도금전해액의 종류에 따른 수지상 구리 분말의 형상 및 표면적 특성 (Effect of Electrolyte Type on Shape and Surface Area Characteristics of Dendritic Cu Powder)

  • We have investigated the effects of applied potential, deposition time and electrolyte types on shapes and physical properties of Cu dendrites by potentiostatic electrodeposition. Finer shape of dendrites was observed at less cathodic potential by 100mV than at the limiting current, due to 'effective overpotential'. The shape of copper dendrite is related to the deposition time, too. The dendrite depositing for 10 min showed the finest shape. The finer dendrite has the less apparent density and the larger specific surface area. Dendrite from chloride solution has the lowest density and the largest surface area among three plating solutions, sulfate, chloride and pyrophosphate.

  • A cytosine deaminase from the cell-free extract of an isolate was examined after ethyl alcohol reactionation. The enzyme catalyzed the conversion of 5-fluorocytosine to 5-fluorouracil by the possession of specificity to the substrate. The optimum temperature and storage time on the stability of the enzyme were at below $50^{\circ}C$ and near 2 days in tris-HCl buffer. The maximum activity was also presented ar 9.0 in pH and $45^{\circ}C$ in temperature. The pHs and temperatures for the enzyme activity ranged from 8.5-9.5 and from 40-$50^{\circ}C$, respectively. the presence of $Ag^{+}, Hg^{2+}, Zn^{2+}$ in the reaction mixture resulted in the marked inhibition in the activity, but 1mM of $Fe^{3+}, K^{+}$, or $Na^{+}$ increased the enzyme activity. The enzyme preparation was vot affected by inhibitors used except N-ethylmaleimide of 1 and 10mM, and considerably activated by 1mM of pyrophosphate and 10mM of phosphate.

탈지조성에 따른 철강용 알칼리탈지제의 합성 및 특성 (Synthesis and Characteristics of Alkaline Degreasing Agents for Steel Containing Degreasing Components)

  • To prepare alkaline degreasing agents(SADAs), tetrasodium pyrophosphate(TSPP), $Na_2CO_3$, Tetronix T-701 (T-701), Na-dioctyl sulfosuccinate(303C), Demol C, and MJU-100A were blended. The prepared degreasing agents were tested with steel specimens and the results were as follows. The degreasing powers of SADA-6($Na_2CO_3$ 50g/TSPP 25g/T-701 10g/303C 15g/Demol C 12g/MJU-100A 8g/water 130g mixture) for press-rust preventing oil were 98% and 99% degreasing at 4wt%, 70 $^{\circ}C$ and 90 $^{\circ}C$, respectively; for quenching oil, the degreasing power of SADA-6 was 92% degreasing at 4wt% and 70 $^{\circ}C$. From these results, it was proved that the SADA-6 exhibits a good degreasing power. Foam heights measured immediately after foaming by Ross & Miles method and Ross & Clark method at 6wt% and 60 $^{\circ}C$ were 16mm and 40mm, respectively. SADA-6 was proved a good low foaming degreasing agent.

Characterization of C-P Lyase gene cluster by in vivo $^{31}$ P-NMR spectroscopy

  • $\^$31/ P-NMR experiment was performed to detect phophonates (Pn) utilization and degradation in the several different C-P lyase mutants of E. coli and in E. aerogenes and the recombinants. The relative peak intensity (RPI) for the standard samples of 0.5 mM methylphosphonate (MPn) and 1.0 mM aminoethylphosphonate in glucose-MOPS medium showed 0.5 : 1.0 ratio. In the case of BW14329 (.DELTA.phnC-P, .delta.phoA), RPI did not change significantly after 24 hrs culturing, which means it nearly could not utilize Pn. In vivo $\^$31/ P-NMR spectrum of E. aerogens (BWKL 16627) during 3 hrs starvation showed two intense peaks at 0-2 ppm and at near-10 ppm which indicate intracellular orthophosphate (Pi) and pyrophosphate (PPi), respectively. Both of them might be released by degradation of inorganic polyphosphate pool. When MPn is supplied to the medium as an unique P source, Pi content in the cell has the constant, but PPi seems to be slightly decreased. Recombinants (BWKL 16954) grew slower than E. aerogenes in the glucose-MOPS media with various P sources. In vivo $\^$31/ P-NMR spectrum of recombinant did not show any intense signal in the cell. Surprisingly, under the cultivation adding with MPn, a few intense peaks in the region of Pi AND phospate monoester were detected.

