해간전(解肝煎)의 항산화(抗酸化) 활성(活性) 및 간세포(肝細胞)의 산화적(酸化的) 손상(損傷)에 대한 보호효과(保護效果) (Antioxidative and Protective Effects of Haeganjeon Extract on Oxidative Damage of Hepatocytes)

  • Objectives: Haeganjeon(HGJ) has been used for the treatment of liver disease in traditional medicine. The present study was carried out to evaluate the antioxidant and protective effects of HGJ extract on oxidative damage of hepatocytes by tert-butyl hydroperoxide(t-BHP). Methods: In the linoleic acid water-alcohol system, the levels of lipid peroxide(LPO) were determined by TBA method. The scavenging effect of HGJ on ${\alpha},{\alpha}-diphenyl-{\beta}-picrylhydrazyl$(DPPH) radical was determined according to the method of Hatano. In the Fenton system(ferrous ion reaction with hydrogen peroxide), the levels of hydroxyl radical induced LPO in rat liver homogenate were determined according to the method of TBA. Inhibitory effect of HGJ on superoxide generation was measured by xanthine-xanthine oxidase system. In order to evaluate antioxidative activity of HGJ in the liver cell, cultured normal rat liver cells(Ac2F) were prepared and incubated with or without HGJ. After 18hr, cells placed in DMEM medium without serum, and then incubated with 1mM tert-butyl hydroperoxide(t-BHP) for 2hrs. Viable cells were detected by MTT assay. Conclusions: In the linoleic acid autoxidation system, HGJ extract significantly inhibited the time course of the lipid peroxidation. These effects were similar to those of BHA HGJ extracts showed about 70% scavenging effect on DPPH radical. And HGJ extract inhibited the lipid peroxide formation in rat liver homogenate induced by hydroxyl radical derived from Fenton system. In addition, HGJ extract protected the cell death induced by t-BHP and significantly increased cell viability in the normal rat liver cell. These result indicated that HGJ extract might playa protective role against oxidative hepatic cell injury by means of free radical scavenger.

Carcinogen (3-methyl-4-dimethyl-aminoazo benzene) 처리후 간세포막에서의 Transferrin Receptor 변동에 관한 연구 (Transferrin Receptors in the Liver Cell Membrane of Carcinogen (3-methyl-4-dimethyl-arninoazobenzene) Treated Rat)

  • 화학물질에 의한 간암 유발과정에서 transferrin receptor (TfR)의 변동을 밝히기 위해 간을 부분절제한 정상백서의 재생간과 발암물질로 3-Me-DAB를 8주간 투여한 백서 또는 약물 투여 후 부분 간절제 수술을 행하여 세포분열을 유도시킨 백서 간조직으로부터 parenchymal cell (PC)과 nonparenchymal cell (NPC)를 분리하고 각각의 세포막을 제조하여 $^{125}I-transferrin$ 결합실험을 실시한 바 다음과 같은 성적을 얻었다. 1. 3-Me-DAB 투여에 의하여 간조직에서 oval cell의 증식, 재생성 변화, 결절형성, 담관의 증식 및 담관세포암 등의 현저한 조직학적 변화가 동반되었다. 그러나 간세포증식을 더욱 촉진시키기 위하여 부분간절제 수술을 하였을 때 수술 후 경과에 따른 형태학적 변동은 큰 차이가 없었다. 2. 정상 재생간의 PC 및 NPC homogenate에서 transferrin 결합량은 부분간 절제 수술 후 1일 및 3일에 증가되었으며 수술 후 7일에 정상으로 회복되었다. 3-Me-DAB 투여에 의해 두세포군에서 모두 정상세포보다 높게 나타났으며 재생기간에 따라 계속 증가되었다. 3. 정상간의 NPC 세포막에서 transferrin 최대 결합량 (Bmax)은 PC 세포막에서 보다 많이 분포되어 있었으며, Kd는 양세포막에서 5.05 또는 6.3nM로 비슷하였다. 4. 재생간의 NPC 및 PC 세포막에서 transferrin 결합량은 부분 간절제 수술 후 1일 및 3일에 $40{\sim}50%$ 증가되었고 수술 후 7일에 정상치로 회복되었다. 5. 3-Me-DAB 처치에 의하여 NPC 및 PC 세포막의 transferrin 결합량은 정상 간세포막에서 보다 약 3배 증가되었고, 3-Me-DAB 투여후 재생간의 NPC 세포막에서는 부분 간절제 수술 후 3일까지 증가된 후 감소되는 양상인데 반해 PC 세포막에서는 수술 후 7일까지 계속 증가되었다. 6. 3-Me-DAB 투여 후 NPC 및 PC 세포막 transferrin binding site에서 Kd치가 $3.1{\sim}4.1\;nM$$25.4{\sim}54.1\;nM$인 두 종류가 존재하는 것으로 나타났다. 이상의 실험성적으로 TfR는 1) 간조직의 PC 및 NPC 세포에 모두 분포되어 있으며, 2) 정상 재생간 및 3-Me-DAB의 처리 후 간세포에서의 세포막 TfR의 증가는 세포내 합성량의 증가에 의하여 일어나며, 3) 정상 재생간의 세포막 TfR는 한 종류의 high affinity site $(Kd,\;<3.1{\sim}7.5\;nM)$에 의하여 증가되나, 3-Me-DAB 처리 후 간세포막에서는 정상에서와 같은 high affinity형 이외에 affinty가 낮은 다른 형태의 TfR $(Kd,\;25.4{\sim}54.1\;nM)$가 세포막으로 출현됨으로써 크게 증가되는 것으로 사료된다.

