Characteristics of Cadmium Accumulating Mutant, Pseudomonas maltophilia H-8M (카드뮴 축적 변이주인 Pseudomonas maltophilia H-8M의 특성)

  • This study was carried out to investigate the characteristics of a mutant, Pseudomonas maltophilia H-8M selected with the treatment of Pseudomonas maltophilia H-8 by N-methyl-N-nitro-N-nitrosoguanidine(MNNG). This mutant showed highest ability of cadmium accumulation. The growth rate of Pseudomonas maltophilia H-8M showed about 80% in 1000ppm Cd containing medium when compare with control for 36h at $30^{\circ}C$. Pseudomonas maltophilia H-8M not inhibited on the growth in addition of various heavy metal such as $Hg^{2+}$, $Zn^{2+}$, $Pb^{2+}$, $Cu^{2+}$, $Cr^{2+}$ and $Co^{2+}$, but inhibited in $Sn^{2+}$ containing medium, respectively. Pseudomonas maltophilia H-8M was accumulated the highest cadmium level of 62.3% on whole cell in the medium containing 50 ppm and 80% of accumulated cadmium was distributed in the cell wall.

  • Pseudomonas maltophilia N246 carrying on OCT plasmid grew on n-alkanes of 6 to 14 carbon atoms, but not on n-alkanes of more carbon atoms. P. maltophilia strains with and without OCT plasmid could utilize primary alcohols. aldehydes and fatty acids derived from n-alkane. The N246 strain could also utilize monocarboxylic and dicarboxylic acids, and terminal branched dimethyloctane. Unlike the genes of alcohol dehydrogenase and aldehyde dehydrogenase which were located on both the chromosome and the OCT plasmid, genes for the alkane hydroxylase components were located only on the OCT plasmid in P. maltophilia N246.

  • Xanthomonns curnpestris M12, Xunthornonas sp. M28, Acinetuhucter Iwofz GI, and Klebsiella pneumoniae L25, Pseudomonas rnaltophiliu N246 were screened to increase the ability for crude oil utilization. All of these could utilize hexadecane and octane with the exception of N246 strain for only octane biodegradation. Thus N246, M12, and M28, strains were specially examined for octane oxidation. Octane biodegradation by three strains showed the optimal conditions at $30^{\circ}C$, pH 7.0~9.0, and 0.2~0.3% octane concentration as a substrate. It was found that P. multofihila N246 and X. curnpestns M12 had plasmid and the cured plasmid from N246 strain lost octane uitilization. Therefore, it was confirmed that certain genes for octane utilization were Iocated on OCT plasmid in N246 strain. The size of OCT plasmid in N246 strain was 118 kb. The N246 strain was resistant to ampicillin.

  • Tn5 lac 삽입으로 채소입고병원균에 길항력이 약화된 T-67 및 고추역병균과 참깨역병균에 길항력이 약화된 T-81의 Tn5 lac 유전자 일부와 오른쪽 말단에 있는 길항관련 유전자의 flanking sequence가 cloning된 pAG67 및 pAG81 clone을 선발하였고, pAG67 및 pAG81 clone된 길항관련 유전자의 flanking sequence를 야생 길항균 Pseudomonas maltophilia B-14의 DNA를 probe로 사용하여 Southern hybridization으로 확인하였으며, 제한효소 지도를 작성하여 8Kb 및 4Kb 크기의 flanking sequence가 cloning되었음을 확인하였다. pAG6 및 pAG81의 flanking sequence를 EcoRi-BglII와 EcoRI-MpaI으로 분리하여 유전자 은행으로부터 길항관련 유전자가 cloning된 cosmid clone 7개주를 선발하였다.

Isolation of Bacteria from Freshwater Plant Roots (담수식물 뿌리로부터 세균의 분리)

  • The isolation and identification of bacteria from freshwater plant root system were performed. Twenty four different strains were identified by microbiological identification system. Gram negative bacteria were isolated three times more than Gram positive ones. The ratio of rod and coccus was 11:1. The similarity above 0.5 was 37.5% from the tested samples in identification. Among these isolated bacteria, three dominant strains Pseudomonas cepacia JH10, Xanthomonas maltophilia JH12, Aeromonas salmonicida JH13 were selected to investigate biochemical properties and optimal growth conditions. These strains reached maximum growth after 24-36 hr of incubation in peptone broth, and their optimal temperature was 28$^{\circ}C$ and that of pH was between 7 and 8.

