Reversal of Immunogenicity in Pediatric Inflammatory Bowel Disease Patients Receiving Anti-Tumor Necrosis Factor Medications

  • Loss of response to anti-tumor necrosis factor (anti-TNF) agents in the treatment of inflammatory bowel disease (IBD) is a major consideration to maintain sustained response. Reversal of immunogenicity can re-establish response and increase the durability of these agents. Strategies to reverse immunogenicity include dose-intensification and/or the addition of an immunomodulator. However, there is a relative paucity of data on the efficacy of such interventions in pediatric IBD patients. Available reports have not strictly utilized homogenous mobility shift assay, which reports on anti-drug antibodies even in the presence of detectable drug, whereas prior studies have been confounded by the use of drug sensitive assays. We report four pediatric inflammatory bowel disease patients with successful reversal of immunogenicity on an anti-TNF agent using dose intensification and/or addition of an immunomodulator.

Immunohistolocalization of Carbonic Anhydrase in Kidney and Intestine of Rainbow Trout, Oncorhynchus mykiss

  • Carbonic anhydrase is essential for the cellular transportation of hydrogen and bicarbonate ions and plays a key role in a wide variety of physiological processes. Rainbow trout, Oncorhynchus mykiss is an important freshwater fish in aquaculture industry and is known to be one of the most susceptible species to environmental contamination. In this study, carbonic anhydrase was detected in the kidney and intestine of rainbow trout. Carbonic anhydrase was isolated from cytosolic proteins and identified by using SDS-PAGE, isoelectric focusing, and immunohistochemical methods. A specific protein band with molecular weight of 30 kDa and pI of 7.0 was detected by Western blotting. The immunohistochemical results showed that carbonic anhydrase was located at various cells in the kidney and intestine of rainbow trout.

송어양식장의 위해요소 관리를 위한 수질 모니터링 (Water Quality Monitoring for Hazard Analysis in Aquaculture Farm of Rainbow Trout)

  • Water quality has been considered to be one of sanitation standard operating procedures (SSOP) for hazard analysis critical control points (HACCP) application in aquaculture farms. This study was conducted to evaluate a hazard caused by water used in aquaculture farm of rainbow trout. The water quality was analyzed to investigate both physiochemical and bacteriological level in water samples collected from aquaculture farm of rainbow trout, Oncorhynchus mykiss. No significant difference were observed on water temperature and pH from season to season. However, the levels of dissolved oxygen were decreased as the outside temperature was increased, even if the levels were adequate for aquaculture. Also, other physiochemical analysis including biochemical oxygen demand (BOD), chemical oxygen demand (COD) and suspended solid (SS) revealed that the waters for aquaculture analyzed in this study was suitable for rainbow trout aquaculture. The bacterial analyses were also revealed that the waters for aquaculture were met to both coliform group (<18 MPN/100mL) and viable cell count (<100 CFU/mL). However, some of waste waters from aquaculture farms showed higher levels of BOD and COD than those of waste water standard (<2 ppm), suggesting that regular cleaning of fish tank and precipitation tank is needed.

Effects of Replacement of Fish Meal with Poultry By-product Meals on Apparent Digestibility, Body Composition and Protein Efficiency Ratio in a Practical Diets for Rainbow Trout, Onchorynchus mykiss

  • This study examined the replacement of the fish meal (FM) with commercial poultry by-product meal (PBM) in practical diets for rainbow trout (Onchorynchus mykiss, Walbaum). Five isocaloric and isonitrogenous diets containing levels 0 (control), 10, 20, 30 and 40% of PBM as a replacement for FM were fed to three replicate groups of rainbow trout with a initial weight $34.50{\pm}0.43g$ (mean${\pm}$SE). Protein efficiency ratio (PER) of diets containing PBM up to 20% were similar to the control while significantly lower values were obtained from the groups receiving higher levels of PBM (p<0.05). Apparent protein digestibility coefficients (ADCs) were significantly lower than that of the control group when PBM was included at level of 20% or more. Similarly, significantly lower values were observed with diets containing 30 and 40% PBM in terms of dry matter, organic matter, ether extract, ash and energy digestibility (p<0.05). Dietary treatments did not significantly affect the body composition of the fish in terms of dry matter, crude protein, ether extract and ash. In conclusion, PBM in a proportion of 20% may replace about 40 % of FM in rainbow trout diet without significant impairment grow.

