Chemical and Microbiological Quality, Capillary Electrophoresis Pattern, and Rennet Coagulation of UHT-treated and Irradiated Milk

  • To see the possibility of irradiation as an alternative to ultra high temperature (UHT) sterilization, the quality characteristics of milk were analyzed. Milk treated by UHT ($135^{\circ}C$ for 4 sec) and irradiation at higher than 3 kGy showed no viable counts after 7 days of storage at $4^{\circ}C$. The contents of certain amino acids of milk, such as Arg, Asp, Glu, Ile, Leu, Lys, Pro, Ser, Thr, and Tyr, were lower in irradiated groups at 10 kGy than in UHT-treated one, but no difference was observed between irradiated milks at less than 5 kGy and UHT. The capillary electrophoresis (CE) patterns of the milk irradiated at 10 kGy showed a similar trend to the raw milk, low temperature long time (LTLT, $63^{\circ}C$ for 30 min), and high temperature short time (HTST, $72^{\circ}C$ for 15 sec) treated. However, the CE pattern of UHT-treated milk was different. Rennet coagulation test agreed with the CE results, showing that all milk samples were coagulated by rennet addition except for UHT-treated milk after 1 hr. These results suggest that irradiation of milk reduce the content of individual amino acids but it may not induce severe conformational change at a protein level when compared with UHT treatment.

대전지역 대학생의 식품섭취빈도에 영향을 미치는 요인 연구 (Study on the Factors Influencing Food Consumption by Food Frequency Qustionnaire of University Students in Taejon)

  • The purpose of this study is to analyze the foods which 424(male 171, female 253) university students surveyed consumed frequently and to evaluate and the factors affecting their food consumption patterns. The survey was conducted at the beginning of nutrition courses each semester, March and September, 1998, with the questionnaire composed of general information, food, drinking and smoking habits, nutrition knowledge/attitude and food frequency questionnaire. The rates of alcohol drinking in male and female students were 89.2% and 78.1% respectively, and the rates of smoking were 68.1% and 1.6% in males and female. The nutrition knowledge score was higher in females than in males, but the nutrition attitude score was not significantly different between the male and female groups. The foods frequently consumed among students were cooked rice(14.3/week), kimchi(11.1/week), coffee(5.7/week), vegetables in soup, jjigae and jorim(3.5/week), carbonated beverages(3.3/week), cooked mixed rice(3.2/week). Ramyun and chocolate·candies·biscuits were frequently consumed, too. There were several factors influencing food consumption patterns. These were gender, regularity of mealtimes, the status of alcohol drinking and smoking, residence type and the levels of nutrition knowledge and attitude. Males ate more frequently cooked rice, coffee, carbonated beverages, ramyun, functional beverages and ham·sausage, while females ate more frequently cooked mixed rice and fruits. Those who had the habits of irregular mealtimes seemed to eat more soft drinks, instant foods and snacks. These trends were also found in the alcohol drinking and smoking groups. High level groups for nutrition knowledge of attitude score chose raw yellow green and green vegetables, cooked mixed rice, soybeans and seaweeds more frequently than the other groups. On the other hand, low level groups for nutrition knowledge or attitude score were apt to eat carbonated beverages and ramyun more frequently. Therefore, more attention should be taken to males, having habits of irregular mealtimes, alcohol drinking and smoking, and low level groups for nutrition knowledge or attitude score so as to improve their health.

국산 5종 포공영(蒲公英)의 항염 효과 및 성분 함량 비교 연구 (Anti-inflammatory effect and contents from the aerial part and root of the various Taraxacum spp. distributed in Korea)

  • Objectives : Taraxaci Herba et Radix (THR) is widely used as a food and medicinal herb in Korea. It has been used for treatment of virus inflammatory disease, liver diseases and gastritis. So far, anti-inflammatory effects and constituents of various species in THR has not been studied for comparison. The aim of this study is to compare the anti-inflammatory effects of the aerial part and root from various THR. Also, we have compared the contents of its known constituents with each. Methods : In this study, we estimated anti-inflammatory effect and compared their constituent by HPLC. For the determination of anti-inflammatory effects, we investigated NO and $PGE_2$ production by ELISA. The expressions of iNOS was determined by western blotting in LPS-induced RAW 264.7 macrophage cells. And, standard compounds which are methyl gallate, gallic acid, syringic acid and esculetin of THR were analyzed by HPLC using a $C_{18}$ column. Results : Methanol extracts of THR decreased NO and $PGE_2$ production. The expressions of iNOS protein were also decreased in methanol extracts of THR. As a result, HPLC analysis showed that they showed similar patterns. Methyl gallate and esculetin showed the highest content. Methyl gallate was included over 10% content in each aerial part and root of THR. Conclusions : These results indicate that most of THR distributed in Korea might represent therapeutic agent for treatment of inflammatory diseases.

