15% of total carbohydrate. The amino acids such as glycine(18.69%), glutamic acid(12.50%) and aspartic acid(9.37%) were noted as major components of total amino acids. The predominant free amino acids were alanine(32.98%), glycine(27.50%), asparagine(19.65%) and taurine(8.29%), and the sum of them were more than 88% to total free amino acids, so they may cause unique taste of Urechis unicinctus with sweetness. As the basis of sugar composition analysis, 56.35% of glucose and 30.49% of N acetylglucosamine were contained respectively, and they might also play an important role in a sweet taste. The leading carbohydrate moiety of glycoprotein from Urechis unicinctus was identified as glucose and N acetylglucosamine similarly to raw muscle, and they occupied more than 50% of total sugar content. Fucose(18.32%) and D glucuronic acid(12.31%) also detected in higher levels com pared to raw muscle. The total amino acid profile of glycoprotein showed a similar trend to raw muscle protein but glycine was detected a significantly lower than that in raw muscle. The glycoprotein from Urechis unicinctus was composed of 4 kinds of subunits, i.e. 25kDa, 20kDa, 18kDa and 12.5kDa of molecular weights through the SDS polyacrylamide gel electrophoresis. On the basis of the IR spectrum of absorptions appeared in 1,240cm-1 and 850cm-1, the glycoprotein had sulfate groups.

Analysis and Comparison of Cerebroside Components from Soybean Fermented Foods (대두발효식품 Cerebroside의 구성성분 분석 및 비교)

  • Cerebroside fatty acids, sugars and long-chain sphingoid bases in raw soybean and soybean fermented foods (chongkukjang and deunjang) were analyzed using gas chromatography-mass spectrometry (GC-MS) and high-pH anion exchange chromatography with pulsed amerometric detection (HPAEC-PAD). Fatty acids of acid-hydrolyzed cerebrosides were derivatized to O-TMS methylester and analysed. The major fatty acids in raw soybean and chongkukjang cerebrosides were identified as 2-hydroxyhexadecanoic acid (16 : 0h), 2-hydroxydocosanoic acid (22 : 0h) and 2-hydroxytetracosanoic acid (24 : 0h). In the case of deunjang cerebroside, 24 : 0h (40.9%) and 22 : 0h (23.4%) were major fatty acids, but 16 : 0h, 23 : 0h, 25 : 0h and 26 : 0h were also detected. Long-chain sphingoid bases of acid-hydrolyzed cerebrosides from raw soybean, chongkukjang and deunjang consisted primarily of 4-tracts, 8-tracts-sphingadienine (dihydroxy base, d18 : 2$\Delta$$^{4trans, 8trans}$) and sis-tracts isomers of 4-hydroxy-sphingenine (trihydroxy base, tl8:1$\Delta$$^{4trans or cis}$) with much less amounts of phytosphingosine (tl8: 0) and isomers of sphingenine (d18 : 1). Although deunjang is a soybean food fermented by fungi and microorganisms for a long period, 2-hydroxyoctadec-3-enoic acid (18 : 1h) and branched 9-methyl-4,8-sphingadienine known as compositional cerebroside fatty acids in Aspergillus species were not detected. Mass spectrum for sugar derivatives in cerebrosides of soybean foods including raw soybean and fermented soybean showed that C-1 of glucose moiety was linked to ceramide backbone as like a monoglucosylceramide.

Effect of Gamma Irradiation on the Conformational Changes of Garlic (Allium sativum) Protein, Allivin (감마선 조사가 마늘(Allium sativum) 단백질 Allivin의 구조적 변화에 미치는 영향)

  • Conformational changes of allivin, a major allergen protein in garlic, by gamma irradiation were observed. Allium solution (3.0 mg/mL in 0.01 M PBS, pH 7.4) and raw garlic were irradiated in a gamma irradiation facility to be absorbed with doses of 2.5, 5.0, 7.5 and 10.0 kGy. After irradiation, a half of irradiated allivin solution was filtered through 0.22 ${\mu}{\textrm}{m}$ filtration kit. UV spectrum and SDS-PAGE were used for observing the conformational changes of allivin by gamma irradiation. Turbidity of allivin solution was increased by optical observation, and the result was validated by the increase of absorbance at 660 m. Turbidity of irradiated and filtered allivin solution significantly (p<0.05) decreased, and the concentration of soluble protein in the solution decreased by increase of the dose. Aggregation among allivin molecules was observed by SDS-PAGE, but irradiated and filtered samples showed decrease of allivin with reducing aggregation trend. SDS-PAGE profiles of irradiated raw garlic were not different. Results indicate that gamma irradiation can decrease the allivin in solution state which may impact on allergenic reaction of garlic.