해양 V. vulnificus의 내독소가 rat의 혈액 성분에 미치는 독성 효과 (The Toxic Effect Of Marine V. Vulnificus Endotoxin OH the Blood Component in Rat)

  • Endotoxin from the cell wall of marine V. vujnificus was .extracted using the hot phenol-water method, injected endotoxin into rat, and tested the toxic effect of endotoxin on the blood component In rat blood. The results showed that blood glucose, blood urea nitrogen, white blood cell, and reticulocyte were Increased and red blood cell was the same as the number of control group(normal blood), but platelet was decreased. Above results suggested that endotoxin induced a malfunction of liver and that the Increase of white blood cell was for the removal of foreign toxic substance.

Effect of Toluene Application to Skin on the Liver Injury in Rats

  • To investigate an effect of the topical toluene application to .at skin on the liver injury, toluene (35 mg/$cm^2$) was sequentially applied for 3 or 5 days to rat skin and then the animals were sacrificed. 5 day toluene-treated rats showed the slight increase of live. weight per body weight(%) compared with control. Serum levels of xanthine oxidase and alanine aminotransferase activity were significantly increased both in 3 days and 5 days toluene-treated animals compared with control. In the histopathological findings, cytoplasmic degeneration of hepatocytes around the central vein was noted in the liver of rats applied with toluene to the skin. These results indicate toluene application to rat skin feds to somewhat slight liver injury. On the other hand, the hepatic benzylalcohol or aldehyde dehydrogenase activities were significantly decreased by toluene application to rat skin. In conclusion, the liver min was induced by toluene application to rat skin, and it can be hypothesized that accumulation of benzaldehyde in liver cell may be responsible for liver injury.

Increased Expression of Epithelial Cell Adhesion Molecule (EpCAM) in Rat Hepatic Tumors Induced by Diethylnitrosamine

  • The epithelial cell adhesion molecule (EpCAM) is a pan-epithelial differentiation antigen that is expressed on almost all carcinomas. However, a role in rat liver carcinogenesis has never been reported previously. Thus, its expression was investigated herein in rat liver tumors induced by diethylnitrosamine (DEN). Twenty male 5-week-old F344 rats were used in this experiment. Mini-osmotic pumps containing doses of 47.5 mg of DEN were inserted into the abdominal cavity of each animal to initiate liver carcinogenesis. All animals were sacrificed at 26 weeks after DEN treatment. At necropsy, hepatic masses were processed for histopathological examination, which revealed forty-four hepatocellular adenomas (HCAs) and twenty hepatocellular carcinomas (HCC). Tumors were immunohistochemically analyzed for EpCAM, proliferating cell nuclear antigen (PCNA) and co-localization of the two. EpCAM expression was mainly detected in hepatic tumor cells, showing a cytoplasmic staining pattern. However, expression was also slightly observed in normally-appearing surrounding hepatic cells. PCNA expression was highly detected in tumor cells, showing nuclear staining. Double staining of EpCAM and PCNA in tumors showed many cells with co-localization. Taken together, EpCAM and PCNA expression were increased in DEN-induced tumors and many tumor cells showed co-expression. It is suggested that EpCAM may increase during DEN-induced tumors, possibly associated with cell proliferation.

Role of Kupffer Cells in Cold/warm Ischemia-Reperfusion Injury or Rat Liver

  • The mechanisms of liver injury from cold storage and reperfusion are not completely under-stood. The aim of the present study was to investigate whether the inactivation of Kupffer cells (KCs) by gadolinium chloride ($GdCl_3$) modulates ischemia-reperfusion injury in the rat liver. Hepatic function was assessed using an isolated perfused rat liver model. In livers subjected to cold storage at $4^{\circ}C$ in University of Wisconsin solution for 24 hrs and to 20 min rewarm-ing ischemia, oxygen uptake was markedly decreased, Kupffer cell phagocytosis was stimulated, releases of purine nucleoside phosphorylase and lactate dehydrogenase were increased as compared with control livers. Pretreatment of rats with $GdCl_3$) , a selective KC toxicant, suppressed kupffer cell activity, and reduced the grade of hepatic injury induced by ischemia-reperfusion. While the initial mixed function oxidation of 7-ethoxycoumarin was not different from that found in the control livers, the subsequent conjugation of its meta-bolite to sulfate and glucuronide esters was suppressed by ischemia-reperfusion, CdCl$_3$restored sulfation and glucuronidation capacities to the level of the control liver. Our findings suggest that Kupffer cells could play an important role in cold/warm ischemia-reperfusion hepatic injury.