  • A survey was conducted to determine the prevalance of infectious pancreatic necrosis virus (IPNV) on fish farms in Korea and the epidemiology of IPNV infection in the farmed rainbow trout. In total, 43 pools of rainbow trout with apparent signs of viral infection from five provinces were obtained and analyzed. Evident cytopathic effects, including karyopycnosis and cell destruction, were observed in CHSE (chinook samlmon embyro)-214 cells infected with the virus isolates. Of these, ten viral isolates were assumed to be IPNV based on biophysical properties. RNA analysis revealed that the isolates contained two-segmented RNA genomes, further indicating that the viral isolates are IPNV. Antigenic comparison of the IPNV isolates identified three distinct serological groups separable by the cross-neutralization test. Of the ten IPNV isolates, six could be classified as strain DRT, two as strain Ab, and two as strain VR299. We were not able to isolate new strain of IPNV or any isolate serologically similar to the standard strain Sp.poraceae and families of the Agaricales, they are genetically more related to the Polyporaceae. These results are consistent with morphological characters observed in those mushrooms. However, it is premature to conclude taxonomic status Ganoderma species in the present study employing small sample size.

Effects of Recombinant Aquaporin 3 and Seawater Acclimation on the Expression of Aquaporin 3 and 8 mRNAs in the Parr and Smolt Stages of Rainbow Trout, Oncorhynchus mykiss

  • This study aimed to examine the role of two aquaporin isoforms (AQP3 and AQP8) in response to the hyperosmotic challenge of transitioning from freshwater (FW) to seawater (SW) during parr and smoltification (smolt) using the rainbow trout, Oncorhynchus mykiss. We examined the changes in the expression of AQPs mRNAs in the gills and intestine of the parr and smolt stages of rainbow trout transferred from FW to SW using quantitative real-time PCR in an osmotically changing environment [FW, SW, and recombinant AQP3 (rAQP3) injection at two dosage rates]. Correspondingly, AQPs were greater during smoltification than during parr stages in the rainbow trout. Plasma osmolality and gill $Na^+/K^+$-ATPase activity increased when the fish were exposed to SW, but these parameters decreased when the fish were exposed to SW following treatment with rAQP3 during the transition to seawater. Our results suggest that AQPs play an important role in water absorbing mechanisms associated with multiple AQP isoforms in a hyperosmotic environment.

Isolation and Characterization of Flavobacterium johnsoniae from Farmed Rainbow Trout Oncorhynchus mykiss

  • Flavobacterium johnsoniae was isolated from farmed rainbow trout Oncorhynchus mykiss in Korea, and its biochemical and molecular characterization was determined. Yellow-pigmented bacterial colonies were isolated from 18 of 64 fish samples (28.1%) on trypticase soy agar plates, and their biochemical profiles were characterized by API 20E and API 20NE test kits. F. johnsoniae was identified by biochemical phenotyping of factors including rapid gliding motility, Gram-negative condition, oxidase- and catalase-positive status, Congo red absorption, nitrate reduction, ${\beta}$-galactosidase production, acid production from glucose, and gelatin and casein hydrolysis. PCR and subsequent sequencing of 16S rRNA confirmed that the yellow-pigmented colonies were most similar to F. johnsoniae. The alignment analysis of 16S rRNA sequences also showed that all 18 rainbow trout isolates had highly similar homologies (97-99% identity). One isolate was selected and named FjRt09. This isolate showed 98% homology with previously reported F. johnsoniae isolates, and in phylogenetic analysis was more closely grouped with F. johnsoniae than with F. psychrophilum, F. columnare, or F. branchiophilum. This is the first report on the occurrence and biochemical characterization of F. johnsoniae isolated from rainbow trout in Korea.

