Cyclic voltammetric measurements were performed for Co(II), Ni(II), Cu(II) and Zn(II) complexes of 1 : 1 alternating copolymer, poly(3-nitrobenzylidene-1-naphthylamine-co-succinic anhydride) (L) and Ni(II) and Cu(II) complexes of 1 : 1 alternating copolymer, poly(3-nitrobenzylidene-1-naphthylamine-co-methacrylic acid) ($L^1$). The in vitro biological screening effects of the investigated compounds were tested against the fungal species including Aspergillus niger, Rhizopus stolonifer, Aspergillus flavus, Rhizoctonia bataicola and Candida albicans and bacterial species including Staphylococcus aureus, Escherichia coli, Klebsiella pneumaniae, Proteus vulgaris and Pseudomonas aeruginosa by well diffusion method. A comparative study of inhibition values of the copolymers and their complexes indicates that the complexes exhibit higher antimicrobial activity. Copper ions are proven to be essential for the growth-inhibitor effect. The extent of inhibition appeared to be strongly dependent on the initial cell density and on the growth medium. The nuclease activity of the above metal complexes were assessed by gel electrophoresis assay and the results show that the copper complexes can cleave pUC18 DNA effectively in presence of hydrogen peroxide compared to other metal complexes. The degradation experiments using Rhodamine B dye indicate that the hydroxyl radical species are involved in the DNA cleavage reactions.
Finding new antimicrobial activities by functional metagenomics has been shown to depend on the heterologous host used to express the foreign DNA. Therefore, efforts are devoted to developing new tools for constructing metagenomic libraries in shuttle vectors replicatable in phylogenetically distinct hosts. Here we evaluated the use of the Escherichia coli-Bacillus subtilis shuttle vector pHT01 to construct a forest-soil metagenomic library. This library was screened in both hosts for antimicrobial activities against four opportunistic bacteria: Proteus vulgaris, Bacillus cereus, Staphylococcus epidermidis, and Micrococcus luteus. A new antibacterial activity against B. cereus was found upon screening in B. subtilis. The new antimicrobial agent, sensitive to proteinase K, was not active when the corresponding DNA fragment was expressed in E. coli. Our results validate the use of pHT01 as a shuttle vector and B. subtilis as a host to isolate new activities by functional metagenomics.
The methanol extract and residual methanol fraction of Excoecaria agallocha L. (Euphorbiaceae) stem bark was investigated in this study by wheat rootlet and shoot growth inhibition, and antimicrobial bioassay. The methanol extract and residual methanol fraction showed high inhibitory effect on both the wheat rootlet (82-89%) and shoot growth (85-90%) compared to control. The methanol extract showed a better and dose related inhibition on both the rootlet and shoot growth compared to residual methanol fraction. The $IC_{50}$ value of methanol extract for rootlet and shoot were $2.88\;{\mu}g/ml$ and $2.32\;{\mu}g/ml$, and of residual methanol fraction for rootlet and shoot were $7.91\;{\mu}g/ml$ and $4.45\;{\mu}g/ml$. The methanol extract and residual methanol fraction did not show any antibacterial activity against the tested microorganisms of clinical isolates Escherichia coli, Staphylococcus aureous, Pseudomonas aeruginosa, Proteus vulgaris and Bacillus subtilis. The plant has the potential to be a source of novel cytotoxic compound(s).
The effect of chitosan on antibacterial activities of cotton, wool and polyester fibers was investigated by shake flask method. Chitosan was treated in 0.1%, 1% and 2% $NaBO_3$ solution to reduce the molecular weight in 4 steps, wadding of cotton, wool and polyester were treated in 0.1%, 0.3% and 0.5% of chitosan solution which were dissolved in 2% acetic acid aqueous solution. The antibacterial activities of the fiber wadding treated and untreated by chitosan against Escherichia coli, Proteus vulgaris and Stephylococcus aureus were measured by shake flask method. On the untreated waddings, cotton showed better antibacterial activities than wool, but on the treated ones, wool showed better than cotton. The antibacterial activity of polyester was better than that of cotton or wool which preserved before and after the chitosan treatment against the three kinds of bacteria. When the chitosan treated cotton waddings was retreated in NaOH aqueous solution, their bacterial activities decreased. After laundering, the antibacterial activities of the treated cotton and wool waddings kept good, but that of the treated polyester reduced by almost half.
Novel copper(II) complexes have been synthesized from the macrocyclic Schiff bases derived from Knoevenagel condensed ${\beta}$-ketoanilides (obtained by the condensation of acetoacetanilide and substituted benzaldehydes), 4-aminoantipyrine and ophenylene diamine. The structural features have been determined from their analytical and spectral data. All the Cu(II) complexes exhibit square planar geometry. Their high molar conductance values support their 1 : 2 electrolytic nature. The magnetic moment data provide evidence for the monomeric nature of the complexes. The X-band ESR spectra of the |$CuL^1$|$(OAc)_2$ in DMSO solution at 300 and 77 K were recorded and their salient features are reported. The 23 representative bacteria were studied for the glucosamine contents which represent the glycolipid content of the cell wall. The distribution of glycolipid in various bacteria was examined and the relationship between the glycolipid contents and the Gram stain was elucidated. The results were as follows: 1. The contents of glucosamine in the glycolipid of Gram negative and variable bacterial cell wall were large ranging from the least 0.04 ${\mu}g$ of Proteus vulgaris to the largest 2.48 ${\mu}g$ of Aerobacter aerogenes. The Gram positive bacteria and only those Gram positive among Bacilli contained less than 0.02 ${\mu}g$ of glucosamine contents. The least glucosamine containing Gram positive bacteria were Corynebacterium sepedonicum and Staphylococcus aureus. It could generally be said that the Gram negative and variable bacteria contain the higher content of glucosamine in the cell wall than the positives. 2. The bacteria were better stained by the Gram solution after the extraction of glycolipid from the cell wall than those without extraction. 3. The four infrared spectra of glycolipids obtained from Aerobacter aerogenes, Bacillus circulans, Pseudomonas fluorescens, and Salmonella typhirurium showed all the similar characteristics. All showed the existence of groups; OH, C-O, C-O-C, $CH_2+CH_3$, amide band, fatty acid ester band and ester carbonyl bond.