Optimization of Ascorbic Acid-2-Phosphate Production from Ascorbic Acid Using Resting Cell of Brevundimonas diminuta

  • With the aim to produce ascorbic acid-2-phosphate(AsA-2-P) from L-ascorbic acid(AsA, Vitamin C), nine bacteria conferring the ability to transform AsA to AsA-2-P were isolated from soil samples alongside known strains from culture collections. Most isolates were classified to the genus Brevundimonas by 16S phylogenetic analysis. Among them, Brevundimonas diminuta KACC 10306 was selected as the experimental strain because of its the highest productivity of AsA-2-P. The optimum set of conditions for the AsA-2-P production from AsA using resting cells as the source of the enzyme was also investigated. The optimum cultivation time was 16 h and the cell concentration was 120g/l(wet weight). The optimum concentrations of AsA and pyrophosphate were 550mM and 450mM, respectively. The most effective buffer was 50mM sodium formate. The optimum pH was 4.5 and temperature was $40^{\circ}C$. Under the above conditions, 27.5g/l of AsA-2-P was produced from AsA after 36 h of incubation, which corresponded to a 19.7% conversion efficiency based on the initial concentration of AsA.

Biochemical Characterization of a Putative Calcium Influx Factor as a Diffusible Messenger in Jurkat Cells, Xenopis Oocytes, and Yeast

  • Highly purified high performance thin layer chromatography (HPTLC) fractions containing a putative calcium influx factor (CIF) were prepared from the Jurkat cells and Xenopus oocytes in which $Ca^{2+}$ stores were depleted by thapsigargin treatment and from the yeast in which intracellular $Ca^{2+}$ stores were also depleted by genetic means. Microinjection of the fractions has been shown to elicit $Ca^{2+}$ dependent currents in Xenopus oocytes. The nature of the membrane currents evoked by the putative CIF appeared to be carried by chloride ions since the current was blocked by the selective chloride channel blocker 1 mM niflumic acid and its reversal potential was about -24 mV. Injection of the calcium chelator 1,2-bis(2-aminophenoxy)ethane-N, N, N',N'-tetraacetic acid (BAPTA) eradicated the current activities, suggesting the current responses are entirely $Ca^2$-dependent. Moreover, the currents were sensitive to the removal of extracellular calcium, indicating the dependence on calcium entry through the plasma membrane calcium entry channels. CIF activities were insensitive to protease, heat, and acid treatments and to Dische-reaction whereas the activities were sensitive to nucleotide pyrophosphatase and hydrazynolysis. The fraction might have a sugar because it was sensitive to Molisch test and Seliwaniff's resorcinol reaction. From the above results, CIF as a small and stable molecule seems to have pyrimidine, pyrophosphate, and a sugar moiety.oiety.

분광법을 이용한 토양 유기물의 간이 측정 방법의 국내 적용 가능성 (Feasibility of a simple determination of soil organic matter content using spectrophotometric method in Korean soils)

  • A method currently used to determine soil organic matter (SOM), Tyurin method, is time consuming and expensive while accurate. Recently, a spectrophotometric determination was reported to be rapid, accurate, stable, easy to execute, and amendable to field use for soil samples obtained from Texas, USA. The objective of this study was to test if the spectrophotometric method is applicable to soils in Korea. Soil organic matter was extracted by 1 M HCl followed by a 0.25 M NaOH-0.1 M sodium pyrophosphate solution at a ratio of 1:250 soil:extractant. Soil organic matter determined by Tyurin method was linearly related to the value based on absorbance at 300 nm of the soil extracts with a coefficient of determination ($r^2$) of 0.81. Therefore, the result imply that this spectrophotometric method can be used to determine the soil organic matter of agricultural soils in Korea.

  • 가성통풍으로 인한 수근관 증후군은 드물게 발생한다. 저자들은 수근관 내 종괴로 인한 급성 정중신경 압박 증세로 내원한 65세 여자환자에게 종괴 절제술과 횡수근 인대 절개술을 시행하고 조직검사를 통해 가성통풍으로 진단된 증례를 경험하여 문헌 고찰과 함께 보고하고자 한다.

제주도(濟州道) 화산회토양(火山灰土壌)의 이화학적(理化学的) 특성(特性) 및 유기물(有機物) 성상(性状)에 관(関)한 연구(硏究) (Studies on the Physico-chemical Properties and Characterization of Soil Organic Matter in Jeju Volcanic Ash Soil)