  • 무지개송어, 산천어, 무지개송어와 산천어간의 잡종 3배체를 대상으로 성비조사와 형태학적 계측형질을 측정하였다. 무지개송어, 산천어의 암.수 성비는 공히 유사한 비율로 나타난 반면, 잡종과 잡종 3배체에서는 수컷이 암컷에 비해 높게 나타났다(p<0.01). 잡종 3배체는 두고/두장, 등지느러미 기부장/체장 및 가슴지느러미 길이/체장의 계측형질에서는 친어로 사용된 무지개송어와 산천어의 중간을 나타낸 반면 상악장/두장의 계측형질에서는 무지개송어 쪽을 다소 닮게 나타났다. 이러한 결과들은 잡종 3배체에서 3배체화에 따른 사용된 친어들에 대비된 성비 그리고 계측형질에서의 특징을 나타내었다.

Early-growth comparison of diploid and triploid rainbow trout (Oncorhynchus mykiss) in South Korea

  • Fast somatic growth is important considerations for successful and competitive aquaculture industry. In rainbow trout reared in South Korea, triploid induction was used to suppress negative influence of reproductive maturation to body growth. However, the effects of triploidy are visible in both mature fish and developing juvenile fish. Thus, it is also important to explicate the effect of triploid induction on growth during the early-life stages of rainbow trout-alevins and fry. Rainbow trout fertilized eggs were subjected to triploid induction and polyploidy was checked by flow cytometry. Diploid and triploid alevins and fry were reared separately in tanks with constant flow of freshwater through flow-through water system and growth measurements were done from zero days after hatching (DAH 0) until DAH 134. The egg-yolk morphometrics of alevins-yolk length, yolk height, yolk volume and yolk weight-were statistically similar (p > 0.05) in both genotypes from DAH 0 to DAH 22. The total length, body height, and body weight of alevins and fry were statistically better (p > 0.05) in both genotypes until DAH 92 but thereafter, triploid had a significantly better growth performance (p < 0.05) over diploid fish until the completion of study at DAH 134. With that, triploid induction did not influence alevin yolk regions and body growth and fry somatic growth until around 3 months after hatching, but considerable growth enhancement was subsequently apparent.

Evaluation of feed types based on growth performance, survival, hematology, and resistance in celebes rainbow (Marosantherina ladigesi)

  • Celebes rainbow (Marosantherina ladigesi) is one of Indonesia's exported ornamental fish commodities, but the exploitation of this fish only relies on wild catches. The rise of unlimited fishing, especially those using poison, has changed the aquatic environment, threatening sustainability and causing fish extinction. This study aimed to evaluate the effectiveness of several types of feed in improving the absolute growth rate (AGR), specific growth rate (SGR), survival rate (SR), feed conversion ratio (FCR), feed efficiency (FE), hematology, and immune response of Celebes rainbow. The fish used in this study were male ornamental Celebes rainbow (M. ladigesi) weighing 1.32 ± 0.21 g/ind, reared in 54 L-aquariums at a stocking density of 30 individuals/aquarium for six weeks. The fish were fed according to the test diet consisting of live Tubifex sp worms, dry Tubifex sp worms, Spirulina platensis, and crumble pellets. The parameters observed were AGR, SGR, SR, FCR, FE, hematology, intestinal histology, liver histology, and a challenge test with the pathogenic bacteria Aeromonas hydrophila. The results showed that fish-fed live Tubifex sp worms had better AGR, SGR, SR, FCR, FE, hematology, and disease resistance compared to all other treatments. These results indicate that live Tubifex sp worms are the best feed for rearing Celebes rainbow.
