치자(梔子) 약침(藥鍼)이 백서(白鼠) 모델 족과 염좌(捻挫) 통증(痛症)에 미치는 영향(影響) (Effect of Frutus gardeniae herbal acupuncture on the rat model of ankle sprain pain)

  • Objective : Frutus gardeniae, seed of Gardenia jasminoides Ellis is one of the crude drugs used for the treatment of inflammatory condition in oriental medicine. Methodes : The present study aimed to examine the analgesic effect and anti-inflammatory effect of Frutus gardeniae extract (FGE) on a rat model of ankle sprain pain, and the relations between FGE-induced effect and endogenous nitric oxide (NO) and inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), and c-Fos protein expression in the spinal cord. As a chronic pain model, ankle sprain pain model was used to test the effect of FCE injection applied to acupuncture point. After the induction of ankle sprain, rats subsequently showed a reduced stepping force of the affected limb for at least the next 4 days. The reduced stepping force of the limb was presumably due to a painful knee. FGE dissolved in normal saline was injected several acupoints. Results : After the treatment, behavioral tests measuring stepping force were periodically conducted during the next 8 hours. FGE produced significant improvement of stepping force of the hindlimb affected by the ankle sprain lasting at least 4 hours. FGE produced the improvement of stepping force of the affected hindlimb in a dose-dependent manner. In addition, FGE injection showed inhibitory effect on the paw edema induced by ankle sprain. Both NO production and iNOS, COX-2 protein expression increased by ankle sprain were suppressed by FGE. FGE on combination with electroacupuncture (EA) produced more powerful and longer lasting improvement of stepping force of the hindlimb affected by the ankle sprain than either FGE or EA did. The present study suggest that FGE produces a potent analgesic effect on the ankle sprain pain model of the rat and that FGE-induced analgesia modulate endogenous NO through the suppression of iNOS/COX-2 protein expression.

Anti-inflammatory Action of Phenolic Compounds from Gastrodia elata Root

  • Previous screening of the pharmacological action of Gastrodia elata (GE) root (Orchidaceae) showed that methanol (MeOH) extracts have significant anti-inflammatory properties. The antiinflammatory agents of GE, however, remain unclear. In this experiment, MeOH extracts of GE were fractionated with organic solvents for the anti-inflammatory activity-guided separation of GE. Eight phenolic compounds from the ether (EtOEt) and ethyl acetate (EtOAc) fractions were isolated by column chromatography: 4-hydroxybenzaldehyde (I), 4-hydroxybenzyl alcohol (II), benzyl alcohol (III), bis-(4-hydroxyphenyl) methane (IV), 4(4'-hydroxybenzyloxy)benzyl-methylether (V), 4-hydroxy-3-methoxybenzyl alcohol (VI), 4-hydroxy-3-methoxybenzaldehyde (VII), and 4-hydroxy-3-methoxybenzoic acid (VIII). To investigate the anti-inflammatory and anti-oxidant activity of these compounds, their effects on carrageenan-induced paw edema, arachidonic acid (AA)-induced ear edema and analgesic activity in acetic acid (HAc)-induced writhing response were carried out in vivo; cyclooxygenase (COX) activity, reactive oxygen species (ROS) generation in rat basophilic leukemia (RBL 2H3) cells and 1,1-diphenyl-2-picryl-hydroazyl (DPPH) scavenging activity were determined in vitro. These phenolic compounds not only had anti-inflammatory and analgesic properties in vivo, but also inhibited COX activity and silica-induced ROS generation in a dose-dependent manner. Among these phenolic compounds, compound VII was the most potent anti-inflammatory and analgesic. Compound VII significantly inhibited silica-induced ROS generation and compound VI significantly increased DPPH radical scavenging activity. Compounds I, II and III significantly inhibited the activity of COX-I and II. These results indicate that phenolic compounds of GE are anti-inflammatory, which may be related to inhibition of COX activity and to anti-oxidant activity. Consideration of the structure-activity relationship of the phenolic derivatives from GE on the anti-inflammatory action revealed that both C-4 hydroxy and C-3 methoxy radicals of benzyl aldehyde play an important role in anti-inflammatory activities.

장수진흙버섯 자실체 추출물의 다양한 생리활성 효과 (Various physiological effects from fruiting body extracts of Phellinus baumii)

  • 본 연구에서는 장수진흙버섯 자실체로부터 메탄올과 열수를 이용해 추출한 물질의 항당뇨, 항콜린에스테라아제, 항염증 효과를 탐색하였다. 항당뇨 실험에서 α-amylase 효소에 대한 메탄올과 열수 추출물의 저해 효과는 1.0~2.0 mg/ml의 농도에서 각각 50.67~61.33%와 52.67~65.00%를 보여 양성대조군인 acarbose의 83.67~96.33%에 비해 크게 낮았으나 α-glucosidase에 대한 메탄올과 열수 추출물의 1.0~2.0 mg/ml의 농도에서의 저해 효과는 각각 74.33~89.67%와 75.67~91.00%를 보여 53.67%와 84.67%를 보인 양성대조군 acarbose에 비해 유의하게 높아 탄수화물 분해효소인 α-glucosidase에 대한 저해 효과가 특이적으로 높았다. 메탄올과 열수 추출물의 아세틸콜린 에스테라아제에 대한 저해 효과는 1.0 mg/ml의 농도에서 각각 96.05%와 94.58%를 보여 양성대조군인 galanthamine (97.80%)과 유사하였으나 butyrylcholinesterase에 대한 저해효과는 1.0 mg/ml의 농도에서 각각 91.05%와 82.27%를 보여 galanthamine의 저해 효과 81.12%에 비해 유의하게 높았다. 염증 저해 효과 실험에서 RAW 264.7 대식세포가 배양되고 있는 배지에 메탄올과 열수 추출물을 각각 전처리 후 염증 매개 물질인 LPS를 1 ㎍/ml 처리하여 24시간 배양 후 NO 생성의 저해 효과를 조사한 결과 각각의 추출물 농도가 증가함에 따라 생성된 NO의 양도 점차 유의하게 감소하는 경향을 나타내었다. Carrageenan의 주사에 의해 흰쥐의 뒷발에 유도된 부종 저해실험에서는 투여한 메탄올 추출물의 농도가 증가함에 따라 뒷발에 유발된 부종의 용적도 농도 의존적으로 점차 감소하는 추세를 나타냈다. 따라서 장수진흙버섯의 자실체에 함유된 물질은 항당뇨, 항콜린에스테라아제 및 항염증 효과를 지니고 있어서 천연 건강식품으로의 개발 가능성이 높은 것으로 사료 된다.
