PREPROCESSING EFFECTS ON ON-LINE SSC MEASUREMENT OF FUJI APPLE BY NIR SPECTROSCOPY

  • The aims of this research were to investigate the preprocessing effect of spectrum data on prediction performance and to develop a robust model to predict SSC in intact apple. Spectrum data of 320 Fuji apples were measured with the on-line transmittance measurement system at the wavelength range of 550∼1100nm. Preprocess methods adopted for the tests were Savitzky Golay, MSC, SNV, first derivative and OSC. Several combinations of those methods were applied to the raw spectrum data set to investigate the relative effect of each method on the performance of the calibration model. PLS method was used to regress the preprocessed data set and the SSCs of samples, and the cross-validation was to select the optimal number of PLS factors. Smoothing and scattering corection were essential in increasing the prediction performance of PLS regression model and the OSC contributed to reduction of the number of PLS factors. The first derivative resulted in unfavorable effect on the prediction performance. MSC and SNV showed similar effect. A robust calibration model could be developed by the preprocessing combination of Savitzky Golay smoothing, MSC and OSC, which resulted in SEP= 0.507, bias=0.032 and R$^2$=0.8823.

Development of an efficient method of radiation characteristic analysis using a portable simultaneous measurement system for neutron and gamma-ray

  • The method of measuring and classifying the energy category of neutrons directly using raw data acquired through a CZT detector is not satisfactory, in terms of accuracy and efficiency, because of its poor energy resolution and low measurement efficiency. Moreover, this method of measuring and analyzing the characteristics of low-energy or low-activity gamma-ray sources might be not accurate and efficient in the case of neutrons because of various factors, such as the noise of the CZT detector itself and the influence of environmental radiation. We have therefore developed an efficient method of analyzing radiation characteristics using a neutron and gamma-ray analysis algorithm for the rapid and clear identification of the type, energy, and radioactivity of gamma-ray sources as well as the detection and classification of the energy category (fast or thermal neutrons) of neutron sources, employing raw data acquired through a CZT detector. The neutron analysis algorithm is based on the fact that in the energy-spectrum channel of 558.6 keV emitted in the nuclear reaction 113Cd + 1n → 114Cd + in the CZT detector, there is a notable difference in detection information between a CZT detector without a PE modulator and a CZT detector with a PE modulator, but there is no significant difference between the two detectors in other energy-spectrum channels. In addition, the gamma-ray analysis algorithm uses the difference in the detection information of the CZT detector between the unique characteristic energy-spectrum channel of a gamma-ray source and other channels. This efficient method of analyzing radiation characteristics is expected to be useful for the rapid radiation detection and accurate information collection on radiation sources, which are required to minimize radiation damage and manage accidents in national disaster situations, such as large-scale radioactivity leak accidents at nuclear power plants or nuclear material handling facilities.

색소첨가 NIR을 이용한 우유 체세포수 측정법 개발 (Development of Rapid Somatic Cell Counting Method by Using Dye Adding NIR Spectroscopy)

  • 본 연구에서는 근적외선 분광광도계를 이용하여 원유의 체세포수 측정에 필요한 최적의 색소를 선정하기 위하여 eosin-Y, methyl red, methylen blue, resazurin 및 amido black 10B등의 5가지 색소를 0.01%첨가 후 근적외선 분광광도계를 이용하여 400-2,500 nm 영역에서 측청하였으며, PLS(partial least squar)분석 결과는 다음과 같다. 교정부 모델에서의 상관도는 raw milk 0.78, eosin-Y 0.65, methyl red 0.63, methylen blue 0.65, resazurin 0.98 그리고 amido black 10B는 0.99 였다. 또한, 검증부 모델에서의 상관도는 raw milk 0.49, eosin-Y 0.21, methyl red 0.36, methylen blue 0.47, resazurin 0.95 그리고 amido black 10B는 0.98 였다. 위 결과를 종합해 보면, amido black 10B를 첨가한 경우 검증부 모델의 상관도는 0.98, 검증부 오차(SEP)는 0.09로 가장 우수한 결과를 보였다.