The monthly distribution of marine bacteria in the coastal waters of Chung-Mu was investigated from April, 1976 to March, 1977. The aim of this study was to obtain basic data for the prevention of food poisoning and for the efficient aquaculture of the area. Samples of sea water, mud, fish and shell fish were taken every month. The results are as follows: 1. One thousand four hundred and twenty-six strains were isolated from 732 samples of sea water, mud, fish and shell fish. They were 450 strains of Pseudomonas fluorescens, 422 strains of Achromobacter liquefacience, 72 strains of Vibrio parahaemolyticus, 234 strain of Vibrio alginolyticus, and 248 strains of Proteus vulgaris. 2. V. parahaemolyticus occupied $9.84\%$ of the total samples and $52\%$ of them were found in the sea water and mud. 3. The muds sampled districtly beneath the aquaculture raft contained much gas. They seemed to originate from the deposition of excretion of shell fish. The fatness of the shell fish was low where the gas was abundant. 4. It was .found that the shell fish with low fatness contained much Vibrio sp. 5. Regional distribution shows that marine bacteria were abundant in the order of station 10, 9, 8, 11, 1. The area around station 9 was polluted by reclamation of the area, and station 8 showed the influence of the excrement treat tank located nearly.

This study was carried out to investigate the antioxidative and antimicrobial activities of internal organs of Aplysia kurodai (AK). The internal organs of AK were extracted with methanol (AKM), which was then further fractionated into four subfractions by using solvent partition method, affording hexane (AKMH), methanol (AKMM), butanol (AKMB), and aqueous (AKMA) soluble fractions. The antioxidative activity of fractions from AK was investigated by measuring the scavenging activities of AK against DPPH radical, peroxynitrite ($ONOO^-$) and reactive oxygen species (ROS). Among the various solvent fractions, AKMB showed a marked scavenging effect against DPPH radical, peroxynitrite ($ONOO^-$) and reactive oxygen species (ROS). The antimicrobial activity was increased in proportion to its concentration by the paper disk method. Among the various solvent fractions, AKMM fractions of AK showed the strongest antimicrobial activities. The methanol extracts exhibited antimicrobial activity against all organisms tested, while the hexane extracts showed antimicrobial activity only against Proteus vulgaris. The results suggest that the AK may be suitable for development as a food preservative and alternative antioxidant.
The antibacterial activity of Poria cocos against the human pathogenic bacteria and general bacteria was observed. The results obtained were as follows; 1. There wasn't any antibacterial activity at the Poria cocos powders, pieces and its extracts in every different kinds and places of origin. 2. Hyphae of Poria cocos in every different kinds and places of origin had the antibacterial activity against four Gram positive bacteria, i.e., Staphylococcus aureus, S. epidermidis, Bacillus subtilis and Streptococcus faecalis, on the other hand, hadn't the antibacterial activity against six Gram negative bacteria, Salmonella typhi, Shigella dysenteriae, Pseudomonas aeruginosa, Escherichia coli, Proteus vulgaria and Klebsiella pneumoniae. 3. The average inhibitory zone was 13.6mm for S. aureus, 14.7mm for S. epidermidis, 12.7mm for B. subtilis, and 13.2mm for S. faecalis at the concentration of $335{\mu}g$ of hyphae of Poria cocos.

A key virulence factor for urinary tract pathogens is the enzyme urease, which catalyzes the hydrolysis of urea into ammonium ions and carbonic acid. Urease activity plays an important role in the pathogenesis of urinary tract infection. In this study, the inhibitory effect of zerumbone against six urease-producing bacteria (Klebsiella oxytoca, K. pneumoniae, Morganella morganii, Proteus mirabilis, P. vulgaris, and Staphylococcus saprophyticus) and their urease activities were evaluated. The results of the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) tests showed that zerumbone had antibacterial effect against these six urease-producing bacteria. The MIC and MBC of zerumbone ranged from 0.5 to 2 mM and 1 to 4 mM, respectively. In the urease inhibitory assay, zerumbone showed better urease inhibition ($56.28{\pm}2.45-37.83{\pm}3.47%$) than the standard urease inhibitor, acetohydroxamic acid ($40.46{\pm}1.94-22.99{\pm}3.53%$). However, zerumbone did not affect the levels of the urease subunit. These results clearly indicated that zerumbone has antibacterial potential against urease-producing bacteria and possesses excellent bacterial urease inhibition properties.