Chemical constituents of Dicentra spectabilis and their anti-inflammation effect

  • Column chromatographic separation of the MeOH extract from the roots of Dicentra spectabilis yielded fourteen compounds, menisdaurin (1), menisdaurilide (2), trans-N-p-coumaroyltyramine (3), trans-N-p-feruloyltyramine (4), 4-O-feruloylquinicacid (5), chlorogenic acid (6), 3-O-feruloylquinicacid (7), ferulic acid (8), protopine (9), Kaempferol 3,7-di-O-${\beta}-{\text\tiny{D}}$-glucopyranoside (10), kaempferol 3-O-${\beta}-{\text\tiny{D}}$-glucopyranosyl-7-O-${\alpha}-{\text\tiny{L}}$-rhamnopyranoside (11), ${\alpha}-rhamnoisorobin$ (12), astragalin (13), and nicotiflorin (14). Their structures were determined on the basis of NMR spectroscopic data. Among them, compound 1, 3-8, and 10-14 isolated from this plant were reported for the first time. The isolated compounds (1-14) were tested for nitric oxide (NO) inhibitory activity on lipopolysaccharide-stimulated RAW 264.7 cells. Compound 3, 4 and 12 significantly inhibited NO production. Moreover, Compound 3 suppressed pro-inflammatory cytokines ($TNF-{\alpha}$, $IL-1{\beta}$ and IL-6) in a dose- dependent manner. These data suggest that compound 3 possess anti-inflammatory activity and might be useful natural materials for development of anti-inflammatory agent.

Effects of feminine cleanser Inclear on sperm motility: A prospective study

  • Objective: The objective of this study is to estimate the effects of Inclear, a feminine cleanser, on sperm motility. Methods: Semen samples were obtained from infertile male patients. Following liquefaction, the raw semen samples were diluted with Ham's F-10 nutrient mixture medium containing 0.4% human serum albumin solution at a ratio of 1:3. The semen samples were subsequently centrifuged to separate the seminal plasma from the serum. The supernatant was discarded, and the pellet was resuspended. The sample was again centrifuged to remove cell debris, and the supernatant was removed. The final pellet was gently loosened by resuspension and incubated in medium alone as a control, and in a 10% solution of the medium plus Inclear. A sampling time of 30 minutes was selected on the basis of sperm transport studies. Sperm motility was evaluated with computer-assisted sperm analysis. Results: A total of 20 samples were analyzed. The mean age of patients was $34.40{\pm}2.96years$. There was no difference in sperm concentration and motility in the two samples at 0 minute and 30 minutes of incubation. In both semen samples, the sperm concentration and motility decreased after an incubation period of 30 minutes. However, there was no statistical difference between the samples. Sperm concentration and motility were not significantly different between the control and Inclear samples after 0 minute and 30 minutes of incubation. Conclusion: Inclear has no negative effects on sperm motility. This product can be recommended to pregnancy planners for vaginal hygiene and as a vaginal lubricant.

액체 첨가가 없는 밀링법을 이용한 ibuprofen과 nicotinamide의 공결정 형성 (Liquid-free milling to prepare a cocrystal of ibuprofen and nicotinamide)

  • 약물의 공결정 형성은 결정구조를 디자인하여 물리적인 성질을 제어하는 방법으로 각광받고 있다. 특히, 밀링 결정화법은 약물 공결정의 형성 여부를 판단하기 위한 스크리닝 기법으로 널리 사용되고 있는데, 대부분의 경우에 소량의 액체를 첨가하는 것이 필요하다. 본 연구에서는 액체의 첨가가 없는 밀링법을 가능하게 하기 위해서, ibuprofen/nicotinamide의 공결정화에 온도가 미치는 영향을 살폈다. 액체질소 냉각 온도와 상온에서의 밀링을 비교하였을 때, 액체질소 냉각 상태에서 공결정화가 상대적으로 효과적으로 진행된 것을 XRD, DSC 등의 분석법을 이용하여 확인하였다. 이러한 결과는 원료 물질의 유리전이 온도 이하에서 밀링하였을 때 분자의 운동성이 제한되어, 원료 결정구조의 파쇄와 공결정 유도가 효과적으로 일어나는 것으로 해석할 수 있다. 본 결과가 약물 밀링 공결정화에 응용되기 위해서는 여러 약물 공결정에 대한 추가 연구가 필요하다.

명일엽 전초 및 생즙의 영양성분 분석 (The Nutritional Components of Aerial Whole Plant and Juice of Angelica keiskei Koidz)

  • 명일엽의 일반성분중 지방과 단백질은 잎에서 가장 많았으며, 수분, 총당 및 환원당은 줄기에서 가장 많았고 회분과 섬유소는 전초에서 가장 많이 나타났다. 무기질 함량은 K와 Ca이 가장 많고, 다음으로 Na, P, Mg의 순으로 많았으며, Mn, P, Zn을 제외한 나머지 성분은 잎에서 가장 많았다. Ascorbic acid, ${\beta}-carotene$, Thiamin, Riboflavin 등의 비타민성분도 잎에서 많은 함량을 나타내었다. 총 아미노산 조성은 17종이 동정되었으며 그 함량별로는 일반적으로 glycine, glutamic acid, alanine, aspartic acid, leucine 순이었으며, 필수아미노산의 비율은 전초, 잎과 줄기에서 각각 39.74, 39.87, 37.10%이었다. 유리아미노산은 9종의 필수아미노산이 포함되어 있었고, 그 함량은 전초에서 leucine, alanine, valine, methionine, 잎에서 alanine, lysine, aspartic acid, leucine, 줄기에서 aspartic acid, methionine, glutamic acid, alanine순으로 많이 함유되어 있었다. 지방산 조성은 각 부위에서 14종이 동정되었으며 그중 linoleic acid, linolenic acid, palmitic acid, margaric acid 등이 주성분이었고, 특히 전초와 줄기에서는 linoleic acid가 각각 31.14, 43.29%이었고, 잎에서는 linolenic acid가 35.14%이었다.