수소자기공명원리를 이용한 비피괴적 농산물 함수율 측정장치 개발을 위한 기초실험 (Development of Nondestructive Moisture Sensor for Agricultural Products Using Hydrogen NMR : Feasibility Experiments)

  • Nuclear magnetic resonance(NMR) was used to study a feasibility test for the possibility to develop a moisture sensor applicable for agricultural products such as raw and powdered grain. Samples of wheat flour and brown rice were tested with a 200MHz hydrogen NMR. The samples were loaded into 5mm NMR glass tubes and $30^{\circ}$ pulses were supplied to get resonance signals. Hydrogen spectrum intensities of the samples were compared to moisture content measured by a drying oven method. High linear correlations were obtained between the hydrogen spectrum intensity and moisture content. Therefore, moisure contents of agricultural products could be measured nondestructively utilizing the NMR principle. A low-cost design concept of the moisture sensor with the NMR principle was proposed for a practical use.

  • HRV(heart rate variability) is the time series data of R-R interval time duration based on ECGs. Power spectral analysis of HRV has recently been used to define the activity of ANS(autonomic nervous system). In this study, 14 rats were divided into two groups, sympathectomy and vagotomy. During the experiments, ECGs of rats were collected three times at each experimental conditions or the duration of 5 minutes, where sampling frequency was set at 2KHz. After the application of the Berger's Serires algorithm to ECG raw data, power spectrum of HRV was obtained via FFT. Results showed that HF/LF were increased or the sympathectomy group and decreased or the vagotomy group. It implies that the variations in HF/LF components could be used or the ANS function classification.

유두컵 라이너의 물리화학적 특성 조사 (A Study on the Physical and Chemical Properties of Teat Cup Liners)

  • This study was carried out to investigate the physical and chemical properties of teat cup liners. The hardness of the liners was $50\~67$, and their fatigue to failure 38-1,185 cycles. The elongation and tensile strength of these liners were about $134 kgf/cm^2\;and\;473\%$, respectively. The infrared spectrum and the gas chromatogram revealed that the liner A was consisted of NR, SBR, and BR, with a composition ratio of 60:20:20 (part per hundred rubber). The raw rubber materials used for liners B to G, on the other hand, were NBR only. However, the liner H was made of silicon rubber. The thermogravimetric analysis showed that the liners tested in this study contained raw rubber material, carbon black, organic compounds and metallic compounds.

은행잎 추출물 유래 Ginkgetin 및 Isoginkgetin의 융복합 특성 연구 (Physicochemical properties of Ginkgetin and Isoginkgetin extracted from Ginko leaves)

  • 은행잎의 Biflavonoid인 ginkgetin과 isoginkgetin을 화장품 소재로 이용 가능성 여부를 확인하기 위해 이를 분리하여 화학 구조 및 물리화학적 특성을 규명하고자 하였다. 음건한 은행나무 단풍잎 1 kg을 순차적으로 ethanol, n-hexane, ethylacetate 및 normal butyl alcohol로 분획 추출하여 최종 ginkgetin과 isoginkgetin을 확보하였다. 두 화합물은 모두 HPLC를 통해 99% 이상 고 순도로 나타났고 MS spectrum을 통한 분자량은 566, 분자식은 $C_{32}H_{22}O_{10}$로 동일한 이성질체적 구조를 나타냈다. UV spectrum으로 flavonoid의 골격과 NMR로 aromatic proton의 존재와 탄소 수 및 구성비를 측정한 결과 문헌에 보고된 flavonoid 화합물의 정보와 일치하였다.

개불(Urechis unicinctus) 당단백질의 성분조성과 특성 (Chemical Composition of Glycoprotein from Urechis unicinctus)

  • To confirm the food quality of Urechis unicinctus which have been favored as a special raw seafood in southern area in Korea, the chemical composition of Urechis unicinctus and those glycoprotein were studied. Freeze dried Urechis unicinctus was composed of more than 70% of crude protein and 15% of total carbohydrate. The amino acids such as glycine(18.69%), glutamic acid(12.50%) and aspartic acid(9.37%) were noted as major components of total amino acids. The predominant free amino acids were alanine(32.98%), glycine(27.50%), asparagine(19.65%) and taurine(8.29%), and the sum of them were more than 88% to total free amino acids, so they may cause unique taste of Urechis unicinctus with sweetness. As the basis of sugar composition analysis, 56.35% of glucose and 30.49% of N acetylglucosamine were contained respectively, and they might also play an important role in a sweet taste. The leading carbohydrate moiety of glycoprotein from Urechis unicinctus was identified as glucose and N acetylglucosamine similarly to raw muscle, and they occupied more than 50% of total sugar content. Fucose(18.32%) and D glucuronic acid(12.31%) also detected in higher levels com pared to raw muscle. The total amino acid profile of glycoprotein showed a similar trend to raw muscle protein but glycine was detected a significantly lower than that in raw muscle. The glycoprotein from Urechis unicinctus was composed of 4 kinds of subunits, i.e. 25kDa, 20kDa, 18kDa and 12.5kDa of molecular weights through the SDS polyacrylamide gel electrophoresis. On the basis of the IR spectrum of absorptions appeared in 1,240cm-1 and 850cm-1, the glycoprotein had sulfate groups.

