Effects of Taeumjowetang on Lipid Peroxidation by Free Radicals and Oxidative Damage of Hepatocytes by tert-Butyl Hydroperoxide (태음조위탕(太陰調胃湯)의 항산화(抗酸化) 효능(效能)에 의한 간세포(肝細胞) 보호(保護) 효과(效果)에 관한 연구(硏究))

  • Effects of Taeumjowetang on Lipid Peroxidation by Free Radicals and Oxidative Damage of Hepatocytes by tert-Butyl Hydroperoxide. 1. Purpose The present study was carried out to evaluate the antioxidant effects of Taeumjowetang in vitro. 2. Methods In this study, antioxidant effects of TJT on lipid peroxidation were determined according to the method of TBA. (Abbreviation) TJT : Taeumjowetang, TBA : 2-thiobarbituric acid. 3. Results : 1) TJT inhibited markedly peroxidation of linoleic acid during the autoxidation. 2) TJT inhibited lipid peroxidation induced by hydroxyl radical derived from H2O2-Fe2+ in rat liver homogenate. 3) TJT showed 66% scavenging effect on DPPH radical. 4) TJT exhibited a 25% inhibitory effect on superoxide generation from xanthine-xan thine oxidase system. 5) To investigate the antioxidative effects of TJT on the hepatocytes, cultured normal rat liver cells(Ac2F) were prepared and incubated with or without TJT. After 16~18hr, cells placed in DMEM medium without serum, and then incubated with 1mM t-BHP for 2hr. Viable cells were detected by MTT assay. In this test, TJT protected the cell death induced by t-BHP and significantly increased cell viability in the normal rat liver cell.   • The protective effect of Bifidobacterium breve K-110, K-111 and B. infantis K-525 on carbon tetrachloride induced hepatotoxicity in rats was investigated. Among them, B. infantis K-525 had the most potent hepatoprotective activity. It reduced serum aspartate aminotransferase and alanine aminotransferase levels to 51% and 80% of the $CCl_{4}-treated$ groups, respectively. In rat liver homogenate intoxificated with $CCl_{4}$, B. infantis K-525 inhibited in vitro as well as in vivo lipid peroxidation more than the other Bifidobacteria.

Effect of Korean Red Ginseng Powder on the Lipid Concentrations and Tissue Lipid Peroxidation in the Rats Fed High Fat Diet (고지방급여 흰쥐의 혈청과 간의 지질 농도 및 조직 과산화지질 농도에 미치는 홍삼분말의 영향)

  • Effect of Korean red ginseng (KRG) on the level of serum and liver lipids and lipid peroxidation was investigated in the rats fed high fat diet. Content of serum total cholesterol was significantly decreased (P<0.05) in KRG I group and KRG II group. Content of HDL-cholesterol was significantly increased by 69.75% and 39.15% in KRG I and KRG II group compared to control group, respectively. Atherogenic index (hi) was also significantly decreased by 74.76% and 37.38% in KRG I and KRG II groups compared to control group, respectively. Serum triglyceride content was significantly decreased (p<0.05) in only KRG II group. Antioxidative activity of KRG on the lipid peroxidation of serum and tissues in rats was also studied in vivo by measuring the formation of thiobarbituric acid reactive substances (TBARS). Contents of TBARS in the serum of both KRG groups were significantly decreased (p<0.05) and that of nonheme iron in serum was significantly increased (p<0.05) in a dose-dependent manner, which suggested that lipid peroxidation contents are inversely correlated with serum nonheme iron content. Content of TBARS in liver was significantly decreased (p<0.05) in KRG I and KRG II groups, without any influence in other tissues. Content of TBIARS in liver microsomal fractions stimulated by Fe$^{2+}$/ascorbate was significantly decreased (p<0.05) in KRG I and KRG II groups, whereas this observation did not occur in liver mitochondrial fractions. When the effect of KRG on TBARS content in the liver fractions of homogenates, microsomes, and mitochodria stimulated by Fe$^{2+}$/ascorbate was tested in vitro experimental model, TBARS of liver three fractions was significantly decreased at 6 mg/mL KRG compared with those of control. These results suggested that KRG powder have hypocholesterolemic effect as well as antioxidative effect in the serum and liver of the rats fed high fat diet.