Rapid and Nondestructive Discrimination of Fusarium Asiaticum and Fusarium Graminearum in Hulled Barley (Hordeum vulgare L.) Using Near-Infrared Spectroscopy

  • Purpose: This study was conducted to discriminate between normal hulled barley and Fusarium (Fusarium asiaticum and Fusarium graminearum) infected hulled barley by using the near-infrared spectroscopy (NIRS) technique. Methods: Fusarium asiaticum and Fusarium graminearum were artificially inoculated in hulled barley and the reflectance spectrum of the barley spike was obtained by using a near-infrared spectral sensor with wavelength band in the range 1,175-2,170 nm. After obtaining the spectrum of the specimen, the hulled barley was cultivated in a greenhouse and visually inspected for infections. Results: From a partial least squares discriminant analysis (PLS-DA) prediction model developed from the raw spectrum data of the hulled barley, the discrimination accuracy for the normal and infected hulled barley was 99.82% (563/564) and 100% (672/672), respectively. Conclusions: NIRS is effective as a quick and nondestructive method to detect whether hulled barley has been infected with Fusarium. Further, it expected that NIRS will be able to detect Fusarium infections in other grains as well.

Effects of Gelatin Hydrolysates Addition on Technological Properties and Lipid Oxidation of Cooked Sausage

  • This study investigated the impacts of gelatin hydrolysate addition on the technological properties and lipid oxidation stability of cooked sausage. Gelatin hydrolysate was prepared from pork and duck skin gelatin, through stepwise hydrolysis using collagenase and pepsin. The cooked sausages were formulated without gelatin (control) or with 1% pork skin gelatin, 1% duck skin gelatin, 1% pork skin gelatin hydrolysate, and 1% duck skin gelatin hydrolysate. The pH, color characteristics, protein solubility, cooking loss, and textural properties of cooked sausages were evaluated, and the 2-thiobarbituric acid reactive substances (TBARS) value was measured weekly to determine lipid oxidation stability during 4 wk of refrigerated storage. Enzymatic hydrolysis of gelatin decreased protein content and CIE L* but increased redness and yellowness (p<0.05). When 1% gelatin or gelatin hydrolysate was incorporated in cooked sausage, however, little to no impacts on pH value, moisture content, protein content, color characteristics, protein solubility, and cooking loss were found (p>0.05). The addition of 1% duck skin gelatin hydrolysate increased the cohesiveness and chewiness of cooked sausages. The inclusion of 1% duck skin gelatin accelerated lipid oxidation of cooked sausages during refrigerated storage (p<0.05), whereas duck skin gelatin hydrolysate caused a lower TBARS value in cooked sausage compared to duck skin gelatin. The results show comparable effects of gelatin and gelatin hydrolysate addition on the technological properties of cooked sausages; however, the oxidative stability of raw materials for gelatin extraction should be evaluated clearly in further studies.

산국 꽃의 항염 활성 연구 (Anti-inflammatory effects of Chrysanthemum boreale flower)

  • Objectives : Chrysanthemum boreale flower is widely distributed in Korea, Japan, China, and Eastern countries. C. boreale flower is also one of the herbs used for the treatment of various inflammatory disease in Korean Medicine. So, this research was designed to study anti-inflammatory effect of C. boreale flower and its mechanism. Methods : We investigated nitro oxide (NO) and prostaglandin $E_2$ ($PGE_2$) production by ELISA. And expressions of inducible nitric oxide synthase (iNOS), Cyclooxygenase-2 (COX-2) and nuclear factor-${\kappa}B$ P50/65 (NF-${\kappa}B$ P50, NF-${\kappa}B$ P65) were measured in RAW 264.7 murine macrophage cells induced by LPS. Results : MeOH ex., EtOAc fr., $CHCl_3$ fr. and Water fr. of C. boreale flower showed anti-inflammatory effect through inhibition of NO and PGE expression respectively. Among them, EtOAc fr. and $CHCl_3$ fr. inhibited production of NO and $PGE_2$ through inhibition of iNOS and COX-2 expression. And MeOH ex., EtOAc fr. and $CHCl_3$ fr. inhibited translocation of NF-${\kappa}B$ P65, NF-${\kappa}B$ P50 by inhibiting phosphrylation of $I{\kappa}B$. Conclusions : MeOH ex. EtOAc fr, $CHCl_3$ fr., and Water fr. of the C. boreale flower have anti-inflammatory activity.