대두발효식품 Cerebroside의 구성성분 분석 및 비교 (Analysis and Comparison of Cerebroside Components from Soybean Fermented Foods)

  • 대두 및 대두발효식품인 청국장과 된장의 cerebroside 구성성분인 지방산, 당 및 장쇄 sphingoid base들을 GC-MS로 분석하였다. Cerebroside 산-가수분해물의 지방산들을 O-TMS methylester 유도체로 만들어 분석하였으며 대두 및 청국장의 경우 2-hydroxyhexadecanoic acid(16 : 0h)가 각각 52.2% 및 62.0%로 가장 많았고 2-hydroxydocosanoic acid(22 : 0h) 및 2-hydroxytetracosanoic acid(24 : 0h)도 15.0~17.6%이었다. 그러나 된장의 경우 16 : 0h는 10.2%로 많은 감소를 보인 반면 24 : 0h는 40.9%, 22 : 0h는 23.4% 었으며 23 : 0h, 25 : 0h 및 26 : 0h도 대두 및 청국장에 비하여 많았다. Cerebroside 산-가수분해물의 장쇄 sphingoid base들은 N-acetyl-O-TMS 유도체로 분석하였으며 대두 및 청국장에서 4- trans, 8-trans-sphingadienine (d18 : 2 $\Delta$$^{4trans, 8trans}$) 는 59.9% 및 44.5%이었으며 4-hydroxy-8-trans, cis-sphingening (t18 : 1$\Delta$$^{8trans or cis}$)는 20.9% 및 35.9% 이었고 된장 cerebroside 역시 dl8 : 2$\Delta$$^{4trans, 8trans}$가 가장 많았다. 그러나 phytosphingosine(t18 : 0) 및 sphingosine 이 성질체(d18 : 1)들은 거의 보이지 않았다. 또한 된장의 경우 오랜 기간 미생물들에 의하여 발효되었지만 Aspergillus 속들의 cerebroside가 가지는 것으로 알려진 2-hydroxyoxtadec-3-enoic acid(18 : 1h) 및 9-methyl-4,8-sphingadienine 등은 발견되지 않았다. 대두, 청국장 및 된장 cerebroside의 당유도체 mass spectrum 분석 결과 99%이상이 glucose 1번 탄소가 ceramide backbone에 결합된 monoglucocerebroside 인 것으로 나타났다.

감마선 조사가 마늘(Allium sativum) 단백질 Allivin의 구조적 변화에 미치는 영향 (Effect of Gamma Irradiation on the Conformational Changes of Garlic (Allium sativum) Protein, Allivin)

  • 감마선 조사에 의 한 마늘의 주요 알러젠 단백질인 allivin의 구조적 변화를 관찰하였다. Allivin 용액 (3.0 mg/mL in 0.01 M PBS, UH 7.4)과 생 마늘에 2.5, 5.0, 7.5, 10.0 kGy의 흡수선량을 갖도록 감마선을 조사하였다. 조사 후 용액의 절반을 0.22$\mu\textrm{m}$ filter로 여과하여 준비하였다. 감마선 조사에 의한 allivin의 변화는 UV spectrum과 SDS-PAGE를 사용하여 관찰하였다. 가시적으로 관찰하였을 때, 조사선량이 증가할수록 allivin 용액의 혼탁도가 증가하였으며 ,660 nm 파장에서의 흡광도의 증가로 확인할 수 있었다. 조사 후 여과한 allivin 용액의 UV spectrum 측정에서 혼탁도의 변화가 크게 감소되었으며, 선량이 증가할수록 용해된 단백질의 농도가 소하였다. SDS-PAGE시험에서 감마선 조사된 allivin의 큰 변화는 관찰되지 않았으나, 분자간 응집이 관찰되었다 그러나, 조사 후 여과한 시료에서 는 allivin의 감소가 관찰되었으며, 분자간 응집으로 발생된 끌림도 감소하였다. 생마늘에 대한 전기영동 결과는 차이가 없었다. 이 결과들은 감마선 조사가 수용액 상태에서 allivin의 알러지성을 감소시킬 수 있음을 시사한다.