The Spermatogenic Effect of Yacon Extract and Its Constituents and Their Inhibition Effect of Testosterone Metabolism

  • We screened the pharmacological effects of a 50% ethanol extract of Yacon tubers and leaves on spermatogenesis in rats. As a result, we found that Yacon tuber extracts increased sperm number and serum testosterone level in rats. It has been reported that the crude extract of Yacon tubers and leaves contain phenolic acids, such as, chlorogenic acid, ferulic acid and caffeic acid by HPLC/MS analysis. We were interested in the contributions made by phenolic acid, particularly chlorogenic acid of Yacon tuber extract to the spermatogenic activity. After administering Yacon tuber extract or chlorogenic acid to rats for 5 weeks, numbers of sperm in epididymis were increased by 34% and 20%, respectively. We also administered ferulic acid, which has been reported to be a metabolite of chlorogenic acid and a constituent of Yacon tuber extract to investigate its spermatogenic activity in rats. Yacon tuber extract and ferulic acid increased sperm numbers by 43% and 37%, respectively. And, Yacon tuber extract, and chlorogenic acid showed significantly inhibition effect of testoeterone degradation in rat liver homogenate. We considered that the spermatogenic effect of Yacon tuber extract might be related to phenolic compounds and their inhibitory effect of testosterone degradation. Yacon showed the possibility as ameliorable agents of infertility by sperm deficiency and late onset hypogonadism syndrome with low level of testosterone.

Effects of Butanol Extract of Aralia elata on Lipid Peroxidation (두릅나무 부탄올 추출물이 지질 과산화에 미치는 영향)

  • It is well known that lipidperoxide, formed in vivo, induced the denaturation of enzyme and destruction of cell membrane to acute injury of tissue. Aralia elata have physiological activates, the improvement of lipid metabolism, antidiabetic activity etc., which was thought to have the relationship to lipid peroxidation. The anti-lipidperoxidative effect of Aralia elata have not yet established. In this study, we examined the anti-lipidperoxidative effects of Aralia elata (Butanol fraction) on CCI$_{4}$ induced lipidperoxidation in rats, and elucidated the anti-lipidperoxidative mechanism. In rat liver homogenate intoxicated with CCI$_{4}$ (0.5 ml/100g), BuOH fraction of Aralia elata (80 mg/Kg/day) exhibited 85.41% anti-lipidperoxidative effect but in serum 69.63% inhibitory effects, respectively. In mitochondrial and microsomal fraction showed inhibition of 55.85% and 69.30%, respectively. In order to elucidate the mechanism of anti-lipidperoxidation effects of Aralia elata, enzymatic (NADPH dependent) and non-enzymatic (Ascorbic acid catalyzed) reaction, in vitro, were performed. In enzymatic reation, Aralia elata exhibited 59.43% anti-lipidperoxidation effects, but in non-enzymatic reaction exhibited 43.27% inhibition. Therefore, it is noteworthy that antioxidative power of them may mainly results from the inhibition by enzymatic reaction.

Inhibition Effect of Testosterone Metabolism of Some Natural Products Containing Yacon and their Ameliorative Effect of Benign Prostatic Hyperplasia Symptom (야콘을 포함한 수종 천연물의 남성호르몬 대사 억제효과 및 전립선비대 증상 개선효과)

  • We reported previously that the extracts of Yacon tuber and leaf have spermatogenic activity and testosterone level increasing effect in men. The aim of this study was to investigate the mechanism of testosterone level increasing effect of Yacon tuber and leaf extracts. In this study, we found that Yacon leaf extracts and chlorogenic acid, a constituent of Yacon extracts showed significantly inhibition effect of testosterone degradation in rat liver homogenate. We suggest that this could be a part of direct mechanism about increasing effect of testosterone level. Futhermore, we evaluated the inhibition effect of testosterone metabolism of Torilis japonica and Coix lacryma-jobi have been reported as $5{\alpha}$-reductase inhibitor. The inhibition ratio of testosterone metabolism of these were Torilis japonica>Coix lacryma-jobi>Yacon. We also evaluated the benign prostatic hyperplasia (BPH) score with the preparation composed of these natural product, Yacon pill. It showed the decreased the BPH score in men. Rats with BPH showed significantly increased dihydrotestosterone (DHT) levels in the serum; however, Yacon pill extracts treated rats showed significant reduction of DHT levels compared with the BPH group. Conclusively, Yacon pill showed the possibility as ameliorable agents of BPH symptoms.