Inductive Effects of Ginseng Saponins on the Rat LDH A-gene and the Synthetic rate of Hepatocyte DNA in Regenerating Rat Liver Cells

  • The effects of ginseng saponins, G-Rbl and G-Rc on the rat liver LDH A-gene transcnptional activity was investigated during pro-replicative phase of rat liver after partial hepatectomy. Changes in LDH A-mRNA levels in regenerating rat liver after intraperitoneal administrations of G-Rbl of G-Rc were tested by slot blot hybridization methods. The results showed that G-Rbl (1 mg/100g B.W) and G-Rc (1 ma/100g B.W) caused marked increases of LDH A-mRNA contents by respectively 1.9- and 1.5-fold in rat liver at 5·hours after partial hepatectomy. Dose dependent effect of G-Rbl and G-Rc (1-25 mg/100g B.W) on the LDH A-mRNA levels on regenerating rat liver were also analyzed. The maximal in- creases of liver LDH A-mRNA levels were observed with the doses of 1 mg for G-Rbl and 5 mg for G-Rc However, when the administration doses of G-Kbl and G-Rc were increased to 20 mg, G-Rbl caused a marked decrease of LDH A-mRNA level to 61% of those in sham-operated rat liver In contrast, G-Rc slightly decreased the liver LDH A-mRNA contents by 30% as compared to those of the maximum value but still maintained 22% higher LDH A-mRNA levels then those of sham-operated rate liver. On the basis of these experimental results, we conclude that ginseng saponin, G-Rb 1 and G·Rc have stimulatory effect at the lower concentration (1 mg/100g B.W) and inhibitory effect at the higher concentration (20 moi loos 5.W) on the LDH A-gene transcription during regeneration of rat liver, Additionally we also investigated the stimulatory effects of ginsenosides on the protein and DNA synthetic activities in hepatocyte primary cell cultures isolated from regenerating rat liver. Both of G·Rc and -Re increased the synthetic rates of hepatocytes proteins and DNA at the administration doses of 50 ug and 100 ug/3 ml/dish respectively representing 1.3-1.6 fold increases. From these results we postulate that G-Rc and -Re may have a mitogen enhancer activity for the hepatocyte proliferation during rat liver regeneration period. 간세포(肝細胞)의 산화적 손상에 대한 백화사설초(白花蛇舌草)의 항산화효과 (The Anti-Oxidative Effects of Oldenlandiae Diffusae Herba Extract on Oxidative Hepatic Injury)

  • Objective : This study was designed to investigate the anti-oxidative effects of Oldenlandiae Diffusae Herba Water extract (ODHW) on lipid peroxidation by free radicals oxidative hepatic injury. Methods : In order to evaluate anti-oxidative activities of ODHW in the liver cell, cultured normal rat liver cells(Ac2F) were incubated with or without ODHW. After 16 hours to 18 hours of experiment, cells were placed in DMEM medium without serum, and then incubated with 1mM tert-butyl hydro-peroxide(t-BHP) for two hours. Viable cells were detected by MTT assay. The levels of LPO induced by hydroxyl radical derived from H2O2-Fe2+ system in rat liver homogenate were determined by means of TBA. Inhibitory effect of ODHW on superoxide generation was measured by xanthine-xanthine oxidase system. Results : In the linoleic acid autoxidation system, ODHW exhibited antioxidant activity, which inhibited 85% of linoleic acid peroxidation. These effects were similar to those of dl-a-tocopherol. ODHW showed scavenging effects on DPPH radical, inhibited superoxide generation in xanthine-xanthine oxidase system, and also inhibited lipid peroxidation of rat liver tissue with hydroxyl radical derived from $H_2O_2-Fe^{2+}$ system. In addition, ODHW protected the cell death induced by t-BHP and it significantly increased cell viability in a normal rat liver cell(Ac2F)

  • The activation of the hepatic stellate cell (HSC) is a key step in liver fibrogenesis. We investigated the changes of global gene expression during activation in hepatic stellate cells using a rat cDNA microarray with 5, 000 sequence-verified clones. We identified osteopontin (OPN), a secreted matrix protein, as one of the upregulated factors. Northern analysis showed OPN mRNA was increasingly expressed during progressive activation of cultured rat HSCs and in models of experimental liver fibrosis. (omitted)