  • 제주도(濟州道) 화산회토양유기물(火山灰土壌有機物)의 화학성(化学性), 유기물(有機物) 성상(性狀) 및 부식산(腐植酸)의 광학적(光学的) 특성(特性)을 알고져 수종(数種)의 화산회토양(火山灰土壌)을 공시(供試)하여 실내시험(室內試験)한 결과(結果)를 요약(要約)하면 다음과 같다. 가. 토양(土壌)의 화학적(化学的) 성질(性質) 1) 화산회토양(火山灰土壌)은 비화산회토양(非火山灰土壌)에 비(比)하여 유기물(有機物)(4~27%), 유효규산(有効珪酸)(291~884ppm), 활성(活性) 알루미늄(150~478ppm) 및 활성철함량(活性鉄含量)(0.77~0.86%)이 많고 반대(反対)로 유효인산(有効燐酸)(4~15ppm) 함량(含量)이 현저(顕著)히 낮았다. 2) 가리(加里), 석화(石火), 고토등(苦土等)은 밭의 경우 화산회토양(火山灰土壌)에서 높은 편이나 삼림지(森林地) 및 유휴지토양(遊休地土壌)은 낮은 경향(傾向)을 보였다. 3) 화산회토양(火山灰土壌)은 규반비(珪礬比) 및 규산(珪酸)/유기물비(有機物比)가 낮은 반면(反面) K/Ca+Mg, Al/Fe(활성(活性)) 및 C/P비(比)가 현저(顕著)히 높았다. 나. 유기물(有機物) 및 질소분별정량(窒素分別定量) 1) 화산회토양(火山灰土壌)은 유기물(有機物)의 총탄소중(総炭素中) Humin-C의 비율(比率), 유기물중(有機物中) Humin산(酸) 그리고 Humin중(中) C/N율(率)이 비화산회토양(非火山灰土壌)보다 현저(顕著)히 높았다. 2) 화산회토양(火山灰土壌)은 비화산회토양(非火山灰土壌)에 비(比)하여 총질소(総窒素), 산(酸) 및 알카리가용성(可溶性) 질소함량(窒素含量)이 현저(顕著)히 높은 반면(反面) 총질소중(総窒素中) 무기태질소(無機態窒素)로 방출(放出)될 수 있는 무기화율(無機化率)은 높지 않았다. 다. 토양부식(土壌腐植)의 형태(形態) 1) 화산회토양(火山灰土壌)은 비화산회토양(非火山灰土壌)에 비(比)하여 광흡(光吸) 수능(收能)이 높고(K600, RF치(値), ${\delta}logK$) 부식화도(腐植化度)가 진전(進展)될수록 색농도(色濃度)가 짙은 것으로 나타났다. 2) 화산회토양(火山灰土壌)은 산화제(酸化劑)에 대(対)한 저항성(抵抗性)이 높고 산(酸) 및 알카리 가수분해성(加水分解性)이 강(强)하여 부식화도(腐植化度)가 높아 부식(腐植)의 자연분해(自然分解)가 극(極)히 어려운 것으로 나타났다. 라. Humin의 관능기조사(官能基調査) 화산회토양(火山灰土壌)의 유출부식산(油出腐植酸)의 관능기(官能基)는 phenolic-OH기(基), Alcoholic-OH기(基) 및 Carboxyl기(基)가 많고 비화산회토양(非火山灰土壌)은 Methoxyl기(基) 및 Carbonyl기(基)가 많았다. 마. 부식산(腐植酸)의 흡광도(吸光度) 1) 공시토양(供試土壌)의 가시광역(可視光域)은 200~500nm부근의 단파장영역(短波長領域)이었으며 주로 350, 420, 450 및 480nm 에서 4개(個)의 흡광곡선(吸光曲線)을 나타내었다. 2) 화산회토양(火山灰土壌)인 흑악통(黑岳統)은 362nm부근에서 단일(單一)의 높은 흡광도(吸光度)를 보였으며 비화산회토양(非火山灰土壌)인 영악통(永楽統)은 360nm와 390nm에서 2개(個)의 단순(單純)한 높은 흡광대(吸光帶)를 나타내었다. 마. 분해촉진제(分解促進剤) 처리효과(處理効果) 화산회토양(火山灰土壌)에 대(対)한 분해촉진효과(分解促進効果)는 이도통(統)은 역분해성(易分解性) 유기물(有機物) 첨가(添加)에 따른 "Priming Effect"가 증가(增加)되었으며 남원(南元)과 흑악통(黑岳統)은 Na-Pyrophosphate의 첨가효과(添加効果)가 있었다.

Expression of orf8 (chlD) as Glucose-1-Phosphate Thymidylyltransferase Gene Involved in Olivose Biosynthesis from Streptomyces antibioticus Tü99 and Biochemical Properties of the Expressed Protein

  • The orf8(chlD) gene cloned from Streptomyces antibioticus T$\"{u}$99 was overexpressed using an E. coli system to confirm its biological function. Induction of the E. coli strain transformed with recombinant plasmid pRFJ 1031 containing orf8 resulted in the production of a 43,000 dalton protein. Glucose-1-phosphate thymidylyltransferase activity of the cell extract obtained from the transformed strain was 4-5 times higher than that of the control strain. The expressed protein was purified 18-fold from E. coli cell lysate using three chromatographic steps with a 17% overall recovery to near homogeneity. The N-terminal amino acid sequence of the purified protein agrees with the nucleotide sequence predicted from the orf8 gene. The SDS-PAGE estimated subunit mass of 43,000 dalton agrees well with that calculated from the amino acid composition deduced from the nucleotide sequence of the orf8 gene (43,000 Da). Also, the native enzyme has a monomeric structure with a molecular mass of 43,000 dalton. The purified protein showed glucose-1-phosphate thymidylyltransferase activity catalyzing a reversible bimolecular group transfer reaction, and was highly specific for dTTP and ${\alpha}$-D-glucose 1-phosphate as substrates in the forward reaction, and for dTDP-D-glucose and pyrophosphate in the reverse reaction.