Microbial Degradation of Alkane Components in Crude Oil (미생물에 의한 원유중 Alkane 성분의 분해)

  • The isolates biodigrading crude oil were examined to characterize thier properties. Isolates which were identified as Acinetobacter lwoffii G1, Klebsiella pneumoniae L25, Pseudomonas maltophilia N246, Xanthomonas campestris M12, and Xanthomonas sp. M28. The optimum concentration of crude oil was 0.15% for the bacterial growth. X. campestris M12, Xanthomonas sp. M28, and K. penumoniae L25 showed the maximal growth at the concentration of 3.5% sodium chloride, indicating that they were derived from sea water. Among the isolates, X. campestris M12, Xanthomonas sp. M28 specially utilized hexadecane and octane, and P. maltophilia N246 utilized octane with optimum concentration of 0.2-0.3% as sole carbon source. The utilization of components of saturate fraction by K. pneumoniae L25 was examined by gas-liquid chromatography. The short-chain saturates are used before the long chain ones although they almost disappear within 8 days of incubation at $30^{\circ}C$.

The Degrdation of Pigment-Producing Furfural in Aquatic Waste (환경오염 유해색소의 미생물학적 분해)

  • Isolated Gram-negative bacteria, being capable of degrading toxic, recalcitrant, and pigment-producing furfural, were tentatively identified as Pseudomonas testosteroni, Pseudomonas maltophilia, Klebsiella Pneumoniae, and Pseudomonas fluorescens. They exhibited synergistic effects between P. testosteroni and the others in the degradation of colourproducing furfural. Synergistic effects and possible sequence of its degradation were attempted by manometric technique. P. testosteroni could degrade furfural to decolourize it and produce ninhydrin-reaction postive substance (NPS) which could be utilized by P. maltophilia and K. pneumoniae and the latter two bacteria could ,degrade furfural to 2-furoic acid as an oxidized form. Finally 2-furoic acid was further oxidized by P. fluorescens. Once NPS and 2-furoic acid were produced, the degradation efficiency was enhanced by competing four bacteria against furfural and 2-furoic acid.

Synergistic Antimicrobial Action of Thymol and Sodium Bisulfate against Burkholderia cepacia and Xanthomonas maltophilia Isolated from the Space Shuttle Water System

  • A combination of thymol and sodium bisulfate was found to be an effective biocidal agent against strains of Burkholderia cepacia and of Xanthomonas maltophilia that were found in the space shuttle water system. Potassium iodide (KI), the biocidal agent used in the past, had a minimum inhibitory concentration (MIC) of 50,000 ppm against the two B. cepacia (541 STS-81 and 1119 STS-91) strains, whereas that of thymol and sodium bisulfate was 2,400 and 950 ppm, which was 21 and 53 times lower than that of KI for B. cepacia, respectively. The MIC value for the combination of thymol and sodium bisulfate was 4 times lower than that for thymol or sodium bisulfate alone against B. cepacia (541 STS-81, 1119 STS-91) or Pseudomonas cepacia (ATCC 31941). The fractional inhibitory concentration (FIC) of the combination of thymol and sodium bisulfate for all organisms tested was less than 0.5, indicating a strong synergistic effect.

Isolation, Identification and Characterization of Bacteria Degrading Crude Oil (원유 분해 미생물의 분리, 동정 및 특성)

  • Crude oil-degrading bacteria were isolated from the sites contaminated by oil products. The isolates were identified as Acinetobacter sp. A132, Pseudomonas putida A422, Pseudomonas aeruginosa F721, F722, and Xanthomonas maltophilia B823. The results of investigation on the degradability of crude oil indicated that the strain A132 had the highest rate of $6.04g/L{\cdot}day$. Also, the strain A132 and F722 almost degraded each of n-alkane compounds between $nC_{10}$ and $nC_{32}$. The strain A422 degraded benzene and xylene but not n-alkane. The strain B823 grew somewhat in crude oil but did not entirely degrade other substrates used in this study. The results of the GC/FID analysis on the degradability of the mixed n-alkane compounds showed that the strain F722 could degrade 100% of the compounds with $nC_7{\sim}nC_{10}$ and more than 80% of those with $nC_{11}{\sim}nC_{24}$.

  • This study was conducted to determine the effect of treatment with the plant-growth-promoting bacteria on the growth of red pepper(Capsicum annuum L.).The eight plant-growth-promoting bacteria were isolated from the humic soil in the forest region. The isolated bacteria(IB) was identified by the method of the biochemical test(API kit) and the composition of the fatty acid(MIDI system).The IBs were inoculated by spray of 17ml at 72 cell tray filled with peatmoss every week. respectively, with mixed liquid eulture of eight strains. The IBs were identified as Micrococcus sp.. Bacillus subtilis. Enterobacter agglomerans, Bacillus megaterium, Pseudomonas putida. Pseudomonas fluorescens, Xanthomonas maltophilia and Staphylococcus xylosus by API kit and MIDI system. The plant height number of leaves and leaf length of red pepper grown on peatmoss treated with the IB were better than those of nontreatment at the 10th day after inoculation.
